| 2 | Combination types between graphene oxide and substrate affect the antibacterial activity显示文摘Duo to their superior physicochemical properties,graphene and its derivatives(GDs),such as graphene oxide(GO)and reduced graphene oxide(rGO),have attracted extensive research interests around the world.In recent years,antibacterial activities of GDs have aroused wide concern and substantial works have been done.However,the underlying antibacterial mechanisms still remain controversial.Antibacterial activities of GDs vary with various factors,such as size,number of layers,oxygen-containing groups,and experimental surroundings.We assume that combination types between graphene oxide and substrate may affect the antibacterial activity.Therefore,in this work,GO was fixed on the titanium surface with three kinds of combination types including drop with gravitational effects(GO-D),electrostatic interaction(GO-APS)and electrophoretic deposition(GO-EPD),and the antibacterial activities in vitro were systematically investigated.Results showed that combination types affected the ability of GO for preventing Staphylococcus aureus(S.aureus)from gathering,sharpness of wrinkles or edges and reactive oxygen spices(ROS)levels.Once S.aureus are in the form of separation without aggregation,GO can effectively interact with them and kill them with sharp wrinkles or edges and high ROS levels.GOEPD could effectively prevent S.aureus from gathering,own sharp wrinkles or edges and could generate higher ROS levels.As a result,GO-EPD exhibited optimal antibacterial activity against S.aureus,followed by GO-APS and GO-D. | Jiajun Qiu Lu Liu Hongqin Zhu Xuanyong Liu | 2018 | Bioactive Materials2018,3,3: | 7 |
| 3 | Indications of stratospheric anomalies in the freezing rain and snow disaster in South China,2008显示文摘NCEP/NCAR reanalysis data were used to characterize stratospheric temperature and water-vapor anomalies before and after the freezing rain and snow disaster of South China in 2008,and the influence of stratospheric circulation anomalies on the troposphere.Stratospheric temperature and water-vapor anomalies provided good leading indicators of this weather event.The period from December 1st 2007 to February 28th 2008 was divided into 18 pentads.During the 6th pentad,temperature decreased significantly at 10 hPa in the near-polar stratospheric region,and the decreasing trend strengthened and extended downward and southward to middle and lower latitudes.During the 14th-18th pentads,the temperature decrease reached its maximum and extended to 30°N.This coincided with the widespread freezing rain and snow event.By the end of January 2008,the temperature decrease ended in the near-polar stratospheric region,but continued in the mid-latitude area of the troposphere as the freezing rain and snow weather persisted.Similar to the temperature variations,positive anomalies of relative humidity in the stratospheric near-polar region also strengthened and extended downward and southward,coinciding with the freezing rain and snow event.Along with the significant relationship between the freezing rain and snow disaster and stratospheric circulation anomalies,the stratospheric polar vortex changed its shape in late December,intensifying and spreading downward from the top of the stratosphere and southward to the Asian continent,resulting in a deepening of the East Asian Trough and a strengthening of meridional circulation.Before the occurrence of the freezing rain and snow event,temperature and vapor increases in the stratosphere transferred downward to the troposphere,along with a stratospheric flow in the near-polar region southward to lower latitudes. | CHEN QuanLiang LI Zhan FAN GuangZhou ZHU KeYun ZHANG Wen ZHU HongQin | 2011 | Science China Earth Sciences2011,54,8: | 5 |
| 4 | Culturing adequate CAR-T cells from less peripheral blood to treat B-cell malignancies显示文摘Objective:Chimeric antigen receptor-modified T(CAR-T)cells have shown impressive results against relapsed/refractory B cell malignancies.However,the traditional manufacture of CAR-T cells requires leukapheresis to isolate large amounts of peripheral blood T cells,thus making some patients ineligible for the procedure.Methods:We developed a simple method for CAR-T cell preparation requiring small volumes of peripheral blood.First,CD3+T cells isolated from 50 mL peripheral blood from patients(B-cell malignancies)were stimulated with immobilized anti-CD3/RetroNectin in 6-well plates and then transduced with CAR-expressing lentiviral vector.After 4 d,the T cells were transferred to culture bags for large-scale CAR-T cell expansion.In vitro and animal experiments were performed to evaluate the activity of the manufactured CAR-T cells.Finally,29 patients with B-cell acute lymphoblastic leukemia(B-ALL)and 9 patients with B-cell lymphoma were treated with the CAR-T cells.Results:The CAR-T cells were expanded to 1–3×10^(8) cells in 8–10 d and successfully killed B cell-derived malignant tumor cells in vitro and in vivo.For patients with B-ALL,the complete remission rate was 93%1 