1Channel morphologic processes of a highly sinuous bend approaching neck cutoff by bank erosion in the middle Yangtze River显示文摘The distal reach of the Lower Jingjiang River(LJR)in the middle of the Yangtze River consists of five adjacent bends,among which the Qigongling Bend is a U-shaped meander with a mean sinuosity of 2.2 and the narrowest neck 525 m in width.This bend is slowly approaching neck cutoff owing to progressive bank erosion.An abnormal phenomenon has occurred in this bend since the Three Gorges Reservoir(TGR)began to operate in 2003 which is erosion in the inner bank zone and deposition in the outer bank zone.This problem has not been fully understood because of the interplay of changes in water-sediment,bank erosion,and artificial bank revetment.In this study,aerial and remote sensing images,hydrological data,channel topography,and an existing bank erosion model are used to reveal channel morphodynamics of this bend and the trend of the potential neck cutoff induced by bank erosion.The study results show that the clear water released from the TGR has provided by forcefully eroded the point bar of inner bank but failed to scour the outer bank due to the protection of bank revetment since the 1990 s.Thus far,the outer bank zone near the bend apex has increasingly widened in conjunction with the formation of 2 emerging sand bars.Consequently,the thalweg of the main channel has laterally shifted toward the inner bank by roughly 800 m.More severely,the rate of bank retreat on the upstream side of the bend neck was about 4.5 m/yr in 2010-2019,but the downstream side of this neck was experienced slight deposition.Bank erosion could be accelerated by progressively increasing erosion and eventually trigger the occurrence of neck cutoff in the next few decades,thereby significantly altering the quasi-equilibrium regime of channel morphodynamics in the LJR.Zhiwei Li Hanyuan Yang Junqiang Xia Meirong Zhou Shanshan Deng Yingzhen Wang 2021International Journal of Sediment Research2021,36,4:5
2Characteristics of the Late Quaternary right-lateral strike-slip movement of Bolokenu-Aqikekuduk fault in northern Tianshan Mountains,NW China显示文摘The characteristics of the Bolokenu-Aqikekuduk(Bo-A) fault,a right-lateral strike-slip fault that runs for more than 700 km long and obliquely cuts North Tianshan Mountains,are evaluated here based on remote sensing data,and through an analysis of the results from field investigations as well as climate-geomorphic events. The fault is composed of a western segment with a NW strike and an eastern segment with a NWW strike.The western segment is nearly 250 km long,extending northwestward into Kazakhstan with a right-lateral strike-slip rate of 5 mm/a.This domain consists of 4-5 rupture sections,with 3-4 deformation belts,caused by ancient or historical earthquakes,and suggesting the potential for the occurrence of further strong earthquakes(with M≈7.5) in future. The eastern segment of the fault shows a right-lateral strike-slip rate of 1-1.4 mm/a,with the development of 3-4 deformation belts caused by ancient or historical earthquakes,and with a potential for future strong earthquake with M≈7.0. 3Telomere-to-telomere and gap-free reference genome assembly of the kiwifruit Actinidia chinensis显示文摘Kiwifruit is an economically and nutritionally important fruit crop with extremely high contents of vitamin C.However,the previously released versions of kiwifruit genomes all have a mass of unanchored or missing regions.Here,we report a highly continuous and completely gap-free reference genome of Actinidia chinensis cv.