| 1 | Construction and characterization of bivalent vaccine candidate expressing HspA and M_r18 000 OMP from Helicobacterpylori显示文摘AIM: To construct a recombinant vector which can express outer membrane protein (OMP) with Mr18 000 and heat shock protein A (HspA) from Helicobacter pylori(H.pylori)in E.coli BL21, and to exploit the possibility for obtaining the vaccine conferring protection from H. pylori infection.METHODS: The target gene of HspA was amplified from H.pylori chromosome by PCR, and then inserted into the prokaryotic expression vector pET32a (+) by restrictive endonuclease enzyme kpn Ⅰ, BamH Ⅰ simultaneously. The recombinant vector was used to sequence, and then together with pET32a (+)/Omp18, digested by restrictive endonuclease enzyme Hind Ⅲ and BamH Ⅰ simultaneously. pET32a(+)/HspA and Omp18 were recovered from 1% agarose gel by gel kit, and ligated with T4 ligase by BarmH Ⅰ digested viscidity end. The recombinant plasmid of pET32a(+)/HspA/Omp18 was transformed and expressed in E. coli BL21 (DE3) under induction of IPTG. After purification, its antigenicity of the fusion protein was detected by Western blot.RESULTS: Enzyme digestion analysis and sequencing showed that the target genes were inserted into the recombinant vector, composed of 891 base pairs, encoded objective polypeptides of 297 amino acid residues. Compared with GenBank reported by Tomb et al, there were 1.3 %and 1.4 % differences in obtained H. pylori nucleotide sequence and amino acid residues, respectively. SDS-PAGE analysis showed that relative molecule mass (Mr) of the expressed product was Mr 51 000,Mr of protein expressed by pET32a (+) was about Mr 20 000, and soluble expression product accounted for 18.96 % of total bacterial protein.After purification with Ni+2-NTA agarose resins, the purification of recombinant fusion protein was about 95 %. Western blot showed that recombinant fusion protein could be recognized by the patients′ serum infected with H. pylori and anti-Omp18 monoclone, suggesting that this protein had good antigenicity.CONCLUSION: The gene coding for H. pylori Mr18 000OMP and HspA was cloned and expressed successfully. | 2 | Chronology and geochemistry of the volcanic rocks in Woruo Mountain region,Northern Qiangtang depression:Implications to the Late Triassic volcanic-sedimentary events显示文摘A suite of sedimentary-volcaniclastic rocks intercalated with the volcanic rocks unconformably overlies the Triassic Xiaochaka Formation in the Woruo Mountain region, Qiangtang Basin, northern Tibet. The vitric tuff from the base of these strata gives a SHRIMP zircon U-Pb age of 216 ± 4.5 Ma, which represents the age of the Late Triassic volcanic-sedimentary events in the Woruo Mountain region, and is consistent with that of the formation of the volcanic rocks from the Nadi Kangri Formation in the Nadigangri-Shishui River zone. There is a striking similarity in geochemical signatures of the volcanic rocks from the Woruo Mountain region and its adjacent Nadigangri-Shishui River zone, indicating that all the volcanic rocks from the Qiangtang region might have the same magmatic source and similar tectonic setting during the Late Triassic. | 3 | Mature Archean continental crust in the Yangtze craton: Evidence from petrology, geochronology and geochemistry显示文摘Previous studies have shown that the Archean basement is widely distributed throughout the Yangtze craton. To date, however, Archean basement terrains have not been found, except for a few Kongling high-grade metamorphic terrains in the Huangling dome that have been confirmed to be of Archean age. To further understand the basement component and crustal evolution of the Yangtze craton, we carried out a petrological, geochronological and geochemical study of the Jinshan K-feldspar granite emplaced within the Yangpo Group, located in Huji Town, Zhongxiang City, Hubei Province. Results indicate that the zircon SHRIMP U-Pb age of the Jinshan granite is 2655±9 Ma, placing it within the middle Neoarchean. Chemically, this pluton