1S19W,T27W,and N33OY mutations in ACE2 enhance SARS-CoV-2 S-RBD binding toward both wild-type and antibody-resistant viruses and its molecular basis显示文摘SARS-CoV-2 recognizes,via its spike receptor-binding domain(S-RBD),human angiotensin-converting enzyme 2(ACE2)to initiate infection.Ecto-domain protein of ACE2 can therefore function as a decoy.Here we show that mutations of S19W,T27W,and N330Y in ACE2 could individually enhance SARS-CoV-2 S-RBD binding.Y330 could be synergistically combined with either W19 or W27,whereas W19 and W27 are mutually unbeneficial.The structures of SARS-CoV-2S-RBD bound to the ACE2 mutants reveal that the enhan ced binding is mainly con tributed by the van der Waals interactio ns mediated by the aromatic side-chai ns from W19,W27,and Y330.While Y330 and W19/W27 are distantly located and devoid of any steric interference,W19 and W27 are shown to orient their side-chains toward each other and to cause steric conflicts,explai ning their in compatibility.Finally,using pseudotyped SARS-CoV-2 viruses,we dem on strate that these residue substitutions are associated with dramatically improved entry-inhibition efficacy toward both wild-type and antibody-resistant viruses.Taken together,our biochemical and structural data have delineated the basis for the elevated S-RBD binding associated with S19W,T27W,and N330Y mutations in ACE2,paving the way for potential application of these mutants in dinical treatment of COVID-19.Fei Ye Xi Lin Zimin Che Fanli Yang Sheng Lin Jing Yang Hua Chen Honglu Sun Lingling Wang Ao Wen Xindan Zhang Yushan Dai Yu Cao Jingyun Yang Guobo Shen Li Yang Jiong Li Zhenling Wang Wei Wang Xiawei Wei Guangwen Lu 2021Signal Transduction and Targeted Therapy2021,6,10:0

3Study on Callus Induction and Tissue Culture Regeneration System of Malus toringoides显示文摘[Objectives]To study the culture of callus produced by the leaves of the ethnic medicine‘E Se’,the induction of adventitious buds and the regeneration system of tissue culture seedlings.[Methods]Multi-factor orthogonal experiment was used to design and analyze.[Results]The best medium for callus induction was MS+2.0 mg/L 6-BA+0.1 mg/L 2,4-D.By comparing the effects of cytokinin 6-BA,KT and TDZ on induction of adventitious bud differentiation,it was found that the optimum medium for adventitious bud regeneration was MS+0.5 mg/L 6-BA+0.5 mg/L KT+0.15 mg/L IBA.Finally,adding 0.2 mg/L IBA(0.1 or 0.2 mg/L)to MS medium could effectively make regenerated seedlings produce roots,and the efficiency was more than 70%.[Conclusions]It laid a foundation for the large-scale cultivation of the seedlings of the ethnic medicine‘E Se’and the promotion of their artificial cultivation.Xiaobo QIN Bei NIU Lijuan FAN Guozhen ZHANG Dongmei HU Bingbing ZHANG Xiaodong SHI Xinyi XU Peng WU Tingwei DAI Siying QIN Xiao LI Yushan DENG Fei ZU 2019Medicinal Plant2019,10,5:0
4Artificial synapses based on organic electrochemical transistors with self-healing dielectric layers显示文摘Organic electrochemical transistors(OECTs)have emerged as one type of promising building block for neuromorphic systems owing to their capability of mimicking the morphology and functions of biological neurons and synapses.Currently,numerous kinds of OECTs have been developed,while self-healing performance has been neglected in most reported OECTs.In this work,the OECTs using self-healing polymer electrolytes as dielectric layers are proposed.Several important synaptic behaviors are simulated in the OECTs by doping the channel layers with ions from the electrolytes.Benefitting from the dynamic hydrogen bonds in the self-healing polymer electrolytes,the OECTs can successfully maintain their electrical performance and the ability of emulating synaptic behaviors after self-healing compared with the initial state.More significantly,the sublinear spatial summation function is demonstrated in the OECTs and their potential in flexible electronics is also validated.These results suggest that our devices are expected to be a vital component in the development of future wearable and bioimplantable neuromorphic systems.Yushan Gao Junyao Zhang Dapeng Liu Tongrui Sun Jun Wang Li Li Shilei Dai Jianhua Zhang Zhenglong Yang Jia Huang 2024Chinese Chemical Letters2024,35,3:0
