1Natural selection maintains the transcribed LTR retrotransposons in Nosema bombycis显示文摘Eight intact LTR retrotransposons(Nbr1?Nbr8)have been previously characterized from the genome of Nosema bombycis,a eu-karyotic parasite with a compact and reduced genome.Here we describe six novel transcribed Nbr elements(Nbr9?Nbr14)identified through either cDNA library or RT-PCR.Like previously determined ones,all of them belong to the Ty3/Gypsy superfamily.Retrotransposon diversity and incomplete domains with insertions(Nbr12),deletions(Nbr11)and in-frame stop codons in coding regions(Nbr9)were detected,suggesting that both defective and loss events of LTR retrotransposon have happened in N.bombycis genome.Analysis of selection showed that strong purifying selection acts on all elements except Nbr11.This implies that selective pressure keeps both these Nbrs and their functions in genome.Interestingly,Nbr11 is under positive selection and some positively selected codons were identified,indicating that new functionality might have evolved in the Nbr11 retrotransposon.Unlike other transposable elements,Nbr11 has integrated into a conserved syntenic block and probably resulted in the inversion of both flanking regions.This demonstrates that transposable element is an important factor for the reshuffling and evolution of their host genomes,and may be maintained under natural selection.Heng Xiang Guoqing Pan Ruizhi Zhang Jinshan Xu Tian Li Wenle Li Zeyang Zhou Zhonghuai Xiang 2010Journal of Genetics and Genomics2010,37,5:7
2Effects of atmospheric pressure fluctuations on hill-side coal fires and surface anomalies显示文摘This paper presents numerical studies on the effects of atmospheric pressure fluctuations on hill-side coal fires and their surface anomalies. Based on the single-particle reaction–diffusion model, a formula to estimate oxygen consumption rate at high temperature controlled by oxygen transport is proposed.Daily fluctuant atmospheric pressure was imposed on boundaries, including the abandoned gallery and cracks. Simulated results show that the effects of atmospheric pressure fluctuations on coal fires and surface anomalies depend on two factors: the fluctuant amplitude and the pressure difference between inlet(s) and outlet(s) of the air ventilation system. If the pressure difference is close to the fluctuant amplitude, atmospheric pressure fluctuations greatly enhance gas flow motion and temperatures of the combustion zone and outtake(s). 3Solution-processable precursor route for fabricating ultrathin silica film for high performance and low voltage organic transistors显示文摘Silica is one of the most commonly used materials for dielectric layer in organic thin-film transistors due to its excellent stability, excellent electrical properties, mature preparation process, and good compatibility with organic semiconductors. However, most of conventional preparation methods for silica film are generally performed at high temperature and/or high vacuum. In this paper, we introduce a simple solution spin-coating method to fabricate silica thin film from precursor route, which possesses a low leakage current, high capacitance, and low surface roughness. The silica thin film can be produced in the condition of low temperature and atmospheric environment. To meet various demands, the thickness of film can be adjusted by means of preparation conditions such as the speed of spin-coating and the concentration of solution. The p-type and n-type organic field effect transistors fabricated by using this film as gate electrodes exhibit excellent electrical performance including low voltage and high performance. 5BmSE, a SINE family with 3′ ends of (ATTT) repeats in domesticated silkworm (Bombyx mori)显示文摘Short interspersed elements (SINEs), which are mainly composed of Bm1, are abundant in the domesticated silkworm. A 294 bp novel SINE family, designated as BmSE, was identified by mining the database of the complete Bombyx mori genome. A representational BmSE element is flanked by an 11 bp target site duplication sequence posterior poly (A) at the 3′ end and has the sequence motifs of an internal promoter of RNA polymerase III, which are similar to that of Bm1. The repetitive elements of BmSE are widely distributed in all 28 chromosomes of the genome and share the common (ATTT) repeats at the ends. GC-content distribution shows that BmSE tends to accumulate preferably in the region of higher AT content than that of Bm1. A high proportion of the BmSEs are mapped to the coding sequence introns, whereas several elements are also present in the UTR of some transcripts, indicating that BmSEs are indeed exonized with UTRs. Of the 615 identified structural variants (SVs) of BmSE among the 40 domesticated and wild silkworms, only 230 SVs were found in the domesticated silkworms, indicating that many recent SV events of BmSE occurred after domestication, which was probably due to its mobilization. 6Advances in the origin of overpressures in sedimentary basins显示文摘Much progress in the studies on overpressuring mechanisms has been made during the past one to two decades.(1)The causes of overpressure are divided into five categories,namely,disequilibrium compaction,fluid expansion,diagenesis,tectonic compression and pressure transfer.The fluid expansion involves hydrocarbon generation,oil cracking to gas and hydrothermal expansion.The diagenesis includes smectite-to-illite transformation.