13D reconstruction and capsid protein characterization of grass carp reovirus显示文摘Grass carp reovirus (GCRV) is a relatively new virus first isolated in China and is a member of the Aquareovirus genus of the Reoviridae family. Recent report of genomic se-quencing showed that GCRV shared high degree of homology with mammalian reovirus (MRV). As a step of our effort to understand the structural basis of GCRV pathogenesis, we determined the three-dimensional (3D) structure of GCRV capsid at 17 ? resolution by electron cryomicro-scopy. Each GCRV capsid has a multilayered organization, consisting of an RNA core, an inner, middle and outer protein layer. The outer layer is made up of 200 trimers that are arranged on an incomplete T=13 icosahedral lattice. A characteristic feature of this layer is the depression re-sulting from the absence of trimers around the peripentonal positions, revealing the underlying trimers on the middle layer. There are 120 subunits in the inner layer arranged with T=1 symme-try. These structural features are common to other members of the Reoviridae. Moreover, SDS-PAGE analysis showed that GCRV virions contain seven structural proteins (VP1-VP7). These structural proteins have a high degree of sequence homology to MRV, consistent with the structural similarities observed in our study. The high structural similarities of isolated GCRV and MRV suggest that future structural studies focusing on GCRV entering into and replicating within its host cell are necessary in order to fully understand the structural basis of GCRV pathogenesis.Shah Sanket 2005Science China(Life Sciences)2005,48,6:35

3Identification and Characterization of a Distinct Strain of Beak and Feather Disease Virus in Southeast China显示文摘Beak and feather disease virus(BFDV) is an infectious agent responsible for feather degeneration and beak deformation in birds. In March 2017, an epidemic of psittacine beak and feather disease(PBFD) struck a farm in Fuzhou in the Fujian Province of southeast China, resulting in the death of 51 parrots. In this study, the disease was diagnosed and the pathogen was identified by PCR and whole genome sequencing. A distinct BFDV strain was identified and named as the FZ strain.This BFDV strain caused severe disease symptoms and pathological changes characteristic of typical PBFD in parrots, for example, loss of feathers and deformities of the beak and claws, and severe pathological changes in multiple organs of the infected birds. Phylogenetic analysis showed that the FZ strain was more closely related to the strain circulating in New Caledonia than the strains previously reported in China. Nucleotide homology between the FZ strain and other 43 strains of BFDV ranged from 80.0% to 92.0%. Blind passage experiment showed that this strain had limited replication capability in SPF Chicken Embryos and DF-1 Cells. Furthermore, the capsid(Cap) gene of this FZ strain was cloned into the p GEX-4 T-1 expression vector to prepare the polyclonal anti-Cap antibody. Western blotting analysis using the anti-Cap antibody further confirmed that the diseased parrots were infected with BFDV. 4Cloning and Expression of the vp39 Gene of Bombyx mori Nuclear Polyhedrosis Virus in E.coli显示文摘The nuclear capsid protein gene (vp39) or Bombyx mori nuclear polykedrosis virus (BmNPV)was amplified successfully by PCR technique and inserted into pGEM 3zf(+). The 5’ and 3’ terminal area orthe amplirled vp39 gene were sequenced with sliver-staining dideoxy method. Bmvp39 gene was sub-cloned into the expression vector pRSET-A, and transformed into E. coli BL21. This gene was highly expressed byIPTG induction. 5Ⅰ-Ⅳ登革病毒基因同源性分析及Ⅱ型C蛋白二级结构预测显示文摘目的对登革病毒(dengue virus,DENV)Ⅰ~Ⅳ基因同源性进行分析,同时对DENVⅡC蛋白的二级结构进行预测,为更深入了解DENV 4个血清型间差异及衣壳蛋白C构想。方法通过DNAMAN分子生物学分析软件和蛋白结构在线预测网站分别对登革病毒基因同源性及C蛋白的二级结构进行预测。结果在登革病毒基因组同源性中,DENVⅠ与Ⅲ同源性最高为72%,发现DENVⅠ与Ⅳ同源性最低为67%;在登革病毒基因中,前膜基因prM的的同源性最高为82.5%,非结构基因NS2a的同源性最低为55.28%。在编码capsid的氨基酸中,占组成比例最多的为是精氨酸,可能存在的蛋白结合位点较多。结论通过基因组同源分析图谱看出Ⅰ~ⅣDENV在结构基因(C、prM、E)之间和非结构基因NS1,NS2b和NS3中的同源性较高,同时蛋白二级结构预测发现DENV ⅡC蛋白可能具有分子识别和蛋白骨架等功能。姜黎明 杨佳佳 罗佳 叶超 文送娇 孙强明 2017医学研究杂志2017,46,6:1
