The role of furin cleavage site in SARS-CoV-2 spike protein-mediated membrane fusion in the presence or absence of trypsin显示文摘Dear Editor,The rapid spread of SARS-CoV-2(also known as 2019-nCoV and HCoV-191),a novel lineage B betacoronavirus(βCoV),has caused a global pandemic of coronavirus disease(COVID-19).It has been speculated that RRAR,a unique furin-like cleavage site(FCS)in the spike protein(S),which is absent in other lineage BβCoVs,such as SARS-CoV,is responsible for its high infectivity and transmissibility. | Shuai Xia Qiaoshuai Lan Shan Su Xinling Wang Wei Xu Zezhong Liu Yun Zhu Qian Wang Lu Lu Shibo Jiang | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 8
RBD-Fc-based COVID-19 vaccine candidate induces highly potent SARS-CoV-2 neutralizing antibody response显示文摘The pandemic of coronavirus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has posed serious threats to global health and economy, thus calling for the development of safe and effective vaccines. The receptorbinding domain(RBD)in the spike protein of SARS-CoV-2 is responsible for its binding to angiotensin-converting enzyme 2(ACE2)receptor.It contains multiple dominant neutralizing epitopes and serves as an important antigen for the development of COVID-19 vaccines. Here,we showed that immunization of mice with a candidate subunit vaccine consisting of SARS-CoV-2 RBD and Fc fragment of human IgG,as an immunopotentiator, elicited high titer of RBD-specific antibodies with robust neutralizing activity against both pseudotyped and live SARS-CoV-2 infections. The mouse antisera could also effectively neutralize infection by pseudotyped SARS-CoV-2 with several natural mutations in RBD and the IgG extracted from the mouse antisera could also show neutralization against pseudotyped SARS-CoV and SARS-related coronavirus(SARSr-CoV).Vaccination of human ACE2 transgenic mice with RBD-Fc could effectively protect mice from the SARS-CoV-2 challenge. These results suggest that SARS-CoV-2 RBD-Fc has good potential to be further developed as an effective and broad-spectrum vaccine to prevent infection of the current SARS-CoV-2 and its mutants,as well as future emerging SARSr-CoVs and re-emerging SARS-CoV. | Zezhong Liu Wei Xu Shuai Xia Chenjian Gu Xinling Wang Qian Wang Jie Zhou Yanling Wu Xia Cai Di Qu Tianlei Ying Youhua Xie Lu Lu Zhenghong Yuan Shibo Jiang | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 7

Altered Motor-Striatal Plasticity and Cortical Functioning in Patients with Schizophrenia显示文摘Patients with schizophrenia undergo changes in brain plasticity. In the present study, we characterized motor cortical-striatal plasticity in such patients. Compared with the potentiation following high-frequency repetitive transcranial magnetic stimulation in the control group, the patients demonstrated impaired plasticity of corticostriatal motor-evoked potentials recorded from hand muscles.Notably, the loss of cortical plasticity was correlated with impaired motor learning in a rotary pursuit task. Moreover, the loss of plasticity was correlated with the symptoms of schizophrenia. The results suggest that the progression of schizophrenia is accompanied by altered cortical plasticity and functioning. | Dongsheng Zhou Feng Pang Shiyan Liu Ying Shen Lingjiang Liu Zezhong Fang Chuang Wang Zhenyu Hu Ti-Fei Yuan | 2017 | Neuroscience Bulletin2017,33,3: | 5
A novel STING agonist-adjuvanted pan-sarbecovirus vaccine elicits potent and durable neutralizing antibody and T cell responses in mice, rabbits and NHPs显示文摘The emergence of SARS-CoV-2 variants and potentially other highly pathogenic sarbecoviruses in the future highlights the need for pan-sarbecovirus vaccines. Here,we discovered a new STING agonist,CF501,and found that CF501-adjuvanted RBD-FC vaccine(CF501/RBD-FC)elicited significantly stronger neutralizing antibody(nAb)and T cell responses than Alum-and cGAMP-adjuvanted RBD-FC in mice.Vaccination of rabbits and rhesus macaques(nonhuman primates,NHPs)with CF501/RBD-FC elicited exceptionally potent nAb responses against SARS-CoV-2 and its nine variants and 41 S-mutants,SARS-CoV and bat SARSr-CoVs.CF501/RBD-FC-immunized hACE2-transgenic mice were almost completely protected against SARS-CoV-2 challenge, even 6 months after the initial immunization.NHPs immunized with a single dose of CF501/RBD-FC produced high titers of nAbs.The immunized macaques also exhibited durable humoral and cellular immune responses and showed remarkably reduced viral load in the upper and lower airways upon SARS-CoV-2 challenge even at 108 days post the final immunization.Thus,CF501/RBD-Fc can be further developed as a novel pan-sarbecovirus vaccine to combat current and future outbreaks of sarbecovirus diseases. | Zezhong Liu Jie Zhou Wei Xu Wei Deng Yanqun Wang Meiyu Wang Qian Wang Ming Hsieh Jingming Dong Xinling Wang Weijin Huang Lixiao Xing Miaoling He Chunlin Tao Youhua Xie Yilong Zhang Youchun Wang Jincun Zhao Zhenghong Yuan Chuan Qin Shibo Jiang Lu Lu | 2022 | Cell Research2022,32,3: | 5
