1Anthropogenic Impact on Surface Water Quality in Taihu Lake Region,China显示文摘Taihu Lake region is one of the most industrialized areas in China, and the surface water is progressively susceptible to anthropogenic pollution. The physicochemical parameters of surface water quality were determined at 20 sampling sites in Taihu Lake region, China in spring, summer, autumn, and winter seasons of 2005-2006 to assess the effect of human activities on the surface water quality. Principal component analysis (PCA) and cluster analysis (CA) were used to identify characteristics of the water quality in the studied water bodies. PCA extracted the first three principal components (PCs), explaining 80.84% of the total variance of the raw data. Especially, PC1 (38.91%) was associated with NH 4 -N, total N, soluble reactive phosphorus, and total P. PC2 (22.70%) was characterized by NO 3 -N and temperature. PC3 (19.23%) was mainly associated with pH and dissolved organic carbon. CA showed that streams were influenced by urban residential subsistence and livestock farming contributed significantly to PC1 throughout the year. The streams influenced by farmland runoff contributed most to PC2 in spring and winter compared with other streams. PC3 was affected mainly by aquiculture in spring, rural residential subsistence in summer, and livestock farming in fall and winter seasons. Further analyses showed that farmlands contributed significantly to nitrogen pollution of Taihu Lake, while urban residential subsistence and livestock farming also polluted water quality of Taihu Lake in rainy season. 3Hepatocyte growth factor gene transfer effects on the femoral and intramuscular nerve in a canine model of lower limb ischemia显示文摘BACKGROUND: Recent advancements in gene therapy have provided new methodology for treating ischemia in lower extremities. Gene transfer of angiogenic factors to ischemic tissues may promote local proliferation of new vessels and form collateral circulation. OBJECTIVE: To observe histopathological changes in the femoral and intramuscular nerve three months after intramuscular injection of hepatocyte growth factor (HGF) into the peripheral skeletal muscle in a canine model of lower limb ischemia. DESIGN: Randomized occlusion modelled and verification animal study. SETTING: Experimental Center, Lanzhou General Hospital of Lanzhou Military Area Command of Chinese PLA. MATERIALS: This study was performed at Animal Experimental Center, Lanzhou General Hospital of Lanzhou Military Area Command of Chinese PLA from September to November 2006. A total of eight male mongrel dogs, weighing 12–15 kg and 1.5–3 years of age, were selected for this study. This experimental study was in accordance with local ethics standards. Recombinant plasmid carrying HGF (pUDKH) and occlusion model plasmid (pUDK) were provided by the Third Laboratory of Radiation Medical Institute, Academy of Military Medical Sciences of PLA. METHODS: Grouping and model establishment: under anesthesia, complete vascular occlusion models were established on the left lower extremities. The experimental dogs were randomly divided into a model group and a pUDKH treatment group, with four dogs in each group. Dogs in the pUDKH group were injected with 0.15 mg/kg pUDKH. Ten minutes later, intramuscular injections were performed at three spots into the peripheral skeletal muscle of the left hind limb, as well as lateral injections at two spots. The injection volume at each spot was 0.2 mL. Dogs in the model group were injected with pUDK, and dosage and injection method were identical to the treatment group. MAIN OUTCOME MEASURES: Histopathological changes in the femoral nerve, as well as internal and external intramuscular nerve tissues in the hind limb of dogs three months after plasmid injection under optic microscope. RESULTS: (1) Histopathological changes in the femoral nerve: tiny nerves from the femoral nerve to the intramuscular nerve exhibited marked degeneration in the model group. The degenerating features included neurites, myelin sheaths, and Schwann cell nuclei. Neuropathy in the pUDKH treatment group was not detected. (2) Histopathological changes of the intramuscular nerve: large and irregular vacuoles were present on several longitudinal sections of intramuscular nerve fibers in the model group, as well as annular-shaped blank regions on transverse sections of peripheral neurites. In the pUDKH treatment group, large, blank regions were present in several segments of partial nerve fibers of the longitudinal intramuscular nerve region, but only a few nerve fibers exhibited annular-shaped blank regions on the transverse section of peripheral neurites. 