| 1 | Effect of resveratrol on activation of nuclear factor kappa-B and inflammatory factors in rat model of acute pancreatitis显示文摘AIM: To observe the effect of resveratrol on nuclear factor Kappa-B (NF-κB) activation and the inflammatory response in sodium taurocholate-induced pancreatitis in rats.METHODS: Seventy-two male SD rats were randomly divided into three groups: sham operation group (control),severe acute pancreatitis (SAP) group, and severe acute pancreatitis group treated with resveratrol (RES). A SAP model was established by injecting 4% sodium taurocholate 1 mL/kg through puncturing the pancreatic duct. In Res group, Res was given at 30 mg/kg b.m. intraperitoneally after the SAP model was successfully established. Eight animals from each group were sacrificed at 3, 6 and 12 h after modeling. The expression of NF-κB activation of pancreas was detected by immunohistochemical staining, whereas the levels of TNF-α and IL-8 in pancreatic tissues were estimated by radioimmunoassay. The pathological changes of pancreas and lungs were examined microscopically.RESULTS: Much less hyperemia, edema, dust-colored necrotic focus and soaps were noticed in pancreas in RES group than in SAP group. In RES group, hemorrhage,exudates and infiltration of inflammatory cells in pancreas and interstitial edema, destruction of alveolar wall in lung were significantly less than in SAP group. In the SAP group,the activation of NF-κB in pancreatic tissues was enhanced significantly at any measure point compared with control group (64.23±10.72% vs2.56±0.65%, 55.86±11.34% vs 2.32±0.42%, 36.23±2.30% vs 2.40±0.36% ,P <0.01), TNF-α,IL-8 were also increased and reached their peak at 6 h and then declined. The activation of NF-κB and the levels of TNF-α and IL-8 in RES group were significantly lower than those in SAP group (P<0.01): activation (52.63±9.45% vs 64.23±10.72%, 40.52±8.40% vs 55.86±11.34%, 29.83±5.37% vs36.23±2.30%), TN-α (132.76±15.68 pg/mL vs 158.36±12.58 pg/mL, 220.32±23.57 pg/mL vs 247.67± 11.62 pg/mL, 175.68±18.43 pg/mL vs 197.35±12.57 pg/mL) and IL-8 (0.62±0.21 μg/L vs0.83±0.10 μg/L, 1.10±0.124 μg/L vs1.32±0.18 μg/L, 0.98±0.16 μg/L vs 1.27±0.23 μg/L).CONCLUSION: The activation of NF-κB is involved in the inflammatory response of rats with SAP. | 2 | Effect of resveratrol on microcirculation disorder and lung injury following severe acute pancreatitis in rats显示文摘AIM: To investigate the mechanism of resveratrol underlying the microcirculation disorder and lung injury following severe acute pancreatitis (SAP).METHODS: Twenty-four rats were divided into 3 groups (SAP, sham and resveratrol groups) randomly. SAP model was established by injecting 4% sodium taurocholate 1 mL/kg through puncturing pancreatic ducts. Sham (control) group (8 rats) was established by turning over the duodenum.Resveratrol was given at 0.1 mg/kg b.m. intraperitoneally.Rats were sacrificed 9 h after SAP was induced. Blood samples were obtained for hemorrheological examination.Lung tissues were used for pathological observation, and examination of microvascular permeability, dry/wet ratio and myeloperoxidase (MPO) activity. Gene expression of intercellular adhesion molecule-1 (ICAM-1) was detected by RT-PCR.RESULTS: Compared with SAP group, resveratrol relieved the edema and infiltration of leukocytes in the lungs. Resveratrol improved markers of hemorrheology:high VTB (5.77±1.18 mPas vs9.49±1.34 mPas), low VTB (16.12±3.20 mPas vs30.91±7.28 mPas), PV (4.69±1.68 mPas vs8.00±1.34 mPas), BSR (1.25±0.42 mm/h vs 0.03±0.03mm/h), VPC (54.67±3.08% vs 62.17±3.39%), fibrinogen (203.2±87.8 g/L vs 51.3±19.1 g/L), original hemolysis (0.45±0.02 vs 0.49±0.02), and complete hemolysis (0.41±0.02 vs0.43±0.02) (P<0.05). Resveratrol decreased the OD ratio of ICAM-1 gene (0.800±0.03 vs 1.188±0.10),dry/wet ratio (0.74±0.02 vs 0.77±0.03), microvascular permeability (0.079±0.006 vs 0.112±0.004) and MPO activity (4.42±0.32 vs 5.03±0.51) significantly (P<0.05).CONCLUSION: Resveratrol can improve the microcirculation disorder of the lung by decreasing leukocyte-endothelial interaction, reducing blood