1Antidiabetic Effects of Gegen Qinlian Decoction via the Gut Microbiota Are Attributable to Its Key Ingredient Berberine显示文摘Gegen Qinlian Decoction(GQD),a traditional Chinese medicine(TCM)formula,has long been used for the treatment of common metabolic diseases,including type 2 diabetes mellitus.However,the main limitation of its wider application is ingredient complexity of this formula.Thus,it is critically important to identify the major active ingredients of GQD and to illustrate mechanisms underlying its action.Here,we compared the effects of GQD and berberine,a hypothetical key active pharmaceutical ingredient of GQD,on a diabetic rat model by comprehensive analyses of gut microbiota,short-chain fatty acids,proinflammatory cytokines,and ileum transcriptomics.Our results show that berberine and GQD had similar effects on lowering blood glucose levels,modulating gut microbiota,inducing ileal gene expression,as well as relieving systemic and local inflammation.As expected,both berberine and GQD treatment significantly altered the overall gut microbiota structure and enriched many butyrate-producing bacteria,including Faecalibacterium and Roseburia,thereby attenuating intestinal inflammation and lowering glucose.Levels of short-chain fatty acids in rat feces were also significantly elevated after treatment with berberine or GQD.Moreover,concentration of serum proinflammatory cytokines and expression of immune-related genes,including Nfkb1,Stat1,and Ifnrg1,in pancreatic islets were significantly reduced after treatment.Our study demonstrates that the main effects of GQD can be attributed to berberine via modulating gut microbiota.The strategy employed would facilitate further standardization and widespread application of TCM in many diseases.Xizhan Xu Zezheng Gao Fuquan Yang Yingying Yang Liang Chen Lin Han Na Zhao Jiayue Xu Xinmiao Wang Yue Ma Lian Shu Xiaoxi Hu Na Lyu Yuanlong Pan Baoli Zhu Linhua Zhao Xiaolin Tong Jun Wang 2020Genomics, Proteomics & Bioinformatics2020,18,6:29
2The temporal dynamics of antimicrobial-resistant Salmonella enterica and predominant serovars in China显示文摘Salmonella enterica is one of the most common bacterial pathogens in humans and animals.Systematic studies on the trends and geographical distribution of antimicrobial-resistant Salmonella and dominant serovars have been well studied in European and American countries while not in China.Here,taking the One-Health strategy,we used>35000 Salmonella enterica isolates to explore the temporal and spatial dynamics of dominant serovars in China.We found that Salmonella Typhimurium was the dominant serovar causing human infection in China,which was consistent with Australia but inconsistent with North American and European countries.The proportion of Salmonella serovars Typhimurium,London,Rissen,Corvallis,Meleagridis,Kentucky,and Goldcoast showed an increasing trend during 2006-2019.We randomly selected 1962 isolates for comparative genomics and antimicrobial resistance studies and found that the number of antibiotic resistance genes(ARGs)per isolate increased 1.84 and 2.69 times of human and non-human origins,respectively,spanning 14 years.The proportion of antimicrobial-resistant Salmonella isolates had an increasing trend during 2006-2019,especially beta-lactam,quinolone,tetracycline,and rifampicin resistance.Moreover,we found that higher diversity of sequence types(STs)in S.Typhimurium than in other serovars,ST34 from pig and ST 19 from chicken origin,were mainly associated with isolates causing child and adult gastro-infection,respectively.Our results fill in the data gap on the trends of dominant serovars and antimicrobial resistance of Salmonella enterica in China.These data provide useful information for public health decision-makers prioritizing interventions for foodborne diseases and food safety.Yanan Wang Yue Liu Na Lyu Zhiyuan Li Sufang Ma Demin Cao Yuanlong Pan Yongfei Hu Hua Huang George F.Gao Xuebin Xu Baoli Zhu 2023National Science Review2023,10,3:4

