| 1 | Famitinib in combination with concurrent chemoradiotherapy in patients with locoregionally advanced nasopharyngeal carcinoma: a phase 1, open-label, dose-escalation Study显示文摘Background:Famitinib is a tyrosine kinase inhibitor against multiple targets,including vascular endothelial growth factor receptor 2/3,platelet-derived growth factor receptor,and stem cell factor receptor(c-kit).Previous studies have demonstrated anti-tumour activities of famitinib against a wide variety of advanced-stage solid cancers.We aimed to determine the safety and efficacy of famitinib with concurrent chemoradiotherapy(CCRT)in patients with locoregionally advanced nasopharyngeal carcinoma(NPC).We also evaluated the feasibility of contrast-enhanced ultrasound(D-CEUS)as a predictor of early tumour response to famitinib and to correlate functional parameters with clinical efficacy.Methods:The trial was conducted in subjects with stage III or IVa-b NPC using a 3+3 design of escalating fami-tinib doses.Briefly,subjects received 2 weeks of famitinib monotherapy followed by 7 weeks of famitinib plus CCRT.D-CEUS of the neck lymph nodes was performed at day 0,8 and 15 after famitinib was administered before starting concurrent chemoradiotherapy.End points included safety,tolerability and anti-tumour activity.Results:Twenty patients were enrolled(six each for 12.5,16.5 and 20 mg and two for 25 mg).Two patients in the 25 mg cohort developed dose-limiting toxicities,including grade 4 thrombocytopenia and grade 3 hypertension.The most common grade 3/4 adverse events were leukopenia,neutropenia and radiation mucositis.D-CEUS tests showed that more than 60%of patients achieved a perfusion parameter response after 2 weeks taking famitinib alone,and the parameter response was associated with disease improvement.In the famitinib monotherapy stage,three patients(15%)showed partial responses.The complete response rate was 65%at the completion of treatment and 95%3 months after the treatment ended.After a median follow-up of 44 months,the 3-year progression-free survival(PFS)and distant metastasis-free survival were 70%and 75%,respectively.Subjects with a decrease of perfusion parameter response,such as peak intensity decreased at least 30%after 1 week of famitinib treatment,had higher 3-year PFS(90.9%vs.44.4%,95%CI 73.7%-100%vs.11.9%-76.9%,P<0.001)than those with an increase or a reduction of less than 30%.Conclusions:The recommended famitinib dose for phase II trial is 20 mg with CCRT for patients with local advanced NPC.D-CEUS is a reliable and early measure of efficacy for famitinib therapies.Further investigation is required to confirm the effects of famitinib plus chemoradiotherapy. | Qiuyan Chen Linquan Tang Na Liu Feng Han Ling Guo Shanshan Guo Jianwei Wang Huai Liu Yanfang Ye Lu Zhang Liting Liu Pan Wang Yingqin Li Qingmei He Xiaoqun Yang Qingnan Tang Yang Li YuJing Liang XueSong Sun Chuanmiao Xie Yunxian Mo Ying Guo Rui Sun Haoyuan Mo Kajia Cao Xiang Guo Musheng Zeng Haiqiang Mai Jun Ma | 2018 | Cancer Communications2018,38,1: | 6 |
| 2 | Regulation of the cellular uptake of nanopartides by the orientation of helical polypeptides显示文摘Controlling the cellular interactio n and internal izatio n of polymer-modified nan oparticles (NPs) is of central importa nee to the developme nt of promisi ng nano medici nes. Here, we describe the use of synthetic polypeptides for NP surface coati ng and regulati on of their cellular uptake behaviors by simply switching the conformation and anchoring orientation. Our results show that gold NPs (AuNPs) coated with a helical poly(Y-(2-(2-(2-methoxyethoxy)ethoxy)ethoxy)esteryl「glutamate)(L-P(EG3Glu)5o) from the C-terminus ((L-C)-AuNPs) exhibit greater zeta potential and more cellular uptake (2.0-5.5 fold higher) than those coated with the same polypeptide but anchored from the N-terminus ((L-N)-AuNPs), or from both the C- and N-terminus at a 1/1 molar ratio ((L-C/L-N)-AuNPs). A similar orientation-regulated cellular internalization pattern is observed in D-P(EG3Glu)50 but not the unstructured DL-P(EG3Glu)5o-rnodified AuNPs, suggesting an important and universal role of the helix-derived macrodipole in cellular uptake. Moreover, this orientation-governed internalization is successfully reproduced in P(EG3Glu)50-coated gold nano rods (AuNRs), and applied to the desig n of doxorubici reloaded polypeptide micelles. Simulation study offers time-resolved in sights regarding the NP-membrane in teracti ons and membrane remodeling. Thus, our study provides a delicate way of regulating the surface chemistry of NPs and the subsequent NP-cell interactions. | 5 | A Comparison Study on the δD Value of Polycyclic Aromatic Hydrocarbons in Hydrous Pyrolysate of Herbaceous Peat with Different Water Medium显示文摘To investigate the influence of diagenetic water media on the hydrogen isotopes of individual sedimentary aromatic compounds,a series of hydrous pyrolyses were conducted on herbaceous peat.Polycyclic aromatic hydrocarbons(PAHs)in hydrous pyrolysed samples and their hydrogen isotopic composition characteristics were studied.The aqueous medium demonstrated a significant influence on the hydrogen isotopic composition of the individual PAHs generated during pyrolysis.The results showed that the PAHs formed after pyrolysis in the presence of a saltwater medium with high δD value from a salt lake had a heavy hydrogen isotopic composition.The PAHs formed after pyrolysis in the presence of a fresh water medium with low δD value from a swamp had a light hydrogen isotopic composition.The difference in the average PAHδD value between the two hydrous experiments varied from -174‰ to -109‰,suggesting that the hydrogen isotopic composition of individual sedimentary PAHs can reflect the source of the diagenetic water medium.In addition,a comparative study found that the hydrogen isotopes of PAHs were superior to those of n-alkanes in the same sample for diagenetic water indications.The results indicated that the exchange of water-derived inorganic hydrogen and organic hydrogen was more intensive in freshwater experiments than in saltwater experiments.With an increase in the simulation temperature,the average δD value of PAHs generated in the hydrous simulation experiments showed an increasing trend,reflecting that the δD value of sedimentary PAHs formed with the participation of diagenetic water media was still closely related to the thermal maturity of organic matter.Comparative studies showed that theδD values of different types of organic compounds produced by hydrous pyrolysis of peat were in the order,PAHs>n-alkanes>methane. | DUAN Yi WU Yingzhong XING Lantian WU Yingqin ZHAO Yang LI Zhongping MA Lanhua DUAN Mingchen | 2023 | Acta Geologica Sinica(English Edition)2023,97,6: | 0 |
