1Research and development of a novel subunit vaccine for the currently circulating pseudorabies virus variant in China显示文摘Pseudorabies(PR)is a devastating viral disease which leads to fatal encephalitis and respiratory disorders in pigs.Commercial gE-deleted live pseudorabies virus(PRV)vaccine has been widely used to control this disease in China.However,the new-emerging variants of PRV compromises the protection provided by current vaccines and lead to the outbreak of PR in vaccinated pig herds.Several killed and live vaccine candidates based on current PRV variants have been reported to be effective to control the disease.A subunit vaccine based on gB protein,one major PRV glycoprotein which elicits strong humoral and cellular immune responses,however,was never evaluated for protection against the current circulating PRV variants.In this study,full-length PRV gB protein was successfully expressed in baculovirus/insect cells in the soluble format and was tested on 3-week-old piglets as a subunit vaccine.Compared with unvaccinated pigs,the gB-vaccinated pigs developed specific antibody-mediated responses and were protected from the virulent PRV HN1201 challenge.All vaccinated pigs survived without showing any PRV-specific respiratory and neurological signs,but all unvaccinated pigs died within 7 days after HN1201 challenge.Hence,this novel gB-based vaccine could be applied as an effective subunit vaccine to control PRV variant in China.Yuzhou WANG Tongyan WANG He YAN Fanli YANG Linghua GUO Qingyuan YANG Xule HU Feifei TAN Yan XIAO Xiangdong LI Kegong TIAN 2015Frontiers of Agricultural Science and Engineering2015,2,3:2

3MST4 kinase regulates immune thrombocytopenia by phosphorylating STAT1-mediated M1 polarization of macrophages显示文摘Primary immune thrombocytopenia(ITP)is an autoimmune hemorrhagic disorder in which macrophages play a critical role.Mammalian sterile-20-like kinase 4(MST4),a member of the germinal-center kinase STE20 family,has been demonstrated to be a regulator of inflammation.Whether MST4 participates in the macrophage-dependent inflammation of ITP remains elusive.The expression and function of MST4 in macrophages of ITP patients and THP-1 cells,and of a macrophage-specific Mst4−/−(Mst4ΔM/ΔM)ITP mouse model were determined.Macrophage phagocytic assays,RNA sequencing(RNA-seq)analysis,immunofluorescence analysis,coimmunoprecipitation(co-IP),mass spectrometry(MS),bioinformatics analysis,and phosphoproteomics analysis were performed to reveal the underlying mechanisms.The expression levels of the MST4 gene were elevated in the expanded M1-like macrophages of ITP patients,and this elevated expression of MST4 was restored to basal levels in patients with remission after high-dose dexamethasone treatment.The expression of the MST4 gene was significantly elevated in THP-1-derived M1 macrophages.Silencing of MST4 decreased the expression of M1 macrophage markers and cytokines,and impaired phagocytosis,which could be increased by overexpression of MST4.In a passive ITP mouse model,macrophage-specific depletion of Mst4 reduced the numbers of M1 macrophages in the spleen and peritoneal lavage fluid,attenuated the expression of M1 cytokines,and promoted the predominance of FcγRIIb in splenic macrophages,which resulted in amelioration of thrombocytopenia.Downregulation of MST4 directly inhibited STAT1 phosphorylation,which is essential for M1 polarization of macrophages.Our study elucidates a critical role for MST4 kinase in the pathology of ITP and identifies MST4 kinase as a potential therapeutic target for refractory ITP.Jingjing Cao Lili Ji Yanxia Zhan Xia Shao Pengcheng Xu Boting Wu Pu Chen Luya Cheng Xibing Zhuang Yang Ou Fanli Hua Lihua Sun Feng Li Hao Chen Zhaocai Zhou Yunfeng Cheng 2023Cellular & Molecular Immunology2023,20,12:0

