| 1 | The combination of PD-1 blockade with interferon-αhas a synergistic effect on hepatocellular carcinoma显示文摘Background The efficacy of immune checkpoint inhibitors(ICIs),such as programmed cell death protein-1(PD-1)or its ligand 1(PD-L1)antibody,in hepatocellular carcinoma(HCC)is limited,and it is recommended that they be combined with other therapies.We evaluated the combination of pegylated interferon-α(Peg-IFNα)with PD-1 blockade in HCC mouse models.Methods We analyzed the effects of Peg-IFNαon tumor-infiltrating immune cells and PD-1 expression in the HCC immune microenvironment and examined the underlying mechanism of its unique effect on the PD-1 pathway.The in vivo efficacy of anti-PD-1 and Peg-IFNαwas evaluated in both subcutaneous and orthotopic mouse models of HCC.Results The combination of Peg-IFNαwith PD-1 blockade dramatically enhanced T-cell infiltration,improved the efficacy of PD-1 antibody and prolonged mouse survival compared with PD-1 antibody monotherapy.Mechanistically,Peg-IFNαcould recruit cytotoxic CD8+T cells to infiltrate the HCC microenvironment by inducing tumor cells to secrete the chemokine CCL4.Nevertheless,the HCC microenvironment quickly overcame the immune responses by upregulating PD-1 expression in CD8+T cells via the IFNα-IFNAR1-JAK1-STAT3 signaling pathway.The combination of PD-1 blockade with Peg-IFNαcould restore the cytotoxic capacity of CD8+T cells and exerted a significant synergistic effect on HCC.Conclusion These results indicate that in addition to initiating the antitumor immune response itself,Peg-IFNαcan also generate a microenvironment favoring PD-1 blockade.Thus,the combination of Peg-IFNαand PD-1 blockade can be a promising strategy for HCC. | Ying Zhu Mo Chen Da Xu Tian-En Li Ze Zhang Jian-Hua Li Xiang-Yu Wang Xin Yang Lu Lu Hu-Liang Jia Qiong-Zhu Dong Lun-Xiu Qin | 2022 | Cellular & Molecular Immunology2022,19,6: | 6 |

| 4 | Bimodality and growth of the spectra of typhoon-generated waves in northern South China Sea显示文摘Buoy-based observations of wave spectra during the passage of three typhoons in the northern South China Sea are examined.Though most spectra of mature typhoon-generated waves are unimodal,double-peaked spectra account for a significant proportion during the growing and decaying stages.This is due either to the superposition of swells on local wind waves or to the mechanism of nonlinear interaction between different wave components.The growth rate of energy density is an effective way to predict spectrum variation.The dominant wave direction depends on the location of the typhoon center to the site,but the direction spread shows no regularity in distant regions.In this study,a new six-parameter spectral formula is proposed to represent doublepeaked spectra and is shown to provide a better fit than previous models.The theoretical relationship between shape parameter and spectral width is still applicable to each peak.The characteristics of the variations of spectral parameters are analyzed.It is demonstrated that the spectral parameters are not only related to the typhoon intensity and typhoon track,but also have strong intercorrelations.Moreover,the growth relation between significant wave height and significant wave period is obtained to fit the typhoon-generated waves. | Dongxue Mo Yahao Liu Yijun Hou Ze Liu | 2019 | Acta Oceanologica Sinica2019,38,11: | 1 |
| 5 | Flexural and longitudinal shear performance of precast lightweight steel-ultra-high performance concrete composite beam显示文摘In this study,the flexural and longitudinal shear performances of two types of precast lightweight steel–ultra-high performance concrete(UHPC)composite beams are investigated,where a cluster UHPC slab(CUS)and a normal UHPC slab(NUS)are connected to a steel beam using headed studs through discontinuous shear pockets and full-length shear pockets,respectively.Results show that the longitudinal shear force of the CUS is greater than that of the NUS,whereas the interfacial slip of the former is smaller.Owing to its better integrity,the CUS exhibits greater flexural stiffness and a higher ultimate bearing capacity than the NUS.To further optimize the design parameters of the CUS,a parametric study is conducted to investigate their effects on the flexural and longitudinal shear performances.The square shear pocket is shown to be more applicable for the CUS,as the optimal spacing between two shear pockets is 650 mm.Moreover,a design method for transverse reinforcement is proposed;the transverse reinforcement is used to withstand the splitting force caused by studs in the shear pocket and prevent the UHPC slab from cracking.According to calculation results,the transverse reinforcement can be canceled when the compressive strength of UHPC is 150 MPa and the volume fraction of steel fiber exceeds 2.0%. | Ze MO Jiangrui QIU Hanbin XU Lanlan XU Yuqing HU | 2023 | Frontiers of Structural and Civil Engineering2023,17,5: | 0 |
