1Structure and function of aggrecan显示文摘Aggrecan is the major proteoglycan in the articular cartilage. This molecule is important in the proper functioning of articular cartilage because it provides a hydrated gel structure (via its interaction with hyaluronan and link protein) that endows the cartilage with load-bearing properties. It is also crucial in chondroskeletal morphogenesis during development. Aggrecan is a multimodular molecule expressed by chondrocytes. Its core protein is composed of three globular domains (G1, G2, and G3) and a large extended region (CS) between G2 and G3 for glycosaminoglycan chain attachment. G1 comprises the amino terminus of the core protein. This domain has the same structural motif as link protein. Functionally, the G1 domain interacts with hyaluronan acid and link protein, forming stable ternary complexes in the extracellular matrix. G2 is homologous to the tandem repeats of G1 and of link protein and is involved in product processing. G3 makes up the carboxyl terminus of the core protein. It enhances glycosaminoglycan modification and product secretion. 2The interaction of versican with its binding partners显示文摘Versican belongs to the family of the large aggregating chondroitin sulfate proteoglycans located primarily within theextracellular matrix (ECM). Versican, like other members of its family, has unique N- and C-terminal globular regions,each with multiple motifs. A large glycosaminoglycan-binding region lies between them. This review will begin byoutlining these structures, in the context of ECM proteoglycans. The diverse binding partners afforded to versican byvirtue of its modular design will then be examined. These include ECM components, such as hyaluronan, type Icollagen, tenascin-R, fibulin-1, and -2, fibrillin-1, fibronectin, P- and L-selectins, and chemokines. Versican also binds tothe cell surface proteins CD44, integrin β1, epidermal growth factor receptor, and P-selectin glycoprotein ligand-1.These multiple interactors play important roles in cell behaviour, and the roles of versican in modulating such processesare discussed.Yao Jiong WU David P. 9Rectal arterio-portal fistula: An unusual cause of persistent bleeding per rectum following a proximal spleno-renal shunt显示文摘Gastrointestinal arterio-venous malformations are a known cause of gastrointestinal bleeding. We present a rare case of persistent rectal bleeding due to a rectal arterio-portal venous fistula in the setting of portal hypertension secondary to portal vein thrombosis. The portal hypertension was initially surgically treated with splenectomy and a proximal splenorenal shunt. However, rectal bleeding persisted even after surgery, presenting us with a diagnostic dilemma. The patient was re-evaluated with a computed tomography mesenteric angiogram which revealed a rectal arterio-portal fistula. Arterio-portal fistulas are a known but rare cause of portal hypertension, and possibly the underlying cause of continued rectal bleeding in this case. This was successfully treated using angiographic localizationand super-selective embolization of the rectal arterioportal venous fistula via the right internal iliac artery. The patient subsequently went on to have a full term pregnancy. 18整合素α5和肌动蛋白在周期性压力下椎间盘细胞中的表达显示文摘目的探讨整合素α5和肌动蛋白在周期性压力下椎间盘细胞中的表达。方法对猪椎间盘组织进行体外细胞的分离和培养,对细胞施加周期性液压,通过对细胞的形态学观察、Western免疫印迹和免疫组化染色,分别检测整合素α5和肌动蛋白在周期性压力下椎间盘细胞中的表达。结果经加压后,纤维环细胞和髓核细胞均可见体积缩小,由多角形转变为细长形,整合素α5和肌动蛋白在纤维环细胞和髓核细胞中的表达均明显减少。结论体外培养的单层椎间盘细胞在受力状态下,整合素α5的表达减少,整合素α5将力信号转换到细胞内时,进一步影响到与其关系密切的肌动蛋白。这可能是椎间盘细胞力学传导的重要机制。于胜吉 邱贵兴 Yang Burton Roth Sandra Whyne Cari Yee Albert 2005中华外科杂志2005,43,24:1
