| 1 | USP19 suppresses inflammation and promotes M2-like macrophage polarization by manipulating NLRP3 function via autophagy显示文摘Macrophage polarization to proinflammatory M1-like or anti-inflammatory M2-like cells is critical to mount a host defense or repair tissue.The exact molecular mechanisms controlling this process are still elusive.Here,we report that ubiquitin-specific protease 19(USP19)acts as an anti-inflammatory switch that inhibits inflammatory responses and promotes M2-like macrophage polarization.USP19 inhibited NLRP3 inflammasome activation by increasing autophagy flux and decreasing the generation of mitochondrial reactive oxygen species.In addition,USP19 inhibited the proteasomal degradation of inflammasome-independent NLRP3 by cleaving its polyubiquitin chains.USP19-stabilized NLRP3 promoted M2-like macrophage polarization by direct association with interferon regulatory factor 4,thereby preventing its p62-mediated selective autophagic degradation.Consistent with these observations,compared to wild-type mice,Usp19−/−mice had decreased M2-like macrophage polarization and increased interleukin-1βsecretion,in response to alum and chitin injections.Thus,we have uncovered an unexpected mechanism by which USP19 switches the proinflammatory function of NLRP3 into an anti-inflammatory function,and suggest that USP19 is a potential therapeutic target for inflammatory interventions. | Tao Liu Liqiu Wang Puping Liang Xiaojuan Wang Yukun Liu Jing Cai Yuanchu She Dan Wang Zhi Wang Zhiyong Guo Samuel Bates Xiaojun Xia Junjiu Huang Jun Cui | 2021 | Cellular & Molecular Immunology2021,18,10: | 15 |
| 2 | Incidence and mortality of primary bone cancers in China, 2014显示文摘Objective: In this study, we aimed to estimate the updated incidence and mortality of primary bone cancers based on population-based cancer registration data in 2014, collected by the National Central Cancer Registry of China(NCCRC).Methods: In 2017, 339 registries' data were qualified based on data quality criteria set down by the NCCRC.Cases of primary bone cancers were retrieved from the national database. We estimated numbers of primary bone cancer cases and deaths in China using age-specific rates and corresponding national population stratified by area,sex, age-group(0, 1-4, 5-9, 10-14, …, 85+). Chinese standard population in 2000 and Segi's World population were applied for the calculation of age-standardized incidence and mortality rates.Results: In 2014, 24,000 primary bone cancer cases and 17,200 deaths attributable to primary bone cancers were estimated to have occurred in China. The crude incidence rate of primary bone cancers was 1.76/100,000, with agestandardized incidence rate by Chinese standard population(ASIRC) and by World standard population(ASIRW)being 1.35/100,000 and 1.32/100,000, respectively. The crude mortality rate of primary bone cancers was1.26/100,000, with age-standardized mortality rate by Chinese standard population(ASMRC) and by World standard population(ASMRW) being 0.88/100,000 and 0.86/100,000, respectively. Age-specific incidence curve was bimodally distributed with age, with the first peak occurring in the second decade of the life and the second peak in the elderly. Males had higher crude and age-standardized rates for both incidence and mortality compared with females. Both crude and age-standardized incidence rates were higher in rural areas than in urban areas, so were the crude and age-standardized mortality rates.Conclusions: This population-based study presents the most recently available estimates on primary bone cancers in China, revealing that the males are 1.34 times as much as females suffering from primary bone cancers and the adolescents in puberty and the elderly are predominantly affected groups by these cancers. | 3 | Defined tumor antigen-specific T cells potentiate personalized TCR-T cell therapy and prediction of immunotherapy response显示文摘Personalized immunotherapy targeting tumor-specific antigens(TSAs)could generate efficient and safe antitumor immune response without damaging normal tissues.Although neoantigen vaccines have shown therapeutic effect in clinic trials,precise prediction of neoantigens from tumor mutations is still challenging.The host antitumor immune response selects and activates T cells recognizing tumor antigens.Hence,T cells engineered with T-cell receptors(TCRs)from these naturally occurring tumor antigen-specific T(Tas)cells in a patient will target personal TSAs in his/her tumor.To establish such a personalized TCR-T cell therapy,we comprehensively characterized T cells in tumor and its adjacent tissues by single-cell mRNA sequencing(scRNA-seq),TCR sequencing(TCR-seq)and in vitro neoantigen stimulation.Compared to bystander T cells circulating among tissues,Tas cells were characterized by tumor enrichment,tumor-specific clonal expansion and neoantigen specificity.We found that CXCL13 is a unique marker for both CD4^(+)and CD8^(+)Tas cells.Importantly,TCR-T cells expressing TCRs from Tas cells showed significant therapeutic effects on autologous patient-derived