Microbial diversity accumulates in a downstream direction in the Three Gorges Reservoir显示文摘Organic and inorganic materials migrate downstream and have important roles in regulating environmental health in the river networks. However,it remains unclear whether and how a mixture of materials(i.e.,microbial species)from various upstream habitats contribute to microbial community coalescence upstream of a dam. Here we track the spatial variation in microbial abundance and diversity in the Three Gorges Reservoir based on quantitative PCR and 16 S rRNA gene high-throughput sequencing data.We further quantitatively assess the relative contributions of microbial species from mainstem, its tributaries, and the surrounding riverbank soils to the area immediately upstream of the Three Gorges Dam(TGD).We found an increase of microbial diversity and the convergent microbial distribution pattern in areas immediately upstream of TGD,suggesting this area become a new confluence for microbial diversity immigrating from upstream.Indeed, the number of shared species increased from upstream to TGD but unique species decreased, indicating immigration of various sources of microbial species overwhelms local environmental conditions in structuring microbial community close to TGD.By quantifying the sources of microbial species close to TGD,we found little contribution from soils as compared to tributaries, especially for sites closer to TGD,suggesting tributary microbes have greater influence on microbial diversity and environmental health in the Three Gorges Reservoir.Collectively, our results suggest that tracking microbial geographic origin and evaluating accumulating effects of microbial diversity shed light on the ecological processes in microbial communities and provide information for regulating aquatic ecological health. | Shang Wang Weiguo Hou Hongchen Jiang Liuqin Huang Hailiang Dong Shu Chen Bin Wang Yongcan Chen Binliang Lin Ye Deng | 2021 | Journal of Environmental Sciences2021,33,3: | 7
Upregulation of β-catenin signaling represents a single common pathway leading to the various phenotypes of spinal degeneration and pain显示文摘Spine degeneration is an aging-related disease, but its molecular mechanisms remain unknown, although elevatedβ-catenin signaling has been reported to be involved in intervertebral disc degeneration. Here,we determined the role ofβ-catenin signaling in spinal degeneration and in the homeostasis of the functional spinal unit(FSU),which includes the intervertebral disc, vertebra and facet joint and is the smallest physiological motion unit of the spine. We showed that pain sensitivity in patients with spinal degeneration is highly correlated withβ-catenin protein levels. We then generated a mouse model of spinal degeneration by transgenic expression of constitutively activeβ-catenin in Col2^(+) cells. We found thatβ-catenin-TCF7 activated the transcription of CCL2,a known critical factor in osteoarthritic pain.Using a lumbar spine instability model,we showed that aβ-catenin inhibitor relieved low back pain. Our study indicates thatβ-catenin plays a critical role in maintaining spine tissue homeostasis, its abnormal upregulation leads to severe spinal degeneration, and its targeting could be an avenue to treat this condition. | Ke Lu Qingyun Wang Hua Jiang Jun Li Zhou Yao Yongcan Huang Jianquan Chen Yejia Zhang Guozhi Xiao Xueyu Hu Zhuojing Luo Liu Yang Liping Tong Di Chen | 2023 | Bone Research2023,11,2: | 0
Enhancement of critical-sized bone defect regeneration by magnesium oxide-reinforced 3D scaffold with improved osteogenic and angiogenic properties显示文摘The healing of critical-sized bone defects(CSD)remains a challenge in orthopedic medicine. In recent years, scaffolds with sophisticated microstructures fabricated by the emerging three-dimensional(3D)printing technology have lighted up the treatment of the CSD due to the elaborate microenvironments and support they may build. Here,we established a magnesium oxide-reinforced 3D-printed biocompos-ite scaffold to investigate the effect of magnesium-enriched 3D microenvironment on CSD repairing. The composite was prepared using a biodegradable polymer matrix, polycaprolactone(PCL),and the disper-sion phase, magnesium oxide(MgO).With the appropriate surface treatment by saline coupling agent, the MgO dispersed homogeneously in the polymer matrix, leading to enhanced mechanical performance and steady release of magnesium ion(Mg^(2+))for superior