| 1 | Emergence and prevalence of naturally occurring lower virulent African swine fever viruses in domestic pigs in China in 2020显示文摘African swine fever virus(ASFV)has been circulating in China for more than two years,and it is not clear whether the biological properties of the virus have changed.Here,we report on our surveillance of ASFVs in seven provinces of China,from June to December,2020.A total of 22 viruses were isolated and characterized as genotype II ASFVs,with mutations,deletions,insertions,or short-fragment replacement occurring in all isolates compared with Pig/HLJ/2018(HLJ/18),the earliest isolate in China.Eleven isolates had four different types of natural mutations or deletion in the EP402R gene and displayed a nonhemadsorbing(non-HAD)phenotype.Four isolates were tested for virulence in pigs;two were found to be as highly lethal as HLJ/18.However,two non-HAD isolates showed lower virulence but were highly transmissible;infection with 106 TCID50 dose was partially lethal and caused acute or sub-acute disease,whereas 103 TCID50 dose caused non-lethal,sub-acute or chronic disease,and persistent infection.The emergence of lower virulent natural mutants brings greater difficulty to the early diagnosis of ASF and creates new challenges for ASFV control. | Encheng Sun Zhenjiang Zhang Zilong Wang Xijun He Xianfeng Zhang Lulu Wang Wenqing Wang Lianyu Huang Fei Xi Haoyue Huangfu Ghebremedhin Tsegay Hong Huo Jianhong Sun Zhijun Tian Wei Xia Xuewu Yu Fang Li Renqiang Liu Yuntao Guan Dongming Zhao Zhigao Bu | 2021 | Science China(Life Sciences)2021,64,5: | 46 |

| 3 | Growth and Mortality of Small Yellow Croaker(Larimichthys polyactis) Inhabiting Haizhou Bay of China显示文摘Length frequency data of small yellow croaker(Larimichthys polyactis) were acquired from the survey vessel in May,July,September and December,2011 in Haizhou Bay of China.In this study,921 fish individuals were analyzed for the estimation of growth and mortality parameters.Between length and weight,the power coefficient b was 2.7321,2.9703,3.0418 and 2.7252 for the 4 surveying months,respectively.The estimated von Bertalanffy growth function parameters were 230mm(L ∞) and 0.26yr-1(K) as were calculated with ELEFAN method equipped in FiSAT computer package.With length-converted catch curve analysis,the total mortality rate(Z) and its 95% confidence interval were 2.16(1.69-2.64) yr-1,0.59(0.15-1.04) yr-1,1.16(0.80-1.52) yr-1 and 0.96(0.70-1.23) yr-1 for the 4 surveying months,respectively,with the pooled data the value was 1.15(0.81-1.48) yr-1.The natural mortality rate(M) was 0.516 yr-1 as was calculated with Pauly's equation(the annual average sea water temperature was 11℃).Therefore,fish mortality rate was 0.634 yr-1.The yield-per-recruit analysis indicated that when t c was 1,F max was 0.7 and F 0.1 was 0.55.Currently,the age at first capture is about 1 year and F current was 0.634.Therefore,F current was larger than F 0.1 and less than F max.This indicates that current fish mortality is at a dangerously high level.With Gulland method,the biological reference point for fishery(F opt) was estimated as 0.516 yr-1,lower than current fish mortality.Accordingly,reducing catch in the region was strongly recommended. | LIU Qun XU Binduo YE Zhenjiang REN Yiping | 2012 | Journal of Ocean University of China2012,11,4: | 17 |

| 6 | Functional analysis of the larval serum protein gene promoter from silkworm,Bombyx mori.显示文摘The regulation region of larval serum protein gene, Bombyx mori. (BmLSP), consisting of the first intron, the first exon, the central promoter region and 5′-upstream region, is cloned from genomic DNA from the silkworm va-riety of Suju譓inghu. Using PCR and restriction endonu-clease methods, a series of luciferase reporter plasmids, driven by different length of BmLSP promoters, are con-structed. Via the transient expression system in BmN cells, the effects of the regulation elements and foreign insect hor-mones on the BmLSP promoter activity are investigated. The results demonstrate that the promoter activity of BmLSP is 5.8- or 4.4-fold higher than that of BmLSPs whose first in-tron or the element in 5′-upstream region harboring the homologous