1Soil water depletion depth by planted vegetation on the Loess Plateau显示文摘Evapotranspiration of much planted vegetation exceeds precipitation, and this can deplete soil water and cause a deep dry layer in the soil profile, which is a serious obstacle to sustainable land use on the Loess Plateau, China. This study aimed to determine water depletion depth of planted grassland, shrub, and forest in a semiarid area on the Loess Plateau. Soil moisture of five vegetation types was measured to >20 m in depth. The vegetation types were crop, natural grasse, seven-year-old planted alfalfa (Medicago sativa L.), 23-year-old planted caragana (Caragana microphylla Lam.) shrub, and 23-year-old planted pine (Pinus tabulaeformis L) forest land. Through comparing moisture of planted alfalfa grass, caragana shrub, and pine forest to crop and natural grassland, the depth and amount of soil water consumed by grassland, caragana brush and pine forest was determined. 2The Chromosome-Level Genome Sequence of the Autotetraploid Alfalfa and Resequencing of Core Germplasms Provide Genomic Resources for Alfalfa Research显示文摘Alfalfa(Medicago sativa)is one of the most important forage crops in the world;however,its molecular genetics and breeding research are hindered due to the lack of a high-quality reference genome.Here,we report a de novo assembled 816-Mb high-quality,chromosome-level haploid genome sequence for‘Zhongmu No.1’alfalfa,a heterozygous autotetraploid.The contig N50 is 3.92 Mb,and 49165 genes are annotated in the genome.The alfalfa genome is estimated to have diverged from M.truncatula approximately 8 million years ago.Genomic population analysis of 162 alfalfa accessions revealed high genetic diversity,weak population structure,and extensive gene flow from wild to cultivated alfalfa.Genome-wide association studies identified many candidate genes associated with important agronomic traits.Furthermore,we showed that MsFTa2,a Flowering Locus T homolog,whose expression is upregulated in salt-resistant germplasms,may be associated with fall dormancy and salt resistance.Taken together,these genomic resources will facilitate alfalfa genetic research and agronomic improvement.Chen Shen Huilong Du Zhuo Chen Hongwei Lu Fugui Zhu Hong Chen Xiangzhao Meng Qianwen Liu Peng Liu Lihua Zheng Xiuxiu Li Jiangli Dong Chengzhi Liang Tao Wang 2020Molecular Plant2020,13,9:21
3Influence of lactic acid bacteria, cellulase, cellulase-producing Bacillus pumilus and their combinations on alfalfa silage quality显示文摘This study assessed the effects of lactic acid bacteria(LAB), cellulase, cellulase-producing Bacillus pumilus and their combinations on the fermentation characteristics, chemical composition, bacterial community and in vitro digestibility of alfalfa silage. A completely randomized design involving a 8(silage additives)×3 or 2(silage days) factorial arrangement of treatments was adopted in the present study. The 8 silage additive treatments were: untreated alfalfa(control); two commercial additives(GFJ and Chikuso-1); an originally selected LAB(Lactobacillus plantarum a214) isolated from alfalfa silage; a cellulase-producing Bacillus(CB) isolated from fresh alfalfa; cellulase(C); and the combined additives(a214+C and a214+CB). Silage fermentation characteristics, chemical composition and microorganism populations were analysed after 30, 60 and 65 days(60 days followed by exposure to air for five additional days). In vitro digestibility was analysed for 30 and 60 days. Compared with the other treatments, selected LAB a214, a214 combined with either C or CB, and Chikuso-1 had the decreased(P<0.001) pH and increased(P<0.001) lactic acid concentrations during the ensiling process, and there were no differences(P>0.05) among them. Fiber degradation was not significant(P≥0.054) in any C or CB treatments. The a214 treatment showed the highest(P=0.009) in vitro digestibility of dry matter(595.0 g kg^(–1) DM) after ensiling and the highest abundance of Lactobacillus(69.42 and 79.81%, respectively) on days 60 and 65, compared to all of other treatments. 