2Genetic diversity and construction of core collection in Chinese wheat genetic resources显示文摘Genetic diversity among 5029 accessions representing a proposed Chinese wheat core collection was analyzed using 78 pairs of fluorescent microsatellite (SSR) primers mapped to 21 chromosomes. A stepwise hierarchical sampling strategy with priority based on 4×105 SSR data-points was used to construct a core collection from the 23090 initial collections. The core collection consisted of 1160 accessions, including 762 landraces, 348 modern varieties and 50 introduced varieties. The core ac-counts for 23.1% of the 5029 candidate core accessions and 5% of the 23090 initial collections, but retains 94.9% of alleles from the candidate collections and captures 91.5% of the genetic variation in the initial collections. These data indicate that it is possible to maintain genetic diversity in a core collection while retaining fewer accessions than the accepted standard, i.e., 10% of the initial collections captured more than 70% of their genetic diversity. Estimated genetic representation of the core con-structed by preferred sampling (91.5%) is much higher than that by random sampling (79.8%). Both mean genetic richness and genetic diversity indices of the landraces were higher than those of the modern varieties in the core. Structure and principal coordinate analysis revealed that the landraces and the modern varieties were two relatively independent subpopulations. Strong genetic differentiation associated with ecological environments has occurred in the landraces, but was relatively weak in the modern cultivars. In addition, a mini-core collection was constructed, which consisted of 231 accessions with an estimated 70% representation of the genetic variation from the initial collections. 3CapOX as neoadjuvant chemotherapy for locally advanced operable colon cancer patients: a prospective single-arm phase Ⅱ trial显示文摘Objective: The aim of this prospective, single-arm phase II trial was to confirm the safety and efficacy of neoadjuvant chemotherapy(NAC) using oxaliplatin plus capecitabine(Cap OX) for patients with operable locally advanced colon cancer(CC).Methods: Patients with computed tomography-defined T4 or lymph node-positive CCs were enrolled. After radiological staging, patients were treated with at least 2 cycles of NAC consisting of 130 mg/m2 oxaliplatin on d 1,plus 1,000 mg/m2 capecitabine twice daily for 14 d every 3 weeks, followed by surgery, and then with the rest cycles of adjuvant chemotherapy. Radiological response was evaluated after 2 cycles of NAC. Tumor response, treatment toxicity, and surgical complications were recorded. The pathological response to therapy was evaluated according to the tumor regression grade(TRG) score. The primary endpoint was pathologic tumor response. This trial is registered in Clinical Trials.gov(No: NCT02415829).Results: Forty-seven patients were enrolled in the study. Forty-two patients completed the planned treatments.The total radiological response rate was 68%(32/47), including complete and partial response rates of 2%(1/47)and 66%(31/47), respectively. Stable disease was observed in 32%(15/47) and progressive disease was observed in none. Complete pathologic response, major regression, and at least moderate regression were achieved in 1(2%), 2(4%), and 29(62%) patients, respectively. Four patients developed grade 3 treatment toxicities. One patient with wound infection occurred after operation(1/47, 2%). 4Modulating macrophage activities to promote endogenous bone regeneration: Biological mechanisms and engineering approaches显示文摘A coordinated interaction between osteogenesis and osteoimmune microenvironment is essential for successful bone healing.In particular,macrophages play a central regulatory role in all stages of bone repair.Depending on the signals they sense,these highly plastic cells can mediate the host immune response against the exterior signals of molecular stimuli and implanted scaffolds,to exert regenerative potency to a varying extent.In this article,we first encapsulate the immunomodulatory functions of macrophages during bone regeneration into three aspects,as sweeper,mediator and instructor.We introduce the phagocytic role of macrophages in different bone healing periods(‘sweeper’)and overview a variety of paracrine cytokines released by macrophages either mediating cell mobilisation,vascularisation and matrix remodelling(‘mediator’),or directly driving the osteogenic differentiation of bone progenitors and bone repair(‘instructor’).Then,we systematically classify and discuss the emerging engineering strategies to recruit,activate and modulate the phenotype transition of macrophages,to exploit the power of endogenous macrophages to enhance the performance of engineered bone tissue.Yiming Niu Zhenzhen Wang Yuchen Shi Lei Dong Chunming Wang 2021Bioactive Materials2021,6,1:14

