1MATRIEX imaging:multiarea two-photon real-time in vivo explorer显示文摘Two-photon laser scanning microscopy has been extensively applied to study in vivo neuronal activity at cellular and subcellular resolutions in mammalian brains.However,the extent of such studies is typically confined to a single functional region of the brain.Here,we demonstrate a novel technique,termed the multiarea two-photon real-time in vivo explorer(MATRIEX),that allows the user to target multiple functional brain regions distributed within a zone of up to 12mm in diameter,each with a field of view(FOV)of ~200μm in diameter,thus performing two-photon Ca2+imaging with single-cell resolution in all of the regions simultaneously.For example,we demonstrate real-time functional imaging of single-neuron activities in the primary visual cortex,primary motor cortex and hippocampal CA1 region of mice in both anesthetized and awake states.A unique advantage of the MATRIEX technique is the configuration of multiple microscopic FOVs that are distributed in three-dimensional space over macroscopic distances(>1 mm)both laterally and axially but that are imaged by a single conventional laser scanning device.In particular,the MATRIEX technique can be effectively implemented as an add-on optical module for an existing conventional single-beam-scanning two-photon microscope without requiring any modification to the microscope itself.Thus,the MATRIEX technique can be readily applied to substantially facilitate the exploration of multiarea neuronal activity in vivo for studies of brain-wide neural circuit function with single-cell resolution.Mengke Yang Zhenqiao Zhou Jianxiong Zhang Shanshan Jia Tong Li Jiangheng Guan Xiang Liao Bing Leng Jing Lyu Kuan Zhang Min Li Yan Gong Zhiming Zhu Junan Yan Yi Zhou Jian K Liu Zsuzsanna Varga Arthur Konnerth Yuguo Tang Jinsong Gao Xiaowei Chen Hongbo Jia 2019Light(Science & Applications)2019,8,1:4
2Effect of methyl tert-butyl ether on adipogenesis and glucose metabolism in vitro and in vivo显示文摘Methyl tert-butyl ether (MTBE),as a widely used gasoline additive,is suspected of being environmentally toxic.MTBE accumulates mainly in adipose tissue,but its effect on obesity or obesity-related metabolic disorders has not been well understood yet.Therefore,we examined the effect of MTBE on the adipose function and the related metabolic processes with both 3T3-L1 cell line and C57BL/6J mice model.We found that exposure to MTBE at the environmental relevant concentration (100 μmol/L) could significantly induce differentiation of preadipocyte and disturb insulin-stimulated glucose uptake of mature adipocyte.The in vivo observation in male mice showed a positive correlation of visceral white adipose tissue (vWAT) expansion and cell size increase with MTBE treatment in 14 weeks.Glucose tolerance and insulin sensitivity tests demonstrated that MTBE at 1000 μg/(kg·day) disturbed the systemic glucose metabolism in a gender-specific manner,which might be partly attributed to the alterations of gut microbiota community at genus level with respect to Akkermansia,Clostridium XIVb,and Megamonas.In summary,our study characterized the effect of MTBE on adipose tissue function and glucose homeostasis in vitro and in vivo,and revealed that systemic disorders of the glucose metabolism might be modulated by the related gut microbiota.Yue Tang Qidong Ren Qing Wen Caixia Yu Xinni Xie Qing Hu Yuguo Du 2019Journal of Environmental Sciences2019,31,11:4
3Methyl tertiary-butyl ether inhibits THP-1 macrophage cholesterol efflux in vitro and accelerates atherosclerosis in ApoE-deficient mice in vivo显示文摘The biosafety of methyl tertiary-butyl ether(MTBE),mainly used as a gasoline additive,has long been a contentious topic.In addition to its routine toxicities,MTBE has been demonstrated to disrupt glucose and lipid metabolism and contribute to the development of type2 diabetes as well as obesity.As one of the morbidities related to dyslipidemia,atherosclerosis is worthy of being investigated under MTBE exposure.Since foam cells derived from macrophages play pivotal roles during atherosclerosis development,we studied the effects of MTBE on macrophages in vitro and assessed the effect of MTBE on atherosclerosis plaque formation with the ApoE^(-/-)mouse model in uiuo for the first time.Our results demonstrated that exposure to MTBE at environmentally relevant concentrations decreased the expression of ABCA1 and ABCG1,which are responsible for macrophage cholesterol efflux,at both mRNA and protein levels in THP-1 macrophages.Consequently,treatment with MTBE inhibited the transport of cholesterol from