| 1 | Handgrip strength and health outcomes: Umbrella review of systematic reviews with meta-analyses of observational studies显示文摘Purpose:The aim of the present study was to assess both the credibility and strength of evidence arising from systematic reviews with meta-analyses of observational studies on handgrip strength and health outcomes.Methods:An umbrella review of systematic reviews with meta-analyses of observational studies was conducted.We assessed meta-analyses of observational studies based on random-effect summary effect sizes and their p values,95%prediction intervals,heterogeneity,small-study effects,and excess significance.We graded the evidence from convincing(Class I)to weak(Class IV).Results:From 504 articles returned in a search of the literature,8 systematic reviews were included in our review,with a total of 11 outcomes.Overall,nine of the 11 of the outcomes reported nominally significant summary results(p<0.05),with 4 associations surviving the application of the more stringent p value(p<106).No outcome presented convincing evidence.Three associations showed Class II evidence(i.e.,highly suggestive):(1)higher handgrip values at baseline were associated with a minor reduction in mortality risk in the general population(n=34 studies;sample size=1,855,817;relative risk=0.72,95%confidence interval(95%CI):0.670.78),(2)cardiovascular death risk in mixed populations(n=15 studies;relative risk=0.84,95%CI:0.780.91),and(3)incidence of disability(n=7 studies;relative risk=0.76,95%CI:0.660.87).Conclusion:The present results show that handgrip strength is a useful indicator for general health status and specifically for early all-cause and cardiovascular mortality,as well as disability.To further inform intervention strategies,future research is now required to fully understand mechanisms linking handgrip strength scores to these health outcomes. | Pinar Soysal Christopher Hurst Jacopo Demurtas Joseph Firth Reuben Howden Lin Yang Mark A.Tully Ai Koyanagi Petre Cristian Ilie Guillermo F.Lopez-Sanchez Lukas Schwingshackl Nicola Veronese Lee Smith | 2021 | Journal of Sport and Health Science2021,10,3: | 5 |

| 3 | Imputation of single-cell gene expression with an autoencoder neural network显示文摘Background:Single-cell RNA-sequencing(scRNA-seq)is a rapidly evolving technology that enables measurement of gene expression levels at an unprecedented resolution.Despite the explosive growth in the number of cells that can be assayed by a single experiment,scRNA-seq still has several limitations,including high rates of dropouts,which result in a large number of genes having zero read count in the scRNA-seq data,and complicate downstream analyses.Methods:To overcome this problem,we treat zeros as missing values and develop nonparametric deep learning methods for imputation.Specifically,our LATE(Learning with AuToEncoder)method trains an autoencoder with random initial values of the parameters,whereas our TRANSLATE(TRANSfer learning with LATE)method further allows for the use of a reference gene expression data set to provide LATE with an initial set of parameter estimates.Results:On both simulated and real data,LATE and TRANSLATE outperform existing scRNA-seq imputation methods,achieving lower mean squared error in most cases,recovering nonlinear gene-gene relationships,and better separating cell types.They are also highly scalable and can efficiently process over 1 million cells in just a few hours on a GPU.Conclusions:We demonstrate that our nonparametric approach to imputation based on autoencoders is powerful and highly efficient. | Md.Bahadur Badsha Rui Li Boxiang Liu Yang ILi Min Xian Nicholas EBanovich Audrey Qiuyan Fu | 2020 | Quantitative Biology2020,8,1: | 1 |