‘Hongyang’,named Hongyang v4.0,which is the first to achieve two de novo haploid-resolved haplotypes,HY4P and HY4A.HY4P and HY4A have a total length of 606.1 and 599.6 Mb,respectively,with almost the entire telomeres and centromeres assembled in each haplotype.In comparison with Hongyang v3.0,the integrity and contiguity of Hongyang v4.0 is markedly improved by filling all unclosed gaps and correcting some misoriented regions,resulting in∼38.6–39.5 Mb extra sequences,which might affect 4263 and 4244 protein-coding genes in HY4P and HY4A,respectively.Furthermore,our gap-free genome assembly provides the first clue for inspecting the structure and function of centromeres.Globally,centromeric regions are characterized by higher-order repeats that mainly consist of a 153-bp conserved centromere-specific monomer(Ach-CEN153)with different copy numbers among chromosomes.Functional enrichment analysis of the genes located within centromeric regions demonstrates that chromosome centromeres may not only play physical roles for linking a pair of sister chromatids,but also have genetic features for participation in the regulation of cell division.The availability of the telomere-to-telomere and gap-free Hongyang v4.0 reference genome lays a solid foundation not only for illustrating genome structure and functional genomics studies but also for facilitating kiwifruit breeding and improvement.Junyang Yue Qinyao Chen Yingzhen Wang Lei Zhang Chen Ye Xu Wang Shuo Cao Yunzhi Lin Wei Huang He Xian Hongyan Qin Yanli Wang Sijia Zhang Ying Wu Songhu Wang Yi Yue Yongsheng Liu 2023Horticulture Research2023,10,2:4
4A key‘foxy’aroma gene is regulated by homologyinduced promoter indels in the iconic juice grape‘Concord’显示文摘‘Concord’,the most well-known juice grape with a parentage of the North American grape species Vitis labrusca L.,possesses a special‘foxy’aroma predominantly resulted from the accumulation of methyl anthranilate(MA)in berries.This aroma,however,is often perceived as an undesirable attribute by wine consumers and rarely noticeable in the common table and wine grape species V.vinifera.Here we discovered homology-induced promoter indels as a major genetic mechanism for species-specific regulation of a key‘foxy’aroma gene,anthraniloyl-CoA:methanol acyltransferase(AMAT),that is responsible for MA biosynthesis.We found the absence of a 426-bp and/or a 42-bp sequence in AMAT promoters highly associated with high levels of AMAT expression and MA accumulation in‘Concord’and other V.labrusca-derived grapes.These promoter variants,all with direct and inverted repeats,were further confirmed in more than 1,300 Vitis germplasm.Moreover,functional impact of these indels was validated in transgenic Arabidopsis.Superimposed on the promoter regulation,large structural changes including exonic insertion of a retrotransposon were present at the AMAT locus in some V.vinifera grapes.Elucidation of the AMAT genetic regulation advances our understanding of the‘foxy’aroma trait and makes it genetically trackable and amenable in grapevine breeding.Yingzhen Yang JoséCuenca Nian Wang Zhenchang Liang Honghe Sun Benjamin Gutierrez Xiaojun Xi Jie Arro YiWang Peige Fan Jason Londo Peter Cousins Shaohua Li Zhangjun Fei Gan-Yuan Zhong 2020Horticulture Research2020,7,1:3

6One-Pot Three-Dimensional Printing Robust Self-Supporting MnO_(x)/Cu-SSZ-13 Zeolite Monolithic Catalysts for NH_(3)-SCR显示文摘Honeycomb cordierite coated with Cu-SSZ-13 zeolite is widely used for the selective catalytic reduction of NO_(x) with NH_(3)(NH_(3)-SCR)to reduce pollutants from vehicle emissions.However,conventional honeycomb catalysts fabricated via coating techniques are limited by low zeolite loadings,loss of the deposited zeolites,and complicated preparation processes.Herein,a facile,one-step three-dimensional(3D)printing strategy is developed to construct MnO_(x)/Cu-SSZ-13 monolithic catalysts with excellent catalytic performance for NH_(3)-SCR.Iron-containing halloysite nanotubes(Fe-HNTs)are introduced as printing ink additives to ensure mechanical stability and modulate the NH_(3)-SCR performance of monolithic catalysts in high temperature conditions.In situ incorporation of Mn into the Cu-SSZ-13 zeolite monoliths during the 3D printing process boosts the mechanical strength of the monolithic structures from 2.54 MPa to 4.33 MPa as well as broadens the temperature window(165-550℃)of the catalysts for NH_(3)-SCR with NO_(x) conversion of above 80%.Such robust multicomponent-integrated 3D-printed selfsupporting catalysts not only possess high zeolite loading and excellent catalytic activity,but also avoid complicated manufacturing processes,which contrasts with conventional honeycomb catalysts fabricated by extrusion coupled with coating.Yingzhen Wei Mengyang Chen Xiaoyu Ren Qifei Wang Jinfeng Han Wenzheng Wu Xiangguang Yang Shuang Wang Jihong Yu 2022CCS Chemistry2022,4,5:1