6Multiple Roles of HIV-1 Capsid during the Virus Replication Cycle显示文摘Human immunodeficiency virus-1 capsid(HIV-1 CA) is involved in different stages of the viral replication cycle. During virion assembly, CA drives the formation of the hexameric lattice in immature viral particles, while in mature virions CA monomers assemble in cone-shaped cores surrounding the viral RNA genome and associated proteins. In addition to its functions in late stages of the viral replication cycle, CA plays key roles in a number of processes during early phases of HIV-1 infection including trafficking, uncoating, recognition by host cellular proteins and nuclear import of the viral preintegration complex. As a result of efficient cooperation of CA with other viral and cellular proteins, integration of the viral genetic material into the host genome, which is an essential step for productive viral infection, successfully occurs. 9AAV8 transduction capacity is reduced by prior exposure to endosome-like pH conditions显示文摘Adeno-associated virus(AAV)is an essential instrument in the neuroscientist’s toolkit,which allows delivery of DNA to provide labeling with fluorescent proteins or genetic instructions to regulate gene expression.In the field of neural regeneration,the transduction of neurons enables the observation and regulation of axon growth and regeneration,and in the future will likely be a mechanism for delivering molecular therapies to promote sprouting and regeneration after central nervous system injury.Traditional formulations of AAV preparations permit efficient viral transduction under physiologic conditions,but an improved understanding of the mechanistic limitations of AAV transduction may facilitate production of more resilient AAV strains for investigative and therapeutic purposes.We studied AAV transduction in the context of prior exposure of AAV serotype 8(AAV8)to environmental pH within the range encountered during endosomal endocytosis(pH 7.4 to pH 4.4),during which low pH-triggered structural and autoproteolytic changes to the viral capsid are believed to be necessary for endosome escape and virus uncoating.Due to the fundamental nature of these processes,we hypothesized that premature exposure of AAV8 particles to acidic pH would decrease viral transduction of HT1080 cells in vitro,as measured by fluorescent reporter gene expression using high-content imaging analysis.We found that increasingly acidic incubation conditions were associated with concomitant reductions in transduction efficiency,and that quantitative levels of reporter gene expression in transduced cells were similarly decreased.The biggest decrease in transduction occurred between pH 7.4 and pH 6.4,suggesting the possible co-occurrence of a pH-associated event and viral inactivation within that range.Taken together,these findings indicate that exposure of AAV8 to acidic pH for as little as 1 hour is deleterious to transduction ability.Future studies are necessary to understand the pH-associated causative mechanisms involved.This study was approved by the University of Miami Institutional Animal Care and Use Committee,USA(Protocol#18-108-LF)on July 12,2018.Jeffrey ALowell Kar Men Mah John LBixby Vance PLemmon 2021Neural Regeneration Research2021,16,5:0
10Elucidating cellular interactome of chikungunya virus identifies host dependency factors显示文摘Chikungunya virus(CHIKV)is a re-emerging mosquito-transmitted RNA virus causing joint and muscle pain.To better understand how CHIKV rewires the host cell and usurps host cell functions,we generated a systematic CHIKV-human protein-protein interaction map and revealed several novel connections that will inform further mechanistic studies.One of these novel interactions,between the viral protein E1 and STIP1 homology and U-box containing protein 1(STUB1),was found to mediate ubiquitination of E1 and degrade E1 through the proteasome.Capsid associated with G3BP1,G3BP2 and AAAþATPase valosin-containing protein(VCP).Furthermore,VCP inhibitors blocked CHIKV infection,suggesting VCP could serve as a therapeutic target.Further work is required to fully understand the functional consequences of these interactions.Given that CHIKV proteins are conserved across alphaviruses,many virus-host protein-protein interactions identified in this study might also exist in other alphaviruses.Construction of interactome of CHIKV provides the basis for further studying the function of alphavirus biology.Peiqi Yin Xia Jian Yihan Liu Yuwen Liu Lu Lv Haoran Cui Leiliang Zhang 2023Virologica Sinica2023,38,4:0
11Nucleotide sequence of maize dwarf mosaic virus capsid protein gene and its expression in Escherichia coli显示文摘The 3’-terminal 1 279 nucleotide sequence of maize dwarf mosaic virus (MDMV) genome has been determined. This sequence contains an open reading frame of 1023 nudeotides and a 3’ -non-coding region of 256 nucleotides. The open reading frame includes all of the coding regions for the viral capsid protein (CP) and part of the viral nuclear inclusion protein (Nib). The predicted viral CP consists of 313 amino acid residues with a calculated molecular weight of 35400. The amino acid sequence of the viral CP derived from MDMV cDNA shows about 47%-54% homology to that of 4 other potyviruses. The viral CP gene was constructed in frame with the lacZ gene in pUC19 plasmid and expressed in E. coli cells.