yields abundant silica and alkalis and is depleted in Ca, Mg and Ti. Furthermore, it is enriched in Rb, Th, Ga, Y and Zr, depleted in Sr, Ba, Nb and Ta, and especially lacking in Eu. High ratios of FeO*/MgO (32.0 to 58.7) and 104 ×Ga/Al (3.19 to 3.41) were also found. The pluton exhibits characteristics typical of A-type granites with crustal source magmas. Moreover, the meta-sedimentary rock association of the Yangpo Group, into which the pluton intruded, clearly shows relatively stable depositional environments of a shallow shelf sequence. Therefore, before the middle Neoarchean, the Yangtze craton contained mature continental crust. | 4 | Metallogenic Epoch of Nonferrous Metallic and Silver Deposits in the Jiaodong Peninsula, China and its Geological Significance显示文摘As China's most important gold-producing district,the Jiaodong Peninsula also contains copper,lead-zinc,molybdenum(tungsten),and other nonferrous metal ore deposits,but the space-time and genetic relationships with gold deposits remain uncertain.To investigate the temporal relationship between these nonferrous metal and gold ore deposits,We collected the samples from a number of nonferrous metallic and silver deposits and metallogenetic rock bodies in the eastern Jiaodong Peninsula for isotopic dating.The results show that the Re-Os isotopic model ages of the Lengjia molybdenum deposit in Rongcheng range from 114.5± 1.8 Ma to 112.6± 1.5 Ma,with an average age of 113.6± 1.6Ma;the LA-ICP-MS ^(206)Pb/^(238)U ages of 33 zircons in the sericitization porphyritic monzogranite that hosts the Tongjiazhuang silver deposit in Rongcheng range between 122 Ma and 109 Ma,with a weighted mean age of 116.04± 0.95 Ma;the LA-ICP-MS ^(206)Pb/^(238)U ages of 31 zircons in the copper metallogenic pyroxene monzodiorite that hosts the Kuangbei copper deposit in Rongcheng range from126 Ma to 106 Ma,with a weighted mean age of 116.6± 1.7 Ma;and the LA-ICP-MS ^(206)Pb/^(238)U ages of19 zircons in the pyroxene monzodiorite surrounding the Dadengge gold and multimetal deposit in Weihai range from 113 Ma to 110 Ma,with a weighted mean age of 111.7± 0.6 Ma.All these results indicate that the metallogenic ages of the silver and nonferrous metallic deposits in the Jiaodong Peninsula are in a limited range from 118 Ma to 111 Ma.Previous studies have demonstrated that the isotopic ages of gold deposits in the Jiaodong Peninsula range from 123 Ma to 110 Ma,while Weideshanian magmatism occurred between 126 Ma to 108 Ma.Both these ranges are grossly consistent with the metallogenic ages of silver and nonferrous metallic deposits in this study,suggesting that the large-scale mineralization occurred in the Early Cretaceous when magmatic activities were strong.This epoch may be linked to the lithosphere thinning and the thermo-upwelling extension in eastern China at that time.In addition,field investigation also shows that gold and nonferrous metallic deposits are distributed nearby the Weideshanian granite,with the nonferrous metallic deposits lying within or surrounding the granite pluton and the gold deposits outside the granite pluton.We propose the following mineralization scenario:In the Early Cretaceous,an intensive lithospheric extension induced partial melting and degassing of the metasomatized lithospheric mantle,which resulted in the formation of mantle-derived fluids enriched in metal elements.During the rapid process of magma ascent and intrusion,crust-derived fluids were activated by the magmatic thermal dome and served to further extract ore-forming materials from the crust.These fluids may have mixed with the mantle-derived fluid to form a crust-mantle mixing-type ore-forming fluid.The high-temperature conditions in the center or in contact with the granitic magmatic thermal dome would have been favorable for the formation of porphyry-type,skarn-type,and hydrothermal-vein-type ores,thus forming a series of Mo(W),Cu,and Pb-Zn deposits in the mid-eastern Jiaodong Peninsula.In contrast,the medium-to low-temperature conditions in