sequence with the first intron of light-chain fib-roin gene (EHIF) is deleted, respectively, suggesting that both the first intron and EHIF contain the main positive cis-acting elements. However, the inactive mariner transposable ele-ment (MTE) in 5′-upstream region presents a negative effect. Furthermore, the effects of juvenile hormone analogue (JHA) on the BmLSP promoter activity show a typical dose-dependent manner, that is, low concentration treat-ments increase the BmLSP promoter activity and high con-centration treatments decrease it. | 7 | Three-dimensional bioprinting of multicell-laden scaffolds containing bone morphogenic protein-4 for promoting M2 macrophage polarization and accelerating bone defect repair in diabetes mellitus显示文摘Critical-sized bone defect repair in patients with diabetes mellitus remains a challenge in clinical treatment because of dysfunction of macrophage polarization and the inflammatory microenvironment in the bone defect region.Three-dimensional(3D)bioprinted scaffolds loaded with live cells and bioactive factors can improve cell viability and the inflammatory microenvironment and further accelerating bone repair.Here,we used modified bioinks comprising gelatin,gelatin methacryloyl(GelMA),and 4-arm poly(ethylene glycol)acrylate(PEG)to fabricate 3D bioprinted scaffolds containing BMSCs,RAW264.7 macrophages,and BMP-4-loaded mesoporous silica nanoparticles(MSNs).Addition of MSNs effectively improved the mechanical strength of GelMA/gelatin/PEG scaffolds.Moreover,MSNs sustainably released BMP-4 for long-term effectiveness.In 3D bioprinted scaffolds,BMP-4 promoted the polarization of RAW264.7 to M2 macrophages,which secrete anti-inflammatory factors and thereby reduce the levels of pro-inflammatory factors.BMP-4 released from MSNs and BMP-2 secreted from M2 macrophages collectively stimulated the osteogenic differentiation of BMSCs in the 3D bioprinted scaffolds.Furthermore,in calvarial critical-size defect models of diabetic rats,3D bioprinted scaffolds loaded with MSNs/BMP-4 induced M2 macrophage polarization and improved the inflammatory microenvironment.And 3D bioprinted scaffolds with MSNs/BMP-4,BMSCs,and RAW264.7 cells significantly accelerated bone repair.In conclusion,our results indicated that implanting 3D bioprinted scaffolds containing MSNs/BMP-4,BMSCs,and RAW264.7 cells in bone defects may be an effective method for improving diabetic bone repair,owing to the direct effects of BMP-4 on promoting osteogenesis of BMSCs and regulating M2 type macrophage polarization to improve the inflammatory microenvironment and secrete BMP-2. | Xin Sun Zhenjiang Ma Xue Zhao Wenjie Jin Chenyu Zhang Jie Ma Lei Qiang Wenhao Wang Qian Deng Han Yang Jinzhong Zhao Qianqian Liang Xiaojun Zhou Tao Li Jinwu Wang | 2021 | Bioactive Materials2021,6,3: | 12 |
| 8 | Spatial and Seasonal Patterns of Ichthyoplankton Assemblages in the Haizhou Bay and Its Adjacent Waters of China显示文摘Surveys were conducted in five voyages in Haizhou Bay and its adjacent coastal area from March to December 2011 during full moon spring tides. The ichthyoplankton assemblages and the environmental factors that affect their spatial and seasonal patterns were determined. Totally 35 and 12 fish egg and larvae taxa were identified, respectively. Over the past several decades, the egg and larval species composition has significantly changed in Haizhou Bay and its adjacent waters, most likely corresponding with the alteration of fishery resources, which are strongly affected by anthropogenic activities and climate change. The Bray-Curtis dissimilarity index identified four assemblages: near-shore bay assemblage, middle bay assemblage and two closely related assemblages(near-shore/middle bay assemblage and middle/edge of bay assemblage). The primary species of each assemblage principally reflected the spawning strategies of adult fish. The near-shore bay assemblage generally occurred in near-shore bay, with depths measuring <20 m, and the middle bay assemblage generally occurred in the middle of bay, with depths measuring 20 to 40 m. Spatial and seasonal variations in ichthyoplankton in each assemblage were determined by interactions between