| 1 | Prioritizing natural-selection signals from the deep-sequencing genomic data suggests multi-variant adaptation in Tibetan highlanders显示文摘Human genetic adaptation to high altitudes(>2500 m)has been extensively studied over the last few years,but few functional adaptive genetic variants have been identified,largely owing to the lack of deep-genome sequencing data available to previous studies.Here,w e build a list of putative adaptive variants,including 63 missense,7 loss-of-function;1,298 evolutionarily conserved variants and 509 expression quantitative traits loci.Notably,the top signal of selection is located in TMEM 247,a transmembrane protein-coding gene.The Tibetan version of TMEM 247 harbors one high-frequency(76.3%)missense variant,rsl 16983452(c.248C>T;p.Ala83Val);with the T allele derived from archaic ancestry and carried by>94%of Tibetans but absent or in low frequencies(<3%)in non-Tibetan populations.The rsl 16983452-T is strongly and positively correlated with altitude and significantly associated with reduced hemoglobin concentration(p=5.78×10^-5),red blood cell count(p=5.72×10^-7)and hematocrit(p=2.57×10^-6).In particular,TMEM247-rs 116983452 shows greater effect size and better predicts the phenotypic outcome than any E P A S1 variants in association with adaptive traits in Tibetans.Modeling the interaction between TM£M247-rsl 16983452 and EPAS1 variants indicates weak but statistically significant epistatic effects.Our results support that multiple variants may jointly deliver the fitness of the Tibetans on the plateau,where a complex model is needed to elucidate the adaptive evolution mechanism. | Lian Deng Chao Zhang Kai Yuan Yang Gao Yuwen Pan Xueling Ge Yaoxi He Yuan Yuan Yan Lu Xiaoxi Zhang Hao Chen Haiyi Lou Xiaoji Wang Dongsheng Lu Jiaojiao Liu Lei Tian Qidi Feng Asifullah Khan Yajun Yang Zi-Bing Jin Jian Yang Fan Lu Jia Qu Longli Kang Bing Su Shuhua Xu | 2019 | National Science Review2019,6,6: | 9 |
| 2 | Tip-assisted ambient electric arc ionization mass spectrometry for rapid detection of trace organophosphorus pesticides in strawberry显示文摘In this study,an ambient mass spectrometry(AMS)based method was developed for rapid detection of organophosphorus pesticides in strawberry.This method combines an electric arc and a microsyringe tip to realize tip-assisted ambient electric arc ionization(TAAEAI).A high-voltage electric arc can be applied to the microsyringe tip to generate an electric field,which results in corona discharge at the microsyringe tip.The juiced strawberry sample loaded on the tip could be directly ionized with TAAEAI and then analyzed by a mass spectrometer.TAAEAI-MS was successfully applied to analyze 6 organophosphorus pesticides in three strawberry samples.Malathion and profenofos were detected from the investigated strawberry samples.This method could quantitatively determine the contents of organophosphorus pesticides in strawberry with high reproducibility,high precision,and high sensitivity.Sample matrices did not interfere with the pesticide analysis.The recoveries of organophosphorus pesticides spiked in strawberry samples varied between 82.6%and 116%with relative standard deviations(RSDs)less than 9.2%.The limits of detection(LODs)varied between 0.0124µg/g and 0.0245µg/g,while the limits of quantification(LOQs)varied between 0.0413µg/g and 0.0817µg/g.The coefficients of determination(R2)of the method were determined to be>0.995.The method established here may have potential application in the detection of organophosphorus pesticides in vegetables and fruits. | Zihan Ma Yuanji Gao Fengjian Chu Yunli Tong Yuwen He Yuan Li Zhan Gao Weiwei Chen Shuheng Zhang Yuanjiang Pan | 2022 | Chinese Chemical Letters2022,33,9: | 2 |

