2Fermented soybean meal improves the growth performance, nutrient digestibility, and microbial flora in piglets显示文摘In order to increase nutritive values of soybean meal(SBM), 3 species of microbes were used to ferment SBM. Through a 3 x 3 orthogonal design and parameter measurements of soybean peptide and antinutritional factor contents in the fermented soybean meal(FSBM), it was estimated that the best microbial proportion of Bacillus subtilis, Hansenula anomala and Lactobacillus casei was 2:1:2 for SBM fermentation(P < 0.05). The further piglet feeding experiment showed that 10% FSBM substitute for SBM had no significant effect on growth performance of suckling piglets(d 7-28)(P> 0.05). However, newlyweaned piglets(d 28-38) fed 10% FSBM and different levels of plasma protein obtained higher average daily gain(ADG) and feed conversion ratio(FCR), compared with those without FSBM but with 6% plasma protein(P < 0.05). Piglets(d 38-68) fed diets supplemented with FSBM and soybean protein concentrate(SBPC) at 3.75% and 7.5% respectively increased nutrient digestibility,fecal enzyme activity and lactic acid bacteria counts, and decreased fecal Escherichia coli counts(P < 0.05), compared with the control. 3Simulation of shock wave buffet and its suppression on an OAT15A supercritical airfoil by IDDES显示文摘In the present paper,extremely unsteady shock wave buffet induced by strong shock wave/boundary-layer interactions (SWBLI) on the upper surface of an OAT15A supercritical airfoil at Mach number of 0.73 and angle of attack of 3.5 degrees is first numerically simulated by IDDES,one of the most advanced RANS/LES hybrid methods.The results imply that conventional URANS methods are unable to effectively predict the buffet phenomenon on the wing surface;IDDES,which involves more flow physics,predicted buffet phenomenon.Some complex flow phenomena are predicted and demonstrated,such as periodical oscillations of shock wave in the streamwise direction,strong shear layer detached from the shock wave due to SWBLI and plenty of small scale structures broken down by the shear layer instability and in the wake.The root mean square (RMS) of fluctuating pressure coefficients and streamwise range of shock wave oscillation reasonably agree with experimental data.Then,two vortex generators (VG) both with an inclination angle of 30 degrees to the main flow directions are mounted in front of the shock wave region on the upper surface to suppress shock wave buffet.The results show that shock wave buffet can be significantly suppressed by VGs,the RMS level of pressure in the buffet region is effectively reduced,and averaged shock wave position is obviously pushed downstream,resulting in increased total lift.HUANG JingBo XIAO ZhiXiang LIU Jian FU Song 2012Science China(Physics,Mechanics & Astronomy)2012,55,2:24
4A pilot phase Ⅱ study of neoadjuvant triplet chemotherapy regimen in patients with locally advanced resectable colon cancer显示文摘Objective: This study aims to investigate the feasibility, safety and efficacy of triplet regimen of neoadjuvant chemotherapy in patients with locally advanced resectable colon cancer.Methods: Patients with clinical stage IIIb colon cancer received a perioperative triple chemotherapy regimen(oxaliplatin 85 mg/m2 and irinotecan 150 mg/m2, combined with folinic acid 200 mg, 5-fluorouracil 500 mg bolus and then 2,400 mg/m2 by 44 h infusion or capecitabine 1 g/m2 or S-1 40–60 mg b.i.d orally d 1–10, repeated at 2-week intervals) for 4 cycles. Complete mesocolic excision was scheduled 2–6 weeks after completion of neoadjuvant treatment and followed by a further 6 cycles of FOLFOXIRI or XELOX. Primary outcome measures of this stage II trial were feasibility, safety, tolerance and efficacy of neoadjuvant treatment.Results: All 23 patients received neoadjuvant chemotherapy and underwent surgery. Twenty-one patients(91.3%) had reductions in tumor volume after neoadjuvant treatment, and 13 patients(56.5%) had grade 3–4toxicity. No patients had severe complications from surgery. 7Synergistic instability of coal pillar and roof system and filling method based on plate model显示文摘The security challenges from room and pillar gobs include land subsidence, spontaneous combustion of coal pillars and mine flood caused by gob water. To explore the instability mechanism of room and pillar gob, we established a mechanical model of elastic plate on elastic foundation in which pillars and hard roofs were considered as continuous Winkler foundations and elastic plates, respectively. The synergetic instability of pillar and roof system was analyzed based on plate bending theory and catastrophe theory. In addition, mechanical conditions and math criterion of roof failure and overall instability of coal pillar and roof system were given. Through analyzing both advantages and disadvantages of some technologies such as induced caving, filling, gob sealing and isolation, we presented a new filling method named box-filling, in view of box foundation theory, to control the disasters of ground collapse, water inrush and mine fire. In a gob's treatment project in Ordos, safety assessment and filling design of a room and pillar gob have been done by the mechanical model. The results show that the gob will collapse when the pillars' average yield band is wider than 0.93 m, and box-filling can control land collapse, mine flood and mine fire economically and efficiently. 