4Alfalfa-containing diets alter luminal microbiota structure and short chain fatty acid sensing in the caecal mucosa of pigs显示文摘Background: Pork produced by outdoor-reared pigs raised mostly on alfalfa pastures attracts increasing population of consumer from most of the world. In China, pigs were raised with alfalfa-containing diets to seek for good quality pork.However, the influence of dietary alfalfa involving high level of insoluble dietary fiber(IDF) on pig intestinal luminal microbiota composition remains unclear. The objective of this study was to investigate the effects of alfalfa on luminal microbiota and short chain fatty acids(SCFA) production, and gene expressions involved in SCFA sensing, transporting and absorbing in pig caecal mucosa.Results: Twenty-four growing pigs were randomly allotted to four diets containing 0%, 5%, 10% and 15% alfalfa meal for a 28-d experiment. Ingestion of alfalfa meal-contained diets significantly increased the ratio of body weight gain to feed consumption. Illumina MiS eq sequencing of the V3 region of the 16 S r RNA genes showed that alfalfa-containing diet significantly decreased the relative abundance of genera Turicibacter, Acidiphilium, Paracoccus, Propionibacterium,Corynebacterium, Pseudomonas, Acinetobacter, and Staphylococcus, and increased the relative abundance of genera Lachnospira, Marvinbryantia, and Desulfovibrio in the caecal digesta. Butyrate concentration was significantly increased in the hindgut by the supplementation of alfalfa meal in diets. 5Effects of alfalfa coverage on runoff,erosion and hydraulic characteristics of overland flow on loess slope plots显示文摘An evaluation of the interactions between vegetation,overland and soil erosion can provide valuable insight for the conservation of soil and water.An experiment was conducted to study water infiltration,runoff generation process,rate of sediment erosion,and hydrodynamic characteristics of overland flow from a sloping hillside with different draw-off discharges from alfalfa and control plots with 20°slope.The effect of alfalfa on runoff and sediment transport reduction was quantitatively analyzed.Alfalfa was discussed for its ability to reduce the overland flow scouring force or change the runoff movement.Compared to the bare-soil plots,alfalfa plots generated a 1.77 times increase in infiltration rate.Furthermore,the down-slope water infiltration rate for the bare soil plots was higher than in the up-slope,while the opposite was found in the alfalfa plots.In addition,alfalfa had a significant effect on runoff and sediment yield.In comparison to the control,the runoff coefficient and sediment transportation rate decreased by 28.3%and 78.4%in the grass slope,respectively.The runoff generated from the alfalfa and bare-soil plots had similar trends with an initial increase and subsequent leveling to a steady-state rate.The transport of sediment reduced with time as a consequence of the depletion of loose surface materials.The maximum sediment concentration was recorded within the first few minutes of each event.The alfalfa plots had subcritical flow while the baresoil plots had supercritical flow,which indicate that the capability of the alfalfa slope for resisting soil erosion and sediment movement was greater than for bare soil plots.Moreover,the flow resistance coefficient and roughness coefficient for the alfalfa plots were both higher than for the bare-soil plots,which indicate that overland flow in alfalfa plots had retarded and was blocked,and the flow energy along the runoff path had gradually dissipated.Finally,the ability to erode and transport sediment had decreased.Shufang WU Pute WU Hao FENG G.P.Merkley 2011Frontiers of Environmental Science & Engineering2011,5,1:11
6卫星搭载对紫花苜蓿当代植株生物量的影响(英文)显示文摘[Objective] The aim of this study was to investigate the effect of space flight factors on plant biomass in the generation of alfalfa carried by the satellite.[Method]Seeds from three lines of alfalfa were carried by the seed-breeding satellite Shijian-8.After the satellite returned to the ground,stem diameter,primary branch number and current-year individual biomass of alfalfa were studied.[Result]After space flight,primary branch number and current-year individual biomass of alfalfa increased significantly,while the stem diameter had no significant change.Using the value over(the mean value of control + three standard deviation)as a criterion to screen,the variants with enlarged stem diameter,increased primary branch number and individual biomass was two,five and twelve respectively.[Conclusion]The obtained variants can be used in the variety improvement of alfalfa and its new variety breeding,but whether its favorable variation can inherit stably to the progenies needs further study.任卫波 赵亮 王蜜 陈立波 郭慧琴 2008Agricultural Science & Technology2008,9,5:10
