1Ferroptosis is essential for diabetic cardiomyopathy and is prevented by sulforaphane via AMPK/NRF2 pathways显示文摘Herein,we define the role of ferroptosis in the pathogenesis of diabetic cardiomyopathy(DCM)by examining the expression of key regulators of ferroptosis in mice with DCM and a new ex vivo DCM model.Advanced glycation end-products(AGEs),an important pathogenic factor of DCM,were found to induce ferroptosis in engineered cardiac tissues(ECTs),as reflected through increased levels of Ptgs2 and lipid peroxides and decreased ferritin and SLC7 A11 levels.Typical morphological changes of ferroptosis in cardiomyocytes were observed using transmission electron microscopy.Inhibition of ferroptosis with ferrostatin-1 and deferoxamine prevented AGE-induced ECT remodeling and dysfunction.Ferroptosis was also evidenced in the heart of type 2 diabetic mice with DCM.Inhibition of ferroptosis by liproxstatin-1 prevented the development of diastolic dysfunction at 3 months after the onset of diabetes.Nuclear factor erythroid 2-related factor 2(NRF2)activated by sulforaphane inhibited cardiac cell ferroptosis in both AGE-treated ECTs and hearts of DCM mice by upregulating ferritin and SLC7 A11 levels.The protective effect of sulforaphane on ferroptosis was AMP-activated protein kinase(AMPK)-dependent.These findings suggest that ferroptosis plays an essential role in the pathogenesis of DCM;sulforaphane prevents ferroptosis and associated pathogenesis via AMPK-mediated NRF2 activation.This suggests a feasible therapeutic approach with sulforaphane to clinically prevent ferroptosis and DCM.Xiang Wang Xinxin Chen Wenqian Zhou Hongbo Men Terigen Bao Yike Sun Quanwei Wang Yi Tan Bradley B.Keller Qian Tong Yang Zheng Lu Cai 2022Acta Pharmaceutica Sinica B2022,12,2:20
2Multi-Modality Video Representation for Action Recognition显示文摘Nowadays,action recognition is widely applied in many fields.However,action is hard to define by single modality information.The difference between image recognition and action recognition is that action recognition needs more modality information to depict one action,such as the appearance,the motion and the dynamic information.Due to the state of action evolves with the change of time,motion information must be considered when representing an action.Most of current methods define an action by spatial information and motion information.There are two key elements of current action recognition methods:spatial information achieved by sampling sparsely on video frames’sequence and the motion content mostly represented by the optical flow which is calculated on consecutive video frames.However,the relevance between them in current methods is weak.Therefore,to strengthen the associativity,this paper presents a new architecture consisted of three streams to obtain multi-modality information.The advantages of our network are:(a)We propose a new sampling approach to sample evenly on the video sequence for acquiring the appearance information;(b)We utilize ResNet101 for gaining high-level and distinguished features;(c)We advance a three-stream architecture to capture temporal,spatial and dynamic information.Experimental results on UCF101 dataset illustrate that our method outperforms other previous methods.Chao Zhu Yike Wang Dongbing Pu Miao Qi Hui Sun Lei Tan 2020Journal on Big Data2020,2,3:1
3Tumor-specific and photothermal-augmented chemodynamic therapy by ferrocene-carbon dot-crosslinked nanoparticles显示文摘Extensive research have been devoted to the exploration of multifunctional theranostic agents for cancer,but the poor tumor specificity and unsatisfactory treatment efficacy are some of the critical obstacles for their clinical translations.Herein,ferrocene-carbon dot-crosslinked nanoparticles(Fc-CD NPs)were designed and fabricated for achieving highly specific and photothermal-augmented chemodynamic therapy(CDT).The Fc-CD NPs were found not only to inherit the immanent fluorescence,photoacoustic,and photothermal properties of carbon dots(CDs),but also be endowed with CDT that could occur selectively in tumor microenvironment(TME)due to the presence of Fc for triggering Fenton reaction.Moreover,the enlarged particle size of Fc-CD NPs facilitated their effective accumulation at tumor sites,thus realizing great improvement for antitumor treatment outcomes.Once docking at tumor and being exposed to 660 nm laser irradiation,significantly amplified CDT effect of Fc-CD NPs was observed due to heataccelerating generation of reactive oxygen species(ROS).More interestingly,since the produced ROS could in turn alleviate the thermal-resistance of photothermal therapy(PTT),the therapeutic efficiency of integrated PTT and CDT was synergized to the maximum extent.This study on the one hand provides a facile approach to fabricate CDs-based multifunctional theranostic nanoplatform with enhanced tumor accumulation and specificity,on the other hand emphasizes the merits of synergizing mutually beneficial therapeutic modalities for more efficient cancer therapy.Shan Sun Qiao Chen Yike Li Yao Yu Zhongjun Li Hengwei Lin 2022SmartMat2022,3,2:1