7Myeloid-specific targeting of Notch ameliorates murine renal fibrosis via reduced infiltration and activation of bone marrowderived macrophage显示文摘Macrophages play critical roles in renal fibrosis.However,macrophages exhibit ontogenic and functional heterogeneities,and which population of macrophages contributes to renal fibrosis and the underlying mechanisms remain unclear.In this study,we genetically targeted Notch signaling by disrupting the transcription factor recombination signal binding protein-JK(RBP-J),to reveal its role in regulation of macrophages during the unilateral ureteral obstruction(UUO)-induced murine renal fibrosis.Myeloid-specific disruption of RBP-J attenuated renal fibrosis with reduced extracellular matrix deposition and myofibroblast activation,as well as attenuated epithelial-mesenchymal transition,likely owing to the reduced expression of TGF-β.Mean while,RBP-J deletion significantly hampered macrophage infiltration and activation in fibrotic kidney,although their proliferation appeared unaltered.By using macrophage clearance experiment,we found that kidney resident macrophages made negligible contribution,but bone marrow(BM)-derived macrophages played a major role in renal fibrogenesis.Further mechanistic analyses showed that Notch blockade reduced monocyte emigration from BM by down-regulating CCR2 expression.Finally,we found that myeloid-specific Notch activation aggravated renal fibrosis,which was mediated by CCR2^+macrophages infiltration.In summary,our data have unveiled that myeloid-specific targeting of Notch could ameliorate renal fibrosis by regulating BM-derived macrophages recruitment and activation,providing a novel strategy for intervention of this disease.Yali Jiang Yuanyuan Wang Pengfei Ma Dongjie An Junlong Zhao Shiqian Liang Yuchen Ye Yingying Lu Peng Zhang Xiaowei Liu Hua Han Hongyan Qin 2019Protein & Cell2019,10,3:10
8On the improved tensile strength and ductility of Mg-Sn-Zn-Mn alloy processed by aging prior to extrusion显示文摘This study investigated the influence of aging prior to extrusion(APE)on the tensile strength and ductility of as-extruded Mg-8.32Sn-1.85Zn-0.17Mn alloy.Results demonstrated that APE treatment dramatically increased the volume fraction of recrystallized grains,thereby decreasing the grain size of the as-extruded alloy.This phenomenon was primarily attributed to the particle-stimulated nucleation and pinning effect induced by large amounts of small Mg 2 Sn precipitates produced by the APE treatment and dynamic precipitation.The tensile yield strength increased from 242.4 MPa to 256.5 MPa after APE treatment.The improved tensile strength can be attributed to the enhanced grain boundary strengthening and precipitation strengthening.The ductility of the as-extruded alloy also markedly increased from 7.1%to 13.5%after the APE treatment.The improved ductility of APE alloy was attributed to the texture randomization,the activity of pyramidalslip and the suppressed formation of{10-11}contraction twins and coarse Mg 2 Sn phases.Liping Zhong Yongjian Wang Yuchen Dou 2019Journal of Magnesium and Alloys2019,7,4:8
9Self-supported high-entropy alloy electrocatalyst for highly efficient H_(2) evolution in acid condition显示文摘Developing non-precious catalysts as Pt substitutes for electrochemical hydrogen evolution reaction(HER)with superior stability in acidic electrolyte is of critical importance for large-scale,low-cost hydrogen production from water.Herein,we report a CoCrFeNiAl high-entropy alloy(HEA)electrocatalyst with self-supported structure synthesized by mechanical alloying and spark plasma sintering(SPS)consolidation.The HEA after HF treatment and in situ electrochemical activation for 4000 cycles of cyclic voltammetry(HF-HEAa2)presents favourable activity with overpotential of 73 mV to reach a current density of 10 mA cm^(2) and a Tafel slope of 39.7 mV dec1.The alloy effect of Al/Cr with Co/Fe/Ni at atomic level,high-temperature crystallization,as well as consolidation by SPS endow CoCrFeNiAl HEA with high stability in 0.5 M H2SO4 solution.The superior performance of HF-HEAa2 is related with the presence of metal hydroxides/oxides groups on HEA.Peiyan Ma Mingming Zhao Long Zhang Heng Wang Junfeng Gu Yuchen Sun Wei Ji Zhengyi Fu 2020Journal of Materiomics2020,6,4:8
