1Coordinated Transcriptional Regulation by the UVB Photoreceptor and Multiple Transcription Factors for Plant UV-B Responses显示文摘Non-damaging ultraviolet B(UV-B)light promotes photomorphogenic development and stress acclimation through UV-B-specific signal transduction in Arabidopsis.UV-B irradiation induces monomerization and nuclear translocation of the UV-B photoreceptor UV RESISTANCE LOCUS 8(UVR8).However,it is not clear how the nuclear localization of UVR8 leads to changes in global gene expression.Here,we reveal that nuclear UVR8 governs UV-B-responsive transcriptional networks in concert with several previously known transcription factors,including ELONGATED HYPOCOTYL 5(HY5)and PHYTOCHROME INTERACTING FACTOR 4(PIF4).Based on the transcriptomic analysis,we identify MYB13 as a novel positive regulator in UV-B-induced cotyledon expansion and stress acclimation.MYB13 is UV-B inducible and is predominantly expressed in the cotyledons.Our results demonstrate that MYB13 protein functions as a transcription factor to regulate the expression of genes involved in auxin response and flavonoid biosynthesis through direct binding with their promoters.In addition,photoactivated UVR8 interacts with MYB13 in a UV-B-dependent manner and differentially modulates the affinity of MYB13 with its targets.Taken together,our results elucidate the cooperative function of the UV-B photoreceptor UVR8 with various transcription factors in the nucleus to orchestrate the expression of specific sets of downstream genes and,ultimately,mediate plant responses to UV-B light.Chongzhen Qian Zhiren Chen Qing Liu Weiwei Mao Yanling Chen Wei Tian Yan Liu Jiupan Han Xinhao Ouyang Xi Huang 2020Molecular Plant2020,13,5:13

7Plant responses to UV‑B radiation:signaling,acclimation and stress tolerance显示文摘Ultraviolet-B(UV-B)light is an intrinsic part of sunlight that reaches the earth’s surface,and affects plant survival and adaptation.How plants respond to UV-B light is regulated by the wavelength,intensity and duration of UV-B radiation,and is also regulated by photosynthetically active radiation perceived by phytochrome and cryptochrome photore-ceptors.Non-damaging UV-B light promotes plant photomorphogenesis and UV-B acclimation which enhances plant tolerance against UV-B stress.However,high-level UV-B radiation induces DNA damage,generates reactive oxygen species(ROS)and impairs photosynthesis.Plants have evolved efficient mechanisms to utilize informational UV-B signal,and protect themselves from UV-B stress.UV RESISTANCE LOCUS8(UVR8)is a conserved plant-specific UV-B photoreceptor.It interacts with CONSTITUTIVELY PHOTOMORPHOGENIC1(COP1)to initiate UV-B-specific light signal-ing and regulate UV-B responsive gene expression.A set of transcription factors such as ELONGATED HYPOCOTYL5(HY5)function downstream of the UVR8-COP1 module to promote seedling de-etiolation for photomorphogenic development and biosynthesis of sunscreen flavonoids for UV-B stress tolerance.In addition to UVR8 signaling path-ways,plants subjected to damaging UV-B radiation initiate stress protection and repair mechanisms through UVR8-independent pathways.In this review,we summarize the emerging mechanisms underlying UV-B stress acclimation and protection in plants,primarily revealed in the model plant Arabidopsis thaliana.Zhiren Chen Yuan Dong Xi Huang 2022Stress Biology2022,2,1:0
8m^(6)A mRNA modification potentiates Th17 functions to inflame autoimmunity显示文摘N6-methyladenosine(m^(6)A),the most common and abundant epigenetic RNA modification,governs mRNA metabolism to determine cell differentiation,proliferation and response to stimulation.m^(6)A methyltransferase METTL3 has been reported to control T cell homeostasis and sustain the suppressive function of regulatory T cells(Tregs).However,the role of m^(6)A methyltransferase in other subtypes of T cells remains unknown.T helper cells 17(Th17)play a pivotal role in host defense and autoimmunity.Here,we found that the loss of METTL3 in T cells caused serious defect of Th17 cell differentiation,and impeded the development of experimental autoimmune encephalomyelitis(EAE).We generated Mettl3f/fIl17aCre mice and observed that METTL3 deficiency in Th17 cells significantly suppressed the development of EAE and displayed less Th17 cell infiltration into central nervous system(CNS).Importantly,we demonstrated that depletion of METTL3 attenuated IL-17A and CCR5 expression by facilitating SOCS3 mRNA stability in Th17 cells,leading to disrupted Th17 cell differentiation and infiltration,and eventually attenuating the process of EAE.Collectively,our results highlight that m^(6)A modification sustains Th17 cell function,which provides new insights into the regulatory network of Th17 cells,and also implies a potential therapeutic target for Th17 cell mediated autoimmune disease.Xuefei Wang Chen Chen Hongwei Sun Kaiqiong Mao Jiameng Yao Weiqiao Zhang Meixiao Zhan Hua-Bing Li Zhiren Zhang Shu Zhu Ligong Lu 2023Science China(Life Sciences)2023,66,11:0
9Optical-field-induced Freeder-icksz transition of a nematic liquid crystal doped with dia-zophenyl显示文摘4[4’-(phenylazo)phenylazo]phenol was synthesized and characterized. Optical-induced Freedericksz transition of 5CB liquid crystal doped with trace diazophenyl was studied. It was observed that the doped 5CB dramatically decreased the optical field threshold, and the Freedericksz transition threshold occurred at optical intensity level of 0.3 mW/mm2 for the 12.5 μm thickness planar alignment doped 5CB, which is three orders of magnitude lower than that for the pure 5CB.