biological behavioral traits and oceanographic features, particularly the variation of local conditions within the constraint of a general reproductive strategy. | 10 | Ligustrazine alleviates acute lung injury in a rat model of acute necrotizing pancreatitis显示文摘BACKGROUND: Acute necrotizing pancreatitis leads to a systemic inflammatory response characterized by widespread leukocyte activation and, as a consequence, distant lung injury. The aim of this study was to evaluate the effect of ligustrazine, extracted from Ligusticum wallichii a traditional Chinese medicine, on lung injury in a rat model of acute necrotizing pancreatitis (ANP). METHODS: A total of 192 rats were randomly divided into three groups: control (C group); ANP without treatment (P group); and ANP treated with ligustrazine (T group). Each group was further divided into 0.5, 2, 6 and 12 hours subgroups. All rats were anesthetized with an intraperitoneal injection of sodium pentobarbital. Sodium taurocholate was infused through the pancreatic membrane to induce ANP. For the T group, sodium taurocholate was infused as above, then 0.6% ligustrazine was administered via the femoral vein. The effects of ligustrazine on the severity of lung injury were assessed by lung wet/dry weight ratio, myeloperoxidase (MPO) activity and histopathological changes. Pulmonary blood flow was determined by the radioactive microsphere technique (RMT). RESULTS: The blood flow in the P group was significantly lower than that of the C group, while the blood flow in the T group was significantly higher than that of the P group but showed no significant difference from the C group. Compared with C group, the lung wet/dry ratios in both the P and T groups were significantly increased, but there was no significant difference between them. The MPO activity in the P group was greatly increased over that of the C group. In the T group, although the MPO activity was also higher than in the C group, it much less increased than in the P group. Moreover, the difference between P and T groups was significant after 0.5 to 12 hours. After induction of the ANP model, the pancreas showed mild edema and congestion; the longer the time, the more severe this became. The pulmonary pathological changes wereaggravated significantly in the P group. Histopathological scores were higher in the P group than in the C group throughout the experimental course. Histopathological scores in the T group were lower than those in the P group at 6 and 12 hours. CONCLUSIONS: Microcirculatory disorder is an important factor of lung injury in ANP. | 11 | Defined tumor antigen-specific T cells potentiate personalized TCR-T cell therapy and prediction of immunotherapy response显示文摘Personalized immunotherapy targeting tumor-specific antigens(TSAs)could generate efficient and safe antitumor immune response without damaging normal tissues.Although neoantigen vaccines have shown therapeutic effect in clinic trials,precise prediction of neoantigens from tumor mutations is still challenging.The host antitumor immune response selects and activates T cells recognizing tumor antigens.Hence,T cells engineered with T-cell receptors(TCRs)from these naturally occurring tumor antigen-specific T(Tas)cells in a patient will target personal TSAs in his/her tumor.To establish such a personalized TCR-T cell therapy,we comprehensively characterized T cells in tumor and its adjacent tissues by single-cell mRNA sequencing(scRNA-seq),TCR sequencing(TCR-seq)and in vitro neoantigen stimulation.Compared to bystander T cells circulating among tissues,Tas cells were characterized by tumor enrichment,tumor-specific clonal expansion and neoantigen specificity.We found that CXCL13 is a unique marker for both CD4^(+)and CD8^(+)Tas cells.Importantly,TCR-T cells expressing TCRs from Tas cells showed significant therapeutic effects on autologous patient-derived xenograft(PDX)tumors.Intratumoral Tas cell levels measured by CXCL13 expression precisely predicted the response to immune checkpoint blockade,indicating a critical role of Tas cells in the antitumor immunity.We further identified CD200 and ENTPD1 as surface markers for CD4^(^(+))and CD8^(^(+))Tas cells respectively,which enabled the isolation of Tas cells from tumor by Fluorescence Activating Cell Sorter(FACS)sorting.Overall,our results suggest that TCR-T cells engineered with Tas TCRs are a promising agent for personalized immunotherapy,and intratumoral Tas cell levels determine the response to immunotherapy. | Jingjing He Xinxin Xiong Han Yang Dandan Li Xuefei Liu Shuo Li Shuangye Liao Siyu Chen Xizhi Wen Kuai Yu Lingyi Fu Xingjun Dong Kaiyu Zhu Xiaojun Xia Tiebang Kang Chaochao Bian Xiang Li Haiping Liu Peirong Ding Xiaoshi Zhang Zhenjiang Liu Wende Li Zhixiang Zuo Penghui Zhou | 2022 | Cell Research2022,32,6: | 10 |
