| 6 | Antioxidant and immunomodulatory activities of a polysaccharide from Flammulina velutipes显示文摘OBJECTIVE: To evaluate the antioxidant and immunomodulatory activities of a unique polysaccharide from the medicinal fungus Flammulina velutipes in vitro.METHODS: Using water extraction and alcohol precipitation, crude polysaccharides were obtained. After purification by DEAE-cellulose 52 ion exchange chromatography and Sephacryl S-300 HR gel filtration chromatography, High performance liquid chromatography equipped with evaporative light-scattering detector, Infrared radiation and Nuclear magnetic resonance were used to evaluate the structure of the polysaccharide. Its immunomodulatory activity was measured by examining the production of nitric oxide(NO) and cytokine secretion, and via lymphocyte proliferation experiments. Its effects on the scavenging activities of hydroxyl radical, superoxide anion and reducing power were also measured.RESULTS: A water-soluble polysaccharide, Flammulina velutipes polysaccharide I-A(FVP I-A), was obtained with a molecular mass of 8.14×104Da determined by high performance gel permeation chromatography. An in vitro antioxidant assay indicated that FVP I-A could scavenge hydroxyl radical, superoxide anion and possessed reducing power and could largely promote NO production and augment the interleukin-1β, interleukin-6, and tumor necrosis factor-α secretion by RAW264.7 macrophages(P<0.05). | 8 | Cryptotanshinone suppresses key onco-proliferative and drugresistant pathways of chronic myeloid leukemia by targeting STAT5 and STAT3 phosphorylation显示文摘C-Myc and signal transducer and activator of transcription(STAT) family proteins have been proposed to be important downstream genes of BCR-ABL, which characterizes most cases of chronic myeloid leukemia(CML). Here, we report a c-Myc pathway-targeted screening of seven natural anticancer compounds, in which we identified cryptotanshinone as a highly promising agent for CML therapy. Cryptotanshinone depletes c-Myc in CML by repressing the phosphorylation of STAT5.Decreased viability of K562 cells correlated with p-STAT5 suppression. Unexpectedly, imatinib activates rather than inhibits the phosphorylation of STAT3 in K562 cells. We demonstrated that cryptotanshinone, as a dual inhibitor of p-STAT5 and p-STAT3,can effectively block IL-6-mediated STAT3 activation and reverse BCR-ABL kinase-independent drug resistance. Moreover, we showed that the epigenetic rebalance between decreased BCR-ABL/STAT5/c-Myc and enhanced STAT3/multi-drug resistance(MDR) pathways is characteristic of the cancer stem cell-like property of K562/ADR. Simultaneously suppressing these two pathways using cryptotanshinone proves to be critical for the malignant network redress and MDR reversal of K562/ADR. | 10 | Neutrophil-mimicking therapeutic nanoparticles for targeted chemotherapy of pancreatic carcinoma显示文摘Due to the critical correlation between inflammation and carcinogenesis, a therapeutic candidate with anti-inflammatory activity may find application in cancer therapy. Here, we report the therapeutic efficacy of celastrol as a promising candidate compound for treatment of pancreatic carcinoma via na?ve neutrophil membrane-coated poly(ethylene glycol) methyl ether-block-poly(lactic-co-glycolic acid)(PEG-PLGA) nanoparticles. Neutrophil membranecoated nanoparticles(NNPs) are well demonstrated to overcome the blood pancreas barrier to achieve pancreas-specific drug delivery in vivo. Using tumor-bearing mice xenograft model, NNPs showed selective accumulations at the tumor site following systemic administration as compared to nanoparticles without neutrophil membrane coating. In both orthotopic and ectopic tumor models,celastrol-loaded NNPs demonstrated greatly enhanced tumor inhibition which significantly prolonged the survival of tumor bearing mice and minimizing liver metastases. | 13 | Multifunctional polymeric micelle-based chemo-immunotherapy with immune checkpoint blockade for efficient treatment of orthotopic and metastatic breast cancer显示文摘Immunotherapy has become a highly promising paradigm for cancer treatment. Herein, a chemo-immunotherapy was developed by encapsulating chemotherapeutic drug doxorubicin(DOX) and Toll-like receptor 7 agonist imiquimod(IMQ) in low molecular weight heparin(LMWH)-D-α-tocopheryl succinate(TOS) micelles(LT). In this process, LMWH and TOS were conjugated by ester bond and they were not only served as the hydrophilic and hydrophobic segments of the carrier, but also exhibited strong anti-metastasis effect. The direct killing of tumor cells mediated by