2Isolation,characterization,and phylogenic analysis of three new severe fever with thrombocytopenia syndrome bunyavirus strains derived from Hubei Province,China显示文摘Hubei Province is a major epidemic area of severe fever with thrombocytopenia syndrome bunyavirus(SFTSV) in China. However, to date, a few SFTSV strains have been isolated from Hubei Province, preventing effective studies of epidemic outbreaks. Here, we report three confirmed patients(2015–2016) with typical symptoms of severe fever with thrombocytopenia syndrome disease(SFTS) who were farmers resident in different regions in Hubei Province. Three new SFTSV strains were isolated from the serum samples of each patient. Characterization of viral growth properties showed that there were no significant differences in virus production. All strains were completely sequenced, and phylogenetic analysis showed that unlike the other strains from Hubei province, which belonged to the SFTSV C3 genotype, one of the three strains belonged to the SFTSV C2 genotype. 4Tick-Borne Viruses显示文摘Ticks are important vectors for the transmission of pathogens including viruses.The viruses carried by ticks also known as tick-borne viruses(TBVs),contain a large group of viruses with diverse genetic properties and are concluded in two orders,nine families,and at least 12 genera.Some members of the TBVs are notorious agents causing severe diseases with high mortality rates in humans and livestock,while some others may pose risks to public health that are still unclear to us.Herein,we review the current knowledge of TBVs with emphases on the history of virus isolation and identification,tick vectors,and potential pathogenicity to humans and animals,including assigned species as well as the recently discovered and unassigned species.All these will promote our understanding of the diversity of TBVs,and will facilitate the further investigation of TBVs in association with both ticks and vertebrate hosts.Junming Shi Zhihong Hu Fei Deng Shu Shen 2018Virologica Sinica2018,33,1:10
5Isolation,Characterization,and Phylogenetic Analysis of Two New Crimean-Congo Hemorrhagic Fever Virus Strains from the Northern Region of Xinjiang Province,China显示文摘Crimean-Congo hemorrhagic fever(CCHF)caused by the CCHF virus(CCHFV)is a tick-borne natural focal disease with a mortality rate of approximately 50%.CCHFV is widely prevalent in Africa,southern Asia,the Middle East,and southeast Europe.CCHF outbreaks have been reported previously in Xinjiang province,China,especially in its southern region.Epidemiological surveys conducted on ticks and animals have revealed the presence of CCHFV strains in ticks,rodents,and infected individuals from cities and counties in southern Xinjiang.Phylogenetic analyses revealed that the Chinese CCHFV strains belong to one genotype,based on complete sequences of the S segments of its negative-stranded RNA genome.The present study reports two new CCHFV strains isolated from Hyalomma asiaticum asiaticum ticks collected from Fukang City and Wujiaqu City in the northern region of Xinjiang.Viral characteristics and their evolutionary relationships were analyzed through metagenomic and reverse-transcription PCR analyses;these analyses indicated that the genotype of both strains was different from that of other Chinese strains.Furthermore,previous reports of CCHFV in Xinjiang were reviewed and phylogenetic analyses were performed.CCHFV was found to prevail in Fukang City in Junggar Basin for more than 20 years,and that Fukang City and Wujiaqu City are considered natural reservoirs of different genotypes of CCHFV strains.Our findings facilitate the understanding of CCHFV distribution in Xinjiang province and provide insights into the evolutionary relationships among Chinese CCHFV strains.Yanfang Zhang Shu Shen Yaohui Fang Jinliang Liu Zhengyuan Su Jinhao Liang Zhong Zhang Qiaoli Wu Cheng Wang Abulikemu Abudurexiti Zhihong Hu Yujiang Zhang Fei Deng 2018Virologica Sinica2018,33,1:8
6Virus like particle-based vaccines against emerging infectious disease viruses显示文摘Emerging infectious diseases are major threats to human health.Most severe viral disease outbreaks occur in developing regions where health conditions are poor.With increased international travel and business,the possibility of eventually transmitting infectious viruses between different countries is increasing.The most effective approach in preventing viral diseases is vaccination.However,vaccines are not currently available for numerous viral diseases.Viruslike particles(VLPs) are engineered vaccine candidates that have been studied for decades.VLPs are constructed by viral protein expression in various expression systems that promote the selfassembly of proteins into structures resembling virus particles.VLPs have antigenicity similar to that of the native virus,but are non-infectious as they lack key viral genetic material.VLP vaccines have attracted considerable research interest because they offer several advantages over traditional vaccines.Studies have shown that VLP vaccines can stimulate both humoral and cellular immune responses,which may offer effective antiviral protection.Here we review recent developments with VLP-based vaccines for several highly virulent emerging or re-emerging infectious diseases.The infectious agents discussed include RNA viruses from different virus families,such as the Arenaviridae,Bunyaviridae,Caliciviridae,Coronaviridae,Filoviridae,Flaviviridae,Orthomyxoviridae,Paramyxoviridae,and Togaviridae families.Jinliang Liu Shiyu Dai Manli Wang Zhihong Hu Hualin Wang Fei Deng 2016Virologica Sinica2016,31,4:7
