5Writing and erasing MYC ubiquitination and SUMOylation显示文摘The transcription factor c-MYC(MYC thereafter)controls diverse transcription programs and plays a key role in the development of many human cancers.Cells develop multiple mechanisms to ensure that MYC levels and activity are precisely controlled in normal physiological context.As a short half-lived protein,MYC protein levels are tightly regulated by the ubiquitin proteasome system.Over a dozen of ubiquitin ligases have been found to ubiquitinate MYC whereas a number of deubiquitinating enzymes counteract this process.Recent studies show that SUMOylation and deSUMOylation can also regulate MYC protein stability and activity.Interestingly,evidence suggests an intriguing crosstalk between MYC ubiquitination and SUMOylation.Deregulation of the MYC ubiquitination-SUMOylation regulatory network may contribute to tumorigenesis.This review is intended to provide the current understanding of the complex regulation of the MYC biology by dynamic ubiquitination and SUMOylation and their crosstalk.Yingxiao Chen Xiao-Xin Sun Rosalie C.Sears Mu-Shui Dai 2019Genes & Diseases2019,6,4:3
6Target localization method of non-cooperative spacecraft on on-orbit service显示文摘With the explosion of the number of meteoroid/orbital debris in terrestrial space in recent years, the detection environment of spacecraft becomes more complex. This phenomenon causes most current detection methods based on machine learning intractable to break through the two difficulties of solving scale transformation problem of the targets in image and accelerating detection rate of high-resolution images. To overcome the two challenges, we propose a novel noncooperative target detection method using the framework of deep convolutional neural network.Firstly, a specific spacecraft simulation dataset using over one thousand images to train and test our detection model is built. The deep separable convolution structure is applied and combined with the residual network module to improve the network’s backbone. To count the different shapes of the spacecrafts in the dataset, a particular prior-box generation method based on K-means cluster algorithm is designed for each detection head with different scales. Finally, a comprehensive loss function is presented considering category confidence, box parameters, as well as box confidence. The experimental results verify that the proposed method has strong robustness against varying degrees of luminance change, and can suppress the interference caused by Gaussian noise and background complexity. The mean accuracy precision of our proposed method reaches 93.28%, and the global loss value is 13.252. The comparative experiment results show that under the same epoch and batchsize, the speed of our method is compressed by about 20% in comparison of YOLOv3, the detection accuracy is increased by about 12%, and the size of the model is reduced by nearly 50%.Yingxiao LI Ju HUO Ping MA Ruiye JIANG 2022Chinese Journal of Aeronautics2022,35,11:2
7Chitosan@Puerarin hydrogel for accelerated wound healing in diabetic subjects by miR-29ab1 mediated inflammatory axis suppression显示文摘Wound healing is one of the major global health concerns in patients with diabetes.Overactivation of pro-inflammatory M1 macrophages is associated with delayed wound healing in diabetes.miR-29ab1 plays a critical role in diabetes-related macrophage inflammation.Hence,inhibition of inflammation and regulation of miR-29 expression have been implicated as new points for skin wound healing.In this study,the traditional Chinese medicine,puerarin,was introduced to construct an injectable and self-healing chitosan@puerarin(C@P)hydrogel.The C@P hydrogel promoted diabetic wound healing and accelerated angiogenesis,which were related to the inhibition of the miR-29 mediated inflammation response.Compared to healthy subjects,miR-29a and miR-29b1 were ectopically increased in the skin wound of the diabetic model,accompanied by upregulated M1-polarization,and elevated levels of IL-1βand TNF-α.Further evaluations by miR-29ab1 knockout mice exhibited superior wound healing and attenuated inflammation.The present results suggested that miR-29ab1 is essential for diabetic wound healing by regulating the inflammatory response.Suppression of miR-29ab1 by the C@P hydrogel has the potential for improving medical approaches for wound repair.Xiaoling Zeng Baohui Chen Luping Wang Yingxiao Sun Zhao Jin Xuanyong Liu Liping Ouyang Yun Liao 2023Bioactive Materials2023,,1:2

11Altered expression of cytochrome P450 and possible correlation with preneoplastic changes in early stage of rat hepatocarcinogenesis显示文摘Aim: Correlation of cytochrome P450 (CYPs) with preneoplastic changes in the early stage of hepatocarcinogenesis is still unclear. To detect the expression of carcinogen-metabolizing related microsomal P450 enzymes, namely the CYP1A1,CYP1A2, CYP2B 1/2, CYP2E1, and CYP3A, we performed the medium-term bioassay of Ito's model in Sprague-Dawley rats. Methods: The amount and activity of CYP were assessed by biochemical and immunohistochemical methods in week 8.The correlation between CYP expression and microsomal oxidative stress was investigated by comparing the generation of microsomal lipid peroxidation in the presence or absence of specific CYP inhibitor. Results: In the DEN-2-AAF and 2-AAF alone groups, the expression of CYP1A1 and CYP2E1 were up-regulated and the expression of CYP2B 1/2 and CYP1 A2 were quite the contrary. Strong staining of CYP2E1 and CYP2BI/2 was found around the centrolobular vein and weak staining in the altered hepatic foci revealed by immunohistochemical procedure.There was no significant change in the activity of CYP3A among the 4 groups.Altered hepatic tissue bore more microsomal NADPH (nicotinamide adenine dimucleotide phosphate,reduced form)-dependent lipid peroxidation than normal tissue. And the difference among the 4 groups disappeared when CYP2E1 was inhibited. More microsomal lipid peroxidation was generated when incubated with CYP1A inhibitor α-naphthoflavone. 