5Mechanism of Inhibitory Effect of Essential Oil from Valerianae Jatamansi Rhizoma et Radix on Activation of Microglia显示文摘[Objectives]To investigate the inhibitory effect and possible mechanism of essential oil from Valerianae Jatamansi Rhizoma et Radix on microglia activation induced by lipopolysaccharide(LPS).[Methods]The LPS-induced microglia activation model was established and treated with different doses of essential oil from Valerianae Jatamansi Rhizoma et Radix.MTT assay was used to detect cell viability,ELISA to detect IL-6 secretion,RT-PCR to detect mRNA expression levels of IL-6,IL-1β,NF-κB,and IκBα,Western blotting to detect the protein expression of IL-6,IL-1β,NF-κB,IκBα,and their phosphorylated products.[Results]Compared with the normal control group,the model group showed increased IL-6 content(P<0.01),upregulated mRNA and protein levels of IL-6,IL-1β,and NF-κB(P<0.01),and elevated ratio of P-IκBα/IκBα(P<0.05).Compared with the model group,4 and 2μg/L essential oil from Valerianae Jatamansi Rhizoma et Radix reduced the content of IL-6(P<0.05),while 4,2,and 1μg/L essential oil from Valerianae Jatamansi Rhizoma et Radix down-regulated the mRNA and protein levels of IL-6,IL-1β,and NF-κB to varying degrees(P<0.05 or P<0.01),up-regulate the mRNA expression of IκBα(P<0.05 or P<0.01),and decreased the ratio of P-IκBα/IκBα(P<0.05 or P<0.01).[Conclusions]Essential oil from Valerianae Jatamansi Rhizoma et Radix can inhibit LPS-induced microglia activation,and its mechanism may be related to the inhibition of the NF-κB/IκBαsignaling pathway.Qingde LONG Nanyun YANG Fanli ZENG Xu ZHANG Xiaoying WU Xiangchun SHEN 2022Medicinal Plant2022,13,1:0

7Ultrathin 2D Covalent Organic Framework Film Fabricated via Langmuir-Blodgett Method with a“Two-in-One”Type Monomer显示文摘In recent years, covalent organic frameworks(COFs) are evolving as a novel kind of porous materials for catalysis and molecular separation, gas adsorption, etc. Various functional building blocks have been explored to tune the pore channels, including the pore size and structures. In this article, a new terphenyl(TP) based COF(TP-COF) was developed via a “two-in-one” strategy by using a symmetric A2B2monomer, i.e., 4,4''-diamino-2',5'-diformyl-1,1':4',1''-terphenyl(DADFTP). The pore size of TP-COF was only 0.99 nm by shortening the arm length of the DADFTP monomer. Freestanding, continuous and ultrathin COF films could be facilely prepared at the air-liquid interface through the modified Langmuir-Blodgett(LB) method. 8S19W,T27W,and N33OY mutations in ACE2 enhance SARS-CoV-2 S-RBD binding toward both wild-type and antibody-resistant viruses and its molecular basis显示文摘SARS-CoV-2 recognizes,via its spike receptor-binding domain(S-RBD),human angiotensin-converting enzyme 2(ACE2)to initiate infection.Ecto-domain protein of ACE2 can therefore function as a decoy.Here we show that mutations of S19W,T27W,and N330Y in ACE2 could individually enhance SARS-CoV-2 S-RBD binding.Y330 could be synergistically combined with either W19 or W27,whereas W19 and W27 are mutually unbeneficial.The structures of SARS-CoV-2S-RBD bound to the ACE2 mutants reveal that the enhan ced binding is mainly con tributed by the van der Waals interactio ns mediated by the aromatic side-chai ns from W19,W27,and Y330.While Y330 and W19/W27 are distantly located and devoid of any steric interference,W19 and W27 are shown to orient their side-chains toward each other and to cause steric conflicts,explai ning their in compatibility.Finally,using pseudotyped SARS-CoV-2 viruses,we dem on strate that these residue substitutions are associated with dramatically improved entry-inhibition efficacy toward both wild-type and antibody-resistant viruses.Taken together,our biochemical and structural data have delineated the basis for the elevated S-RBD binding associated with S19W,T27W,and N330Y mutations in ACE2,paving the way for potential application of these mutants in dinical treatment of COVID-19.Fei Ye Xi Lin Zimin Che Fanli Yang Sheng Lin Jing Yang Hua Chen Honglu Sun Lingling Wang Ao Wen Xindan Zhang Yushan Dai Yu Cao Jingyun Yang Guobo Shen Li Yang Jiong Li Zhenling Wang Wei Wang Xiawei Wei Guangwen Lu 2021Signal Transduction and Targeted Therapy2021,6,10:0
