1Analytical validation of GMEX rapid point- of care CYP2C19 genotyping system for the CHANCE-2 trial显示文摘Background and purpose Rapid genotyping is useful for guiding early antiplatelet therapy in patients with high-risk nondisabling ischaemic cerebrovascular events(HR-NICE).Conventional genetic testing methods used in CYP2C19 genotype-guided antiplatelet therapy for patients with HR-NICE did not satisfy the needs of the Clopidogrel in High-Risk Patients with Acute Nondisabling Cerebrovascular Events(CHANCE)-2 trial.Therefore,we developed the rapid-genotyping GMEX(point-of care)system to meet the needs of the CHANCE-2 trial.Methods Healthy individuals and patients with history of cardiovascular diseases(n=408)were enrolled from six centres of the CHANCE-2 trial.We compared the laboratory-based genomic test results with Sanger sequencing test results for accuracy verification.Next,we demonstrated the accuracy,timeliness and clinical operability of the GMEX system compared with laboratory-based technology(YZY Kit)to verify whether the GMEX system satisfies the needs of the CHANCE-2 trial.Results Genotypes reported by the GMEX system showed 100%agreement with those determined by using the YZY Kit and Sanger sequencing for all three CYP2C19 alleles(*2,*3 and*17)tested.The average result’s turnaround times for the GMEX and YZY Kit methods were 85.0(IQR:85.0-86.0)and 1630.0(IQR:354.0-7594.0)min(p<0.001),respectively.Conclusions Our data suggest that the GMEX system is a reliable and feasible point-of care system for rapid CYP2C19 genotyping for the CHANCE-2 trial or related clinical and research applications.Xia Meng Anxin Wang Guojun Zhang Siying Niu Wei Li Sifei Han Fang Fang Xingquan Zhao Kehui Dong Zening Jin Huaguang Zheng Kelin Chen Hao Li Chengyuan Yang Yongjun Wang 2021Stroke & Vascular Neurology2021,6,2:7
2Preparation and Characteristics of a pH-sensitive Glucose-based Hydrogel显示文摘With glucose as the template compound,a p H-sensitive hydrogel was prepared by polymerization of the modified glucose,acrylamide,and acrylic acid.The porous hydrogel showed the highest swelling ratio of 42.7 g/g at p H=7.4 and the best adsorption of methylene blue at p H=7.The Langmuir isotherm fitted very well to the equilibrium adsorption data with the maximum adsorption capacity of 49.1 mg/g.The adsorption kinetics were well described by the pseudo 2^(nd) order model.Adsorption studies suggested that the p H-sensitive glucose-based hydrogel could be used as an adsorbent for the removal of methylene blue from wastewater.Other applications of the hydrogel are on the way,such as scaffolding in the biomedical field and soil conditioning in agriculture.Yan Lin GuiGan Fang YongJun Deng KuiZhong Shen Ting Wu Man Li 2018Paper And Biomaterials2018,3,3:5

The era of genomics and big data has brought the need for collaboration and data sharing in order to make effective use of this new knowledge. Here, we describe the web resources for cancer genomics research and rate them on the basis of the diversity of cancer types, sample size, omics data comprehensiveness, and user experience. 5Brood sex ratio in the Yellow-bellied Prinia(Prinia flaviventris)显示文摘Background:The adjustment of sex ratios in birds can occur at the egg and nestling stages. Previous studies showed that the sex ratio was affected by environmental factors and parental condition; it may result in seasonal and ecosystem differences.Methods:In this study, the brood sex ratio of the Yellow-bellied Prinia(Prinia flaviventris) in the Nonggang area, Guangxi, southwestern China, was investigated during the breeding season from May to June in 2013 using PCR amplification from whole-genome DNA extracted from blood samples. A total of 31 nests of Yellow-bellied Prinia, including 132 brood fledglings and 31 pairs, were sampled.Results:The results showed that the brood sex ratio of the Yellow-bellied Prinia was 1:1, and sex ratios of different nests were evenly distributed within the study area. No significant relationship was found between parental quality and nest characteristics with the brood sex ratio.Conclusions:The present study indicated that no brood sex ratio bias in the Yellow-bellied Prinia highlighted the complexity of sex ratio adjustment in birds. 7Computational prediction and experimental verification of miRNAs in Panicum miliaceum L.显示文摘MicroRNAs (miRNAs) are a class of non-coding RNAs that play critical roles in post-transcriptional regulation. Their target genes are involved in a variety of biological processes such as development, metabolism, and stress response. Panicum miliaceum L. (Panicum) is an important grain crop, but, until now, no miRNAs have been identified in this plant. Using a homology search based on expressed sequence tag (EST) analysis and miRNA precursor secondary structure, a total of 43 new miRNAs were identified. The miRNAs were found to be unevenly distributed among 11 miRNA families. Target analysis using the plant small RNA target analysis server psRNATarget showed that the newly identified miRNAs can potentially regulate 68 target genes. Ten of the 11 miRNA families were annotated as involved in RNA regulation, suggesting they may play an essential role in post-transcriptional regulation in Panicum. 