1Revalidation of a prognostic score model based on complete blood count for nasopharyngeal carcinoma through a prospective study显示文摘Objective: In our previous work, we incorporated complete blood count(CBC) into TNM stage to develop a new prognostic score model, which was validated to improve prediction efficiency of TNM stage for nasopharyngeal carcinoma(NPC).The purpose of this study was to revalidate the accuracy of the model, and its superiority to TNM stage, through data from a prospective study.Methods: CBC of 249 eligible patients from the 863 Program No.2006AA02Z4B4 was evaluated.Prognostic index(PI) of each patient was calculated according to the score model.Then they were divided by the PI into three categories: the low-, intermediate-and high-risk patients.The 5-year disease-specific survival(DSS) of the three categories was compared by a log-rank test.The model and TNM stage(7th edition) were compared on efficiency for predicting the 5-year DSS, through comparison of the area under curve(AUC) of their receiver-operating characteristic curves.Results: The 5-year DSS of the low-, intermediate- and high-risk patients were 96.0%, 79.1% and 62.2%,respectively.The low- and intermediate-risk patients had better DSS than the high-risk patients(P<0.001 and P<0.005, respectively).And there was a trend of better DSS in the low-risk patients, compared with the intermediate-risk patients(P=0.049).The AUC of the model was larger than that of TNM stage(0.726 vs.0.661,P=0.023).Conclusions: A CBC-based prognostic score model was revalidated to be accurate and superior to TNM stage on predicting 5-year DSS of NPC.Xiaohui Li Hui Chang Yalan Tao Xiaohui Wang Jin Gao Wenwen Zhang Chen Chen Yunfei Xia 2016Chinese Journal of Cancer Research2016,28,5:4

3Flexible,video-rate,and aberration-compensated axial dual-line scanning imaging with field-of-view jointing and stepped remote focusing显示文摘Parallel dual-plane imaging with a large axial interval enables the simultaneous observation of biological structures and activities in different views of interest.However,the inflexibility in adjusting the field-of-view(FOV)positions in three dimensions and optical sectioning effects,as well as the relatively small effective axial range limited by spherical aberration,have hindered the application of parallel dual-plane imaging.Herein,we propose a flexible,video-rate,and defocus-aberration-compensated axial dual-line scanning imaging method.We used a stepped mirror to remotely generate and detect dual axial lines with compensation for spherical aberration and FOV-jointing to rearrange into a head-to-head line for high-speed optical sectioning acquisition.The lateral and axial positions of the two FOVs could be flexibly adjusted before and during imaging,respectively.The method also allows the adjustment of optical sectioning effects according to specific experimental requirements.We experimentally verified the consistent imaging performance over an axial range of 300μm.We demonstrated high throughput by simultaneously imaging Brownian motions in two 250μm×250μm FOVs with axial and lateral intervals of 150μm and 240μm,respectively,at 24.5 Hz.We also showed potential application in functional imaging by simultaneously acquiring neural activities in the optic tectum and hindbrain of a zebrafish brain.The proposed method is,thus,advantageous compared to existing parallel dual-plane imaging and potentially facilitates intravital biological study in large axial range.RUI JIN YALAN YU DAN SHEN QINGMING LUO HUI GONG JING YUAN 2021Photonics Research2021,9,8:2

5Binding of HIV-1 virions to α_4β_7 expressing cells and impact of antagonizing α_4β_7 on HIV-1 infection of primary CD4^+ T cells显示文摘HIV-1 envelope glycoprotein is reported to interact with α4β7, an integrin mediating the homing of lymphocytes to gut-associated lymphoid tissue, but the significance of α4β7 in HIV-1 infection remains controversial. Here, using HIV-1 strain Ba L, the gp120 of which was previously shown to be capable of interacting with α4β7, we demonstrated that α4β7 can mediate the binding of whole HIV-1 virions to α4β7-expressing transfectants. We further constructed a cell line stably expressing α4β7 and confirmed the α4β7-mediated HIV-1 binding. In primary lymphocytes with activated α4β7 expression, we also observed significant virus binding which can be inhibited by an anti-α4β7 antibody. Moreover, we investigated the impact of antagonizing α4β7 on HIV-1 infection of primary CD4+ T cells. In α4β7-activated CD4+ T cells, both anti-α4β7 antibodies and introduction of shorthairpin RNAs specifically targeting α4β7 resulted in a decreased HIV-1 infection. Our findings indicate that α4β7 may serve as an attachment factor at least for some HIV-1 strains.