7The characteristics and sources of natural gases from Ordovician weathered crust reservoirs in the Central Gas Field in the Ordos Basin显示文摘The Central Gas Field is a famous large-sized gas field in the Ordos Basin of China. However, identification of main gas sources of the Ordovician reservoirs in this gas field remains puzzling. On the basis of a lot of geochemical data and geological research on natural gases, the characteristics and sources of natural gases from Ordovician weathered crust reservoirs in the Central Gas Field in the Ordos Basin were studied. The results indicated that natural gases from Ordovician weathered crust reservoirs in the Central Gas Field in the Ordos Basin have similar chemical and isotopic compositions to highly mature and over-mature dry gases. Both coal-derived gases and oil-type gases coexist in the Central Gas Field in the Ordos Basin. The former was derived mainly from Carboniferous-Permian coal measures and the latter from Lower Paleozoic marine carbonates. 9Genome analysis of Heliothis virescens ascovirus 3h isolated from China显示文摘No ascovirus isolated from China has been sequenced so far. Therefore, in this study, we aimed to sequence the genome of Heliothis virescens ascovirus 3h(Hv AV-3h) using the 454 pyrosequencing technology. The genome was found to be 190,519-bp long with a G+C content of 45.5%. We also found that it encodes 185 hypothetical open reading frames(ORFs) along with at least 50 amino acids, including 181 ORFs found in other ascoviruses and 4 unique ORFs. Gene-parity plots and phylogenetic analysis revealed a close relationship between Hv AV-3h and three other Hv AV-3a strains and a distant relationship with Spodoptera frugiperda ascovirus 1a(Sf AV-1a), Trichoplusia ni ascovirus 6a(Tn AV-6a), and Diadromus pulchellus ascovirus 4a(Dp AV-4a). Among the 185 potential genes encoded by the genome, 44 core genes were found in all the sequenced ascoviruses. In addition, 25 genes were found to be conserved in all ascoviruses except Dp AV-4a.In the Hv AV-3h genome, 24 baculovirus repeat ORFs(bros) were present, and the typical homologous repeat regions(hrs) were absent. 10Prevalence and Phylogenetic Analysis of Crimean-Congo Hemorrhagic Fever Virus in Ticks from Different Ecosystems in Xinjiang,China显示文摘The Crimean-Congo hemorrhagic fever virus(CCHFV),a member of the genus Orthonairovirus and family Nairoviridae,is transmitted by ticks and causes severe hemorrhagic disease in humans.To study the epidemiology of CCHFV in different ecosystems in Xinjiang,China,a total of 58,932 ticks were collected from Tarim Basin,Junggar Basin,Tianshan Mountain,and Altai Mountain from 2014 to 2017.Hyalomma asiaticum asiaticum was the dominant tick species in Tarim and Junggar basins,whereas Dermacentor nuttalli and Hyalomma detritum were found in Tianshan Mountain and Altai Mountain,respectively.Reverse transcription-polymerase chain reaction of the CCHFV small(S)genome segment was used for the molecular detection.The CCHFV-positive percentage was 5.26%,6.85%,1.94%,and 5.56% in Tarim Basin,Junggar Basin,Tianshan Mountain,and Altai Mountain,respectively.Sequences of the S segment were used for phylogenetic analysis and the results showed that the newly identified CCHFV strains belonged to two clades.Our study confirms that H.asiaticum asiaticum is the major vector of CCHFV in desert habitats which is consistent with previous studies,and also suggests that H.detritum and D.nuttalli are emerging vectors for CCHFV in Xinjiang.Moreover,this study reports the presence of CCHFV in the mountain habitat of Xinjiang for the first time,suggesting that future surveillance of CCHFV should also include mountainous areas.Abulimiti Moming Xihong Yue Shu Shen Chenchen Chang Cheng Wang Tao Luo Yanfang Zhang Rong Guo Zhihong Hu Yujiang Zhang Fei Deng Surong Sun 2018Virologica Sinica2018,33,1:5