(2)Six methods for identifying overpressure origin are proposed,including log curves combination analysis,Bowers method(loading-unloading diagram),velocitydensity crossplotting,correlation of porosities,pressure calculation and correlation,and comprehensive analyses.(3)With more and more application of empirical methods in the study of overpressure formation,almost all of the overpressure cases that are traditionally thought to be caused by disequilibrium compaction are denied totally or partly.Instead,the hydrocarbon generation is demonstrated to be the most significant mechanism for overpressure formation;the clay diagenesis(especially the smectiteillite transformation)as well as tectonic compression and pressure transfer are also important for overpressure formation.In addition,the overpressure formation in many basins is thought to be influenced by the combination of two or more overpressuring mechanisms.(4)Causes of overpressuring differ in lithology;for mudstones,the overpressure formation in source rocks is usually different from that of non-source rocks,the former of which is frequently related to hydrocarbon generation and sometimes also affected by diagenesis,while the later of which is commonly related to disequilibrium compaction,diagenesis and pressure transfer;for permeable rocks such as sandstones,overpressure is mainly caused by pressure transfer.(5)Because organic matter has an obvious influence on logging parameters such as density and acoustic velocity,an appropriate correction on the content of organic matter is needed when these logging data are used to analyze overpressure formation in organic-rich mudstones.It has been revealed that the cause of overpressuring based on the corrected log data can be quite different from that without correction.Jingzhou Zhao Jun Li Zeyang Xu 2018Petroleum Research2018,3,1:0
7Cloning of Immune System-related Gene ApTOLL1 and Its Expression in Nosema-infected Antheraea pernyi显示文摘[Objective]This study aimed to identify the immune system-related TOLL-like receptor family gene of Antherea pernyi,to lay the foundation for further investigating the immune mechanism of Antherea pernyi.[Method] Immune system-related TOLL-like receptor family gene of Antherea pernyi was cloned for sequencing and phylogenetic analysis;in addition,expression variations of TOLL-like receptor family gene in Antherea pernyi infected with Nosema pernyi and Nosema bombycis were detected to analyze the differences in immunological reactivity of Antherea pernyi to Nosema infection.[Result]Based on cDNA cloning and sequencing,an immune system-related gene fragment was isolated from Antherea pernyi,which is the most homologous to the Toll1 gene in Toll signaling pathway of Bombyx mori according to the sequencing result and phylogenetic analysis,which is named ApToll1 gene.Subsequently,Antherea pernyi pupae were injected respectively with Nosema bombycis and Nosema pernyi,fluorescent real-time quantitative PCR analysis showed that ApToll1 was abundantly expressed in Antherea pernyi pupa 2 h after Nosema bombycis injection,which began to express in Antherea pernyi pupa 11 h after Nosema pernyi injection,indicating that the immune response time induced by various Nosema strains varies in Toll signaling pathway of Antherea pernyi.[Conclusion]ApToll1 gene of Antherea pernyi was first cloned in this study,which provides reference for further investigating the immune mechanism of Antherea pernyi.Lijun WANG Jinshan XU Zeyang ZHOU 2012Agricultural Biotechnology2012,1,3:0
8Mitochondrial pyruvate dehydrogenase E1 of Nosema bombycis:A marker in Microsporidian evolution显示文摘Microsporidia are a group of intracelluar eukaryotic parasites,which can infected almost all animals,including human beings.Till now,no mitochodria but mitosome,a remnant of mitochondria was discovered in this phylum.We present here the mitochondrial pyruvate dehydrogenase E1(PDH,including PDHα and PDHβ)of the microsporidian Nosema bombycis,the pathogen of silkworm pébrine.Compared with PDH of microsporidian Encephalitozoon cuniculi and Antonospora locustae,both subunits are conserved.The phylogeny indicated that both subunits are mitochondrial.The syntenic maps revealed the subunits organization of NbPDH is distributed in different scaffolds,similar to that of EcPDH but different with AlPDH,and the relationship between phylogeny tree and organization of PDH suggest that the AlPDH subunits organization is the ancestral style of microsporidia,and through the genome evolution,the reshuffling of the chromosome of microsporidia occurred,the adjacent style of ALPDHE1 organization changed,and the two subunits separated and located to different chromosomes in E.cuniculi.For N.bombycis and N.ceranae,they locate to different scaffolds.In order to determine NbPDH subcellular localizations,we prepared the polyclonal antibodies against NbPDH prokaryotic fusion proteins,and adopted the colloidal gold immunological electron microscopy,the expression signals of NbPDH were observed in spores however,the subcellular localization were not definited.In general,through comparison of three microsporidian PDH molecular phylogeny,subunits organization in chromosomes,localization indicated that PDH is an interesting marker in microsporidia evolution.Tian LI Xiaoqun DANG Jinshan XU Handeng LIU Guoqing PAN Zeyang ZHOU 2009Current Zoology2009,55,6:0