viscosity, improving the decrease of blood flow, and stabilizing erythrocytes in SAP rats. | 3 | Evaluation of measurement uncertainty based on grey system theory for small samples from an unknown distribution显示文摘To evaluate measurement uncertainty for small sample size and measurement data from an unknown distribution, we propose a grey evaluation method of measurement uncertainty based on the grey relation coefficient. The uncertainty of measurement is analyzed using grey system theory, and the defects of the grey evaluation model of measurement uncertainty (GEMU) are studied. We then establish an improved grey evaluation model of measurement uncertainty (IGEMU). Simulations show that the precision of IGEMU is greater than that of GEMU, and that sample size has only a small effect on the precision of IGEVU. In particular, IGEMU is applied to evaluating measurement uncertainty for small sample size and measurement data from an unknown distribution. | 4 | 0.04 degree-per-hour MEMS disk resonator gyroscope with high-quality factor (510 k) and long decaying time constant(74.9 s)显示文摘The disk resonator gyroscope is an attractive candidate for high-performance MEMS gyroscopes.This gyroscope consists of a sensor and readout electronics,and the characteristics of the sensor directly determine the performance.For the sensor,a high-quality factor and long decaying time constant are the most important characteristics required to achieve high performance.We report a disk resonator gyroscope with a measured quality factor of 510 k and decaying time constant of 74.9 s,which is a record for MEMS silicon disk resonator gyroscopes,to the best of our knowledge.To improve the quality factor of the DRG,the quality factor improvement mechanism is first analyzed,and based on this mechanism two stiffness-mass decoupled methods,i.e.,spoke length distribution optimization and lumped mass configuration design,are proposed and demonstrated.A disk resonator gyroscope prototype is fabricated based on these design strategies,and the sensor itself shows an angle random walk as low as 0.001°/√h,demonstrating true potential to achieve navigation-grade performance.The gyroscope with readout electronics shows an angle random walk of 0.01°/√h and a bias instability of 0.04°/h at room temperature without compensation,revealing that the performance of the gyroscope is severely limited by the readout electronics,which should be further improved.We expect that the quality factor improvement methods can be used in the design of other MEMS gyroscopes and that the newly designed DRG can be further improved to achieve navigation-grade performances for high-end industrial,transportation,aerospace,and automotive applications. | Qingsong Li Dingbang Xiao Xin Zhou Yi Xu Ming Zhuo Zhanqiang Hou Kaixuan He Yongmeng Zhang Xuezhong Wu | 2018 | Microsystems & Nanoengineering2018,4,1: | 16 |
| 5 | Modeling and Analysis of Mechanical Quality Factor of the Resonator for Cylinder Vibratory Gyroscope显示文摘Mechanical Quality factor(Q factor) of the resonator is an important parameter for the cylinder vibratory gyroscope(CVG). Traditional analytical methods mainly focus on a partial energy loss during the vibration process of the CVG resonator, thus are not accurate for the mechanical Q factor prediction. Therefore an integrated model including air damping loss, surface defect loss, support loss, thermoelastic damping loss and internal friction loss is proposed to obtain the mechanical Q factor of the CVG resonator. Based on structural dynamics and energy dissipation analysis, the contribution of each energy loss to the total mechanical Q factor is quantificationally analyzed. For the resonator with radius ranging from 10 mm to 20 mm, its mechanical Q factor is mainly related to the support loss, thermoelastic damping loss and internal friction loss, which are fundamentally determined by the geometric sizes and material properties of the resonator. In addition, resonators made of alloy 3J53(Ni42Cr Ti Al), with different sizes, were experimentally fabricated to test the mechanical Q factor. | 6 | 0.79 ppm