4Scission of C–O and C–C linkages in lignin over RuRe alloy catalyst显示文摘The performance of lignin depolymerization is basically determined by the interunit C–O and C–C bonds.Numerous C–O bond cleavage strategies have been developed, while the cleavage of C–C bond between the primary aromatic units remains a challenging task due to the high dissociation energy of C–C bond.Herein, a multifunctional Ru Re alloy catalyst was designed, which exhibited exceptional catalytic activity for the cleavage of both C–O and C–C linkages in a broad range of lignin model compounds(β-1, a-5, 5–5,β-O-4, 4-O-5) and two stubborn lignins(kraft lignin and alkaline lignin), affording 97.5% overall yield of monocyclic compounds from model compounds and up to 129% of the maximum theoretical yield of monocyclic products based on C–O bonds cleavage from realistic lignin. Scanning transmission electron microscopy(STEM) characterization showed that Ru Re(1:1) alloy particles with hexagonal close-packed structure were homogeneously dispersed on the support. 6雷替曲塞经动脉灌注化疗的疗效分析(英文)显示文摘Objective: The aim of this study was to analysis raltitrexed by the safety and efficacy of intra-arterial infusion chemotherapy. Methods: Forty-seven cases in different parts of cancer patients, interventional methods, based on digital subtraction angiography(DSA) prompt, raltitrexed-based chemotherapy, the tumor nourish artery infusion chemotherapy.Patients with postoperative chemotherapy side effects and complications, and efficacy assessments. Results: Forty-seven patients, 2 patients had II-III degree of bone marrow suppression, the rate was 4.26%, 34 cases of I to II degree gastrointestinal reactions, 1 case of grade III gastrointestinal reactions, the incidence of 2.13%, and 1 case mild diarrhea. I degrees or more chemotherapy-related adverse reactions did not occur in this group of patients. All patients received at least 2 times arterial infusion chemotherapy, complete remission(CR) 3 cases, partial remission(PR) 11 cases, stable disease(SD) 17cases, disease progression(PD) 6 cases, total effective rate of treatment 87.23%. Conclusion: Raltitrexed for intra-arterial chemotherapy is safe and effective. 8Detection of Plasmid-Mediated Tigecycline Resistance Gene tet(X4) in a Salmonella enterica Serovar Llandoff Isolate显示文摘The emergence and spread of plasmid-mediated tigecycline resistance genes have attracted extensive attention worldwide.We investigated the distribution of mobile tigecycline resistance genes in Salmonella genomes generated by both our laboratory and public bacterial genomes downloaded from the NCBI GenBank.The tet(X4)-positive strains were subjected to susceptibility testing and conjugation assays.The genetic features of the tet(X4)-bearing plasmid sequence were analyzed.Here,we report the identification of the plasmid-mediated tigecycline resistance gene tet(X4)in a conjugative plasmid of the Salmonella enterica serovar Llandoff strain SH16G3606,isolated from a man in China in 2016,the first reported serovar Llandoff in China as a novel sequence type ST8300.The tet(X4)-mediated resistance phenotype was successfully transferred from the Salmonella Llandoff strain into Escherichia coli J53,resulting in a 32-fold increase in the minimal inhibitory concentration of tigecycline.The tet(X4)gene was located between two copies of ISCR2 in the plasmid pSal21GXH-tetX4.To our knowledge,this is the first report of the plasmid-mediated tigecycline resistance gene tet(X4)in a Salmonella Llandoff strain isolated from a human stool sample in China.In addition,our findings demonstrated that a total of 171 isolates are carrying tet(X)-like genes distributed in 21 countries or areas across 6 continents,posing a serious threat to humans and public health.Overall,our timely discovery of the recent emergence of the tet(X4)gene in Salmonella isolates and other Enterobacteriaceae bacteria species supports the need for rapid surveillance to prevent the tet(X)-like gene from spreading.Yanan Wang Fei Liu Xuebin Xu Hua Huang Na Lyu Sufang Ma Luping Chen Mengyu Mao Yongfei Hu Xiaofeng Song Jing Li Yuanlong Pan Aiping Wang Gaiping Zhang Baoli Zhu George F.Gao 2021Infectious Microbes & Diseases2021,3,4:0
9Identification of candidate genes for aphid resistance in upland cotton by QTL mapping and expression analysis显示文摘Lignin is one of the main components of cell walls and is essential for resistance to insect pests in plants.Cotton plants are damaged by aphid(Aphis gossypii) worldwide but resistant breeding is undeveloped due to scarce knowledge on resistance genes and the mechanism. This study reported a lignin biosynthesisrelated gene identified in the F_(2) population derived from the cross between cotton cultivars Xinluzao 61(resistant to aphid) and Xinluzao 50(susceptible to aphid). A quantitative trait locus was mapped on chromosome D04 with a logarithm of odds(LOD) score of 5.99 and phenotypic effect of 27%. RNA-seq analysis of candidate intervals showed that the expression level of GH_D04G1418 was higher in the resistant cultivar than in the susceptible cultivar. This locus is close to AtLAC4 in the phylogenetic tree and contains a conserved laccase domain. Hence, it was designated GhLAC4-3. Silencing of GhLAC4-3 in Xinluzao 61 via virus-induced gene silencing(VIGS) resulted in decreased lignin content and increased susceptibility to aphids.