8Fault zone structures of northern and southern portions of the main central fault generated by the 2008 Wenchuan earthquake using fault zone trapped waves显示文摘The rupture process of the May 12,2008 MS8.0 Wenchuan earthquake was very complex. To study the rupture zones generated by this earthquake,four dense temporary seismic arrays across the two surface breaking traces of the main-shock were deployed in July and recorded a great amount of aftershocks. This paper focuses on the data interpretation of two arrays across the central main fault,the northern array line 1 and southern array line 3. The fault zone trapped waves recorded by the two arrays were used to study the structure of the central main fault and the difference between the northern and southern portions. The results show that the widths of the rupture zone are about 170?200 m and 200?230 m for northern and southern portions respectively. And the corresponding dip angles are 80° and 70°. The seismic velocity inside the fracture zone is about one half of the host rock. By comparison,the northern portion of the rupture zone is slightly narrower and steeper than the southern portion. Besides these differences,one more interesting and important difference is the positions of the rupture zone with respect to surface breaking traces. At the northern portion,the rupture zone is centered at the surface breaking trace,while at the southern portion it is not but is shifted to the northwest. This difference reflects the difference of rupture behaviors between two portions of the central main fault. The width of the rupture zone is smaller than that of M8.1 Kunlun earthquake though these two earthquakes have almost the same magnitudes. 12Numerical investigation of unsteady vortex breakdown past 80°/65° double-delta wing显示文摘An improved delayed detached eddy simulation(IDDES) method based on the k-x-SST(shear stress transport) turbulence model was applied to predict the unsteady vortex breakdown past an 80 /65 double-delta wing(DDW), where the angles of attack(AOAs) range from 30 to 40. Firstly, the IDDES model and the relative numerical methods were validated by simulating the massively separated flow around an NACA0021 straight wing at the AOA of 60.The fluctuation properties of the lift and pressure coefficients were analyzed and compared with the available measurements. For the DDW case, the computations were compared with such measurements as the mean lift, drag, pitching moment, pressure coefficients and breakdown locations.Furthermore, the unsteady properties were investigated in detail, such as the frequencies of force and moments, pressure fluctuation on the upper surface, typical vortex breakdown patterns at three moments, and the distributions of kinetic turbulence energy at a stream wise section.Two dominated modes are observed, in which their Strouhal numbers are 1.0 at the AOAs of30, 32 and 34 and 0.7 at the AOAs of 36, 38 and 40. The breakdown vortex always moves upstream and downstream and its types change alternatively. 13Tribocorrosion behavior in artificial seawater and anti-microbiologically influenced corrosion properties of TiSiN-Cu coating on F690 steel显示文摘The TiSiN-Cu nanocomposite coating was deposited on F690 steel substrate by arc ion plating. The structure and composition, tribocorrosion behavior and anti-microbiologically influenced corrosion(MIC)properties of TiSiN-Cu coating were investigated. The results show that the TiSiN-Cu coating has unique nanocomposite structures. The results of tribocorrosion show that the potential and current change of F690 steel and TiSiN-Cu coatings tend to be opposite. The reason is that the F690 steel is non-passivated metal and the TiSiN-Cu coating has passivation phenomenon. The TiSiN-Cu coating possesses excellent tribocorrosion resistance. 15The effects of autophagy on vascular endothelial cells induced by airborne PM2.5显示文摘The purpose of this study was to examine the direct toxicity of PM2.5 collected from Beijing on human umbilical vein endothelial cells(HUVEC). A Cell Counting Kit 8(CCK8) assay demonstrated that PM2.5 exposure decreased the proliferation of HUVECs in a dosedependent manner. We also found that PM2.5 exposure induced autophagy in HUVECs, as evidenced by:(1) an increased number of double-membrane vesicles;(2) enhanced conversion and punctuation of the microtubule-associated protein light chain 3(LC3); and(3) decreased levels of the selective autophagy substrate p62 in a time-dependent manner.Furthermore, promoting autophagy in PM2.5-exposed HUVECs with rapamycin increased the cell survival rate, whereas inhibiting autophagy via 3-methyladenine significantly decreased cell survival. These results demonstrate that PM2.5 exposure can induce cytotoxicity and autophagy in HUVECs and that autophagy play a protective role against PM2.5-induced cytotoxicity. 