11Machine-learning-based high-resolution DOA measurement and robust directional modulation for hybrid analog-digital massive MIMO transceiver显示文摘At hybrid analog-digital(HAD)transceiver,an improved HAD estimation of signal parameters via rotational invariance techniques(ESPRIT),called I-HAD-ESPRIT,is proposed to measure the direction of arrival(DOA)of a desired user,where the phase ambiguity due to HAD structure is dealt with successfully.Subsequently,a machine-learning(ML)framework is proposed to improve the precision of measuring DOA.Meanwhile,we find that the probability density function(PDF)of DOA measurement error(DOAME)can be approximated as a Gaussian distribution by the histogram method in ML.Then,a slightly large training data set(TDS)and a relatively small real-time set(RTS)of DOA are formed to predict the mean and variance of DOA/DOAME in the training stage and real-time stage,respectively.To improve the precisions of DOA/DOAME,three weight combiners are proposed to combine the-maximum-likelihood-learning outputs of TDS and RTS.Using the mean and variance of DOA/DOAME,their PDFs can be given directly,and we propose a robust beamformer for directional modulation(DM)transmitter with HAD by fully exploiting the PDF of DOA/DOAME,especially a robust analog beamformer on RF chain.Simulation results show that:(1)the proposed I-HAD-ESPRIT can achieve the HAD Cramer-Rao lower bound(CRLB);(2)the proposed ML framework performs much better than the corresponding real-time one without training stage;(3)the proposed robust DM transmitter can perform better than the corresponding non-robust ones in terms of secrecy rate.Zhihong ZHUANG Ling XU Jiayu LI Jinsong HU Linlin SUN Feng SHU Jiangzhou WANG 2020Science China(Information Sciences)2020,63,8:5
12A new strain of Crimean-Congo hemorrhagic fever virus isolated from Xinjiang, China显示文摘Crimean-Congo hemorrhagic fever virus(CCHFV) is a highly pathogenic tick-borne virus with a fatality rate of up to 50% in humans. CCHFV is widely distributed in countries around the world.Outbreaks of CCHFV infection in humans have occurred in prior years in Xinjiang Province, China.Epidemiological surveys have detected CCHFV RNA in ticks and animals; however, few isolates were identified. In this study, we identified and isolated a new CCHFV strain from Hyalomma asiaticum asiaticum ticks collected from north of Tarim Basin in Xinjiang, China. A preliminary investigation of infection and antigens expression of CCHFV was performed in newborn mice. The target tissues for CCHFV replication in newborn mice were identified. The analysis of the phylogenetic relationships with other Chinese strains suggested that diverse genotypes of CCHFV have circulated in Xinjiang for years. 13Establishment of a Reverse Genetic System of Severe Fever with Thrombocytopenia Syndrome Virus Based on a C4 Strain显示文摘Severe fever with thrombocytopenia syndrome virus(SFTSV)is an emerging tick-borne bunyavirus that causes hemorrhagic fever-like disease(SFTS)in humans with a case fatality rate up to 30%.To date,the molecular biology involved in SFTSV infection remains obscure.There are seven major genotypes of SFTSV(C1-C4 and J1-J3)and previously a reverse genetic system was established on a C3 strain of SFTSV.Here,we reported successfully establishment of a reverse genetics system based on a SFTSV C4 strain.First,we obtained the 5’-and 3’-terminal untranslated region(UTR)sequences of the Large(L),Medium(M)and Small(S)segments of a laboratory-adapted SFTSV C4 strain through rapid amplification of cDNA ends analysis,and developed functional T7 polymerase-based L-,M-and S-segment minigenome assays.Then,fulllength cDNA clones were constructed and infectious SFTSV were recovered from co-transfected cells.Viral infectivity,growth kinetics,and viral protein expression profile of the rescued virus were compared with the laboratory-adapted virus.Focus formation assay showed that the size and morphology of the foci formed by the rescued SFTSV were indistinguishable with the laboratory-adapted virus.However,one-step growth curve and nucleoprotein expression analyses revealed the rescued virus replicated less efficiently than the laboratory-adapted virus.Sequence analysis indicated that the difference may be due to the mutations in the laboratory-adapted strain which are more prone to cell culture.The results help us to understand the molecular biology of SFTSV,and provide a useful tool for developing vaccines and antivirals against SFTS.Mingyue Xu Bo Wang Fei Deng Hualin Wang Manli Wang Zhihong Hu Jia Liu 2021Virologica Sinica2021,36,5:5
14Genome Analysis of Dasineura jujubifolia Toursvirus 2,A Novel Ascovirus显示文摘So far,ascoviruses have only been identified from Lepidoptera host insects and their transmission vectors-endoparasitic wasps.Here,we reported the first finding of a complete novel ascovirus genome from a Diptera insect,Dasineura jujubifolia.Initially,sequence fragments with homology to ascoviruses were incidentally identified during metagenomic sequencing of the mitochondria of D.jujubifolia(Cecidomyiidae,Diptera)which is a major pest on Ziziphus jujuba.Then a full circular viral genome was assembled from the metagenomic data,which has an A+T percentage of 74%and contains 142,600 bp with 141 open reading frames(ORFs).Among the 141 ORFs,37 were conserved in all sequenced ascoviruses(core genes)including proteins predicted to participate in DNA replication,gene transcription,protein modification,virus assembly,lipid metabolism and apoptosis.Multi-gene families including those encode for baculovirus repeated open reading frames(BROs),myristylated membrane proteins,RING/U-box E3 