9Evolution of the magmatic–hydrothermal system and formation of the giant Zhuxi W–Cu deposit in South China显示文摘The Zhuxi deposit is a recently discovered W–Cu deposit located in the Jiangnan porphyry–skarn W belt in South China. The deposit has a resource of 3.44 million tonnes of WO3, making it the largest on Earth,however its origin and the evolution of its magmatic–hydrothermal system remain unclear, largely because alteration–mineralization types in this giant deposit have been less well-studied, apart from a study of the calcic skarn orebodies. The different types of mineralization can be classified into magnesian skarn, calcic skarn, and scheelite–quartz–muscovite(SQM) vein types. Field investigations and mineralogical analyses show that the magnesian skarn hosted by dolomitic limestone is characterized by garnet of the grossular–pyralspite(pyrope, almandine, and spessartine) series, diopside, serpentine,and Mg-rich chlorite. The calcic skarn hosted by limestone is characterized by garnet of the grossular–andradite series, hedenbergite, wollastonite, epidote, and Fe-rich chlorite. The SQM veins host highgrade W–Cu mineralization and have overprinted the magnesian and calcic skarn orebodies. Scheelite is intergrown with hydrous silicates in the retrograde skarn, or occurs with quartz, chalcopyrite, sulfide minerals, fluorite, and muscovite in the SQM veins.Fluid inclusion investigations of the gangue and ore minerals revealed the evolution of the ore-forming fluids, which involved:(1) melt and coexisting high–moderate-salinity, high-temperature, high-pressure(>450 ℃and >1.68 kbar), methane-bearing aqueous fluids that were trapped in prograde skarn minerals;(2) moderate–low-salinity, moderate-temperature, moderate-pressure(~210–300 ℃and ~0.64 kbar),methane-rich aqueous fluids that formed the retrograde skarn-type W orebodies;(3) low-salinity,moderate–low-temperature, moderate-pressure(~150–240 ℃and ~0.56 kbar), methane-rich aqueous fluids that formed the quartz–sulfide Cu(–W) orebodies in skarn;(4) moderate–low-salinity,moderate-temperature, low-pressure(~150–250 ℃and ~0.34 kbar) alkanes-dominated aqueous fluids in the SQM vein stage, which led to the formation of high-grade W–Cu orebodies. The S–Pb isotopic compositions of the sulfides suggest that the ore-forming materials were mainly derived from magma generated by crustal anatexis, with minor addition of a mantle component. The H–O isotopic compositions of quartz and scheelite indicate that the ore-forming fluids originated mainly from magmatic water with later addition of meteoric water. The C–O isotopic compositions of calcite indicate that the ore-forming fluid was originally derived from granitic magma, and then mixed with reduced fluid exsolved from local carbonate strata. Depressurization and resultant fluid boiling were key to precipitation of W in the retrograde skarn stage. Mixing of residual fluid with meteoric water led to a decrease in fluid salinity and Cu(–W) mineralization in the quartz–sulfide stage in skarn. 11Mid-temperature thermoelectric performance of zone-melted Sb_(2)(Te,Se)_(3) alloys near phase transition boundary显示文摘(Bi,Sb)_(2)(Te,Se)_(3) alloys are widely used commercial thermoelectric(TE)materials for solid-state refrigeration around room temperature.The composition-induced structural phase transition could be realized by varying the compositions in these alloys,which may largely alter the electronic structure and phonon dispersion.Among them,the Se-alloyed Sb_(2)Te_(3) accompanied with structural transition is seldom reported.Herein,the interrelations of Se-alloying induced changes in structural phase transition,band structure and TE properties of p-type zone-melted Sb_(2)Te_(3-x)Se_(x)(x=1.5-2.4)alloys near phase transition boundary are systematically investigated.The results demonstrate that Sb_(2)Te_(3-x)Se_(x) shows a structural transition from a rhombohedral phase to mixed structure at x=2.0.The carrier concentration and bandgap at room temperature of Sb_(2)Te_(3)-xSex(x=1.5-2.4)constantly decrease with increasing Se contents x.The zT peak of the Sb_(2)TeSe_(2) matrix is improved and shifted to higher temperature by optimizing carrier concentration via Ag doping.A maximum zT of~0.4 is obtained at 680 K in Sb_(1.97)Ag_(0.03)TeSe_(2) alloy,about 100% enhancement compared with the undoped sample.Beibei Li Renshuang Zhai Teng Fang Kaiyang Xia Yongjun Wu Tiejun Zhu 2019Journal of Materiomics2019,5,4:1

