| 2 | miR-21 promotes the differentiation of hair folliclederived neural crest stem cells into Schwann cells显示文摘Hair follicle-derived neural crest stem cells can be induced to differentiate into Schwann cells in vivo and in vitro. However, the underlying regulatory mechanism during cell differentiation remains poorly understood. This study isolated neural crest stem cells from human hair follicles and induced them to differentiate into Schwann cells. Quantitative RT-PCR showed that microRNA(miR)-21 expression was gradually increased during the differentiation of neural crest stem cells into Schwann cells. After transfection with the miR-21 agonist(agomir-21), the differentiation capacity of neural crest stem cells was enhanced. By contrast, after transfection with the miR-21 antagonist(antagomir-21), the differentiation capacity was attenuated. Further study results showed that SOX-2 was an effective target of miR-21. Without compromising SOX2 mRNA expression, miR-21 can down-regulate SOX protein expression by binding to the 3′-UTR of miR-21 mRNA. Knocking out the SOX2 gene from the neural crest stem cells significantly reversed the antagomir-21 inhibition of neural crest stem cells differentiating into Schwann cells. DPIs commonly contain micronized drug particles which, because of their cohesiveness and strong propensity to aggregate, have poor aerosolization performance. Thus carriers with a larger particle size are added to address this problem. However, the performance of DPIs is profoundly influenced by the physical properties of the carrier, particularly their particle size, morphology/shape and surface roughness. The great difference with the reality is 'cold drift' of the simulated surface temperature. The coupled model hascertain ability to simulate the atmosphere geopotential height fields, precipitation and low-level southwest wind from May to August in 1998. It can display the process of summer monsoon onset during the third dekad of May and the evolution features after the onset. | 6 | Prognostic value of interim ^(18)F-FDG PET/CT in diffuse large B-cell lymphoma显示文摘Objective: Diffuse large B-cell lymphoma (DLBCL) is a heterogeneous disease. The prognostic factor currently used is not accurate enough to predict the outcomes of patients with DLBCL. The prognostic significance of interim PET/CT in DLBCL remains controversial. The aim of this study is to determine the predictive value of interim 18F-FDG PET/CT after first-line treatment in patients with DLBCL. Methods: Thirty-two patients with DLBCL underwent baseline, interim and post-treatment 18F-FDG PET/CT scans. Imaging results were analyzed for the survival of patients via software SPSS 13.0, retrospectively. Results: Thirty-one of the 32 patients were treated with R-CHOP regimen, and interim 18F-FDG PET/CT of 24 patients was performed after 2 cycles of treatment. After a median follow-up period of 16.7 months, the 2-year progression-free survival (PFS) rates were significantly different between the groups above and below SUV max cut-off value of 2.5 (P=0.039). No significant differences were found in the 2-year PFS rates if SUV max cut-off values were set as 2.0 and 3.0, respectively (P=0.360; P=0.113). | 7 | Atmospheric eddy anomalies associated with the wintertime North Pacific subtropical front strength and their influences on the seasonal-mean atmosphere显示文摘This study investigates transient eddy activity anomalies in the mid-latitude upper troposphere associated with intensity variability of the wintertime North Pacific subtropical front. Our results show that the meridional gradient of air temperature and baroclinic instability in the mid-latitude atmosphere become stronger as the subtropical front intensifies, and the mid-latitude westerly jet accelerates with barotropic structure. We further divide the mid-latitude atmospheric eddy activities into high-(2–7 days) and low-frequency(10–90 days) eddy activities according to their life periods. We find that, when the oceanic subtropical front intensifies, the high-frequency atmospheric eddy activity in the mid-latitudes strengthens while the low-frequency eddy activity weakens. The stronger high-frequency eddy activity tends to moderate the air temperature gradient and baroclinicity in the mid-latitudes. High-frequency eddy anomalies accelerate the westerly jet on the northern side and downstream of the westerly jet, and enhance the jet with equivalent barotropic structure. In contrast, the weaker low-frequency eddy activity has a negative contribution to zonal wind speed tendency and attenuates the zonal homogenization of the jet. | 8 | Jurassic Black Shales Facies from Qiangtang Basin (Northern Tibet): Rare Earth and Trace Elements for Paleoceanographic Implications显示文摘The Biluo Co and Amdo 114 station, northern Tibet, cropping out the Early Toarcian and Middle-Late Tithonian (Jurassic) organic-rich black shales, have been a focus to petroleum geologists in discussing their oil-producing potential. This paper first reports the trace elements and