12Rare case of intussusception in an adult with acute myeloid leukemia显示文摘Intussusception is rarely reported in adult patients with acute leukemia. We report a case of intussusception in a 29-year-old woman with acute myeloid leukemia(AML). She developed right lower quadrant pain, fever, and vomiting on day 16 of induction chemotherapy.Physical examination showed tenderness and guarding at the right lower quadrant of the abdomen. Abdominal computed tomography(CT) showed distension of the cecum and ascending colon, which were filled with loops of small bowel, and herniation of the ileocecal valve into the cecum. We proceeded to laparotomy and revealed ileocecal intussusception with the ileocecal valve as the leading point. The terminal ileum was thickened and invaginated into the cecum, which showed gangrenous changes. Right hemicolectomy was performed and microscopic examination of the colonic tissue showed infiltration of leukemic cells. The patient recovered after the operation and was subsequently able to continue treatment for AML. This case demonstrates that the diagnosis of intussusception is difficult because the presenting symptoms can be non-specific, but abdominal CT can be informative for preoperative diagnosis. Resection of the involved bowel is recommended when malignancy is suspected or confirmed. Intussusception should be considered in any leukemia patients presenting with acute abdomen. 16Lead acetate in drinking water is toxic to hippocampal tissue Measuring relative protein changes using tissue array detection显示文摘BACKGROUND: Lead can cause structural changes in the hippocampus, followed by damage to learning and memory functions, but its specific mechanisms are not yet clear. OBJECTIVE: To observe long-term toxicity of high-dose lead in drinking water on hippocampal tissue in rats, and analyze the potential association of oxidative damage, cell apoptosis, and pathology. DESIGN, TIME AND SETTING: A randomized, controlled animal experiment was performed at the Center for Medical Experiment, Lanzhou General Hospital of Lanzhou Military Area Command of Chinese PLA from May 2007 to October 2008. MATERIALS: Rabbit anti Bcl-2, Bax, and inducible nitric oxide synthase (iNOS) polyclonal antibodies were purchased from Santa Cruz Biotechnology, USA. An streptavidin-peroxidase immunohistochemistry kit and concentrated DAB kit were purchased from Beijing Zhongshan Biotechnology Company Limited, China. Crystal violet was purchased from Sigma, USA. METHODS: A total of 72 Wistar rats, aged 3 months, were randomly divided into control, low-, middle-, and high-dose lead groups, with 18 rats per group. Animal models were established through free drinking water contaminated by Pb2+ for 1, 3, and 6 months, respectively. MAIN OUTCOME MEASURES: The general toxicity of lead was dynamically observed; the levels of Pb2+ in the blood and brain tissue homogenate were detected using atomic absorption method; pathological changes were observed using hematoxylin-eosin staining and tigroid body staining; the protein expression levels of Bcl-2, Bax, and iNOS were dynamically observed using streptavidin-peroxidase immunohistochemistry of the hippocampus. RESULTS: Lead exposure reduced autonomic activities, produced a slumped appearance, slow responses, and lusterless fur, especially in the high-dose group. The amount of ingestion and hydroposia showed a decreasing trend, especially in middle- and high-dose groups. Lead levels in whole blood and brain homogenate were higher than controls (P < 0.01). Lead caused degeneration of hippocampal neurons and pyknosis, with fewer tigroid bodies, especially in high-dose lead group. Bcl-2 expression decreased with increasing lead dose (P < 0.01), whereas lead dose-dependently increased Bax levels (P < 0.01) and iNOS levels (P < 0.05). CONCLUSION: High levels of Pb2+ may disrupt hippocampal structure by passing through the blood brain barrier. 17Pharmacokinetics of Idazoxan in Deers Following Intramuscular Administration显示文摘The aim of this experiment was to determine the pharmacokinetics of hydrochloric idazoxan in deers plasma after intramuscular(IM) dosing.Six clinical healthy Cervus nippon Temmincks were injected with the idazoxan solution at the dose of 0.44 mg ? kg-1.Eight mL blood sample was taken from a jugular vein and plasma was separated for drug determination by using liquid chromatography with tandem mass spectrometric detection.Idazoxan pharmacokinetic parameters were simulated by noncompartmental analyses.The results showed that the absorption and elimination of hydrochloric idazoxan in plasma was quick by route of administration,the half-lives of absorption(t1/2Ka) and elimination(t1/2Ke) were(0.2094 ± 0.0341) min,and(13.1842±0.2353) min,respectively,the area under the plasma drug concentration-time curve from 0 to ∞(AUC) was(0.0700±0.0035)(mg ? mL-1) ? min,the maximum concentration in the plasma(Cmax) was(0.0047±0.0005) mg ? mL-1,peaking at(12.4618±0.1198) min after dosing.In conclusion,these data indicated that the kinetics of hydrochloric idazoxan were fitted to one compartment model with first order absorption,which was characterized by rapid drug action,and fast metabolism with few residue in the blood.YIN Baishuang GAO Li WANG Hongbin HA Da 2011Journal of Northeast Agricultural University(English Edition)2011,18,3:0