DOX-loaded micelles(LT-DOX)generated tumor-associated antigens, initiating tumor-specific immune responses in combination with IMQ-loaded micelles(LT-IMQ). Furthermore, the blockade of immune checkpoint with programmed cell death ligand 1(PD-L1) antibody further elevated the immune responses by up-regulating the maturation of DCs as well as the ratios of CD8+ CTLs/Treg and CD4+ Teff/Treg. | 16 | Rhizosphere and bulk soil enzyme activities in a Nothotsuga longibracteata forest in the Tianbaoyan National Nature Reserve,Fujian Province,China显示文摘The rhizosphere, distinct from bulk soil, is defined as the volume of soil around living roots and influenced by root activities. We investigated protease,invertase, cellulase, urease, and acid phosphatase activities in rhizosphere and bulk soils of six Nothotsuga longibracteata forest communities within Tianbaoyan National Nature Reserve, including N. longibracteata + either Phyllostachys pubescens, Schima superba, Rhododendron simiarum, Cunninghamia lanceolata, or Cyclobalanopsis glauca, and N. longibracteata pure forest. Rhizosphere soils possessed higher protease, invertase, cellulase, urease,and acid phosphatase activities than bulk soils. The highest invertase, urease, and acid phosphatase activities were observed in rhizosphere samples of N. longibracteata+ S.superba. Protease was highest in the N. longibracteata + R. simiarum rhizosphere, while cellulase was highest in the pure N. longibracteata forest rhizosphere.All samples exhibited obvious rhizosphere effects on enzyme activities with a significant linear correlation between acid phosphatase and cellulase activities(p \ 0.05) in rhizosphere soils and between protease and acid phosphatase activities(p \ 0.05) in bulk soils. | 19 | Investigation and Improvement of the Staggered Labyrinth Seal显示文摘Recent studies on staggered labyrinth seals have focused on the effects of different parameters,such as the pressure ratio and rotational speed on the leakage flow rate.However,few investigations pay sufficient attention to flow details and the sealing mechanism,which would be of practical importance in designing seals having higher performance.This paper establishes a theoretical model to study the seal mechanism,thus revealing that leakage is determined by the pressure ratio and geometric structure.Numerical simulation is implemented to illustrate details of the flow field within the seal structure.Viscous dissipation is used to quantitatively investigate the contribution that each location makes to the seal performance,revealing that orifices and stagnation points are the most important positions in the seal structure,generating the most dissipation.The orifice is carefully studied by using the theoretical model.Experiments for different pressure ratios are conducted and the results match well with those of the theoretical model and numerical simulation,verifying the theoretical model and analysis of the seal mechanism.Three new designs,based on a good understanding of the seal mechanism,are presented,with one reducing leakage by 24.5%. | LIN Zhirong WANG Xudong YUAN Xin SHIBUKAWA Naoki NOGUCHI Taro | 2015 | Chinese Journal of Mechanical Engineering2015,28,2: | 7 |
| 20 | Related gene expressions in anti-keratinocyte aging induced by Ganoderma lucidum polysaccharides显示文摘Objective:To examine the level of expression of anti skin aging gene of Ganoderma lucidum polysacchayides and clarify its mechanism with anti aging of this ancient Chinese medicine.Methods:HacaT cell of keratinocytes lines were cultured and treated with the polysaccharides.The total RNA was extracted with Trizol reagent and cDNA was synthesized by reverse thanscription.The obtained cDNAs were then fluorescently labeled with cy3 and cy5 respectively and hybridized with gene expressing pedigree cDNA chip.The images were scanned and analyzed with special software.The scan data were analyzed with software and checked by real time PCR.Results:Among total 18 346 human genes,the expression of 103 ones was up-regulated and 378 ones down-regulated.It was demonstrated evidently that Ganoderma lucidum polysaccharides affected the expression of genes of anti skin aging.Two ways are anastomotic.Conclusion:it is concluded by analysis of function of these up-regulation and down-regulation genes that Ganoderma lucidum polysaccharides may play an important role in boosting cell growth and against skin aging.It shows that the results of gene array reliable by real time PCR. | Xie Shaoqiong Liao Wanqing Yao Zhirong Wang Zhidong | 2008 | Journal of Medical Colleges of PLA(China)2008,23,3: | 6 |
