| 4 | Potential rat model of anxiety-like gastric hypersensitivity induced by sequential stress显示文摘AIM To establish a rat model of anxiety-like gastric hyper-sensitivity(GHS) of functional dyspepsia(FD) induced by novel sequential stress.METHODS Animal pups were divided into two groups from postnatal day 2: controls and the sequential-stress-treated. The sequential-stress-treated group received maternal separation and acute gastric irritation early in life and restraint stress in adulthood; controls were reared undisturbed with their mothers. Rats in both groups were followed to adulthood(8 wk) at which point the anxietylike behaviors and visceromotor responses to gastric distention(20-100 mm Hg) and gastric emptying were tested. Meanwhile, alterations in several anxiety-related brain-stomach modulators including 5-hydroxytryptamine(5-HT), γ-aminobutyric acid(GABA), brain-derived neurotrophic factor(BDNF) and nesfatin-1 in the rat hippocampus, plasma and gastric fundus and the 5-HT1 A receptor(5-HT1 AR) in the hippocampal CA1 subfield and the mucosa of the gastric fundus were examined.RESULTS Sequential-stress-treated rats simultaneously demonstrated anxiety-like behaviors and GHS in dose-dependent manner compared with the control group. Although rats in both groups consumed similar amount of solid food, the rate of gastric emptying was lower in the sequentialstress-treated rats than in the control group. Sequential stress significantly decreased the levels of 5-HT(51.91 ± 1.88 vs 104.21 ± 2.88, P < 0.01), GABA(2.38 ± 0.16 vs 5.01 ± 0.13, P < 0.01) and BDNF(304.40 ± 10.16 vs 698.17 ± 27.91, P < 0.01) in the hippocampus but increased the content of nesfatin-1(1961.38 ± 56.89 vs 1007.50 ± 33.05, P < 0.01) in the same site; significantly decreased the levels of 5-HT(47.82 ± 2.29 vs 89.45 ± 2.61, P < 0.01) and BDNF(257.05 ± 12.89 vs 536.71 ± 20.73, P < 0.01) in the plasma but increased the content of nesfatin-1 in it(1391.75 ± 42.77 vs 737.88 ± 33.15, P < 0.01); significantly decreased the levels of 5-HT(41.15 ± 1.81 vs 89.17 ± 2.31, P < 0.01) and BDNF(226.49 ± 12.10 vs 551.36 ± 16.47, P < 0.01) in the gastric fundus but increased the content of nesfatin-1 in the same site(1534.75 ± 38.52 vs 819.63 ± 38.04, P < 0.01). METHOD: Chromatographic separations by silica gel, C18 reversed phase silica gel, and high-performance liquid chromatography(HPLC) were used. All of the structures were elucidated on the basis of spectroscopic analysis and chemical methods. RESULTS: Five 9, 19-cycloartane triterpenes,(3β, 12β, 15α, 24R)-12, 2'-diacetoxy-24, 25-epoxy-15-hydroxy-16, 23-dione-3-O-α-L-arabinopyranoside(1), actein(2), 23-epi-26-deoxyactein(3), asiaticoside B(4), and 12β-hydroxycimigenol(5) were isolated from the roots of Cimicifuga foetida. | 7 | Delayed improvements in visual memory task performance among chronic schizophrenia patients after high-frequency repetitive transcranial magnetic stimulation显示文摘BACKGROUND Cognitive impairments are core characteristics of schizophrenia,but are largely resistant to current treatments.Several recent studies have shown that highfrequency repetitive transcranial magnetic stimulation(rTMS)of the left dorsolateral prefrontal cortex(DLPFC)can reduce negative symptoms and improve certain cognitive deficits in schizophrenia patients.However,results are inconsistent across studies.AIM To examine if high-frequency rTMS of the DLPFC can improve visual memory deficits in patients with schizophrenia.METHODS Forty-seven chronic schizophrenia patients with severe negative symptoms on stable treatment regimens were randomly assigned to receive active rTMS to the DLPFC(n=25)or sham stimulation(n=22)on weekdays for four consecutive weeks.Patients performed the pattern recognition memory(PRM)task from the Cambridge Neuropsychological Test Automated Battery at baseline,at the end of rTMS treatment(week 4),and 4 wk after rTMS treatment(week 8).Clinical symptoms were also measured at these same time points using the Scale for the Assessment of Negative Symptoms(SANS)and the Positive and Negative Syndrome Scale(PANSS).RESULTS There were no significant differences in PRM performance metrics,SANS total score,SANS subscores,PANSS total score,and PANSS subscores between active and sham rTMS groups at the end of the 4-wk treatment period,but PRM performance metrics(percent correct and number correct)and changes in these metrics from baseline were significantly greater in the active rTMS group at week 8 compared to the sham group(all P<0.05).Active rTMS treatment also significantly reduced SANS score at week 8 compared to sham treatment.Moreover,the improvement in visual memory was correlated with the reduction in negative symptoms at week 8.In contrast,there were no between-group differences in PANSS total score and subscale scores at either week 4 or week 8(all P>0.05).CONCLUSION High-frequency transcranial magnetic stimulation improves visual memory and reduces negative symptoms in schizophrenia,but these effects are delayed,potentially due to the requirement for extensive neuroplastic changes within DLPFC networks. | Xiang-Dong Du Zhe Li Nian Yuan Ming Yin Xue-Li Zhao Xiao-Li Lv Si-Yun Zou Jun Zhang Guang-Ya Zhang Chuan-Wei Li Hui Pan Li Yang Si-Qi Wu Yan Yue Yu-Xuan Wu Xiang-Yang Zhang | 2022 | World Journal of Psychiatry2022,12,9: | 3 |