A highly potent and stable pan-coronavirus fusion inhibitor as a candidate prophylactic and therapeutic for COVID-19 and other coronavirus diseases显示文摘The development of broad-spectrum antivirals against human coronaviruses(HCoVs)is critical to combat the current coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and its variants,as well as future outbreaks of emerging CoVs.We have previously identified a polyethylene glycol-conjugated(PEGylated)lipopeptide,EK1C4,with potent pan-CoV fusion inhibitory activity. However,PEG linkers in peptide or protein drugs may reduce stability or induce anti-PEG antibodies in vivo. Therefore,we herein report the design and synthesis of a series of dePEGylated lipopeptide-based pan-CoV fusion inhibitors featuring the replacement of the PEG linker with amino acids in the heptad repeat 2 C-terminal fragment(HR2-CF)of HCoV-OC43.Among these lipopeptides,EKL1C showed the most potent inhibitory activity against infection by SARS-CoV-2 and its spike(S)mutants,as well as other HCoVs and some bat SARS-related coronaviruses(SARSr-CoVs)tested.The dePEGylated lipopeptide EKL1C exhibited significantly stronger resistance to proteolytic enzymes, better metabolic stability in mouse serum, higher thermostability than the PEGylated lipopeptide EK1C4,suggesting that EKL1C could be further developed as a candidate prophylactic and therapeutic for COVID-19 and other coronavirus diseases. | Jie Zhou Wei Xu Zezhong Liu Chao Wang Shuai Xia Qiaoshuai Lan Yanxing Cai Shan Su Jing Pu Lixiao Xing Youhua Xie Lu Lu Shibo Jiang Qian Wang | 2022 | Acta Pharmaceutica Sinica B2022,12,4: | 5
An automated approach to calculating the maximum diameters of multiple cutters and their paths for sectional milling of centrifugal impellers on a 4_(1/2)-axis CNC machine显示文摘Power generators and chemical engineering compressors include heavy and large centrifugal impellers. To produce these impellers in high-speed machining, a 4?-axis milling machine(or a 4-axis machine plus an indexing table) is often used in the industry, which is more rigid than a5-axis milling machine. Since impeller blades are designed with complex B-spline surfaces and impeller channels spaces vary significantly, it is more efficient to use multiple cutters as large as possible to cut a channel in sections and a blade surface in patches, instead of only using a small cutter to machine a whole blade and a channel. Unfortunately, no approach has been established to automatically calculate the largest diameters of cutters and their paths, which include the indexing table angles. To address this problem, an automated and optimization approach is proposed. Based on the structure of a 4?-axis machine, a geometric model for a cutter gouging/interfering the impeller is formulated, and an optimization model of the cutter diameter in terms of the indexing table angle is established at a cutter contact(CC) point on a blade surface. Then, the diameters of the tools, their orientations, and the indexing table angles are optimized, and each tool's paths are generated for machining its corresponding impeller section. As a test, an impeller is efficiently machined with these tools section by section; thus, this approach is valid. It can be directly used in the industry to improve efficiency of machining centrifugal impellers. | Baohai WU Zezhong C.CHEN Ming LUO Dinghua ZHANG Feiyan HAN | 2019 | Chinese Journal of Aeronautics2019,32,4: | 5