7Biological mechanism of controlling cotton aphid (Homoptera: aphididae) by the marginal alfalfa zone surrounding cotton field显示文摘At the beginning stage of cotton aphids (Aphis gossypii Glover) invading the cotton field, large numbers of insects have accumulated in the marginal alfalfa (Medicago sativa L.) zone surrounding it. They are 6.94 times (excluded natural enemies) as many as the aphids in cotton field. Most of these insects are Therioaphid maculata (Buckton), and occur in fields 10-15 d earlier than cotton aphids. Consequently, abundant natural enemies have bred in alfalfa zone when cotton aphids increase rapidly in cotton fields in the middle of June in Xinhe County, Xinjiang Autonomous Region. The total number of the major natural enemies, including Coccinellids, Chrysopids and Syphid flies in alfalfa zone, is 13.65 times that in cotton field within the same acreage of fields. 8Recent progress in alfalfa(Medicago sativa L.)genomics and genomic selection显示文摘Alfalfa(M. sativa L.) is a highly valuable forage crop, providing >58 Mt of hay, silage, and pasture each year in the United States. As alfalfa is an outcrossing autotetraploid crop,however, breeding for enhanced agronomic traits is challenging and progress has historically not been rapid. Methods that make use of genotypic information and statistical models to generate a genomic estimated breeding value(GEBV) for each plant at a young age hold a great deal of promise to accelerate breeding gains. An emerging genomic breeding pipeline employs SNP chips or genotyping-by-sequencing(GBS) to identify SNP markers in a training population, followed by the use of a statistical model to find associations between the discovered SNPs and traits of interest, followed by genomic selection(GS), a breeding program utilizing the trained model to predict breeding values and making selections based on the estimated breeding value(EBV). Much work has been done in recent years in all of these areas, to generate marker sets and discover SNPs associated with desirable traits, and the application of these technologies in alfalfa breeding programs is under way. However, GBS/GWAS/GS is still a new breeding paradigm,and work is ongoing to evaluate different models, software, and methods for use in such programs. 9Proteomic analysis of salt and osmotic-drought stress in alfalfa seedlings显示文摘Alfalfa is widely grown and is one of the most important forage crops in the world, but its growth and biomass production are markedly reduced under salt and drought stress, particularly during the early seedling stage. To identify the mechanisms behind salt and drought responsiveness at the alfalfa seedling stage, the proteins expressed were analyzed under no-treatment, 200 mol L–1 Na Cl and 180 g L–1 PEG treatment conditions during the seedling stage. Out of more than 800 protein spots detected on two-dimensional electrophoresis(2-DE) g els, 35 proteins showed statistically significant responses(P<0.05) to Na Cl and PEG stress, which were selected for tandem mass spectrometric identification, owing to their good resolution and abundance levels, and 32 proteins were positively identified. The identified proteins were divided into seven functional categories: photosynthetic metabolism, protein biosynthesis, folding and assembly, carbohydrate metabolism-associated proteins, stress defense related protein, metabolism of nucleic acid, other function categories and unknown proteins. Our results suggested that these proteins may play roles in alfalfa adaptation to salt and drought stress. 10China’s alfalfa market and imports: Development, trends, and potential impacts of the U.S.-China trade dispute and retaliations显示文摘This study examines the development and trends of China’s alfalfa market and imports, identifies key factors for the rapid increase in China’s alfalfa imports, and discusses potential impacts of the U.S.-China trade dispute and retaliations on the alfalfa markets and trade in both nations. China’s rapid transition toward larger-scale commercial dairy production, with enhanced feed and cost management as well as quality and safety control, and its limited resources for high-quality alfalfa production are key factors for the dramatic increase in its alfalfa imports, from 19 601 metric tons in 2008 to 1.38 million metric tons(mmt) in 2018. While the United States dominated China’s alfalfa imports with an average share of 97.01% from 2007 to 2017, the share dropped to 83.76% in 2018 and 63.28% in January 2019 due to the trade dispute and retaliations started in 2018. China will likely remain a large importer of alfalfa because of both its growing demand and the comparative advantages of imported alfalfa in quality and price, but the imports from the United States will be highly affected by the ongoing trade dispute and negotiations. 