the periphery of the magmatic thermal dome would have favored the deposition of gold(silver) ores under the appropriate physiochemical and structural conditions.The metallogenic epoch of the molybdenum,copper,and silver deposits,and their spatio-temporal and genetic relations to the gold deposits,as demonstrated in this study,not only provide important insights to the study of regional metallogeny,our understanding of the metallogenesis of the Jiaodong type gold deposit,and the geodynamic background of the large-scale mineralization in the Jiaodong Peninsula,but also have practical value in guiding the mineral exploration. | SONG Mingchun WANG Shanshan YANG Lixin LI Jie LI Shiyong DING Zhengjiang | 2017 | Acta Geologica Sinica(English Edition)2017,91,4: | 12 |
| 5 | An oases dynamics model and its application in the Manas oasis显示文摘This study applies theories of systems and system dynamics to ecological economic systems in the oasis environment, with the intention of finding the basic characteristics of the oasis environment and its feedback structures. This study explores the inner mechanisms of economic development in the oasis environment and its relationship to policy variables. This model is applied to economic development in the Manas oasis of the Xinjiang Uygur Autonomous Region of China. System dynamics method starts from studying feedback structure within the system study and then searches for the best solution through identifying the right 'policy point' and through model simulation in the computer. It embodies the cream of 'systems cybernetics theory'. This recombinant plasmid of Top10 was sent out for nucleotide sequence analysis. Finally this sequence AF479754 was compared with HP0638 and JHP0581.RESULTS: The sequence of the aim gene was obtained. It had 924 base pairs. The identity was 95.32% against HP0638, 95.02% against JHP0581, which was higher than the identity between HP0638 and JHP0581.CONCLUSION: Although the aim gene was obtained, but it was different from the published sequence of GenBank. It is not clear what makes this difference. Maybe it is because different strain was used or because there were some variations. | 12 | Exhumation history and preservation of the Jiaojia giant gold deposit, Jiaodong Peninsula显示文摘The Jiaojia giant gold deposit is the largest gold deposit in China, with a total gold reserve of approximately 1200 t.Until now, the knowledge of the exhumation history of post-mineralization period is limited, in particular for the low-temperature thermochronology studies of samples below-1000 m. In this work, we combined zircon fission-track(ZFT) and apatite fission-track(AFT) dating of samples between-1100 and-2000 m to determine the post-mineralization cooling and exhumation history of the Jiaojia giant gold deposit. The ZFT ages ranged from 144.2±6.3 to 124.4±5.5 Ma, representing the cooling period and the disturbance of ore-forming fluid. The AFT ages ranged from 28.1±2.6 to 16.2±1.0 Ma, recording the exhumation and cooling processes. With reference to previous low-temperature thermochronology studies in the Jiaojia goldfield, we estimated the exhumation rate and amount of the Jiaojia giant gold deposit and reconstructed its exhumation and preservation history. The exhumation history was divided into four stages, rapid exhumation(~120–95 Ma), relatively slow exhumation(~95–50 Ma),slow exhumation(~50–30 Ma) and relatively rapid exhumation(since 30 Ma). Each stage corresponds to geological events related to the basin-mountain coupling that have occurred since the Cretaceous in the Jiaodong area, namely, a strong tectonic extension and volcanic eruption in the Jiaolai Basin, subsidence of the Jiaolai Basin and Wangshi Group molasse sedimentary,tectonic quiescence, and the Linqu Group basalt eruption of the Jiaobei uplift. Our results show that the exhumation of the Jiaojia giant gold deposit is ~5.2±1.2 km and the orebody erosion degree is relatively low, indicating huge prospecting potential deep in the Jiaojia giant gold deposit.