13Effect of Co-Culture of Chinese Shrimp(Fenneropenaeus chinensis) and Sea Cucumber(Apostichopus japonicus Selenka) on Pond Environment显示文摘Monoculture of sea cucumber(pond S) and polyculture of shrimp with sea cucumber(pond SS) were established to evaluate the effect of shrimp on the environmental conditions of sea cucumber farming pond. Contributions of sediment organic matter(SOM2) resuspended from benthic sediment and the suspended particulate organic matter(SPOM) deposited from the water column to the precipitated organic matter(SOM1) collected with sediment traps were estimated with carbon stable isotope analysis. The results showed that the levels of SPOM and SOM2 in pond SS significantly decreased in comparison with those in pond S at the end of experiment(P < 0.05), indicating that co-culturing shrimp in sea cucumber farming pond could purify the farming water. Carbon stable isotope analysis showed that the proportion of SOM2 in SOM1 in pond SS(84.97% ± 0.38%) was significantly lower than that in pond S(95.20% ± 0.30%)(P < 0.05), suggesting that the resuspension of organic matter from benthic sediment into overlying water was reduced in polyculture pond. 17Genomic scanning enabling discovery of a new antibacterial bicyclic carbamate-containing alkaloid显示文摘Non-ribosomal peptides are a group of structurally diverse natural products with various important therapeutic and agrochemical applications.Bacterial pyrrolizidine alkaloids(PAs),containing a scaffold of two fused five-membered ring system with a nitrogen atom at the bridgehead,have been found to originate from a multidomain non-ribosomal peptide synthetase to generate indolizidine intermediates,followed by multistep oxidation,catalysed by single Bayer-Villiger(BV)enzymes,to yield PA scaffolds.Although bacterial PAs are rare in natural product inventory,bioinformatics analysis suggested that the biosynthetic gene clusters(BGCs)that are likely to be responsible for the production of PA-like metabolites are widely distributed in bacterial genomes.However,most of the strains containing PA-like BGCs are not deposited in the public domain,therefore preventing further assessment of the chemical spaces of this group of bioactive metabolites.Here,we report a genomic scanning strategy to assess the potential of PA metabolites production in our culture collection without prior knowledge of genome information.Among the strains tested,we found fifteen contain the key BV enzymes that are likely to be involved in the last step of PA ring formation.Subsequently one-strain-many-compound(OSMAC)method,supported by a combination of HR-MS,NMR,SMART 2.0 technology,and GNPS analysis,allowed identification and characterization of a new[5+7]heterobicyclic carbamate,legoncarbamate,together with five known PAs,bohemamine derivatives,from Streptomyces sp.CT37,a Ghanaian soil isolate.The absolute stereochemistry of legoncarbamate was determined by comparison of measured and calculated ECD spectra.Legoncarbamate displays antibacterial activity against E.coli ATCC 25922 with an MIC value of 3.1μg/mL.Finally,a biosynthetic model of legoncarbamate and other bohemamines was proposed based on the knowledge we have gained so far.Qing Fang Linrui Wu Caroline Urwald Morgane Mugat Shan Wang Kwaku Kyeremeh Carol Philips Samantha Law d Yongjun Zhou Hai Deng 2021Synthetic and Systems Biotechnology2021,6,1:1
18Gene panel for Mendelian strokes显示文摘background Mendelian stroke causes nearly 7% of ischaemic strokes and is also an important aetiology of cryptogenic stroke.Identifying the genetic abnormalities in Mendelian strokes is important as it would facilitate therapeutic management and genetic counselling.Next-generation sequencing makes large-scale sequencing and genetic testing possible.Methods A systematic literature search was conducted to identify causal genes of Mendelian strokes,which were used to construct a hybridization-based gene capture panel.Genetic variants for target genes were detected using Illumina HiSeq X10 and the Novaseq platform.The sensitivity and specificity were evaluated by comparing the results with Sanger sequencing.results 53 suspected patients of Mendelian strokes were analysed using the panel of 181 causal genes.According to the American College of Medical Genetics and Genomics standard,16 likely pathogenic/variants of uncertain significance genetic variants were identified.Diagnostic testing was conducted by comparing the consistency between the results of panel and Sanger sequencing.Both the sensitivity and specificity were 100%for the panel.Conclusion This panel provides an economical,time-saving and labour-saving method to detect causal mutations of Mendelian strokes.Fang Fang Zhe Xu Yue Suo Hui Wang Si Cheng Hao Li Wei Li Yongjun Wang 2020Stroke & Vascular Neurology2020,5,4:1

20Study on Pinus elliottii Plantations in Pengchang Forest Farm of Jingmen City显示文摘[Objectives]This study was conducted to improve the ecological and economic benefits of Pinus elliottii plantations in Pengchang Forest Farm, Jingmen City. [Methods] P. elliottii plantations in Pengchang Forest Farm in Jingmen City were selected as the research object, and the appearance and quantitative characteristics of P. elliottii community were studied and analyzed by a sampling method. [Results] Phanerophytes were the most in the P. elliottii community, accounting for 50%. In the community, except P. elliottii, the importance values of native plants were higher. The order of species diversity of P. elliottii community in Pengchang Forest Farm was herb layer, tree layer and shrub layer. The stress resistance of the community was weak, and strong intraspecific competition existed in P. elliottii. It is recommended to thin P. elliottii trees with insufficient lighting, poor growth potential and severely damaged tree body, and interplant Phoebe zhennan after thinning the P. elliottii plantations, so as to create a coniferous and broad-leaved evergreen mixed forest.