rare earth elements to discuss the paleoenvironments, redox conditions and sedimentary mechanisms of those black shales. Both sections exhibit variation in trace element abundances with concentrations <0.1 ppm to 760 ppm, mostly enriched in V, Cr, Ni, Cu, Zn, Mo, Ba and U. Element ratios of Ni/Co, V/Cr, U/Th and V/(V+Ni) plus U were used to identify redox conditions. The shale-normalized rare earth element (REE) patterns are characterized by the flat-shale type with instable Ce anomalies and very weekly positive Eu anomalies. Positive Ceanom values are significant with values varying between - 0.064 and 0.029 in Biluo Co, which may be interpreted as release of REE and input of riverine terrestrial matter with rich Ce (resulting in pH change) during the anoxic conditions. In the middle parts of Amdo 114 station, distinct negative Ceanom values are observed (-0.238 to -0.111) and associated surface water warming were interpreted as being related to a major sea level rise. In contrast, the formation of the black shales in the lower and upper part of the studied succession took place during a cooler (Ceanom values >-0.10), lower surface water productivity, and lower sea-level stage. Thus, we emphasize the role of different factors that control the formation of local and regional black shales. | 9 | Prognostic value of pre-and post-transplantation 18F-fluorodeoxyglucose positron emission tomography results in non-Hodgkin lymphoma patients receiving autologous stem cell transplantation显示文摘Objective: High-dose chemotherapy(HDC) followed by autologous stem cell transplantation(ASCT) is the standard of care in the upfront or relapsed/refractory setting in some patients with non-Hodgkin lymphoma(NHL).However,a proportion of patients do not respond to ASCT.^(18)F-fluorodeoxyglucose(FDG) positron emission tomography(PET)/computed tomography(CT) has been widely used for staging,response evaluation,and prognosis prediction.Here,we investigated the prognostic role of PET/CT in NHL patients before and after ASCT.Methods: A retrospective study was conducted at Peking University Cancer Hospital.All NHL patients who underwent ASCT between March 2010 and July 2016 were identified.Patients who had PET/CT scan before and after ASCT were included.Deauville criteria(5-point scale) were used to interpret PET scans.Univariate and multivariate survival analyses were performed using Cox regression.The predictive value of PET scanning was estimated by comparing the area under the receiver operating characteristic(ROC) curve.Results: In total,79 patients were enrolled in this study.In univariate analysis,pre-and post-ASCT PET result was identified as prognostic factors for 3-year progression-free survival(PFS) and overall survival(OS).Patients with negative pre-ASCT PET result demonstrated significantly better PFS(84.2% vs.54.2%) and OS(89.2% vs.63.6%) than patients with positive pre-ASCT PET result.PFS(91.6% vs.25.3%) and OS(96.5% vs.36.8%) were also significantly different between patients with negative and positive post-ASCT PET result.Multivariate analysis also showed a significant association between survival and post-ASCT PET result.ROC analysis revealed that the predictive value of post-ASCT PET result was superior to that of pre-ASCT PET result alone.Combined pre-and post-ASCT PET result is better for predicting outcomes in patients with NHL receiving transplantation.Deauville criteria score >3 was identified as the best cutoff value for post-ASCT PET.Conclusions: Post-ASCT PET result was more important than pre-ASCT PET result in predicting outcomes for NHL patients who underwent ASCT.The prognostic significance can be improved when combining preASCT PET result with post-ASCT PET result.Deauville criteria can be used for interpreting PET scans in this scenario. | Zhitao Ying Lan Mi Xuejuan Wang Yuewei Zhang Zhi Yang Yuqin Song Xiaopei Wang WenZheng Ningjing Lin Meifeng Tu Yan Xie Lingyan Ping Chen Zhang Weiping Liu LijuanDeng Jun Zhu | 2017 | Chinese Journal of Cancer Research2017,29,6: | 3 |

| 11 | Subseasonal Zonal Oscillation of the Western Pacific Subtropical High during Early Summer显示文摘This study examines the features and dynamical processes of subseasonal zonal oscillation of the western Pacific subtropical high(WPSH) during early summer, by performing a multivariate empirical orthogonal function(MVEOF) analysis on daily winds and a diagnosis on potential vorticity(PV) at 500 hPa for the period 1979–2016. The first MV-EOF mode is characterized by an anticyclonic anomaly occupying southeastern China to subtropical western North Pacific regions. It has a period of 10–25 days and represents zonal shift of the WPSH. When the WPSH stretches more westward, the South Asian high(SAH) extends more eastward. Above-normal precipitation is observed over the Yangtze–Huaihe River(YHR) basin. Suppressed convection with anomalous descending motion is located over the subtropical western North Pacific. The relative zonal movement of the SAH and the WPSH helps to establish an anomalous local vertical