| 12 | Protective effect of clodronate-containing liposomes on intestinal mucosal injury in rats with severe acute pancreatitis显示文摘BACKGROUND: Severe acute pancreatitis (SAP) can result in intestinal mucosal injury. This study aimed to demonstrate the protective effect of clodronate-containing liposomes on intestinal mucosal injury in rats with SAP. METHODS: Liposomes containing clodronate or phosphate buffered saline (PBS) were prepared by the thin-film method SAP models were prepared by a uniform injection of sodium taurocholate (2 mL/kg body weight) into the subcapsular space of the pancreas. Sprague-Dawley rats were randomly divided into a control group (C group), a SAP plus PBS-containing liposomes group (P group) and a SAP plus clodronate-containing liposomes group (T group). At 2 and 6 hours after the establishment of SAP models, 2 mL blood samples were taken from the superior mesenteric vein to measure the contents of serum TNF-α and IL-12. Pathological changes in the intestine and pancreas were observed using hematoxylin and eosin staining, while apoptosis was detected using TUNEL staining. In addition, the macrophage markers cluster of differentiation 68 (CD68) in the intestinal tissue was assessed with immunohistochemistry. RESULTS: At the two time points, the levels of TNF-α and IL-12 in the P group were higher than those in the C group (P<0.05) Compared with the P group, the levels of TNF-α and IL-12 decreased in the T group (P<0.05). The pathological scores of the intestinal mucosa and pancreas in the T group were lower than those of the P group. In the T group, large numbers of TUNEL-positive cells were observed, but none or few in the C and P groups. The number of CD68-positive macrophages decreased in the T group.CONCLUSIONS: Clodronate-containing liposomes have prote- ctive effects against intestinal mucosal injury in rats with SAP. | 14 | Dynamic Analysis of Propulsion Mechanism Directly Driven by Wave Energy for Marine Mobile Buoy显示文摘Marine mobile buoy(MMB) have many potential applications in the maritime industry and ocean science.Great progress has been made,however the technology in this area is far from maturity in theory and faced with many difficulties in application.A dynamic model of the propulsion mechanism is very necessary for optimizing the parameters of the MMB,especially with consideration of hydrodynamic force.The principle of wave-driven propulsion mechanism is briefly introduced.To set a theory foundation for study on the MMB,a dynamic model of the propulsion mechanism of the MMB is obtained.The responses of the motion of the platform and the hydrofoil are obtained by using a numerical integration method to solve the ordinary differential equations.A simplified form of the motion equations is reached by omitting terms with high order small values.The relationship among the heave motion of the buoy,stiffness of the elastic components,and the forward speed can be obtained by using these simplified equations.The dynamic analysis show the following:The angle of displacement of foil is fairly small with the biggest value around 0.3 rad;The speed of mobile buoy and the angle of hydrofoil increased gradually with the increase of heave motion of buoy;The relationship among heaven motion,stiffness and attack angle is that heave motion leads to the angle change of foil whereas the item of speed or push function is determined by vertical velocity and angle,therefore,the heave motion and stiffness can affect the motion of buoy significantly if the size of hydrofoil is kept constant.The proposed model is provided to optimize the parameters of the MMB and a foundation is laid for improving the performance of the MMB. | YU Zhenjiang ZHENG Zhongqiang YANG Xiaoguang CHANG Zongyu | 2016 | Chinese Journal of Mechanical Engineering2016,29,4: | 8 |