10Prognostic value of programmed death.1, programmed death-ligand 1, programmed death-ligand 2 expression, and CD8(+) T cell density in primary tumors and metastatic lymph nodes from patients with stage T1.4N+M0 gastric adenocarcinoma显示文摘Background: Anti-programmed death-1/programmed death-ligand 1(PD-1/PD-L1) immunotherapy has been proved to be effective on gastric cancer in ongoing clinical trials. However, the value of PD-L1 in predicting responses of patients with gastric cancer to anti-PD-1/PD-L1 immunotherapy is controversial. Some studies suggested that intra-and inter-tumoral heterogeneity of PD-L1 expression might explain the controversy.This study aimed to analyze the expression of PD-L1, PD-L2, and PD-1 as well as CD8(+) T-cell density in primary tumors and lymph nodes from patients with stage T1-4 N+M0 gastric adenocarcinoma to explore the heterogeneity of PD-1 signaling pathway molecules.Methods: In primary tumors and metastatic as well as non-metastatic lymph nodes from patients with stage T1-4 N+M0 gastric adenocarcinoma, we detected PD-L1 and PD-L2 expression with immunohistochemistry. CD8(+)T-cell density in primary tumors and PD-1 expression on CD8(+)T cells were detected with immunofluorescence. Univariate analysis was used to determine the prognostic values of them. Cox proportional hazard regression model was used to identify independent risk factors that affect patients' overall survival and disease-free survival.Results: Among 119 eligible patients who had undergone surgical resection, the positive rate of PD-L1 was higher in metastatic lymph nodes than in primary tumors(45.4% vs. 38.7%, P = 0.005); the positive rate of PD-1 on CD8(+)T cells was significantly higher in primary tumors and metastatic lymph nodes than in tumor-free lymph nodes(both P < 0.001). The intensity of PD-1 expression on CD8(+) T cells in primary tumors and in metastatic lymph nodes were stronger than that in tumor-free lymph nodes from the same patient. Beside, the positive rate of PD-L2 did not show any differences between primary tumors and metastatic lymph nodes. 11Thermal adaptation in overheated residential buildings in severe cold area in China显示文摘The winter in the severe cold area of China is long and cold. The mean outdoor temperature is about-10. 0 ℃ during the winter in Harbin,while the indoor air temperature is often above24 ℃. How does the indoor environment influence human thermal comfort and adaptation in such an overheated environment?A combined approach of spot-reading measurements and occupant interview s w as adopted in nine residential buildings of five communities during the heating period in 2013-2014. Tw enty residents w ere chosen as respondents. Totally 308 valid questionnaires w ere collected. The heating periods w ere separated into three phases based on the outdoor temperature.The results show that the mean indoor air temperatures in theearly-,mid-and late-heating periods w ere 23. 6 ℃,24. 3 ℃and 25. 0 ℃,respectively,w hich w ere larger than or close to the upper limit recommended by thermal comfort standards, and slightly higher than the related thermal neutral temperatures. With the heating process,the mean clothing insulation of residents decreased. Opening w indow s and reducing clothing w ere mainly taken by the residents to adapt to the overheated environment.The thermal neutral temperature has an upw ard tendency w ith the increasing indoor air temperature. On the other hand,overheating in residential buildings w ould make residents open w indow s,w hich may cause thermal discomfort and energy w aste. 14Functional analysis of a novel C-glycosyltransferase in the orchid Dendrobium catenatum显示文摘Flavonoids,which are a diverse class of phytonutrients,are used by organisms to respond to nearly all abiotic stresses and are beneficial for human health.Glycosyltransferase,used during the last step of flavonoid biosynthesis,is important in flavonoid enrichment.However,little is known about glycosyltransferase in the orchid Dendrobium catenatum(D.officinale).In this study,we isolated a novel C-glycosyltransferase(designated DcaCGT)from the orchid D.catenatum by identifying and analyzing 82 putative genes in the GT1 family.DcaCGT could specifically catalyze not only di-C-glycosylation but also O-glycosylation.Apart from the normal function of catalyzing 2-hydroxynaringenin and phloretin to the respective di-C-glycosides,DcaCGT also catalyzes apigenin to cosmosiin.Targeted metabolic profiling of the substrates(2-hydroxynaringenin,phloretin,and apigenin)and products(vitexin,isovitexin,vicenin-2,nothofagin,3’,5’-di-C-glucosylphloretin,and cosmosiin)in different tissues showed that vicenin-2 was the most abundant product of this novel enzyme.Cosmosiin was detected in flowers and flower buds.We also established that DcaCGT functions expanded throughout the evolution of D.catenatum.Residual OGT activity may help D.catenatum resist drought stress.Our study illustrates the function,origin,and differentiation of DcaCGT and provides insights into glycosylation and molecular propagation processes,which can be used to improve the production of flavonoids by the cultivated medicinal plant D.catenatum.Zhiyao Ren Xiaoyu Ji Zhenbin Jiao Yingyi Luo Guo-Qiang Zhang Shengchang Tao Zhouxi Lei Jing Zhang Yuchen Wang Zhong-Jian Liu Gang Wei 2020Horticulture Research2020,7,1:5
