1Geothermal data analysis at the high-temperature hydrothermal area in Western Sichuan显示文摘The western Sichuan hydrothermal area is located at the northeastern margin of the eastern syntaxis of the Qinghai-Tibet Plateau, which is also the eastern end of the Mediterranean-Himalayan geothermal activity zone. There are 248 warm or hot springs in this area, and 11 have temperatures beyond the local boiling temperature. Most of these hot springs are distributed along the Jinshajiang, Dege-Xiangcheng, Ganzi-Litang, and Xianshuihe faults, forming a NW-SE hydrothermal belt. A geothermal analysis of this high-temperature hydrothermal area is an important basis for understanding the deep geodynamic process of the eastern syntaxis of the Qinghai-Tibet Plateau. In addition, this study offers an a priori view to utilize geothermal resources, which is important in both scientific research and application. We use gravity, magnetic, seismic, and helium isotope data to analyze the crust-mantle heat flow ratio and deep geothermal structure. The results show that the background terrestrial heat flow descends from southwest to northeast. The crustal heat ratio is not more than 60%. The high temperature hydrothermal active is related to crustal dynamics processes. Along the Batang-Litang-Kangding line, the Moho depth increases eastward, which is consistent with the changing Qc/Qm(crustal/mantle heat flow) ratio trend. The geoid in the hydrothermal zone is 4–6 km higher than the surroundings, forming a local 'platform'. The NW-SE striking local tensile stress zone and uplift structure in the upper and middle crust corresponds with the surface hydrothermal active zone. There is an average Curie Point Depth(CPD) of 19.5–22.5 km in Batang, Litang, and Kangding. The local shear-wave(S-wave) velocity is relatively low in the middle and lower crust. The S-wave shows a low velocity trap(Vs<3.2 km s.1) at 15–30 km, which is considered a high-temperature partial melting magma, the crustal source of the hydrothermal active zone. We conclude that the hydrothermal system in this area can be divided into Batang-type and Kangding-type, both of which rely on a crustal heating cycle of atmospheric precipitation and surface water along the fracture zone. 3Effect of TiO_2 on Crystallization of the Glass Ceramics Prepared from Granite Tailings显示文摘The effect of TiO2 on the crystallization behaviors of the glass ceramics prepared from granite tailings was investigated by differential scanning calorimetry(DSC), X-ray diffraction(XRD), and field emission scanning electron microscopy(FESEM). The results showed that the crystallization peak temperature decreased firstly, and then increased with the increase of TiO2 content. The optimum addition amount of TiO2 was 8 wt%. With a single-step heat treatment at 924 ℃ for 1 h, augite precipitated as the only crystalline phase both on the surface and in the interior. The avrami parameter of the sample was 3.25, suggesting a twodimensional crystallization mechanism. 4Responses of vegetation cover to the Grain for Green Program and their driving forces in the He-Long region of the middle reaches of the Yellow River显示文摘The implementation of the Grain for Green Program is a great breakthrough in the history of China's ecological environment construction,which can control soil erosion effectively,increase land productivity and improve the ecological environment.To investigate the eco-environmental benefits brought by the Grain for Green Program,the spatiotemporal variations of vegetation cover in the growing season from 2000 to 2010 across the Hekou-Longmen(He-Long) region were analyzed by using remote sensing information,meteorological data and land use data.Moreover,the impacts of climate and human activities on vegetation change were evaluated objectively.Annual vegetation cover in the growing season increased very significantly.Increased vegetation cover occurred in 98.7% of the region,of which the area for vegetation cover improved slightly constituted 79.8% of the whole area.Vegetation moderately improved was mainly distributed in the south of the He-Long region,covering 9.6% of the area,and the area for vegetation basically unchanged concentrated in the middle and upper reaches of the Wuding River.Precipitation was found to be an important natural factor influencing vegetation cover change.The area of vegetation cover showing a significantly positive correlation with precipitation occupied 22.14% of the region.As driven by policies from the Grain for Green Program,forestland increased significantly and land use structure became more intensive.Human activities played a positive and effective role in the protection,restoration and improvement of vegetation in the places where vegetation cover was basically unchanged,even though precipitation declined greatly,and vegetation improved moderately with massive increases of forestland and grassland.JianXiang LIU ZhiGuang LI XiaoPing ZHANG Rui LI XianChun LIU HanYang ZHANG 2013Journal of Arid Land2013,5,4:7