However, what remains a conundrum is how these cell-surface receptors end up in the nucleus. In this study, we reported that the transmembrane receptor phosphorylated TrkA was located in a series of carrier vesicles, including ring-like vesicles near the plasma membrane, large core vesicles and small dense core vesicles around the nuclei, as well as in the nucleus in human glioma cell line U251 using immunocytochemistry and immunofluorescence staining. Meanwhile, we also showed that small dense core vesicles budded from large core vesicles, and interacted with the nuclear envelope. | 17 | Design optimization of cast Cu-Al-Be-B alloys for high damping capacity显示文摘This paper investigated high-damping Cu-Al-Be-B cast alloys using metallographic analysis, X-ray diffraction (XRD) and electrical resistance measurements for transformation temperatures. The results showed that beryllium can stabilize β phase, resulting in a thermo-elastic martensite microstructure leading to high-damping capacity in cast Cu-Al-Be-B alloys. Trace additions of boron to Cu-Al-Be alloys can significantly refine the grains, providing high strength and ductility to the alloys. A factorial design of experiment method was used to optimize the composition and properties of cast Cu-Al-Be-B alloys. The optimal microstructure for thermo-elastic martensite can be obtained by adjusting the amounts of aluminum and beryllium to eutectoid or pseudo-eutectoid compositions. | 18 | Equilibrium equations for nonlinear buckling analysis of drill-strings in 3D curved well-bores显示文摘With the development of drilling technology, the oil/gas well has evolved from its early vertical straight form to the inclined, horizontal, plane curved, or even 3D curved well-bore. Understanding of the buck- ling behavior of a drill-string in a well-bore is crucial for the success of a drilling operation. Therefore, equilibrium equations for analyzing the buckling behavior of a drill-string in a 3D curved well-bore are required. Based on Love’s equilibrium equations for a curved and twisted rod in space, a set of equi- librium equations for the nonlinear buckling analysis of a drill-string in a 3D curved well-bore are de- rived by introducing a radial constraint of the well-bore. The proposed formulae can account for the well curvature and tortuosity. Thus, it can be used to analyze the buckling behaviors of a drill-string constrained in a well-bore and subjected to axial compression, torsion at its upper end, and gravity simultaneously. It is worth noting that the existing equations in the literature for a drill-string in a straight and plane curved well-bore with a constant curvature are a special case of the proposed model. | 20 | A reduced order aerothermodynamic modeling framework for hypersonic vehicles based on surrogate and POD显示文摘Aerothermoelasticity is one of the key technologies for hypersonic vehicles. Accurate and efficient computation of the aerothermodynamics is one of the primary challenges for hypersonic aerothermoelastic analysis. Aimed at solving the shortcomings of engineering calculation, computation fluid dynamics(CFD) and experimental investigation, a reduced order modeling(ROM)framework for aerothermodynamics based on CFD predictions using an enhanced algorithm of fast maximin Latin hypercube design is developed. Both proper orthogonal decomposition(POD) and surrogate are considered and compared to construct ROMs. Two surrogate approaches named Kriging and optimized radial basis function(ORBF) are utilized to construct ROMs. Furthermore,an enhanced algorithm of fast maximin Latin hypercube design is proposed, which proves to be helpful to improve the precisions of ROMs. Test results for the three-dimensional aerothermodynamic over a hypersonic surface indicate that: the ROMs precision based on Kriging is better than that by ORBF, ROMs based on Kriging are marginally more accurate than ROMs based on PODKriging.