11Overexpression of vacuolar proton pump ATPase(V-H^+-ATPase) subunits B, C and H confers tolerance to salt and saline-alkali stresses in transgenic alfalfa(Medicago sativa L.)显示文摘The vacuolar proton pump ATPase(V-H^+-ATPase), which is a multi-subunit membrane protein complex, plays a major role in the activation of ion and nutrient transport and has been suggested to be involved in several physiological processes, such as cell expansion and salt tolerance. In this study, three genes encoding V-H^+-ATPase subunits B(Sc VHA-B, Gen Bank: JF826506), C(Sc VHA-C, Gen Bank: JF826507) and H(Sc VHA-H, Gen Bank: JF826508) were isolated from the halophyte Suaeda corniculata. The transcript levels of Sc VHA-B, Sc VHA-C and Sc VHA-H were increased by salt, drought and saline-alkali treatments. V-H^+-ATPase activity was also examined under salt, drought and saline-alkali stresses. The results showed that V-H^+-ATPase activity was correlated with salt, drought and saline-alkali stress. Furthermore, V-H^+-ATPase subunits B, C and H(Sc VHA-B, Sc VHA-C and Sc VHA-H) from S. corniculata were introduced separately into the alfalfa genome. The transgenic alfalfa was verified by Southern and Northern blot analysis. During salt and saline-alkali stresses, transgenic lines carrying the B, C and H subunits had higher germination rates than the wild type(WT). More free proline, higher superoxide dismutase(SOD) activity and lower malondialdehyde(MDA) levels were detected in the transgenic plants under salt and saline-alkali treatments. Moreover, the Sc VHA-B transgenic lines showed greater tolerance to salt and saline-alkali stresses than the WT. 12A global alfalfa diversity panel reveals genomic selection signatures in Chinese varieties and genomic associations with root development显示文摘Alfalfa(Medicago sativa L.) is an important forage crop worldwide. However, little is known about the effects of breeding status and different geographical populations on alfalfa improvement. Here, we sequenced 220 alfalfa core germplasms and determined that Chinese alfalfa cultivars form an independent group, as evidenced by comparisons of FSTvalues between different subgroups, suggesting that geographical origin plays an important role in group differentiation. By tracing the influence of geographical regions on the genetic diversity of alfalfa varieties in China, we identified 350 common candidate genetic regions and 548 genes under selection. We also defined 165 loci associated with 24 important traits from genome-wide association studies. Of those, 17 genomic regions closely associated with a given phenotype were under selection, with the underlying haplotypes showing significant differences between subgroups of distinct geographical origins. Based on results from expression analysis and association mapping,we propose that 6-phosphogluconolactonase(MsPGL) and a gene encoding a protein with NHL domains(MsNHL) are critical candidate genes for root growth. 13A chromosome-scale genome assembly of a diploid alfalfa,the progenitor of autotetraploid alfalfa显示文摘Alfalfa(Medicago sativa L.)is one of the most important and widely cultivated forage crops.It is commonly used as a vegetable and medicinal herb because of its excellent nutritional quality and significant economic value.Based on Illumina,Nanopore and Hi-C data,we assembled a chromosome-scale assembly of Medicago sativa spp.caerulea(voucher PI464715),the direct diploid progenitor of autotetraploid alfalfa.The assembled genome comprises 793.2 Mb of genomic sequence and 47,202 annotated protein-coding genes.The contig N50 length is 3.86 Mb.This genome is almost twofold larger and contains more annotated protein-coding genes than that of its close relative,Medicago truncatula(420 Mb and 44,623 genes).The more expanded gene families compared with those in M.truncatula and the expansion of repetitive elements rather than whole-genome duplication(i.e.,the two species share the ancestral Papilionoideae whole-genome duplication event)may have contributed to the large genome size of M.sativa spp.caerulea.Comparative and evolutionary analyses revealed that M.sativa spp.caerulea diverged from M.truncatula~5.2 million years ago,and the chromosomal fissions and fusions detected between the two genomes occurred during the divergence of the two species.In addition,we identified 489 resistance(R)genes and 82 and 85 candidate genes involved in the lignin and cellulose biosynthesis pathways,respectively.The near-complete and accurate diploid alfalfa reference genome obtained herein serves as an important complement to the recently assembled autotetraploid alfalfa genome and will provide valuable genomic resources for investigating the genomic architecture of autotetraploid alfalfa as well as for improving breeding strategies in alfalfa.Ao Li Ai Liu Xin Du Jin-Yuan Chen Mou Yin Hong-Yin Hu Nawal Shrestha Sheng-Dan Wu Hai-Qing Wang Quan-Wen Dou Zhi-Peng Liu Jian-Quan Liu Yong-Zhi Yang Guang-Peng Ren 2020Horticulture Research2020,7,1:3