5Inclusion of the Eastern Asia endemic genus Sorolepidium in Polystichum (Dryopteridaceae):Evidence from the chloroplast rbcL gene and morphological characteristics显示文摘The taxonomic status of the Eastern Asia endemic Sorolepidium is controversial. Some authors accept it as member of the large diverse genus Polystichum, whereas others suggest that it is an independent genus separated from the later by the exindusiate sorus and the absence of aristate teeth at the pinnae margins. Here we infer phylogenetic relationship of Sorolepidium using DNA sequences of the chloro-plast rbcL gene. Phylogenies were inferred using maximum-parsimony, maximum-likelihood, and Bayesian inference methods. Molecular data establish that Sorolepidium is deeply nested within the large genus Polystichum and has a close relationship with P. duthiei and P. lachenense in the model-based analyses. The Kimura 2-parameter distances of the rbcL sequences between S. glaciale and P. duthiei and P. lachenense were 0.1 and 0.2%, respectively. Furthermore, S. glaciale differed from P. duthiei by a single nucleotide in their rbcL sequences. 6Endothelial cells-targeted soluble human Delta-like 4 suppresses both physiological and pathological ocular angiogenesis显示文摘Due to its essential roles in angiogenesis, Notch pathway has emerged as an attractive target for the treatment of pathologic angiogenesis. Although both activation and blockage of Notch signal can impede angiogenesis, activation of Notch signal may be more promising because it was shown that long-term Notch signal blockage resulted in vessel neoplasm. However, an in vivo deliverable Notch ligand with highly efficient Notch-activating capacity has not been developed. Among all the Notch ligands, Delta-like4(Dll4) is specifically involved in angiogenesis. In this study, we generated a novel soluble Notch ligand h D4 R, which consists of the Delta-Serrate-Lag-2 fragment of human Dll4 and an arginine-glycine-aspartate(RGD) motif targeting endothelial cells(ECs). We demonstrated that h D4 R could bind to ECs through its RGD motif and effectively triggered Notch signaling in ECs. Further, we confirmed that h D4 R could suppress angiogenesis in vitro as manifested by network formation assay and sprouting assay. More importantly, h D4 R efficiently repressed neonatal retinal angiogenesis and laser-induced choroidal neovascularization(CNV) as well in vivo. 7Genome-wide association mapping of vitamins B1 and B2 in common wheat显示文摘Vitamin B is essential for maintaining normal life activities in humans and animals who have to intake the microelement from the outside, especially from cereal products. In the present study 166 Chinese and foreign wheat cultivars planted in two environments were characterized for variation in vitamin B1 and B2 contents. A genome-wide association study(GWAS) using the wheat 90 K SNP assay identified 17 loci for vitamin B1 and 7 for vitamin B2 contents. Linear regression analysis showed a significantly positive correlation of the number of favorable alleles with vitamin B1 and B2 contents. Marker-trait associations(MTAs) at IWB43809(6AS, 0cM) and IWB69903(6AS, 13cM) were new and stable, and significantly associated with vitamin B1 content across two environments. 