scale-factor nonlinearity whole-angle microshell gyroscope realized by real-time calibration of capacitive displacement detection显示文摘Whole-angle gyroscopes have broad prospects for development with inherent advantages of excellent scale factor,wide bandwidth and measurement range,which are restrictions on rate gyroscopes.Previous studies on the whole-angle mode are based mostly on the linear model of Lynch,and the essential nonlinearity of capacitive displacement detection is always neglected,which has significant negative effects on the performance.In this paper,a novel realtime calibration method of capacitive displacement detection is proposed to eliminate these nonlinear effects.This novel method innovatively takes advantage of the relationship between the first and third harmonic components of detective signals for calibration.Based on this method,the real-time calibration of capacitive displacement detection is achieved and solves the problems of traditional methods,which are usually related to the vibration amplitude,environmental variations and other factors.Furthermore,this novel calibration method is embedded into a whole-angle control system to restore the linear capacitive response in real time and then combined with a microshell resonator for the first time to exploit the enormous potential of an ultrahigh Q factor and symmetric structure.The effectiveness is proven because the angle drift is reduced significantly to improve the scale-factor nonlinearity by 14 times to 0.79 ppm with 0.0673/h bias instability and a 0.0017s rate threshold,which is the best reported performance of the MEMS whole-angle gyroscope thus far.More importantly,this novel calibration method can be applied for all kinds of resonators with the requirement of a linear capacitive response even under a large resonant amplitude. | Jiangkun Sun Sheng Yu Yongmeng Zhang Qingsong Li Xiang Xi Kun Lu Xuezhong Wu Dingbang Xiao | 2021 | Microsystems & Nanoengineering2021,7,5: | 1 |

| 9 | Electrosynthesis of ^(15)N-labeled amino acids from ^(15)N-nitrite and ketonic acids显示文摘^(15)N isotope-labeled amino acids(^(15)N-amino acids)are crucial in the fields of biology,medicine,and chemistry.^(15)N-amino acids are conventionally synthesized through microbial fermentation and chemical reductive amination of ketonic acids methodologies,which usually require complicated procedures,high temperatures,or toxic cyanide usage,causing energy and environmental concerns.Here,we report a sustainable pathway to synthesize ^(15)N-amino acids from readily available ^(15)N-nitrite(^(15)NO_(2)-)and biomass-derived ketonic acids under ambient conditions driven by renewable electricity.A mechanistic study demonstrates a ^(15)N-nitrite→^(15)NH_(2)OH→^(15)N-pyruvate oxime→^(15)N-alanine reaction pathway for ^(15)N-alanine synthesis.Moreover,this electrochemical strategy can synthesize six ^(15)N-amino acids with 68%–95%yields.Furthermore,a ^(15)N-labeled drug of ^(15)N-tiopronin,the most commonly used hepatitis treatment drug,is fabricated using ^(15)N-glycine as the building block.Impressively,^(15)N sources can be recycled by the electrooxidation of ^(15)NH4^(+) to ^(15)NO_(2)-with a method economy.This work opens an avenue for the green synthesis of ^(15)N-labeled compounds or drugs. | Yongmeng Wu Mengyang Li Tieliang Li Jinghui Zhao Ziyang Song Bin Zhang | 2023 | Science China Chemistry2023,66,6: | 0 |

| 11 | Inhibitory effect of survivin-shRNA on A431 cutaneous squamous cell carcinoma显示文摘Objective:To observe the effect of survivin-shRNA on A431 cutaneous squamous cell carcinoma(SCC)and explore the molecular biological mechanism of nuclear factorκB(NF-κB)in the inhibition of survivin-shRNA on cutaneous SCC.Methods:Survivin-shRNA adenovirus vector was constructed to screen out the best interference sequence.Nude mice were subcutaneously inoculated with the cultured A431 cell suspension to duplicate A431 transplanted tumor models.These mice were randomly divided into the blank control group,the rAd-EGFP negative control group,the rAd-survivin-shRNA transfection group and the Res positive control