6An Analysis of the Development Process of the Liberal Democratic Party-Taking the Japanese Prime Minister Election as an Example显示文摘This paper combs the development process of the Japanese Liberal Democratic Party,and summarizes some characteristics of the Liberal Democratic Party from the development process.The paper holds that the Liberal Democratic Party is historically inherited and conservative,and is affected by external forces in the process of development,including the influence of the United States,the zaibatsu,and other domestic political parties.Within the LDP,there is factional struggle.The LDP simulates the competition between political parties through factional struggle to update policies and realize its own political ideas.This factional struggle is closely related to the zaibatsu.Politicians rely on the zaibatsus to provide themselves with economic support during the election,and make the zaibatsus profitable through policies after they are elected prime minister.Zaibatsus meet their own interests by supporting different factions.There is a deep binding between zaibatsus and politicians.When the power money proposal is too obvious,it will arouse national resentment,lead to a decline in their own support rate,and give other factions and even other political parties an opportunity to take advantage of it.SUN Yike 2023Journal of US-China Public Administration2023,20,3:0
7Molecular mechanisms of seasonal photoperiod effects of the pineal gland on the hippocampus in rats显示文摘Background:Based on the theory of“five Zang-organs corresponding to the seasons”in traditional Chinese medicine(TCM),physiological functions including emotions vary with the seasons.We aimed to investigate the seasonal photoperiod effects of melatonin(MT)released from the pineal gland on the MT receptor(MTR)-Gs/Gi-cyclic adenosine monophosphate(cAMP)-protein kinase A(PKA)-cAMP response element-binding protein(CREB)signaling pathway in the hippocampus.Methods:Rats were divided into three groups:control,operation(surgery with pineal gland removal),and pseudo-operation groups(same surgery as operation group but without removing pineal gland),and fed at specific time across the four seasons.The levels of MTR,adenylate cyclase(AC),cAMP,PKA,and CREB in the hippocampus were analyzed using ELISA.The concentrations of Gs and Gi were analyzed using Western blot.The expression of CREB mRNA was detected by PCR.Results:For intragroup comparisons,compared with spring,the levels of Gs/Gi in the control group were higher in summer,autumn,and winter(P=.009 in summer;P<.001 in autumn and winter);the levels of MTR,cAMP,PKA,and CREB in the control group were significantly higher in autumn and winter than in spring(all P<.001).The levels of MTR,cAMP,PKA,and CREB in each season were significantly lower in the operation group than in the control group(all P<.05).Significant differences were noted in Gs/Gi levels between the operation group and control group in spring,autumn,and winter(all P<.05).Conclusion:Our findings suggest that MTR-Gs/Gi-cAMP-PKA-CREB signaling pathway is involved in the seasonal photoperiod effects of the pineal gland on the hippocampus and may underpin seasonal changes in emotions.It can support the prevention and treatment of the seasonal onset of mental disorders,and enrich the theory of“five Zang-organs corresponding to the seasons”.Qi Han Guowen Du Leilei Liu Lepeng Wang Wenna Li Hairong Zhang Yike Sun Pei Zhu Ruisen Hao Shuran Ma 2021Journal of Traditional Chinese Medical Sciences2021,8,2:0
8Nanofiber self-consistent additive manufacturing process for 3D microfluidics显示文摘3D microfluidic devices have emerged as powerful platforms for analytical chemistry,biomedical sensors,and microscale fluid manipulation.3D printing technology,owing to its structural fabrication flexibility,has drawn extensive attention in the field of 3D microfluidics fabrication.However,the collapse of suspended structures and residues of sacrificial materials greatly restrict the application of this technology,especially for extremely narrow channel fabrication.In this paper,a 3D printing strategy named nanofiber self-consistent additive manufacturing(NSCAM)is proposed for integrated 3D microfluidic chip fabrication with porous nanofibers as supporting structures,which avoids the sacrificial layer release process.In the NSCAM process,electrospinning and electrohydrodynamic jet(E-jet)writing are alternately employed.The porous polyimide nanofiber mats formed by electrospinning are ingeniously applied as both supporting structures for the suspended layer and percolating media for liquid flow,while the polydimethylsiloxane E-jet writing ink printed on the nanofiber mats(named