9In vitro differentiation of rabbit bone marrow mesenchymal stem cells into Schwann cells Induction effect of platelet-rich plasma显示文摘BACKGROUND: Bone marrow mesenchymal stem cells (BMSCs) differentiate into Schwann cells via specific inducers. However, the induction and culture procedures are complicated. Various growth factors have been used for induction and culture, and are likely affected by their environment. OBJECTIVE: To explore the differentiation feasibility of platelet-rich plasma-induced rabbit BMSCs, supplemented with various growth factors, into Schwann-like cells in vitro, and to examine the secretory function of Schwann-like cells. DESIGN, TIME AND SETTING: This comparison study was performed at the Experimental Animal Center, Affiliated Hospital of Qingdao University Medical College, China in October 2008. MATERIALS: Platelet-rich plasma and BMSCs were respectively obtained from the femoral vein and bone marrow of 2-month old New Zealand rabbits. The rabbit nerve growth factor ELISA kit and rabbit nerve growth factor PCR kit (Jingmei Biotech, China), as well as anti-rabbit S-100 immunofluorescence staining kit (Boster, China) were used in the present study. METHODS: BMSCs at passage three were harvested and were induced by 1 mmol/L β-mercaptoethanol and Dulbecco's-modified eagle's medium complete medium containing 35 ng/mL retinoic acid and 10% fetal bovine serum for the combination induction group, followed by incubation in complete medium supplemented with platelet-rich plasma. BMSCs in the single induction group were induced by β-mercaptoethanol and retinoic acid, followed by incubation in L-DMEM complete medium. BMSCs in the control group were incubated in L-DMEM complete medium. MAIN OUTCOME MEASURES: Cell growth in each group was observed under an inverted microscope. Following induction, S-100 protein expression was identified by immunofluorescence staining. Nerve growth factor protein concentrations in BMSC supernatant were subsequently measured by ELISA. In addition, nerve growth factor mRNA expression in the BMSCs was determined by reverse transcription-polymerase chain reaction. RESULTS: Overall BMSC morphology was similar to Schwann cells in the combination induction group, and morphology of the majority of BMSCs was similar to Schwann cells in the single induction group 9 days after induction. In the control group, significant changes in cell morphology were not observed 9 days after induction. S-100 protein expression was detected in the combination induction and single induction groups following induction. At days 7, 9, and 11, the expression rate of S-100-positive cells was greater in the combination induction group (P < 0.05). At days 4, 7, 9, and 11, nerve growth factor levels were significantly greater in BMSC supernatant in the combination induction and single induction groups compared with the control group (P < 0.05). At days 7, 9, and 11, nerve growth factor levels were significantly greater in the combination induction group compared with the single induction group (P < 0.05). At day 11, nerve growth factor mRNA expression in BMSCs was significantly greater in the combination induction group compared with the single induction group (P < 0.05). 10Fine-tuning brassinosteroid biosynthesis via 3′UTR-dependent decay of CPD mRNA modulates wood formation in Populus显示文摘Brassinosteroids(BRs)are plant hormones that regulate wood formation in trees.Currently,little is known about the post-transcriptional regulation of BR synthesis.Here,we show that during wood formation,fine-tuning BR synthesis requires 3′UTR-dependent decay of Populus CONSTITUTIVE PHOTOMORPHOGENIC DWARF 1(PdCPD1).Overexpression of PdCPD1 or its 3′UTR fragment resulted in a significant increase of BR levels and inhibited secondary growth.In contrast,transgenic poplars repressing PdCPD13′UTR expression displayed moderate levels of BR and promoted wood formation.We show that the Populus GLYCINE-RICH RNA-BINDING PROTEIN1(PdGRP1)directly binds to a GU-rich element in 3′UTR of Pd CPD1,leading to its mRNA decay.We thus provide a post-transcriptional mechanism underlying BRs synthesis during wood formation,which may be useful for genetic manipulation of wood biomass in trees.Dian Wang Xiaoning Hao Li Xu Mengyan Zhao Congpeng Wang Xihao Yu Yingzhen Kong Mengzhu Lu Gongke Zhou Guohua Chai Xianfeng Tang 2023Journal of Integrative Plant Biology2023,65,8:0