macrophages to High-density lipoprotein.ApoE^(-/-)mice exposed to MTBE at environmentally relevant concentrations(100,1000μg/kg)displayed significant increases in lesion area in the aorta and aortic root compared to vehicletreated ones.Further analysis indicated that MTBE exposure enhanced the macrophagespecific marker Mac-2 contents within plaques in the aortic root,implying that MTBE could promote macrophage-derived foam cell formation and thus accelerate atherosclerosis plaque formation.We for the first time demonstrated the pro-atherogenic effect of MTBE via eliciting disruption of macrophage cholesterol efflux and accelerating foam cell formation and atherosclerosis plaque development.Qidong Ren Xinni Xie Yue Tang Qing Hu Yuguo Du 2021Journal of Environmental Sciences2021,33,3:1
4High-fidelity structured illumination microscopy by point-spread-function engineering显示文摘Structured illumination microscopy(SIM)has become a widely used tool for insight into biomedical challenges due to its rapid,long-term,and super-resolution(SR)imaging.However,artifacts that often appear in SIM images have long brought into question its fidelity,and might cause misinterpretation of biological structures.We present HiFi-SIM,a high-fidelity SIM reconstruction algorithm,by engineering the effective point spread function(PSF)into an ideal form.HiFi-SIM can effectively reduce commonly seen artifacts without loss of fine structures and improve the axial sectioning for samples with strong background.In particular,HiFi-SIM is not sensitive to the commonly used PSF and reconstruction parameters;hence,it lowers the requirements for dedicated PSF calibration and complicated parameter adjustment,thus promoting SIM as a daily imaging tool.Gang Wen Simin Li Unbo Wang Xiaohu Chen Zhenglong Sun Yong Liang Xin Jin Yifan Xing Yaming jiu Yuguo Tang Hui Li 2021Light(Science & Applications)2021,10,5:1
5An ultrasensitive aptasensor for prostate specific antigen assay based on Exonuclease T-aided cyclic cleavage显示文摘An ultrasensitive electrochemical aptasensor is presented for prostate specific antigen(PSA) detection. DNA tetrahedronaptamer is designed, which not only facilitates the molecular self-assembly events,but also improves the recognition efficiency between PSA and aptamer sequence on the electrode interface. The DNA conformation on top of DNA tetrahedron changes accordingly, which can be further digested by Exonuclease T(Exo T), a type of single-strand specific nuclease. Electrochemical species are removed synchronously and the initial PSA level can thus be determined. A linear range from 0.5 pg mL^(-1) to50 ng mL^(-1) is achieved with the limit of detection(LOD) as low as 0.15 pg mL^(-1). 11A novel predictor of unsustained return of spontaneous circulation in cardiac arrest patients through a combination of capnography and pulse oximetry: a multicenter observational study显示文摘BACKGROUND:Unsustained return of spontaneous circulation(ROSC)is a critical barrier to survival in cardiac arrest patients.This study examined whether end-tidal carbon dioxide(ETCO_(2))and pulse oximetry photoplethysmogram(POP)parameters can be used to identify unsustained ROSC.METHODS:We conducted a multicenter observational prospective cohort study of consecutive patients with cardiac arrest from 2013 to 2014.Patients’general information,ETCO_(2),and POP parameters were collected and statistically analyzed.RESULTS:The included 105 ROSC episodes(from 80 cardiac arrest patients)comprised 51 sustained ROSC episodes and 54 unsustained ROSC episodes.The 24-hour survival rate was significantly higher in the sustained ROSC group than in the unsustained ROSC group(29.2%vs.9.4%,P<0.05).The logistic regression analysis showed that the difference between after and before ROSC in ETCO_(2)(ΔETCO_(2))and the difference between after and before ROCS in area under the curve of POP(ΔAUCp)were independently associated with sustained ROSC(odds ratio[OR]=0.931,95%confi dence interval[95%CI]0.881-0.984,P=0.011 and OR=0.998,95%CI 0.997-0.999,P<0.001).The area under the receiver operating characteristic curve ofΔETCO_(2),ΔAUCp,and the combination of both to predict unsustained ROSC were 0.752(95%CI 0.660-0.844),0.883(95%CI 0.818-0.948),and 0.902(95%CI 0.842-0.962),respectively.CONCLUSION:Patients with unsustained ROSC have a poor prognosis.The combination ofΔETCO_(2) andΔAUCp showed signifi cant predictive value for unsustained ROSC.Jing Yang Hanqi Tang Shihuan Shao Feng Xu Yangyang Fu Shengyong Xu Chen Li Yan Li Yang Liu Joseph Harold Walline Huadong Zhu Yuguo Chen Xuezhong Yu Jun Xu 2024World Journal of Emergency Medicine2024,15,1:0