12Probability Response of Shape Memory Alloy Beam Subjected to Noise Excitation显示文摘Shape Memory Alloy(SMA)is a typical material with memory effect,and it is widely used in many engineering fields.Based on the elastic theory and Galerkin method,a vibration system of SMA beam with rigid constraints is proposed.The non⁃smooth transformation was employed to deal with the discontinuous position,and the original system was turned into an approximate equivalent system associated with the Dirac function.Then,using the stochastic averaging method,the drift and diffusion coefficients of the corresponding Fokker Planck Kolmogorov equation were described.Lastly,the approximate probability response of the system was formulated analytically.Meanwhile,numerical simulation was carried out to verify the effectiveness of analytical results.Furthermore,stochastic bifurcation was discussed.Results show that the stationary probability response of the system was affected by the increase of noise amplitude and restitution force,and a certain restitution value and damping could induce P⁃bifurcation.Yuting Li Jinqian Feng Yingxiao Wang 2020Journal of Harbin Institute of Technology(New Series)2020,27,1:1
13CRISPR ribonucleoprotein-mediated genetic engineering in plants显示文摘CRISPR-derived biotechnologies have revolutionized the genetic engineering field and have been widely applied in basic plant research and crop improvement.Commonly used Agrobacterium-or particle bombardment-mediated transformation approaches for the delivery of plasmid-encoded CRISPR reagents can result in the integration of exogenous recombinant DNA and potential off-target mutagenesis.Editing efficiency is also highly dependent on the design of the expression cassette and its genomic insertion site.Genetic engineering using CRISPR ribonucleoproteins(RNPs)has become an attractive approach with many advantages:DNA/transgene-free editing,minimal off-target effects,and reduced toxicity due to the rapid degradation of RNPs and the ability to titrate their dosage while maintaining high editing efficiency.Although RNP-mediated genetic engineering has been demonstrated in many plant species,its editing efficiency remains modest,and its application in many species is limited by difficulties in plant regeneration and selection.In this review,we summarize current developments and challenges in RNPmediated genetic engineering of plants and provide future research directions to broaden the use of this technology.Yingxiao Zhang Brian Iaffaldano Yiping Qi 2021Plant Communications2021,2,2:1

16Effects of Dietary Intervention on Hyperlipidemia in Eight Communities of Beijing, China显示文摘To examine the effect of community-based dietary intervention on hyperlipidemia. Methods A total of 180 hyperlipidemia individuals with TG>2.26 mmol稬-1(200 mg穌L-1)and/or TC>5.72 mmol稬-1 (220 mg穌L-1) were selected from 428 eligible subjects in eight communities of Beijing. They were randomly divided into intervention group (n=108) and control group (n=72). Dietary intervention was provided for the intervention group for 6 months. Information on dietary intakes, physical examinations and blood samples was collected. Serum lipids were assayed at baseline and endpoint of the study period. Results Respective decrease in dietary intake of total calories, fat, cholesterol and cooking oil by 13.62%, 24.75%, 24.40%, and 22.43%, in the intervention group was observed. The percentages of total calories from fat, carbohydrate and protein appeared to be desirable after study. Reduced body weight and BMI were also observed. There was a respective 5.61% and 7.06 % decrease in total serum cholesterol and low-density lipoprotein cholesterol in the intervention group, while no significant changes were found in the control group. 19Human-Centric Composite-Quality Modeling and Assessment for Virtual Desktop Clouds显示文摘There are several motivations, such as mobility, cost, and security, that are behind the trend of traditional desktop users transitioning to thin-client-based virtual desktop clouds (VDCs). Such a trend has led to the rising importance of human-centric performance modeling and assessment within user communities that are increasingly making use of desktop virtualization. In this paper, we present a novel reference architecture and its easily deployable implementation for modeling and assessing objective user quality of experience (QoE) in VDCs. This architecture eliminates the need for expensive, time-consuming subjective testing and incorporates finite-state machine representations for user workload generation. It also incorporates slow-motion benchmarking with deep-packet inspection of application task performance affected by QoS variations. In this way, a 'composite-quality' metric model of user QoE can be derived. We show how this metric can be customized to a particular user group profile with different application sets and can be used to a) identify dominant performance indicators and troubleshoot bottlenecks and b) obtain both absolute and relative objective user QoE measurements needed for pertinent selection of thin-client encoding configurations in VDCs. We validate our composite-quality modeling and assessment methodology by using subjective and objective user QoE measurements in a real-world VDC called VDPilot, which uses RDP and PCoIP thin-client protocols. 20ERp44 C160S/C212S mutants regulate IP_(3)R_(1) channel activity显示文摘Previous studies have indicated that ERp44 inhibits inositol 1,4,5-trisphosphate(IP3)-induced Ca2+release(IICR)via IP3R1,but the mechanism remains largely unexplored.Using extracellular ATP to induce intracellular calcium transient as an IICR model,Ca2+image,pull down assay,and Western blotting experiments were carried out in the present study.We found that extracellular ATP induced calcium transient via IP3Rs(IICR)and the IICR were markedly decreased in ERp44 overexpressed Hela cells.The inhibitory effect of C160S/C212S but not C29S/T396A/ΔT(331–377)mutants of ERp44 on IICR were significantly decreased compared with ERp44.However,the binding capacity of ERp44 to L3V domain of IP3R1(1L3V)was enhanced by ERp44 C160S/C212S mutation.Taken together,these results suggest that the mutants of ERp44,C160/C212,can more tightly bind to IP3R1 but exhibit a weak inhibition of IP3R1 channel activity in Hela cells.Congyan Pan Ji Zheng Yanyun Wu Yingxiao Chen Likun Wang Zhansong Zhou Wenxuan Yin Guangju Ji 2011Protein & Cell2011,2,12:0