xenograft(PDX)tumors.Intratumoral Tas cell levels measured by CXCL13 expression precisely predicted the response to immune checkpoint blockade,indicating a critical role of Tas cells in the antitumor immunity.We further identified CD200 and ENTPD1 as surface markers for CD4^(^(+))and CD8^(^(+))Tas cells respectively,which enabled the isolation of Tas cells from tumor by Fluorescence Activating Cell Sorter(FACS)sorting.Overall,our results suggest that TCR-T cells engineered with Tas TCRs are a promising agent for personalized immunotherapy,and intratumoral Tas cell levels determine the response to immunotherapy. | Jingjing He Xinxin Xiong Han Yang Dandan Li Xuefei Liu Shuo Li Shuangye Liao Siyu Chen Xizhi Wen Kuai Yu Lingyi Fu Xingjun Dong Kaiyu Zhu Xiaojun Xia Tiebang Kang Chaochao Bian Xiang Li Haiping Liu Peirong Ding Xiaoshi Zhang Zhenjiang Liu Wende Li Zhixiang Zuo Penghui Zhou | 2022 | Cell Research2022,32,6: | 10 |
| 4 | Cancer incidence and mortality in Gansu province,2012显示文摘Objective: Population-based cancer registration data in 2012 from all available cancer registries in Gansu province were collected by the Central Cancer Registry of Gansu. The numbers of new cancer cases and cancer deaths in Gansu province with compiled cancer incidence and mortality rates were estimated.Methods: In 2015, data from 7 registries in Gansu province were qualified. The pooled data were stratified by area(urban/rural), gender, age group(0, 1-4, 5-9, 10-14, …, 85+) and cancer type. New cancer cases and deaths were estimated using age-specific rates and corresponding population of Gansu province in 2012. The Chinese census data in 2000 and Segi's population were applied for age-standardized rates. All the rates were expressed per 100,000 person-years.Results: Qualified 7 cancer registries(3 urban and 4 rural registries) covered 2,956,560 populations of Gansu province in 2012. The percentage of cases morphologically verified(MV%) and death certificate-only cases(DCO%) were 72.41% and 1.65%, respectively, and the mortality to incidence rate ratio(M/I) was 0.63. It was estimated that there were 575,600 new cancer cases and 331,300 cancer deaths in Gansu province in 2012. The incidence rate was 223.29/100,000(244.14/100,000 in males and 201.50/100,000 in females), the age-standardized incidence rates by Chinese standard population(ASIRC) and by world standard population(ASIRW) were 208.95/100,000 and 206.41/100,000 with the cumulative incidence rate(0-74 years old) of 22.49%. The crude incidence rate in urban areas was equal to that in rural areas. However, after adjusted by age, the cancer incidence rate in urban was the same as that of rural areas. The crude mortality in Gansu province was 128.54/100,000(135.04/100,000 in males and 124.43/100,000 in females), the age-standardized mortality rates by Chinese standard population(ASMRC) and by world standard population(ASMRW) were 109.54/100,000 and 108.44/100,000, respectively, and the cumulative mortality rate(0-74 years old) was 12.91%. The crude cancer mortality rate in urban areas was higher than that of rural areas. However, the agestandardized rate in urban areas was the same as that of rural areas. Cancers of stomach, liver, lung, esophagus, female breast, colorectum, cervix, brain, pancreas and ovary were the most common cancers, accounting for about 83.56% of all cancer new cases. Stomach cancer, liver cancer, lung cancer, esophageal cancer, female breast cancer, colorectal cancer, brain cancer, lymphoma, pancreatic cancer and cervix cancer were the leading causes of cancer death, accounting for about 85.58% of all cancer deaths. The burden between urban and rural, males and females was different.Conclusions: Registration data of Gansu province were qualified to provide basic information on population-based cancer incidence, mortality for cancer prevention and control. The digestive tract cancer burden in Gansu province, especially for males in rural areas, was higher. The incidence rate of female breast cancer was higher in urban areas. | 11 | Simple hybrid dithiafulvenes-triphenylamine systems as dopant-free hole-transporting materials for efficient perovskite solar cells显示文摘Two extended hybrid conjugated systems based on a triphenylamine(TPA) core with two and three peripheral 1,4-dithiafulvenes(DTF) units coded WH-2 and WH-3 as hole-transporting materials(HTMs) applied in perovskite solar cells(PSCs) are synthesized by facile one-step reaction in good yield over 75%. DTF unit as electron donor can enhance the electron donating ability and the fusion of benzenic ring of TPA with DTF unit may lead to reinforced intermolecular interactions in the solid state. In addition,WH-2 and WH-3 exhibit a pyramid shape containing partial planarity and quasi three-dimensionality features, which is also conducive to enhancing the π-π stacking of molecules in the solid state. The above-mentioned structural characteristics make the two HTMs have good hole mobilities. As a result,WH-2 and WH-3 obtained the high intrinsic hole mobilities of 4.69 × 10^(-4)and 2.18 × 10^(-3)cm^(2)V^(-1)s^(-1)respectively.