8Morphological and physiological analysis of narrow and striped leaf 1(nsl1) mutant of rice(Oryza sativa L.) and the gene mapping显示文摘The shape and color of rice leaves are important agronomic traits that directly influence the proportion of sunlight energy utilization and ultimately affect the yield and quality.A new mutant exhibiting stable inheritance was identified as derived from ethyl methane sulfonate(EMS)-treated restorer Jinhui 10,tentatively named as narrow and striped leaf 1(nsl1).The nsl1 displayed pale white leaves at the seeding stage and then white striped leaves in parallel to the main vein at the jointing stage.Meanwhile,its leaf blades are significantly narrower than the control group of Jinhui 10.The chloroplast structures of cells in the white striped area of the nsl1 mutant break down,and the photosynthetic pigments are significantly lower than that of the wild type.Moreover,fluorescence parameters,such as F0,F v/F m,U psII,qp,and ETR,in the nsl1 mutant are significantly lower than those of the wild type,and the photosynthetic efficiency is also significantly decreased.These changes in leaf color and shape,together with physiological changes in the nsl1,result in smaller plant height and a decrease in the most important agronomic traits,such as the number of grains per panicle,grain weight,etc.Genetic analysis shows that the narrow and striped traits of the nsl1 mutant are controlled by a single recessive nuclear gene,which is located between InDel 16and InDel 12 in chromosome 3.The physical distance is204 kb.So far,no similar genes of such leaf color and shape in this area have been reported.This study has laid a solid foundation for the gene cloning and function analysis of NSL1.Fangming Zhao Yong Zhang Yanru Wu Tongming Wang Ling Ma Zhenglin Yang Xia Wei Xianchun Sang Yinghua Ling Nan Wang Changwei Zhang Guanghua He 2014Chinese Science Bulletin2014,59,9:3
9A novel akermanite/poly (lactic-co-glycolic acid) porous composite scaffold fabricated via a solvent casting-particulate leaching method improved by solvent self-proliferating process显示文摘Desirable scaffolds for tissue engineering should be biodegradable carriers to supply suitablemicroenvironments mimicked the extracellular matrices for desired cellular interactions and to providesupports for the formation of new tissues. In this work, a kind of slightly soluble bioactiveceramic akermanite (AKT) powders were aboratively selected and introduced in the PLGA matrix,a novel L-lactide modified AKT/poly (lactic-co-glycolic acid) (m-AKT/PLGA) composite scaffold wasfabricated via a solvent casting-particulate leaching method improved by solvent self-proliferatingprocess. The effects of m-AKT contents on properties of composite scaffolds and on MC3T3-E1 cellularbehaviors in vitro have been primarily investigated. The fabricated scaffolds exhibited threedimensionalporous networks, in which homogenously distributed cavities in size of 300–400 lmwere interconnected by some smaller holes in a size of 100–200 lm. Meanwhile, the mechanicalstructure of scaffolds was reinforced by the introduction of m-AKT. Moreover, alkaline ionic productsreleased by m-AKT could neutralize the acidic degradation products of PLGA, and the apatitemineralizationability of scaffolds could be largely improved. 1040Ar-39Ar Geochronology and Tectonic Implications of the Blueschist from Northwestern Qiangtang, Northern Tibet, Western China显示文摘Blueschist exposed in the northwestern Qiangtang terrane, northern Tibet, western China (~84°30′E, 34°24′N), provides new constraints on the tectonic evolution of Qiangtang as well as northern Tibet. The blueschist represented by lawsonite-and glaucophane-bearing assemblages equilibrated at 375-400 °C and ~11 kbar. 40Ar-39Ar analysis on mineral separate from one blueschist sample yielded a well-defined plateau age of ~242 Ma. Geochemical studies show the blueschist is metamorphosed within-plate basalts. 11Identification of QTL for adult-plant resistance to powdery mildew in Chinese wheat landrace Pingyuan 50显示文摘Powdery mildew caused by Blumeria graminis f. sp. tritici is one of the major wheat diseases worldwide. The Chinese wheat landrace Pingyuan 50 has shown adult-plant resistance(APR)to powdery mildew in the field for over 60 years. To dissect the genetic basis of APR to powdery mildew in this cultivar, a mapping population of 137 double haploid(DH) lines derived from Pingyuan 50/Mingxian 169 was evaluated in replicated field trials for two years in Beijing(2009–2010 and 2010–2011) and one year in Anyang(2009–2010). A total of 540 polymorphic SSR markers were genotyped on the entire population for construction of a linkage map and QTL analysis. Three QTL were mapped on chromosomes 2BS(QPm.caas-2BS.2), 3BS(QPm.caas-3BS),and 5AL(QPm.caas-5AL) with the resistance alleles contributed by Pingyuan 50 explaining 5.3%,10.2%, and 9.1% of the phenotypic variances, respectively, and one QTL on chromosome 3BL(QPm.caas-3BL) derived from Mingxian 169 accounting for 18.1% of the phenotypic variance.QPm.caas-3BS, QPm.caas-3BL, and QPm.caas-5AL appear to be new powdery mildew APR loci.QPm.caas-2BS.2 and QPm.caas-5AL are possibly pleiotropic or closely linked resistance loci to stripe rust resistance QTL. 12QTL mapping of starch granule size in common wheat using recombinant inbred lines derived from a PH82-2/Neixiang188 cross显示文摘Starch is a crucial component determining the processing quality of wheat(Triticum aestivum L.)