Petrology,Geochemistry, and Sr-Nd-S Isotopic Compositions of the Ore-Hosting Biotite Monzodiorite in the Luanjiahe Gold Deposit,Jiaodong Peninsula,China显示文摘The Jiaodong Peninsula is one of the most important Au ore provinces in China. There is an ongoing debate on the correlation between ore formation and magmatism in this province, because few intrusive rocks exhibit a clear association with ore deposits.A mineralized biotite monzodiorite(BM)stock, with disseminated ore, pervasive phyllic alteration, and no deformation, was found in a borehole in the footwall of the Zhaoping fault within the Luanjiahe Au deposit, which may shed light on this debate. The biotite monzodiorite contains explosion breccias, miarolitic cavities, skeletal and den-dritic quartz, and late-stage evolved aplite dikes, and the in-situδ34S values of the disseminated pyrite which is associated with Au mineralization are-1.7‰ to 7.3‰(mean=3.5‰),indicative of a magmatic-hydrothermal system. These findings, combined with the reported age of 123 Ma,show that the intru-sion has close spatial, temporal, and geochemical relationships with Au mineralization in the area. The biotite monzodiorite is metaluminous,high-K calc-alkaline and shoshonitic, with enrichment in light rare earth elements(REEs)and large-ion lithophile elements(LILEs),depletion in high-field-strength elements(HFSEs),and enriched Sr-Nd isotopic compositions. The intrusion may be the product of par-tial melting of enriched lithospheric mantle with a small lower crustal component. The hydrous,Au-bearing,enriched mantle source, and the strongly oxidized magma that was generated, created fa-vorable conditions for Au mineralization. | Xiaofeng Yao Zhizhong Cheng Zezhong Du Zhenshan Pang Yuquan Yang Kun Liu | 2021 | Journal of Earth Science2021,32,1: | 4
A pan-sarbecovirus vaccine induces highly potent and durable neutralizing antibody responses in non-human primates against SARS-CoV-2 Omicron variant显示文摘Dear Editor,Since the outbreak of Coronavirus Disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection,numerous SARS-CoV-2 variants of concern(VOCs),such as Alpha(B.1.1.7),Beta(B.1.351),Gamma(P.1),and Delta(B.1.617.2),have emerged.Recently, the newly identified SARS-CoV-2 VOC,Omicron(B.1.1.529),has rapidly spread in many countries1 and is expected to replace Delta as the dominant variant circulating in the world.Compared to other VOCs,Omicron contains at least 32 mutations in spike(S)protein, including 15 mutations in the receptor-binding domain(RBD),and these mutations are known to confer resistance to neutralizing antibodies(nAbs)and sera of convalescent patients and people who have received COVID-19 vaccines.2–5 Therefore, vaccines able to induce potent and durable neutralizing antibodies against Omicron and other VOCs are urgently needed. | Zezhong Liu Jasper Fuk-Woo Chan Jie Zhou Meiyu Wang Qian Wang Guangxu Zhang Wei Xu Kenn Ka-Heng Chik Yilong Zhang Youchun Wang Kwok-Yung Yuen Lu Lu Shibo Jiang | 2022 | Cell Research2022,32,5: | 4
What Is Semantic Communication?A View on Conveying Meaning in the Era of Machine Intelligence显示文摘In the 1940s,Claude Shannon developed the information theory focusing on quantifying the maximum data rate that can be supported by a communication channel.Guided by this fundamental work, the main theme of wireless system design up until the fifth generation(5G)was the data rate maximization. In Shannon’s theory, the semantic aspect and meaning of messages were treated as largely irrelevant to communication. The classic theory started to reveal its limitations in the modern era of machine intelligence, consisting of the synergy between Internet-of-things(IoT)and artificial intelligence(AI).By broadening the scope of the classic communication-theoretic framework,in this article,we present a view of semantic communication(SemCom)and conveying meaning through the communication systems. We address three communication modalities:human-to-human(H2H),human-to-machine(H2M),and machine-to-machine(M2M)communications.The latter two represent the paradigm shift in communication and computing, and define the main theme of this article.H2M SemCom refers to semantic techniques for conveying meanings understandable not only by humans but also by machines so that they can have interaction and“dialogue”.On the other hand,M2M SemCom refers to effective techniques for efficiently connecting multiple machines such that they can effectively execute a specific computation task in a wireless network. The first part of this article focuses on introducing the SemCom principles including encoding, layered system architecture, and two design approaches:1)layer-coupling design;and 2)end-to-end design using a neural network. The second part focuses on the