| 11 | Identification of Fucans from Four Species of Sea Cucumber by High Temperature ~1H NMR显示文摘Acidic polysaccharide, which has various biological activities, is one of the most important components of sea cucumber. In the present study, crude polysaccharide was extracted from four species of sea cucumber from three different geographical zones, Pearsonothuria graeffei(Pg) from Indo-Pacific, Holothuria vagabunda(Hv) from Norwegian Coast, Stichopus tremulu(St) from Western Indian Ocean, and Isostichopus badionotu(Ib) from Western Atlantic. The polysaccharide extract was separated and purified with a cellulose DEAE anion-exchange column to obtain corresponding sea cucumber fucans(SC-Fucs). The chemical property of these SC-Fucs, including molecular weight, monosaccharide composition and sulfate content, was determined. Their structure was compared simply with fourier infrared spectrum analyzer and identified with high temperature 1H nuclear magnetic resonance spectrum analyzer(NMR) and room temperature 13 C NMR. The results indicated that Fuc-Pg obtained from the torrid zone mainly contained 2,4-O-disulfated and non-sulfated fucose residue, whereas Fuc-Ib from the temperate zone contained non-, 2-O- and 2,4-O-disulfated fucose residue; Fuc-St from the frigid zone and Fuc-Hv from the torrid zone contained mainly non-sulfated fucose residue. The proton of SC-Fucs was better resolved via high temperature 1H NMR than via room temperature 1H NMR. The fingerprint of sea cucumber in different sea regions was established based on the index of anomer hydrogen signal in SC-Fucs. | 14 | Nano-TiO_2 Decorated Radial-Like Mesoporous Silica: Preparation,Characterization, and Adsorption-Photodegradation Behavior显示文摘Supported nanocrystalline Ti_O2 with a diameter of 15–30 nm was prepared from previously synthesized radial mesoporous silica(RMS) by a post-synthesis method. In addition, their adsorption-photocatalytic activity toward the degradation of methylene blue(MB) was determined. RMS was tailor-made with the main template of CTAB and the Si_O2 precursor of TEOS through a facile self-assembly process. The structural, morphological and textural properties of the well-designed Ti_O2/RMS samples were characterized.The RMS structure was retained after loading Ti_O2, but its surface area and pore diameter decreased as a result of partial pore blocking. The removal activity of MB for Ti_O2/RMS was significantly higher than that of commercial Ti_O2 nanoparticles. The optimal Ti_O2 loading(20 wt%) on the support could achieve the complete removal of MB within 70 min. | 18 | SNX17 regulates Notch pathway and pancreas development through the retromer-dependent recycling of Jag1显示文摘Background:Notch is one of the most important signaling pathways involved in cell fate determination.Activation of the Notch pathway requires the binding of a membrane-bound ligand to the Notch receptor in the adjacent cell which induces proteolytic cleavages and the activation of the receptor.A unique feature of the Notch signaling is that processes such as modification,endocytosis or recycling of the ligand have been reported to play critical roles during Notch signaling,however,the underlying molecular mechanism appears context-dependent and often controversial.Results:Here we identified SNX17 as a novel regulator of the Notch pathway.SNX17 is a sorting nexin family protein implicated in vesicular trafficking and we find it is specifically required in the ligand-expressing cells for Notch signaling.Mechanistically,SNX17 regulates the protein level of Jag1a on plasma membrane by binding to Jag1a and facilitating the retromer-dependent recycling of the ligand.In zebrafish,inhibition of this SNX17-mediated Notch signaling pathway results in defects in neurogenesis as well as pancreas development.Conclusions:Our results reveal that SNX17,by acting as a cargo-specific adaptor,promotes the retromer dependent recycling of Jag1a and Notch signaling and this pathway is involved in cell fate determination during zebrafish neurogenesis and pancreas development. | Wenguang Yin Dapeng Liu Nian Liu Liangliang Xu Song Li Shuo Lin Xiaodong Shu Duanqing Pei | 2012 | Cell Regeneration2012,1,1: | 1 |