11A Preliminary Study on Seismicity and Stages of Seismic Energy Accumulation in Seismotectonic Regions of Tianshan显示文摘Using seismic parameters, the characteristics of the seismic activity in various seismotectonic regions of Tianshan were studied in this paper. These regions are going through different stages of seismic energy accumulation. Current seismic risk levels of these areas were analyzed synthetically by the tectonic movement rates, as well as the characteristics of the seismic activity and the recurrence intervals of strong earthquakes. We preliminarily studied the characteristics of seismic activity in different seismic energy accumulating stages. The result shows that the characteristics of the seismic activity in various seismotectonic regions of the Tianshan area are influenced, not only by the regional tectonic movement, but also by the energy accumulating stage of various seismic tectonics. In the intense tectonic movement areas, it is important to estimate its stage of energy accumulating in order to predict the upper limit of the potential earthquake magnitude. In the less intense tectonic movement areas, the estimating of the stage of energy accumulation will help us recognize the dangerous level of the potential strong earthquake.The study shows that the seismotectonic regions in southern Tianshan have reached the mid-stage and late-stage of energy accumulation, with a higher seismic activity and thus a higher seismic dangerous level than those in the northern and middle Tianshan. 12quarTeT: a telomere-to-telomere toolkit for gap-free genome assembly and centromeric repeat identification显示文摘A high-quality genome is the basis for studies on functional,evolutionary,and comparative genomics.The majority of attention has been paid to the solution of complex chromosome structures and highly repetitive sequences,along with the emergence of a new‘telomere-to-telomere(T2T)assembly’era.However,the bioinformatic tools for the automatic construction and/or characterization of T2T genome are limited.Here,we developed a user-friendly web toolkit,quarTeT,which currently includes four modules:AssemblyMapper,GapFiller,TeloExplorer,and CentroMiner.First,AssemblyMapper is designed to assemble phased contigs into the chromosome-level genome by referring to a closely related genome.Then,GapFiller would endeavor to fill all unclosed gaps in a given genome with the aid of additional ultra-long sequences.Finally,TeloExplorer and CentroMiner are applied to identify candidate telomere and centromere as well as their localizations on each chromosome.These four modules can be used alone or in combination with each other for T2T genome assembly and characterization.As a case study,by adopting the entire modular functions of quarTeT,we have achieved the Actinidia chinensis genome assembly that is of a quality comparable to the reported genome Hongyang v4.0,which was assembled with the addition of manual handling.Further evaluation of CentroMiner by searching centromeres in Arabidopsis thaliana and Oryza sativa genomes showed that quarTeT is capable of identifying all the centromeric regions that have been previously detected by experimental methods.Collectively,quarTeT is an efficient toolkit for studies of large-scale T2T genomes and can be accessed at http://www.atcgn.com:8080/quarTeT/home.html without registration.Yunzhi Lin Chen Ye Xingzhu Li Qinyao Chen Ying Wu Feng Zhang Rui Pan Sijia Zhang Shuxia Chen Xu Wang Shuo Cao Yingzhen Wang Yi Yue Yongsheng Liu Junyang Yue 2023Horticulture Research2023,10,8:0