discussion of specific techniques for different application areas of H2M SemCom[including human and AI symbiosis, recommendation, human sensing and care, and virtual reality(VR)/augmented reality(AR)]and M2M SemCom(including distributed learning, split inference, distributed consensus, and machine-vision cameras). Finally,we discuss the approach for designing SemCom systems based on knowledge graphs. We believe that this comprehensive introduction will provide a useful guide into the emerging area of SemCom that is expected to play an important role in sixth generation(6G)featuring connected intelligence and integrated sensing, computing, communication, and control. | Qiao Lan Dingzhu Wen Zezhong Zhang Qunsong Zeng Xu Chen Petar Popovski Kaibin Huang | 2021 | Journal of Communications and Information Networks2021,6,4: | 4
Nitrogen partitioning and microbial protein synthesis in lactating dairy cows with different phenotypic residual feed intake显示文摘Background: Residual feed intake(RFI) is an inheritable measure of feed efficiency that is independent on level of production. However, physiological and metabolic mechanisms underlying divergent RFI are not fully elucidated. This study was conducted to investigate dietary nitrogen(N) partitioning and microbial protein synthesis in lactating dairy cows divergent in phenotypic RFI.Results: Thirty Holstein dairy cows(milk yield = 35.3 ± 4.71 kg/d;milk protein yield = 1.18 ± 0.13 kg/d;mean ± standard deviation) were selected for the experiment to derive RFI. After the RFI measurement period of 50 d, the 10 lowest RFI cows and 8 highest RFI cows were selected. The low RFI cows had lower dry matter intake(DMI, P < 0.05) than the high RFI cows, but they produced similar energy-corrected milk. The ratios of milk to DMI(1.41 vs. 1.24, P < 0.01) and energy-corrected milk to DMI(1.48 vs. 1.36, P < 0.01) were greater in low RFI cows than those in the high RFI cows. The low RFI cows had lower milk urea nitrogen than that in the high RFI cows(P = 0.05). Apparent digestibility of nutrients did not differ between two groups(P > 0.10). Compared with high RFI animals, the low RFI cows had a lower retention of N(5.72 vs. 51.4 g/d, P < 0.05) and a higher partition of feed N to milk N(29.7% vs. 26.5%, P < 0.05).Conclusions: The results suggest that differences in N partition, synthesis of microbial protein, and utilization of metabolizable protein could be part of the mechanisms associated with variance in the RFI. | Yunyi Xie Zezhong Wu Diming Wang Jianxin Liu | 2019 | Journal of Animal Science and Biotechnology2019,10,4: | 4

Late Triassic Biotite Monzogranite from the Western Litang Area,Yidun Terrane,SW China: Petrogenesis and Tectonic Implications显示文摘The Late Triassic igneous rocks in the Yidun terrane can provide vital insights into the evolution of Plaeo-Tethys in western China. We present new zircon U-Pb, whole-rock geochemistry, and Sr-Nd-Pb-Hf isotopic data for the Litang biotite monzogranites, Yidun terrane. The biotite monzogranites have a zircon U-Pb age of 206.1±1.0 Ma(MSWD=1.9,n=30), which indicates Late Triassic magmatism. The biotite monzogranites display I-type affinity, high Na_2O(3.38-3.60 wt%) contente, medii SiO_2(67.12-69.13 wt%), and low P_2 O_5 contents(0.10~0.12 wt%). They enriched in Rb,and Ba and depleted in Nb and Ta, with negative Eu anomalies(Eu/Eu*=0.74—0.81). They have evolved Sr-Nd-Pb-Hf isotopic composition, i.e.,(^(87) Sr/^(86 )Sr)i=0.714225 to 0.714763, negative ?_(Nd(t)) values of -2.0 to-2.6 with two-stage Nd model ages ranging from 1.01 to 1.05 Ga, negative ?_(Ht)(t)) values o f-3.4 to-4.1 with two-stage Hf model ages of 1.85 to1.88 Ga, suggesting a matured crustal sources. Their low Al_2O_3/TiO_2 ratios and medium Cao/Na_2O ratios, medium Mg~# and SiO_2 contents, low [molar Al_2O_3/(MgO+FeO^T)] values, and high [molar Cao/(MgO+FeO^T)] values indicate that the Litang biotite monzogranite was formed by partial melting of metabasaltic rocks. Based on the previous studies, we propose that the Litang biotite monzogranite derived from the westward subduction and closure of the Ganzi-Litang ocean during the Late Triassic-The mantle wedge-derived mafic melts provided sufficient heat for partial melting of ancient metabasalt protolith within the middle-lower crust. | ZHU Yu LAI Shaocong QIN Jiangfeng ZHANG Zezhong ZHANG Fangyi | 2019 | Acta Geologica Sinica(English Edition)2019,93,2: | 1