month after CAR-T cell infusion;after 12 months,the overall survival(OS)and leukemia-free survival rates were 69%and 31%,respectively.For patients with lymphoma,the objective response rate(including complete and partial remission)was 78%2 months after CAR-T cell infusion,and after 12 months,the OS and progression-free survival rates were 71%and 43%,respectively.Cytokine-release syndrome(CRS)occurred in 65.51%and 55.56%of patients with B-ALL and B-cell lymphoma,respectively;severe CRS developed in 20.69%of patients with B-ALL and in no patients with lymphoma.Conclusions:Our novel method can generate sufficient numbers of CAR-T cells for clinical use from 50–100 mL peripheral blood,thus providing an alternative means of CAR-T cell generation for patients ineligible for leukapheresis. | Lu Han Jian Zhou Linlin Li Keshu Zhou Lingdi Zhao Xinghu Zhu Qingsong Yin Yufu Li Hongqin You Jishuai Zhang Yongping Song Quanli Gao | 2021 | Cancer Biology & Medicine2021,18,4: | 2 |

| 10 | Cavitation erosion resistance of 316L stainless steel fabricated using selective laser melting显示文摘Cavitation erosion degrades the performance and reliability of hydraulic machinery.Selective laser melting(SLM)is a type of metal additive manufacturing technology that can fabricate metal parts directly and provide lightweight design in various industrial applications.However,the cavitation erosion behaviors of SLM-fabricated parts have rarely been studied.In this study,SLM 316L stainless steel samples were fabricated via SLM technology considering the scanning strategy,scanning speed,laser power,and build orientation.The effect of the process parameters on the cavitation erosion resistance of the SLM-fabricated 316L stainless steel samples was illustrated using an ultrasonic vibratory cavitation system.The mass loss and surface topography were employed to evaluate the surface cavitation damage of the SLM-fabricated 316L stainless steel samples after the cavitation test.The cavitation damage mechanism of the SLM-fabricated samples was discussed.The results show that the degree of cavitation damage of the sample fabricated via SLM with a few defects,anisotropic build direction,and columnar microstructure is significantly decreased.Defects such as pores,which are attributed to low laser power and high scanning speed,may severely aggravate the cavitation damage of the SLM-fabricated samples.The sample fabricated via SLM with a low laser power and exposure time exhibited the highest porosity and poor cavitation erosion resistance.The cellular structures are more prone to cavitation damage compared with the columnar structures.A sample with a high density of grain boundaries will severely suffer cavitation damage. | Hongqin DING Qing TANG Yi ZHU Chao ZHANG Huayong YANG | 2021 | Friction2021,9,6: | 1 |
| 11 | Atherosis-associated lnc_000048 activates PKR to enhance STAT1-mediated polarization of THP-1 macrophages to M1 phenotype显示文摘Our previous study has demonstrated that lnc_000048 is upregulated in large-artery atherosclerotic stroke and promotes atherosclerosis in ApoE^(-/-)mice.However,little is known about the role of lnc_000048 in classically activated macrophage(M1)polarization.In this study,we established THP-1-derived testing state macrophages(M0),M1 macrophages,and alternately activated macrophages(M2).Real-time fluorescence quantitative PCR was used to verify the expression of marker genes and the expression of lnc_000048 in macrophages.Flow cytometry was used to detect phenotypic proteins(CD11b,CD38,CD80).We generated cell lines with lentivirus-mediated upregulation or downregulation of lnc_000048.Flow cytometry,western blot,and real-time fluorescence quantitative PCR results showed that down-regulation of lnc_000048 reduced M1 macrophage polarization and the inflammation response,while over-expression of lnc_000048 led to the opposite effect.Western blot results indicated that lnc_000048 enhanced the activation of the STAT1 pathway and mediated the M1 macrophage polarization.Moreover,catRAPID prediction,RNA-pull down,and mass spectrometry were used to identify and screen the protein kinase RNA-activated(PKR),then catRAPID and RPIseq were used to predict the binding ability of lnc_000048 to PKR.Immunofluorescence(IF)-RNA fluorescence in situ hybridization(FISH)double labeling was performed to verify the subcellular colocalization of lnc_000048 and PKR in the cytoplasm of M1 macrophage.We speculate that lnc_000048 may form stem-loop structure-specific binding and activate PKR by inducing its phosphorylation,leading to activation of STAT1 phosphorylation and thereby enhancing STAT1 pathway-mediated polarization of THP-1 macrophages to M1 and inflammatory factor expression.Taken together,these results reveal that the lnc_000048/PKR/STAT1 axis plays a crucial role in the polarization of M1 macrophages and may be a novel therapeutic target for atherosclerosis alleviation in stroke. | Yuanyuan Ding Yu Sun Hongyan Wang Hongqin Zhao Ruihua Yin Meng Zhang Xudong Pan Xiaoyan Zhu | 2024 | Neural Regeneration Research2024,19,11: | 0 |