circulation of ascending motion with upper-level divergence over the YHR basin and descending motion with upper-level convergence over the subtropical western Pacific. The above local vertical circulation provides a dynamic condition for persistent rainfall over the YHR basin. An enhanced southwest flow over the WPSH's western edge transports more moisture to eastern China, providing a necessary water vapor condition for the persistent rainfall over the YHR basin. A potential vorticity diagnosis reveals that anomalous diabatic heating is a main source for PV generation. The anomalous cooling over the subtropical western Pacific produces a local negative PV center at 500 hPa. The anomalous heating over the YHR basin generates a local positive PV center. The above south–north dipolar structure of PV anomaly along with the climatological southerly flow leads to northward advection of negative PV. These two processes are conducive to the WPSH's westward extension. | 13 | A 18FDG PET/CT-based volume parameter is a predictor of overall survival in patients with local advanced gastric cancer显示文摘Objective: The present study investigated the prognosis value of preoperative fluorodeoxyglucose(FDG)positron emission tomography/computed tomography(PET/CT) in patients with local advanced gastric cancer(LAGC).Methods: In total, 144 patients [median age 63(range: 48-80) years old] with LAGC underwent 18 F-FDG PET/CT prior to any treatment. The maximum standardized uptake values(SUVmax), mean standardized uptake values(SUVmean), metabolic tumor volume(MTV) and total lesion glycolysis(TLG) of the primary lesion were measured on PET/CT and correlated with clinicopathological features and survival.Results: Significant differences in SUVmean, SUVmax, MTV and TLG were found according to Lauren’s classification, histologic grade and T category(P<0.05). During the 26.5-month follow-up, 51(35.4%) patients died and 70(48.6%) exhibited disease progression. The optimal thresholds of MTV and TLG were 15.1 cm^3 and 47.3 cm^3, respectively. The 3-year progression-free survival(PFS) and overall survival(OS) for patients with high TLG values were 30% and 38% compared to 38% and 47% for low TLG values, respectively(P<0.05). Univariate and multifactor analyses demonstrated that lymph node metastasis and T stage were independent prognostic factors for PFS;T stage, histologic grade and TLG were independent prognostic factors for OS(P<0.05). | 15 | Preclinical characterization and validation of a dual-labeled trastuzumab-based imaging agent for diagnosing breast cancer显示文摘Objective: The combination of both nuclear and fluorescent reporters provides unique opportunities for noninvasive nuclear imaging with subsequent fluorescence image-guided resection and pathology. Our objective was to synthesize and optimize a dual-labeled trastuzumab-based imaging agent that can be used to validate an optical imaging agent with potential use in identifying tumor metastases in human epidermal growth factor receptor 2(HER2) positive breast cancer patients.Methods: [111In]-DTPA-trastuzumab-IRDye 800 was synthesized by a three-step procedure. Purity, stability, immunoreactivity, internalization and biodistribution were explored in HER2+ SKBR-3 cells. Biodistribution of [111In]-DTPA-trastuzumab-IRDye 800 was performed in a SKBR-3 xenograft model.Results: [111In]-DTPA-trastuzumab-IRDye 800 demonstrated high purity by both chemical and fluorometric determinations. Both flow cytometry and the Lindmo assay demonstrated a high binding affinity of [111In]-DTPA-trastuzumab-IRDye 800 to HER2-overexpressing cells. The dual-labeled conjugate was stable in PBS, but not in serum after 24 h at 37 ℃. Larger molecules(>150 k D) were seen after a 24 h-incubation in human serum. Biodistribution studies revealed tumor-specific accumulation of [111In]-DTPAtrastuzumab-IRDye 800 in SKBR-3 tumors, and tumor uptakes at 24 and 48 h were(12.42±1.72)% and(9.96±1.05)%, respectively, following intravenous administration. The tumor-to-muscle ratio was 9.13±1.68 at 24 h, and increased to 12.79±2.13 at 48 h. Liver and kidney showed marked uptake of the dual-labeled imaging agent.Conclusions: [111In]-DTPA-trastuzumab-IRDye 800 is an effective diagnostic biomarker that can be used to validate dual-labeled, molecularly targeted imaging agents and can allow these agents to be translated into clinical practice for identifying HER2+ lesions. | Xuejuan Wang Melissa B. | 16 | Chemo-and Biostratigraphy of the Jurassic Oil Shales from the Qiangtang Basin,Northern Tibet,China:A Case Study of the Toarcian Oceanic Anoxic Event显示文摘The marine oil shales of the Qiangtang Basin, northern Tibet, exposed in the Biluo Co, Tuonamu, Shenglihe and Changsheshan areas are believed to be important petroleum source rocks. This work comprehensively analyzed the carbon isotopes, trace elements, and calcareous nannofosills, ammonites and bivalves of the Biluo Co section in the Qiangtang Basin. The organic carbon isotopes show a positive excursion close to 2.17‰(relative to PDB), which, albeit