15Spatiotemporal variations of nitrate sources and dynamics in a typical agricultural riverine system under monsoon climate显示文摘Nitrogen pollution is a serious environmental issue in the Danjiangkou Reservoir region(DRR),the water source of the South-to-North Water Diversion Project of China.In this research,seasonal surveys and a bi-weekly time series survey were conducted in the Qihe River Basin,one of the most densely populated agricultural basins in the DRR.Hydrochemical compositions(NO3^-Cl^-),dual isotopes(δD-H2O,δ^18O-H2O,δ^15N-NO3^-,andδ^18O-NO3^-),and a Markov Chain Monte Carlo isotope mixing model were jointly applied to unravel the sources,migrations,and transformations of the nitrate(NO3^-)in the basin.It was revealed that the mixing between different sources was the main process controlling the isotopic compositions of the riverine NO3the upper-middle reaches.In contrast,denitrification occurred in the lower reaches.For the first time,the sources of NO3^-quantified at a basin scale in the DRR.Overall,the river transported 484.2 tons/year of NO3^-N to the reservoir,of which 32.6%,36.4%,28.0%,and 3.0%was from soil organic nitrogen,chemical fertilizer,residential sewage and atmospheric precipitation,respectively.The NO3^-N fluxes of the different sources were regulated by the monsoon climate and anthropogenic activities.For example,high precipitation and intense fertilization resulted in severe nonpoint source pollution.Denitrification thrived in soils and reservoirs in wet seasons.Temperature could regulate the migration,nitrification and denitrification processes.Based on the results,we suggest that the management strategies dealing with nitrogen pollution issue in the DRR should follow the specific spatiotemporal characteristics of NO3migration and transformation mechanisms.Hao Jiang Wenjing Liu Jiangyi Zhang Li Zhou Xiaode Zhou Ke Pan Tong Zhao Yuchen Wang Zhifang Xu 2020Journal of Environmental Sciences2020,32,7:5
16Investigating spatial and temporal variations of soil moisture content in an arid mining area using an improved thermal inertia model显示文摘Mining operations can usually lead to environmental deteriorations. Underground mining activities could cause an extensive decrease in groundwater level and thus a dramatic variation in soil moisture content(SMC). In this study, the spatial and temporal variations of SMC from 2001 to 2015 at two spatial scales(i.e., the Shendong coal mining area and the Daliuta Coal Mine) were analyzed using an improved thermal inertia model with a long-term series of Landsat TM/OLI(TM=Thematic Mapper and OLI=Operational Land Imager) data. Our results show that at large spatial scale(the Shendong coal mining area), underground mining activities had insignificant negative impacts on SMC and that at small spatial scale(the Daliuta Coal Mine), underground mining activities had significant negative impacts on SMC. 17Engineering osteoarthritic cartilage model through differentiating senescent human mesenchymal stem cells for testing disease-modifying drugs显示文摘Significant cellular senescence has been observed in cartilage harvested from patients with osteoarthritis(OA).In this study,we aim to develop a senescence-relevant OA-like cartilage model for developing disease-modifying OA drugs(DMOADs).Spe-cifically,human bone marrow-derived mesenchymal stromal cells(MSCs)were expanded in vitro up to passage 10(P10-MSCs).Following their senescent phenotype formation,P10-MSCs were subjected to pellet culture in chondrogenic medium.Results from qRT-PCR,histology,and immunostaining indicated that cartilage generated from P10-MSCs displayed both senescent and OA-like phenotypes without using other OA-inducing agents,when compared to that from normal passage 4(P4)-MSCs.Interestingly,the same gene expression differences observed between P4-MSCs and P10-MSC-derived cartilage tissues were also observed between the preserved and damaged OA cartilage regions taken from human samples,as demonstrated by RNA sequencing data and other analysis methods.Lastly,the utility of this senescence-initiated OA-like cartilage model in drug development was assessed by testing several potential DMOADs and senolytics.The results suggest that pre-existing cellular senescence can induce the generation of OA-like changes in cartilage.The P4-and P10-MSCs derived cartilage models also represent a novel platform for predicting the efficacy and toxicity of potential DMOADs on both preserved and damaged cartilage in humans.Ning Wang Yuchen He Silvia Liu Meagan J.Makarcyzk Guanghua Lei Alexander Chang Peter G Alexander Tingjun Hao Anne-Marie Padget Nuria de Pedro Tsapekos Menelaos Hang Lin 2022Science China(Life Sciences)2022,65,2:5