16Regeneration of intergeneric somatic hybrids by protoplast fusion between Onobrychis viciaefolia and Medicago sativa显示文摘Protoplast fusion was induced between sainfoin and alfalfa by an improved polyethyleneglycol (PEG) method. The intergeneric somatic calluses were selected based on complementation of hydroxyproline-resistance of sainfoin and hormone autonomy growth of alfalfa transformation cell line. 17 somatic hybrid plantlets were regenerat-ed. PEG could induce the tight agglutination of protoplasts. During diluting and washing process, cyclization of the linked membrane and formation of vesicle-like structures were observed, resulting in protoplast fusion. 5%-10% glycerol supplemented in the fusion inducing solution markedly increased the frequency of heterogeneous fusion. Better fusion results were obtained when mixed protoplast suspension was dripped in petri dishes in which PEG solution was previously placed. Chromosome number of regenerated hybrid buds varied from 30 to 60. The genome of hybrids in-cluded the small chromosome from sainfoin and two chromosomes with two clear constrictions from alfalfa. 18Quantitative trait locus mapping of yield and plant height in autotetraploid alfalfa(Medicago sativa L.)显示文摘Alfalfa(Medicago sativa L.)is the most widely grown forage legume crop worldwide.Yield and plant height are important agronomic traits influenced by genetic and environmental factors.The objective of this study was to identify quantitative trait loci(QTL)and molecular markers associated with alfalfa yield and plant height.To understand the genetic basis of these traits,a full-sib F1 population composed of 392 individuals was developed by crossing a low-yielding precocious alfalfa genotype(male parent)with a high-yielding latematuring alfalfa cultivar(female parent).The linkage maps were constructed with 3818 single-nucleotide polymorphism(SNP)markers on 64 linkage groups.QTL for yield and plant height were mapped using phenotypic data for three years.Sixteen QTL associated with yield and plant height were identified on chromosomes 1 to 8.Six QTL explained more than 10%of phenotypic variation,representing major loci controlling yield and plant height.One locus on chromosome 1 controlling yield traits had not been identified in previous studies.Three QTL co-located with other QTL(qyield-1 and qheight-7,qheight-5 and qyield-4,qheight-6,and qyield-6).With further validation,the markers closely linked with these QTL may be used for marker-assisted selection in breeding new alfalfa varieties with high yield.Fei He Ruicai Long Tiejun Zhang Fan Zhang Zhen Wang Xijiang Yang Xueqian Jiang Changfu Yang Xuxin Zhi Mingna Li Longxi Yu Junmei Kang Qingchuan Yang 2020The Crop Journal2020,8,5:1

20Estimation of alfalfa fall dormancy using spaced plant and sward trials across multiple environments显示文摘Background:Alfalfa is a globally important forage crop.Cultivars are characterized by fall dormancy(FD).FD affects biomass yield and winter survival and is used to identify appropriate growing regions of cultivars.It has historically been assessed by measuring the natural height of regrowth in autumn of spaced plants in the field.Because commercial alfalfa is normally grown as a solid planted sward,FD could be different if plants were grown under real production conditions.The objective of this experiment was to assess whether FD ratings obtained from swards were similar to those obtained on spaced plants.Methods:We evaluated 20 alfalfa cultivars in field trials established in 2015 at five locations in the United States.We harvested the trials in early autumn and measured regrowth plant height approximately 3 weeks later in 2015 and 2016.Results:Autumn plant height responded as expected for the established check cultivars in all locations.Fall dormancy ratings for cultivars under both management systems were highly correlated.Conclusions:Estimating FD from spaced plant height measurements in autumn is very robust,and using height data from sward plots gives equivalent results to that measured in spaced plant nurseries.This finding has many practical advantages.Scott A.Newell Daniel J.Undersander Donald Viands Julie Hansen Steve Orloff Daniel H.Putnam E.Charles Brummer 2023Grassland Research2023,2,1:1
