| 1 | Application of bioactive hydrogels combined with dental pulp stem cells for the repair of large gap peripheral nerve injuries显示文摘Due to the limitations in autogenous nerve grafting or Schwann cell transplantation,large gap peripheral nerve injuries require a bridging strategy supported by nerve conduit.Cell based therapies provide a novel treatment for peripheral nerve injuries.In this study,we first experimented an optimal scaffold material synthesis protocol,from where we selected the 10%GFD formula(10%GelMA hydrogel,recombinant human basic fibroblast growth factor and dental pulp stem cells(DPSCs))to fill a cellulose/soy protein isolate composite membrane(CSM)tube to construct a third generation of nerve regeneration conduit,CSM-GFD.Then this CSM-GFD conduit was applied to repair a 15-mm long defect of sciatic nerve in a rat model.After 12 week post implant surgery,at histologic level,we found CSM-GFD conduit could regenerate nerve tissue like neuron and Schwann like nerve cells and myelinated nerve fibers.At physical level,CSM-GFD achieved functional recovery assessed by a sciatic functional index study.In both levels,CSM-GFD performed like what gold standard,the nerve autograft,could do.Further,we unveiled that almost all newly formed nerve tissue at defect site was originated from the direct differentiation of exogeneous DPSCs in CSM-GFD.In conclusion,we claimed that this third-generation nerve regeneration conduit,CSM-GFD,could be a promising tissue engineering approach to replace the conventional nerve autograft to treat the large gap defect in peripheral nerve injuries. | Lihua Luo Yan He Ling Jin Yanni Zhang Fernando P.Guastaldi Abdullkhaleg A.Albashari Fengting Hu Xiaoyan Wang Lei Wang Jian Xiao Lingli Li Jianming Wang Akon Higuchi Qingsong Ye | 2021 | Bioactive Materials2021,6,3: | 11 |
| 2 | VNF-FG design and VNF placement for 5G mobile networks显示文摘Network function virtualization(NFV) is envisioned as one of the critical technologies in 5thGeneration(5G) mobile networks. This paper investigates the virtual network function forwarding graph(VNFFG) design and virtual network function(VNF) placement for 5G mobile networks. We first propose a two-step method composed of flow designing and flow combining for generating VNF-FGs according to network service requests. For mapping VNFs in the generated VNF-FG to physical resources, we then modify the hybrid NFV environment with introducing more types of physical nodes and mapping modes for the sake of completeness and practicality, and formulate the VNF placement optimization problem for achieving lower bandwidth consumption and lower maximum link utilization simultaneously. To resolve this problem, four genetic algorithms are proposed on the basis of the frameworks of two existing algorithms(multiple objective genetic algorithm and improved non-dominated sorting genetic algorithm). Simulation results show that Greedy-NSGA-II achieves the best performance among our four algorithms. Compared with three non-genetic algorithms(random, backtracking mapping and service chains deployment with affiliation-aware), Greedy-NSGA-II reduces 97.04%, 87.76% and88.42% of the average total bandwidth consumption, respectively, and achieves only 13.81%, 25.04% and 25.41%of the average maximum link utilization, respectively. | 3 | Proteomic Analysis of Macrophages:A Potential Way to Identify Novel Proteins Associated with Activation of Macrophages for Tumor Cell Killing显示文摘One major mechanism through which macrophages effectively kill tumor cells requires cell to cell contact,indicating that certain molecules expressed on cell surface of activated macrophages may mediate the tumoricidal capability. Tumor necrosis factor (TNF) and nitric oxide (NO) are the two classical mediators of tumor cell death. However,evidence of discrepancy is accumulating indicating these known mediators do not appear to account for the broad and potent tumoricidal activity of macrophages. To obtain a full repertoire of tumoricidal activation-associated membrane proteins,we combined one-dimensional SDS-PAGE with capillary liquid chromatography-tandem mass spectrometry (LC-MS/MS). Using this technique,we identified 454 activated macrophage specifically expressed proteins with extremely high confidence,including most known activation markers of macrophages,such as NO synthase (iNOS),Ym1,cyclooxygenase,etc. Membrane bound TNF-α was also identified on activated macrophages. However,it was also detected on thioglycolate elicited macrophages,indicating this molecule may not play a key role in conjugation-dependent tumor cell killing. In contrast,although NO has not been assigned as an effector molecule of conjugation-dependent tumoricidal pathway,iNOS was identified from membrane fraction of activated macrophages,suggesting NO may be involved in conjugation-dependent tumoricidal mechanism,because iNOS association with plasma membrane is ideally suited to deliver NO directly into the contacted tumor cells. | 4 | Novel circular RNA expression profile of uveal melanoma revealed