-based products. Wheat starch generally contains A-type and B-type starch granules, having different effects on starch properties and end-use qualities. In the present study, 240 recombinant inbred lines(RILs) derived from a PH82-2/Neixiang 188 cross were grown in Anyang, Henan, China, during three cropping seasons. A-type and B-type granule contents were determined using a laser diffraction particle size analyzer, defined as the percentage of total starch volume. A total of 195 SSR and STS markers were used to construct a genetic map. QTL analysis was performed by composite interval mapping. Three QTL for A-type starch granule content were mapped on chromosomes 1DL, 7BL and 4AL, explaining5.6%, 5.2% and 3.8% of the phenotypic variation, respectively. 13A dual-luciferase reporter system for characterization of small RNA target genes in both mammalian and insect cells显示文摘MicroRNAs(miRNAs)are regulatory RNA molecules that bind to target messenger RNAs(mRNAs)and affect the stability or translational efficiency of the bound mRNAs.Single or dual-luciferase reporter systems have been successfully used to identify miRNA target genes in mammalian cells.These reporter systems,however,are not sensitive enough to verify miRNA-target gene relationships in insect cell lines because the promoters of the target luciferase(usually Renilla)used in these reporter systems are too weak to drive sufficient expression of the target luciferase in insect cells.In this study,we replaced the SV40 promoter in the psiCHECK-2 reporter vector,which is widely used with mammalian cell lines,with the HSV-TK or AC5.1 promoter to yield two new dual-luciferase reporter vectors,designated psiCHECK-2-TK and psiCHECK-2-AC5.1,respectively.Only psiCHECK-2 and psiCHECK-2-AC5.1 had suitable target(7?enz7/a)/reference(firefly)luciferase activity ratios in mammalian(HeLa and HEK293)and insect(Sf9,S2,Helicoverpa zea fat body and ovary)cell lines,while psiCHECK-2-TK had suitable Renilla/firefly luciferase activity ratios regardless of the cell line.Moreover,psiCHECK-2-TK successfully detected the interaction between Helicoverpa armigera miRNA9a and its target,the 3'-untranslated region of heat shock protein 90,in both mammalian and H.zea cell lines,but psiCHECK-2 failed to do so in IT.zea cell lines.Furthermore,psiCHECK-2-TK with the target sequence,HzMasc(H.zea Masculinizer),accurately differentiated between H.zea cell lines with or without the negative regulation factor(miRNA or piRNA)of HzMasc.These data demonstrate that psiCHECK-2-TK can be used to functionally characterize small RNA target genes in both mammalian and insect cells.Zhongyuan Deng Yuting Zhang Leyao Li Xingcheng Xie Jinyong Huang Min Zhang Xinzhi Ni Xianchun Li 2022Insect Science2022,29,3:2
14Dissecting conserved cis-regulatory modules of Glu-1 promoters which confer the highly active endosperm-specific expression via stable wheat transformation显示文摘Wheat high-molecular-weight glutenin subunits(HMW-GS) determine dough elasticity and play an essential role in processing quality. HMW-GS are encoded by Glu-1 genes and controlled primarily at transcriptional level, implemented through the interactions between cis-acting elements and trans-acting factors. However, transcriptional mechanism of Glu-1 genes remains elusive. Here we made a comprehensive analysis of cis-regulatory elements within 1-kb upstream of the Glu-1 start codon(-1000 to-1) and identified 30 conserved motifs. Based on motif distribution pattern, three conserved cis-regulatory modules(CCRMs), CCRM1(-300 to-101), CCRM2(-650 to-400), and CCRM3(-950 to-750), were defined, and their functions were characterized in wheat stable transgenic lines transformed with progressive 5′ deletion promoter::GUS fusion constructs. GUS staining, qP CR and enzyme activity assays indicated that CCRM2 and CCRM3 could enhance the expression level of Glu-1, whereas the 300-bp promoter(-300 to-1), spanning CCRM1 and core region(-100 to-1), was enough to ensure accurate Glu-1 initiation at 7 days after flowering(DAF) and shape its spatiotemporal expression pattern during seed development. Further transgenic assays demonstrated that CCRM1-2(-300 to-209) containing Complete HMW Enhancer(-246 to-209) was important for expression level but had no effect on expression specificity in the endosperm. In contrast, CCRM1-1(-208 to-101) was critical for both expression specificity and level of Glu-1. 