significantly smaller, may also be associated with other Early Toarcian Oceanic Anoxic Events(T-OAE) in the European epicontinental seas and the Tethyan continental margins. Coinciding with the Early Toarcian transgression, the oxygen deficiency in bottom water had led to dysoxic-anoxic conditions and deposition of black shales lacking benthic fauna. Under such condition, the redox-sensitive trace metals such as Mo, V, Ni, Cr, and U were enriched, in conjunction with high planktonic productivity of Watznaueriaceae calcareous nannofossils. | 17 | Sources,dynamics in vivo,and application of astaxanthin and lutein in laying hens:A review显示文摘Astaxanthin(Ax)and lutein are important fat-soluble pigments and essential nutrients for human and animal health.Haematococcus pluvialis microalga and Phaffia rhodozyma yeast are ideal species for commercial Ax production.Marigold flowers are a main source of commercial lutein.Dynamics of dietary Ax and lutein in the gastrointestinal tract are similar to lipids,but their activities are tremendously challenged by many physiological and dietary factors;few data are available about these in poultry.Dietary Ax and lutein have insignificant effects on egg production and egg physical properties,but have pronounced effects on yolk color,nutrition,and functionality.The two pigments can also enhance antioxidative capacity and immune function of laying hens.A few studies have shown that Ax and lutein can improve fertilization and hatchability of laying hens.Considering the pigmentation and health benefits of Ax and lutein from hen feed to human food,the commercial availability,chicken yolk improvement,and immune function of Ax and lutein are the focuses of this review.The potential roles of carotenoids in the cytokine storm and gut microbiota are also briefly presented.The bioavailability,metabolism,and deposition of Ax and lutein in laying hens are suggested for future research. | Hanyi Shi Xuejuan Deng Xiaoyu Ji Ning Liu Huiyi Cai | 2023 | Animal Nutrition2023,,2: | 2 |

| 19 | Structure and functional interactions of IN080 actin/Arp module显示文摘The presence and functions of nuclear actin have been controversial due to the lack of molecular mechanisms.Nuclear actin and actin?『elated proteins (Arps) are subunits of several chromatin remodelers, including the evolutionarily conserved IN080 chromatin-remodeling complex. Here, we present an improved cryo-EM structure of the yeast IN080 complex and the first 3D reconstruction of the IN080 actin/Arp module.The modular and subunit architecture is defined using a combination of subunit deletion analysis and published crosslinking-mass spectrometry. The functional interactions of the IN080 actin/Arp module with a nucleosome is 3D EM reconstructed in two different binding states. Nucleosomes initially bind to the Arp8 subunit and the substantial conformational changes maximize nucleosome contacts of the actin/Arp module, which could promote the bound nucleosome to be engaged onto the IN080 ATPase domain. | 20 | Prognostic value of ^(18)F-fluorodeoxyglucose positron emission tomography using Deauville criteria in diffuse large B cell lymphoma treated with autologous hematopoietic stem cell transplantation显示文摘Objective: High-dose chemotherapy(HDC) followed by autologous hematopoietic stem cell transplantation(auto-HSCT) plays an important role in improving outcomes of diffuse large B cell lymphoma(DLBCL) patients.18 F-fluorodeoxyglucose(18 F-FDG) positron emission tomography(PET)/computed tomography(CT) has been widely accepted in response assessment and prediction of prognosis in DLBCL. Here, we report the value of 18 FFDG PET/CT pre-and post-HSCT in predicting outcomes of patients with DLBCL.Methods: DLBCL patients who had PET/CT scan before and after HSCT were included. PET results were interpreted based upon Deauville criteria. The prognostic value of 18 F-FDG PET/CT in auto-HSCT was evaluated.Results: Eighty-four patients were enrolled. In univariate analysis, pre-and post-HSCT PET findings were correlated with 3-year progression-free survival(PFS) [hazard ratio(HR)=4.391, P=0.001; HR=7.607, P<0.001] and overall survival(OS)(HR=4.792, P=0.008; HR=26.138, P<0.001). Patients receiving upfront auto-HSCT after firstline treatment had better outcomes than relapsed/refractory DLBCL patients(3-year PFS, P<0.001; 3-year OS,P<0.001). In the relapsed/refractory patients, pre-and post-HSCT PET findings were also associated with 3-year PFS(P=0.003 vs. P<0.001) and OS(P=0.027 vs. P<0.001). A significant correlation was observed between clinical response to chemotherapy before auto-HSCT and outcomes of patients in the entire cohort(3-year PFS, P<0.001;3-year OS, P<0.001) and in the subgroup of 21 patients with positive pre-HSCT PET(3-year PFS, P=0.084; 3-year OS, P=0.240). A significant association between survival and post-HSCT PET findings was observed in multivariate analysis(HR=5.168, P<0.001).Conclusions: PET results before and after HSCT are useful prognostic factors for DLBCL patients receiving HSCT.