18Inducing Flow Velocities to Manage Fish Reproduction in Regulated Rivers显示文摘Conservation of endangered or economic fish and control of invasive fish is a great challenge of hydraulic engineering worldwide.Flow velocity has been recognized to affect the spawning of fishes delivering drifting eggs in rivers.However,solid scientific supports and associated mechanisms to establish quantitative relations between flow velocity and fish reproduction,taking into account spawning,fertilizing,hatching,as well as surviving,are lacking.In this paper,we quantified the relationship between flow velocity and reproduction of Chinese carps through both lab and field experiments.The results showed that a minimum velocity was required to trigger Hypophthalmichthys molitrix(H.molitrix)releasing eggs,and a velocity range was preferred to sustain spawning activity.However,the embryo incubation and larvae development of H.molitrix were found to be inhibited with the increase in flow velocity.Considering the requirements of spawning and hatching,as well as larvae surviving,an optimized flow velocity processes was identified for the reproduction of H.molitrix in rivers.These findings were of great significance to the adaptive operation of reservoirs to create reasonable and precise ecological flows for managing fish reproduction,as shown by the promising results in the engineering application to the Three Gorges Reservoir.Qiuwen Chen Jianyun Zhang Yuchen Chen Kangle Mo Jun Wang Lei Tang Yuqing Lin Lei Chen Yong Gao Wei Jiang Yuxin Zhang 2021Engineering2021,7,2:4
19Phytophthora Effectors Modulate Genome-wide Alternative Splicing of Host mRNAs to Reprogram Plant Immunity显示文摘Alternative splicing(AS)of pre-mRNAs increases transcriptome and proteome diversity,regulates gene expression through multiple mechanisms,and plays important roles in plant development and stress responses.However,the prevalence of genome-wide plant AS changes during infection and the mechanisms by which pathogens modulate AS remain poorly understood.Here,we examined the global AS changes in tomato leaves infected with Phytophthora infestans,the infamous Irish famine pathogen.We show that more than 2000 genes exhibiting significant changes in AS are not differentially expressed,indicating that AS is a distinct layer of transcriptome reprogramming during plant-pathogen interactions.Furthermore,our results show that P.infestans subverts host immunity by repressing the AS of positive regulators of plant immunity and promoting the AS of susceptibility factors.To study the underlying mechanism,we established a luminescence-based AS reporter system in Nicotiana benthamiana to screen pathogen effectors modulating plant AS.We identified nine splicing regulatory effectors(SREs)from 87 P.infestans effectors.Further studies revealed that SRE3 physically binds U1-70K to manipulate the plant AS machinery and subsequently modulates AS-mediated plant immunity.Our study not only unveils genome-wide plant AS reprogramming during infection but also establishes a novel AS screening tool to identify SREs from a wide range of plant pathogens,providing opportunities to understand the splicing regulatory mechanisms through which pathogens subvert plant immunity.Jie Huang Xinyu Lu Hongwei Wu Yuchen Xie Qian Peng Lianfeng Gu Juyou Wu Yuanchao Wang Anireddy SNReddy Suomeng Dong 2020Molecular Plant2020,13,10:4
20Controllable synthesis of nitrogen-doped porous carbon from metal-polluted miscanthus waste boosting for supercapacitors显示文摘High-value reclamation of metal-polluted plants involved in phytoremediation is a big challenge.In this study,nitrogen-doped nanoporous carbon with large specific area of 2359.1 m^(2)g^(-1) is facilely fabricated from metal-polluted miscanthus waste for efficient energy storage.The synergistic effect of KOH,urea and ammonia solution greatly improve the nitrogen quantity and surface area of the synthesized carbon.Electrodes fabricated with this carbon exhibit the excellent capacitance performance of 340.2 F g^(-1) at 0.5 A g^(-1) and a low combined resistance of 0.116Ω,which are competitive with most of previously reported carbon-based electrodes.In addition,the as-obtained carbon electrode shows a high specific capacitance retention of over 99.6%even after 5000 cycles.Furthermore,the symmetric supercapacitor fabricated using the synthesized carbon achieves a superior energy density of 25.3 Wh kg^(-1)(at 400 W kg^(-1))in 1 mol L^(-1) Na_(2)SO_(4)aqueous solution.This work provides an efficient route to upcycle metal-polluted plant waste for supercapacitor applications.Zuo Chen Man Zhang Yuchen Wang Zhiyu Yang Di Hu Yetao Tang Kai Yan 2021Green Energy & Environment2021,6,6:4