16Thermodynamic performance of Dual-Miller cycle(DMC) with polytropic processes based on power output, thermal efficiency and ecological function显示文摘This study reports a new model of an air standard Dual-Miller cycle(DMC) with two polytropic processes and heat transfer loss.The two reversible adiabatic processes which could not be realized in practice are replaced with two polytropic processes in order to more accurately reflect the practical working performance. The heat transfer loss is taken into account. The expressions of power output, thermal efficiency, entropy generation rate(EGR) and ecological function are addressed using finite-time thermodynamic theory. Through numerical calculations, the influences of compression ratio, cut-off ratio and polytropic exponent on the performance are thermodynamically analyzed. The model can be simplified to other cycle models under specific conditions, which means the results have an certain universality and may be helpful in the design of practical heat engines. 17Optimization of clay material mixture ratio and filling process in gypsum mine goaf显示文摘Because there is neither waste rock nor mill tailings in the gypsum mine, and the buildings on the goaf of gypsum mine are needed to be protected, the research proposed the scheme of the clay filling technology. Gypsum, cement, lime and water glass were used as adhesive, and the strength of different material ratios were investigated in this study. The influence factors of clay strength were obtained in the order of cement, gypsum, water glass and lime. The results show that the cement content is the determinant influence factor, and gypsum has positive effects, while the water glass can enhance both clay strength and the fluidity of the filing slurry. Furthermore, combining chaotic optimization method with neural network, the optimal ratio of composite cementing agent was obtained. The results show that the optimal ratio of water glass, cement, lime and clay (in quality) is 1.17:6.74:4.17:87.92 in the process of bottom self-flow filling, while the optimal ratio is 1.78:9.58:4.71:83.93 for roof-contacted filling. A novel filling process to fill in gypsum mine goaf with clay is established. 18Fluorination vs. chlorination: a case study on high performance organic photovoltaic materials显示文摘Halogenation is a very efficient chemical modification method to tune the molecular energy levels, absorption spectra and molecular packing of organic semiconductors. Recently, in the field of organic solar cells(OSCs), both fluorine-and chlorinesubstituted photovoltaic materials, including donors and acceptors, demonstrated their great potentials in achieving high power conversion efficiencies(PCEs), raising a question that how to make a decision between fluorination and chlorination when designing materials. Herein, we systemically studied the impact of fluorination and chlorination on the properties of resulting donors(PBDB-T-2 F and PBDB-T-2 Cl) and acceptors(IT-4 F and IT-4 Cl). The results suggest that all the OSCs based on different donor and acceptor combinations can deliver good PCEs around 13%–14%. Chlorination is more effective than fluorination in downshifting the molecular energy levels and broadening the absorption spectra. The influence of chlorination and fluorination on the crystallinity of the resulting materials is dependent on their introduction positions. 19Defined tumor antigen-specific T cells potentiate personalized TCR-T cell therapy and prediction of immunotherapy response显示文摘Personalized immunotherapy targeting tumor-specific antigens(TSAs)could generate efficient and safe antitumor immune response without damaging normal tissues.Although neoantigen vaccines have shown therapeutic effect in clinic trials,precise prediction of neoantigens from tumor mutations is still challenging.The host antitumor immune response selects and activates T cells recognizing tumor antigens.Hence,T cells engineered with T-cell receptors(TCRs)from these naturally occurring tumor antigen-specific T(Tas)cells in a patient will target personal TSAs in his/her tumor.To establish such a personalized TCR-T cell therapy,we comprehensively characterized T cells in tumor and its adjacent tissues by single-cell mRNA sequencing(scRNA-seq),TCR sequencing(TCR-seq)and in vitro neoantigen stimulation.Compared to bystander T cells circulating among tissues,Tas cells were characterized by tumor enrichment,tumor-specific clonal expansion and neoantigen specificity.We found that CXCL13 is a unique marker for both CD4^(+)and CD8^(+)Tas cells.Importantly,TCR-T cells expressing TCRs from Tas cells showed significant therapeutic effects on autologous patient-derived xenograft(PDX)tumors.Intratumoral Tas cell levels measured by CXCL13 expression precisely predicted the response to immune checkpoint blockade,indicating a critical role of Tas cells in the antitumor immunity.We further identified CD200 and ENTPD1 as surface markers for CD4^(^(+))and CD8^(^(+))Tas cells respectively,which enabled the isolation of Tas cells from tumor by Fluorescence Activating Cell Sorter(FACS)sorting.Overall,our results suggest that TCR-T cells engineered with Tas TCRs are a promising agent for personalized immunotherapy,and intratumoral Tas cell levels determine the response to immunotherapy.Jingjing He Xinxin Xiong Han Yang Dandan Li Xuefei Liu Shuo Li Shuangye Liao Siyu Chen Xizhi Wen Kuai Yu Lingyi Fu Xingjun Dong Kaiyu Zhu Xiaojun Xia Tiebang Kang Chaochao Bian Xiang Li Haiping Liu Peirong Ding Xiaoshi Zhang Zhenjiang Liu Wende Li Zhixiang Zuo Penghui Zhou 2022Cell Research2022,32,6:10