ubiquitin ligases,and ATP-binding cassette(ABC)transporters were found in the virus genome.Phylogenetic analysis showed that the newly identified virus belongs to genus Toursvirus of Ascoviridae,and is therefore named as Dasineura jujubifolia toursvirus 2(DjTV-2a).The virus becomes the second reported species of the genus after Diadromus pulchellus toursvirus 1(DpTV-1a).The genome arrangement of DjTV-2a is quite different from that of DpTV-1a,suggesting these two viruses separated in an early time of evolution.The results suggest that the ascoviruses may infect a much broader range of hosts than our previous knowledge,and shed lights on the evolution of ascoviruses and particularly on that of the toursviruses.Jun Wang Minglu Yang Haibing Xiao Guo-Hua Huang Fei Deng Zhihong Hu 2020Virologica Sinica2020,35,2:4

16Phylogenetic analysis revealed the central roles of two African countries in the evolution and worldwide spread of Zika virus显示文摘Recent outbreaks of Zika virus(ZIKV) infections in Oceania's islands and the Americas were characterized by high numbers of cases and the spread of the virus to new areas. To better understand the origin of ZIKV, its epidemic history was reviewed. Although the available records and information are limited, two major genetic lineages of ZIKV were identified in previous studies.However, in this study, three lineages were identified based on a phylogenetic analysis of all virus sequences from Gen Bank, including those of the envelope protein(E) and non-structural protein 5(NS5) coding regions. The spatial and temporal distributions of the three identified ZIKV lineages and the recombination events and mechanisms underlying their divergence and evolution were further elaborated. The potential migration pathway of ZIKV was also characterized. Our findings revealed the central roles of two African countries, Senegal and Cote d'Ivoire, in ZIKV evolution and genotypic divergence. Furthermore, our results suggested that the outbreaks in Asia and the Pacific islands originated from Africa. 18Pathological features of COVID-19-associated lung injury:a preliminary proteomics report based on clinical samples显示文摘The COVID-19 pandemic has emerged as a global health emergency due to its association with severe pneumonia and relative high mortality.However,the molecular characteristics and pathological features underlying COVID-19 pneumonia remain largely unknown.To characterize molecular mechanisms underlying COVID-19 pathogenesis in the lung tissue using a proteomic approach,fresh lung tissues were obtained from newly deceased patients with COVID-19 pneumonia.After virus inactivation,a quantitative proteomic approach combined with bioinformatics analysis was used to detect proteomic changes in the SARS-CoV-2-infected lung tissues.We identified significant differentially expressed proteins involved in a variety of fundamental biological processes including cellular metabolism,blood coagulation,immune response,angiogenesis,and cell microenvironment regulation.Several inflammatory factors were upregulated,which was possibly caused by the activation of NF-κB signaling.Extensive dysregulation of the lung proteome in response to SARS-CoV-2 infection was discovered.Our results systematically outlined the molecular pathological features in terms of the lung response to SARS-CoV-2 infection,and provided the scientific basis for the therapeutic target that is urgently needed to control the COVID-19 pandemic.Ling Leng Ruiyuan Cao Jie Ma Danlei Mou Yunping Zhu Wei Li Luye Lv Dunqin Gao Shikun Zhang Feng Gong Lei Zhao Bintao Qiu Haiping Xiang Zhongjie Hu Yingmei Feng Yan Dai Jiang Zhao Zhihong Wu Hongjun Li Wu Zhong 2020Signal Transduction and Targeted Therapy2020,5,1:4

20Characterization of two monoclonal antibodies, 38F10 and 44D11, against the major envelope fusion protein of Helicoverpa armigera nucleopolyhedrovirus显示文摘The envelope fusion protein F of baculoviruses is a class I viral fusion protein which play a significant role during virus entry into insect cells. F is initially synthesized as a precursor(F_0) and then cleaved into a disulfide-linked F_1 and F_2 subunits during the process of protein maturation and secretion. To facilitate further investigation into the structure and function of F protein during virus infection, monoclonal antibodies(mAbs) against the F_2 subunit of Helicoverpa armigera nucleopolyhedrovirus(HearNPV)(Ha F) were generated. Two kinds of mAbs were obtained according to their different recognition epitopes: one kind of mAbs, as represented by 38F10,recognizes amino acid(aa) 85 to 123 of F_2 and the other kind, represented by 44D11, recognizes aa148 to 173 of F_2. Western blot and immunofluorescence assay confirmed that both of the mAbs recognized the F protein expressed in HearNPV infected cells, however, only 44D11 could neutralize HearNPV infection. The results further showed that 44D11 may not interact with a receptor binding epitope, rather it was demonstrated to inhibit syncytium formation in cells expressing the Ha F protein.