| 6 | Genomic diversity and post-admixture adaptation in the Uyghurs显示文摘Population admixture results in genome-wide combinations of genetic variants derived from different ancestral populations of distinct ancestry, thus providing a unique opportunity for understanding the genetic determinants of phenotypic variation in humans. Here, we used whole-genome sequencing of 92 individuals with high coverage(30–60×) to systematically investigate genomic diversity in the Uyghurs living in Xinjiang, China(XJU), an admixed population of both European-like and East-Asian-like ancestry.The XJU population shows greater genetic diversity, especially a higher proportion of rare variants,compared with their ancestral source populations, corresponding to greater phenotypic diversity of XJU.Admixture-induced functional variants in EDAR were associated with the diversity of facial morphology in XJU. Interestingly, the interaction of functional variants between SLC24A5 and OCA2 likely influences the diversity of skin pigmentation. Notably, selection has seemingly been relaxed or canceled in several genes with significantly biased ancestry, such as HERC2–OCA2. Moreover, signatures of post-admixture adaptation in XJU were identified, including genes related to metabolism(e.g. CYP2D6), digestion(e.g.COL11A1), olfactory perception(e.g. ANO2) and immunity(e.g. HLA). | 8 | Strategies for improving adipose-derived stem cells for tissue regeneration显示文摘Adipose-derived stem cells(ADSCs)have promising applications in tissue regeneration.Currently,there are only a few ADSC products that have been approved for clinical use.The clinical application of ADSCs still faces many challenges.Here,we review emerging strategies to improve the therapeutic efficacy of ADSCs in tissue regeneration.First,a great quantity of cells is often needed for the stem cell therapies,which requires the advanced cell expansion technologies.In addition cell-derived products are also required for the development of‘cell-free’therapies to overcome the drawbacks of cell-based therapies.Second,it is necessary to strengthen the regenerative functions of ADSCs,including viability,differentiation and paracrine ability,for the tissue repair and regeneration required for different physiological and pathophysiological conditions.Third,poor delivery efficiency also restricts the therapeutic effect of ADSCs.Effective methods to improve cell delivery include alleviating harsh microenvironments,enhancing targeting ability and prolonging cell retention.Moreover,we also point out some critical issues about the sources,effectiveness and safety of ADSCs.With these advanced strategies to improve the therapeutic efficacy of ADSCs,ADSC-based treatment holds great promise for clinical applications in tissue regeneration. | Xin Yuan Li Li Haofan Liu Jing Luo Yongchao Zhao Cheng Pan Xue Zhang Yuwen Chen Maling Gou | 2022 | Burns & Trauma2022,10,1: | 1 |

| 10 | Simulation of the water level influence on the difference within the water-tube tiltmeter in Shuangyang Lake显示文摘This article analyzes the relationship between the water level and the water-tube tilting in Shuangyang lake,based on the differential deformation features reflected by the NS and EW components of the water-tube tiltmeter.The results show a good spatiotemporal consistency between the variation of water level and the NS tilt component,which is considered to be affected by the magnitude and duration of the water level variation in Shuangyang Lake.The article uses Landsat remote sensing image data to extract the water boundary of Shuan-gyang Lake,and takes advantage of the finite element numerical simulation method to build three-dimensional models for different geological structural conditions of the Shuangyang seismostation.The simulation results show that when the underground medium is granite,the effect of water level variation on the vertical displacement of the surface is non-directional.With a 50-m soil layer in Model 2,the simulated NS tilt variation is equivalent to the actual observed water-tube tiltmeter NS component when the water level variation is 0.44 m and 0.8m.When the variation of water level reaches 2.0m,the simulation result of the NS component is 79.6 ms,which is slightly larger than the observed result of 60.32 ms.However,the simulation results show that the variation of the EW component is significantly smaller than that of the NS one.Due to the fact that the Shuan-gyang lake is long in the NS direction and short in the EW direction,the existence of the soil layer tends to generate ground deformation along the NS direction in the vicinity of the lake after the increase of water level,thereby resulting in the difference of the ground deformation in the two directions. | Chong Yue Chuncheng Tang Wei Yan Xiaodong Pan Xueme Li Yuwen Tan Zongfeng Zhang Tianlong Yu | 2021 | Earthquake Research Advances2021,1,2: | 1 |

| 15 | Lineage-specific positive selection on ACE2 contributes to the genetic susceptibility of COVID-19显示文摘The Angiotensin-Converting Enzyme-2(ACE2)gene,located on Xp22.2,attracts a great deal of attention because the protein it encodes is believed to be the functional cellular receptor for the new coronavirus(SARS-CoV-2).However,recent studies are controversial,especially concerning the intrinsic link between ACE2 diversity and COVID-19 susceptibility.Here,we conduct a population genetic study on ACE2 in6354 individuals representing 210 present-day populations and 5329 individuals of ancient or archaic groups.We dissected the genetic architecture ofACE2 and identified two major haplogroups(hg)in East Asians,i.e.ACE2-hgl(43%)and ACE2-hg2(53%),while other populations harbor more diverse A CE2-hgs.Accordingly,there was a significant loss of ACE2 common variations in East Asians in contrast to the X-chromosome-wide and genome-wide patterns.Notably,association analysis between ACE2-hgs and COVID-19 severity in 1229 Han Chinese individuals with various levels of COVID-19 severity showed a higher risk ofACE2-hgl(odds ratio=1.56,P<0.01)and a lower risk of ACE2-hg2(odds ratio=0.65,P<0.01).Interestingly,ACE2-hgl is in strong linkage disequilibrium with rs1849863-C,which is an assumed risk factor of elevated plasma ACE2 level and is related to a higher risk of COVID-19 severity,hospitalization and infection.Strikingly,remarkable signatures of positive selection were detected,especially on ACE2-hg2,and were traced back to 100000 years ago(but rose to a strong level during the Bronze Age,5000~3000 years ago,in East Asians).The selection pressures could have stemmed from multiple sources,but pre-COVID-19 viral epidemics and pandemics might have been potential driving forces,which consequently contributed to the genetic susceptibility to COVID-19 within and between populations. | Yuwen Pan Panhong Liu Fang Wang Peng Wu Fanjun Cheng Xin Jin Shuhua Xu | 2022 | National Science Review2022,9,9: | 0 |