group,with 5 mice in each group.The corresponding reagents were injected into the tumors.All the mice were sacrificed on the 20th day.The tumor tissues were isolated,with tumor volume and tumor weight measured,the tumor growth curves were accordingly plotted and the tumor inhibition rate was consequently calculated.Hematoxylin-eosin(HE)staining was used to observe the cellular morphology of the tumors.TUNEL was applied to the detection of cell apoptosis.Western blot was applied to the detection of the expression of Survivin,inhibitor of NF-κB(IκB),P65,P53 and Caspase-3.Results:As to the transplanted tumors,tumor volume and tumor weight were decreased in nude mice in the rAd-survivinshRNA transfection group and the Res positive control group,in comparison with the blank control group and the rAd-EGFP negative control group,the differences were of statistical significance(p<.05);The results of TUNEL showed that the apoptosis rates were significantly increased in the rAd-survivin-shRNA transfection group and the Res positive control group,in comparison with the blank control group and the rAd-EGFP negative control group,the difference was statistically significant(p<.05);In the rAd-survivin-shRNA transfection group and the Res positive control group,the microscopic observation showed a small amount of sparsely-distributed tumor cells,degenerative liquefactive necrosis,cancer cell shrinkage and round-shape,and karyopycnosis;The expressions of Survivin and P65 proteins were decreased in the rAd-survivin-shRNA transfection group and the Res positive control group,in comparison with the blank control group and the rAd-EGFP negative control group,the differences were statistically significant(p<.05),while the expressions of IκB,P53 and Caspase-3 proteins were significantly increased,in comparison with the blank control group and the rAd-EGFP negative control group,the differences were statistically significant(p<.05).Conclusions:Survivin-shRNA can inhibit the growth of the transplanted tumors in cutaneous SCC nude mice,one of the mechanisms is to promote the apoptosis of tumor cells and inhibit the growth of SCC transplanted tumors by inhibiting the NF-κB signaling pathway and then activating the tumor suppressor gene P53. | Yuqin Hao Xia Liu Shuxia Kang Xin Zhang Kun Zhang Yongmeng Zhu | 2018 | Discussion of Clinical Cases2018,5,4: | 0 |
| 12 | Influence of thymopentin-5 on renal pathology and relevant indexes of serum in rats with acute kidney injury caused by sepsis显示文摘Objective:To discuss thymopentin-5 on renal pathology and relevant indexes of serum with sepsis-caused acute kidney injury(AKI)caused by cecal ligation and puncture(CLP).Methods:90 cases of healthy and male SD rats were randomly divided into 3 groups:control group(C group),sepsis group(S group),thymopentin group(T group).These groups were divided into five time point including 1 h,6 h,12 h,24 h and 48 h with 6 rats in each time point.The sepsis model was made by CLP.The blood and kidney tissue were collected in each time point.Changes of renal pathology were observed under light microscope and relevant indexes like serum creatinin(Cr),blood urea nitrogen(Bun),tumor necrosis factorα(TNF-α),interleukin-10(IL-10),CD4+/CD8+were tested and analyzed.Results:In T group,concentrations of Cr,Bun in 6 h after CLP started to rise,reached peak in 24 h and decreased in 48 h,which were all lower than S group(p<.05)and higher than C group(p<.05).Compared with C group,concentrations of TNF-αin 1 h significantly improved,reached peak in 12 h and decreased in 24 h,which were all lower than S group(p<.05);and concentrations of IL-10 in 1 h significantly decreased,reached peak in 12 h and rised in 24 h,which were all more than S group(p<.05).In T group,CD4+/CD8+ratio in 6 h after CLP started to decrease,reached the lowest in 24 h and rised in 48 h,which were all lower than S group(p<.05)and significantly lower than C group(p<.05).Conclusions:Thymopentin-5 plays the role of renal protection in AKI caused by sepsis. | Yongmeng Zhu Lingfeng Wang Weidong Guo | 2015 | Discussion of Clinical Cases2015,2,3: | 0 |