construction fluid in this paper)controllably permeates through the porous mats.After curing,the resultant construction fluid–nanofiber composites are formed as 3D channel walls.As a proof of concept,a microfluidic pressure-gain valve,which contains typical features of narrow channels and movable membranes,was fabricated,and the printed valve was totally closed under a control pressure of 45 kPa with a fast dynamic response of 52.6 ms,indicating the feasibility of NSCAM.Therefore,we believe NSCAM is a promising technique for manufacturing microdevices that include movable membrane cavities,pillar cavities,and porous scaffolds,showing broad applications in 3D microfluidics,soft robot drivers or sensors,and organ-on-a-chip systems.Bin Qiu Xiaojun Chen Feng Xu Dongyang Wu Yike Zhou Wenchang Tu Hang Jin Gonghan He Songyue Chen Daoheng Sun 2022Microsystems & Nanoengineering2022,8,5:0

10Effects of seasonal changes on T-helper 1/T-helper 2 immune balance and eczema onset in rats显示文摘Objective: To explore the impact of seasonal changes on the T-helper 1(Th1)/T-helper 2(Th2) immune balance of the body in order to elucidate the internal immunological mechanisms underlying seasonal eczema.Methods: Eighty 4-week-old male Sprague-Dawley rats were divided into 5 season groups kept in corresponding season simulation environments, and subdivided into the model group and the control group. The Eczema Area and Severity Index(EASI) and scratching frequency were evaluated. The serum levels of immunoglobulin E(IgE), interleukin-2(IL-2), interleukin-12(IL-12), and interferon-gamma(IFN-γ), interleukin-4(IL-4), interleukin-25(IL-25), and interleukin-31(IL-31), and melatonin(MT), as well as the MT receptor(MTR) levels in the spleen, were detected by enzyme-linked immunosorbent assay analysis. The mRNA expression levels of T-bet and GATA3 were detected by quantitative real-time polymerase chain reaction.Results: EASI scores and the scratching frequency of the model group were higher in the long summer than in the other 4 seasons. Meanwhile, the serum levels of IgE and the Th2 cytokines IL-4, IL-25, and IL-31, as well as GATA3 mRNA expression levels, were high during the long summer in both groups. However, the serum levels of the Th1 cytokines IL-2, IL-12, and IFN-γ, as well as MT, MTR, and T-bet mRNA levels, were lower during the long summer. In all 5 seasonal groups(spring, summer, long summer, autumn, and winter), the levels of all immune factors, especially IL-4 and IL-31, were higher in the model group than those in the control group,while the concentrations of MT and MTR were lower.Conclusion: Under long light, hot, and humid conditions in the long summer, the body is more likely to suffer from Th2-dominated immune imbalance. This is the internal mechanism behind the high incidence and severity of eczema during the long summer. 11Seasonal photoperiodic influence of pineal melatonin on hypothalamic-pituitary-adrenal axis-hippocampal-receptor in male rats显示文摘Background: Based on the effect of seasonal changes on human visceral function, this study investigated the impact of seasonal photoperiod of the pineal body on hypothalamic-pituitary-adrenal axis-hippocampal-receptor in rats, aiming to reveal the mechanism by which pineal gland melatonin regulates the seasonal secretion of hippocampal neurotransmitters.Methods: Vernal equinox, summer solstice, autumn equinox, and winter solstice were selected as four experimental time points, and rats were randomly divided into normal control group, sham operation group, and pinealectomized group. The seasonal changes in corticotropin-releasing hormone(CRH),adrenocorticotropic hormone(ACTH), corticosterone, hypothalamic melatonin receptor(MTR), and hippocampal corticosterone receptor(CORTR) were examined by enzyme-linked immunosorbent assay.Results: Comparing the same group between different seasons, we showed that in the normal control group, CRH, ACTH, corticosterone, and MTR were higher, while CORTR was lower in autumn and winter than in spring(all P <.05). Compared with the normal control group, the pinealectomized group showed higher levels of corticosterone(P =.01), MTR(P =.01), and CORTR(P =.03) during spring;reduced levels of MTR and CORTR(both P <.001) during summer;higher levels of ACTH(P =.001) and MTR(P <.001),and lower levels of CRH(P =.001), corticosterone(P <.001), and CORTR(P =.003) during autumn;and lower levels of CRH(P <.001) and MTR(P =.004), and higher level of ACTH(P <.001) in winter.Conclusions: Seasonal photoperiod acts on the pineal gland to secrete different levels of melatonin,resulting in seasonal changes in the hypothalamic-pituitary-adrenal axis-hippocampal-receptor, which may be the pathophysiological basis for the onset of seasonal affective disorder.Wenna Li Ting Li Leilei Liu Qi Han Hairong Zhang Yike Sun Ruisen Hao Shuran Ma 2022Journal of Traditional Chinese Medical Sciences2022,9,2:0