cytocompatibility, higher cell viability, advanced osteogenic differentiation, and cell mineralization capabilities in comparison with the pure PCL.The in-vivo femoral implantation and critical-sized cranial bone defect studies demonstrated the importance of the 3D magnesium microenvironment,as a scaffold that released appropriate Mg^(2+) exhibited remarkably increased bone volume, enhanced angiogenesis, and almost recovered CSD after 8-week implantation.Overall, this study suggests that the magnesium-enriched 3D scaffold is a potential candidate for the treatment of CSD in a cell-free therapeutic approach. | Bo Chen Zhengjie Lin Qimanguli Saiding Yongcan Huang Yi Sun Xinyun Zhai Ziyu Ning Hai Liang Wei Qiao Binsheng Yu Kelvin W.K.Yeung Jie Shen | 2023 | Journal of Materials Science & Technology2023,,4: | 0
Function-oriented design:A novel strategy for advanced biomedical materials显示文摘It has always been a dream to construct tissues and even organs for transplantation to replace those with defects caused by diseases or injuries.Tissue engineering is another milestone in the developmental history of life science after cellular and molecular bioscience.Nevertheless, despite decades of rapid de-velopment,tissue-engineered biomaterials have not been widely used clinically.Biomaterials constructed by physical and chemical methods have lots of difficulty in precisely mimicking the macroscopic and mi-croscopic structures of human tissues. The ultimate way to build organoid tissue for regeneration is to enable the cells to take the initiative and build suitable functions.Based on the thoughts of tissue engi-neering,organoid technology holds great potential as a research tool for a wide range of fields, including developmental biology, disease pathology, cell biology, precision medicine, and drug toxicity and efficacy testing. This technology also holds tremendous potential for regenerative medicine,as organoids present the possibility for autologous and allogeneic cell therapy through the replacement of damaged or dis-eased tissues with organoid-propagated tissue or stem cell populations. In this review work,we briefly outlook the development history of organoid technology, summarize the current bottlenecks and the un-derlying reasons, and propose the unified term“function-oriented design in tissue engineering”,a new topic that may provide a solution to overcome these bottlenecks. | Zhiyu Zhou Wentao Wang Jianmin Wang Hongshui Wang Yi Xia Wei Zhang Yuxiao Lai Xiao Lin Yongcan Huang Xuenong Zou Martin J.Stoddart Zhen Li Wei Tian Shaoyu Liu Xinbao Wu Manman Gao Junhong Li Lei Yang Dafu Chen | 2023 | Journal of Materials Science & Technology2023,,14: | 0
Study on Image Recognition Algorithm for Residual Snow and Ice on Photovoltaic Modules显示文摘The accumulation of snow and ice on PV modules can have a detrimental impact on power generation, leading to reduced efficiency for prolonged periods. Thus,it becomes imperative to develop an intelligent system capable of accurately assessing the extent of snow and ice coverage on PV modules. To address this issue, the article proposes an innovative ice and snow recognition algorithm that effectively segments the ice and snow areas within the collected images.Furthermore, the algorithm incorporates an analysis of the morphological characteristics of ice and snow coverage on PV modules, allowing for the establishment of a residual ice and snow recognition process. This process utilizes both the external ellipse method and the pixel statistical method to refine the identification process. The effectiveness of the proposed algorithm is validated through extensive testing with isolated and continuous snow area pictures. The results demonstrate the algorithm’s accuracy and reliability in identifying and quantifying residual snow and ice on PV modules. In conclusion, this research presents a valuable method for accurately detecting and quantifying snow and ice coverage on PV modules. This breakthrough is of utmost significance for PV power plants,as it enables predictions of power generation efficiency and facilitates efficient PV maintenance during the challenging winter conditions characterized by snow and ice.By proactively managing snow and ice coverage,PV power plants can optimize energy production and minimize downtime, ensuring a sustainable and reliable renewable energy supply. | Yongcan Zhu JiawenWang Ye Zhang Long Zhao Botao Jiang Xinbo Huang | 2024 | Energy Engineering2024,121,4: | 0