by microarray显示文摘Objective: The present study aimed to investigate circular RNA(circRNA) expression in uveal melanoma(UM).Methods: First,we used microarray to compare the expression profiles of circRNA in five UM samples and five normal uvea tissues.Next,bioinformatics analyses,including gene ontology(GO) analysis and pathway analysis,were applied to study these differentially expressed circRNAs to predict pathogenic pathways that may be involved.Quantitative real-time polymerase chain reaction(qRT-PCR) in 20 UM samples and 20 normal uvea samples was used to confirm the circRNA expression profiles obtained from the microarray data.Finally,we analyzed the interaction between validated circRNAs and their potential cancer-associated miRNA targets.Results: In total,50,579 circRNAs [fold change(FC) ≥2.0; P<0.05],including 20,654 up-regulated and 29,925 down-regulated circRNAs,were identified as differentially expressed between UM tissues and normal uvea tissues.We used qRT-PCR to verify seven dysregulated circRNAs indicated by the microarray data,including hsa_circ_0119873,hsa_circ_0128533,hsa_circ_0047924,hsa_circ_0103232,hsa-circRNA10628-6,hsa_circ_0032148 and hsa_circ_0133460,which may be promising candidates to study future molecular mechanisms.Conclusions: This study explored,for the first time,the abnormal expression of circRNAs in UM and described the expression profile of circRNAs,providing a new potential target for the mechanism of UM and future treatment of UM. | Xuan Yang Yang Li Yueming Liu Xiaolin Xu Yingzhi Wang Yanni Yan Wenjia Zhou Jingyan Yang Wenbin Wei | 2018 | Chinese Journal of Cancer Research2018,30,6: | 6 |

| 8 | Effective E.coli inactivation of core-shell ZnO@ZIF-8 photocatalysis under visible light synergize with peroxymonosulfate:Efficiency and mechanism显示文摘How to utilize inexhaustible solar light as a means of disinfection technology for its cheap and green remains a challenge.In this work,core-shell ZnO@ZIF-8 was synthesized and used for bacterial inactivation synergizing with peroxymonosulfate(PMS)under visible light irradiation.It took 50 min to achieve thorough sterilization for 7.5-log Escherichia coli(E.coli)cells in vis/PMS/ZnO@ZIF-8 system,compared with that 4.5-log reduction completed in vis/PMS/ZnO system under the same conditions.The enhanced photocatalytic disinfection mechanisms of fabricated ZnO@ZIF-8 were investigated by UV-vis diffuse reflectance spectra,electrochemical impedance spectra and Mott-Schottky plots.The promoted bactericidal efficiency was attributed to higher charge-separation efficiency and stronger oxidation ability of photo-generated holes.Moreover,it was found that^(1)O_(2)and·OH induced bacterial cell lesion process,and the former was the main active species.The external reactive oxygen species(ROS)caused a series of cell wall damage,intercellular ROS up-regulation and genome DNA unwinding,finally resulted in irreversible bacterial death.A two-route mechanism in vis/PMS/ZnO@ZIF-8 system was proposed,in which the generation of^(1)O_(2)was supposed as the product of the oxygen oxidation of photo-generated holes and PMS dissociation.Our work is expected to provide advanced information about a low-cost water disinfection technology of visible light photocatalysis. | Yanni Jiang Zhaokun Xiong Jianbo Huang Feng Yan Gang Yao Bo Lai | 2022 | Chinese Chemical Letters2022,33,1: | 2 |

| 15 | Seed filling dynamic traits of oil flax in response to nitrogen and phosphorus显示文摘Crop yield is primarily seed-filling-limited in production system under field conditions.This study was aimed to determine whether seed filling traits of oil flax(Linum usitatissimum L.)could be controlled by phosphorus(P),nitrogen(N),and phosphorus and nitrogen(NP)supply.Effects on seed filling traits were investigated in 2 years including capsule diameter,capsule height,capsule dry matter(DM),seed DM per capsule,pericarp DM per capsule,protein content and oil content.DM translocation from pericarps to seed,translocation efficiency,and contribution of photoassimilates during seed filling period were also detected.In a randomized complete block design,cultivar'Longyaza 1'was grown under P(33 kg P/ha),N(75 kg N/ha),and NP(33 kg P/ha and 75 kg N/ha)along with a zerofertilizer(CK)treatment in 2013 and 2014.Results suggested that DM translocation efficiency and contribution efficiency increased to different extent due to P,N or NP application.At 42 DAA(days after anthesis),seed DM per capsule reached the greatest,while protein content and pericarp DM obtained the least level.However,the highest oil content was detected at 35 DAA.A significant positive linear relationship was observed between seed DM,capsule DM and DM translocation in both years.Protein content showed inconsistent relation with oil content.The results indicated that appropriate N and P management could be an effective approach to increase oil flax production. | Yaping Xie Bin Wang Limin Wang Junyi Niu Wei Zhao Bin Yan Li Zhao Zhao Dang Wenjuan Li Yanni Qi Jianping Zhang | 2019 | Oil Crop Science2019,4,3: | 1 |