15Partial anammox achieved in full scale biofilm process for typical domestic wastewater treatment显示文摘The slow initiation of anammox for treating typical domestic wastewater and the relatively high footprint of wastewater treatment infrastructures are major concerns for practical wastewater treatment systems.Herein,a 300 m^(3)/d hybrid biofilm reactor(HBR)process was developed and operated with a short hydraulic retention time(HRT)of 8 h.The analysis of the bacterial community demonstrated that anammox were enriched in the anoxic zone of the HBR process.The percentage abundance of Candidatus Brocadia in the total bacterial community of the anoxic zone increased from 0 at Day 1 to 0.33%at Day 130 and then to 2.89%at Day 213.Based upon the activity of anammox bacteria,the removal of ammonia nitrogen(NH_(4)^(+)-N)in the anoxic zone was approximately 15%.This showed that the nitrogen transformation pathway was enhanced in the HBR system through partial anammox process in the anoxic zone.The final effluent contained 12 mg/L chemical oxygen demand(COD),0.662 mg/L NH_(4)^(+)-N,7.2 mg/L total nitrogen(TN),and 6 mg/L SS,indicating the effectiveness of the HBR process for treating real domestic wastewater.Feng Hou Ting Zhang Yongzhen Peng Xiaoxin Cao Hongtao Pang Yanqing Shao Xianchun Lu Ju Yuan Xi Chen Jin Zhang 2022Frontiers of Environmental Science & Engineering2022,16,3:2
16QTL mapping of adult plant resistance to stripe rust and leaf rust in a Fuyu 3/Zhengzhou 5389 wheat population显示文摘Stripe or yellow rust(YR)and leaf rust(LR)cause large losses in wheat production worldwide.Resistant cultivars curtail the levels of losses.The present study aimed to identify quantitative trait loci(QTL)for YR and LR resistance in 147 F2:6 recombinant inbred lines(RIL)derived from the cross Fuyu 3/Zhengzhou 5389.The RIL population and parents were genotyped with the Wheat55 K single nucleotide polymorphism(SNP)array and simple sequence repeat(SSR)markers.All materials were also phenotyped for YR severity at Mianyang in Sichuan province and Baoding in Hebei province in the 2015/2016,2016/2017,and 2017/2018 cropping seasons,and LR severity at Zhoukou in Henan province and at Baoding in 2017/2018.Eleven QTL for YR resistance and five for LR resistance were detected using inclusive composite interval mapping(Ici Mapping).Four of these QTL on chromosomes 1 BL,2 BS,3 AL,and 5 AL conferred resistance to both YR and LR.The QTL on 1 BL was Lr46/Yr29,and that on 7 BL might be Lr68.The QTL on chromosome 2 BS was detected at a similar position to previously detected loci.QYr.hebau-3 AL/QLr.hebau-3 AL,QYr.hebau-5 AL/QLr.hebau-5 AL,QYr.hebau-7 DL,QYr.hebau-4 BS,QYr.hebau-6 DL,and QYr.hebau-2 AS are likely to be new.An SSR marker for QYr.hebau-7 DL was developed and validated in a diverse wheat panel from China,suggesting effectiveness in different genetic backgrounds.These QTL with closely linked SNP and SSR markers could be useful for marker-assisted selection in wheat breeding programs targeting durable resistance to both diseases.Takele Weldu Gebrewahid Peipei Zhang Yue Zhou Xiaocui Yan Xianchun Xia Zhonghu He Daqun Liu Zaifeng Li 2020The Crop Journal2020,8,4:2

18Evaluation of strategy for analyzing mouse liver plasma membrane proteome显示文摘Plasma membrane (PM) proteome is one of the major subproteomes present in the cell,and is very important in liver function. In the present work, C57 mouse liver PM was purified by density-gradient centrifugation. The purified PM was verified by electron microscope analysis and Western blotting. The results showed that the PM was enriched by more than 20-fold and the contamination of mitochondria was reduced by 2-fold compared with the homogenization fraction. Proteins were separated by 2DE and 1DE, trypsin-digested and submitted to ESI-Q-TOF and MALDI-TOF-TOF mass spectrometry or directly digested in solution and analyzed by LC-ESI ion trap mass spectrometry. In all, 547 non-redundant mouse liver PM proteins were identified, of which 34% contributed to plasma membrane or plasma membrane-related proteins.
