2食细菌线虫Caenorhabditis elegans的取食偏好性显示文摘通过设置平板培养试验,以模式种线虫Caenorhabditis elegans为材料,观察了食细菌线虫的取食行为。结果表明:C.elegans在取食细菌时对原位土壤中分离的一种Pseudomonas sp细菌存在最大的取食偏好性。这种取食偏好性表现在大部分C.elegans在24 h内都直接朝Pseudomonas sp迁移,说明C.elegans能通过某种机制识别Pseudomonas sp。距离迁移试验及C.elegans迁移率表明它可能是通过辨别细菌所发出的气味识别它喜欢的食物。C.elegans的繁殖率跟其取食的偏好性是相关的,在迁移率较高的细菌培养基中线虫表现出更高的繁殖率。结果还表明:相对于G+细菌而言,C.elegans偏好取食G-细菌。为进一步了解土壤生态系统中食细菌线虫与细菌群落结构间相互关系提供了帮助。肖海峰 焦加国 胡锋 李辉信 2010生态学报2010,30,24:5
3Anti-parasitic effects of water-soluble alkaloid fractions from ethanolic extracts of Sophora moorcroftiana seeds in Caenorhabditis elegans显示文摘Parasite infections of humans and animals remain a major global health problem, with limited choice of drugs being available to the treatment of parasitosis in the clinic. Sophora moorcroftiana(S. moorcroftiana) is a shrub that grows in Tibet Plateau of China. Decoction of the seeds has been used as a traditional Tibetan medicine to treat parasitosis for years. But the anti-parasitic effects of water-soluble fractions in the seeds need further investigation. In the present study, the water-soluble alkaloid fractions(E2) were obtained from S. moorcroftiana seeds by refluxing extraction with 60% ethanol and low polarity fraction(E2-a) and high polarity fraction(E2-b) were subsequently isolated from E2 using column chromatography. As a parasite model, Caenorhabditis elegans(C. elegans) were treated with different fractions and their survivals were recorded. The results showed that that E2-a induced a lower survival rate in C. elegans than E2-b and E2. The protoscoleces of Echinococcus granulosus(E. granulosus) were cultured in the presence of E2-a. Compared with E2-b and E2, protoscoleces exhibited decreased survival rate following E2-a treatment. Furtherly, the effects of E2-a on the behavior, brood size, and lifespan of the worms were investigated. Body bend frequencies of the worms treated with the high concentration of E2-a were reduced by two-thirds compared with the control group(P < 0.01). Compared with non-E2-a-treated group, exposure of nematodes to E2-a led to a decrease in head thrashes and pharyngeal pumps frequency(P < 0.01). E2-a treatment resulted in a significantly lower brood size(P < 0.01). Additional E2-a treatment induced a significantly shortened lifespan, compared with the control(P < 0.05). 4高功率毫米波辐照对秀丽线虫行为学和细胞形态学的影响显示文摘采用角锥喇叭天线,使用不同输出平均功率的毫米波激光辐照秀丽线虫,采用实时显微摄像系统记录线虫行为学变化,并使用体视显微镜观察线虫细胞形态学变化。受照射后自由活动的秀丽线虫能够快速逃离高功率毫米波的辐照区域。当受输出平均功率为10 W和12 W辐照后,线虫先后出现运动速度增加、运动速度减慢直至身体僵直不动的变化过程。当输出平均功率为5 W时,线虫出现热耐受现象。细胞形态学观察结果显示:辐照后的线虫腹中卵细胞核结构出现不同程度的异常。高功率毫米波辐照可显著影响线虫的行为学特征及细胞形态学,同时说明秀丽线虫是可适用于高功率毫米波生物效应研究的模式生物之一。任长虹 袁广江 高艳 吴永红 徐志伟 徐寿喜 粟亦农 刘濮鲲 张成岗 2010强激光与粒子束2010,22,12:2
5线虫脂肪沉积中组织蛋白酶B的表达及功能研究显示文摘文章利用高糖/高胆固醇饮食诱导了线虫,油红O染色结果表明线虫脂肪沉积显著增加。为了研究组织蛋白酶B在诱导线虫过程中的作用,利用实时荧光定量聚合酶链式反应(polymerase chain reaction,PCR)研究了组织蛋白酶B同源基因的表达。选取其中的关键基因利用RNA干扰(RNA interference,RNAi)技术,研究其在线虫脂肪沉积中的作用。结果表明:在线虫脂肪沉积改变后,组织蛋白酶B同源基因表达随之改变;而利用RNA干扰组织蛋白酶B关键基因cpr-3的表达,线虫的脂肪沉积未受影响。因此,组织蛋白酶B同源基因可能参与线虫脂肪沉积改变后的生理变化,而未直接参与线虫脂肪沉积的调控。赵丽娟 鲍斌 2016合肥工业大学学报(自然科学版)2016,39,6:2
6C.elegans-based screen identifies lysosome-damaging alkaloids that induce STAT3-dependent lysosomal cell death显示文摘Lysosomes are degradation and signaling centers within the cell,and their dysfunction impairs a wide variety of cellular processes.To understand the cellular effect of lysosome damage,we screened natural smallmolecule compounds that induce iysosomal abnormality using Caenorhabditis elegans (C.elegans)as a model system.A group of vobasinyl-ibogan type bisindole alkaloids (ervachinines A-D)were identified that caused lysosome enlargement in C.elegans macrophage-like cells.Intriguingly,these compounds triggered cell death in the germ line independently of the canonical apoptosis pathway.In mammalian cells, ervachinines A-D induced lysosomal enlargement and damage,leading to leakage of cathepsin proteases, inhibition of autophagosome degradation and necrotic cell death.Further analysis revealed that this ervachinine-induced lysosome damage and lysosomal cell death depended on STAT3 signaling,but not RIP1 or RIP3 signaling.These findings suggest that lysosomedamaging compounds are promising reagents for dissecting signaling mechanisms underlying lysosome homeostasis and lysosome-related human disorders.Yang Li Yu Zhang Qiwen Gan Meng Xu Xiao Ding Guihua Tang Jingjing Liang Kai Liu Xuezhao Liu Xin Wang Lingli Guo Zhiyang Gao Xiaojiang Hao Chonglin Yang 2018Protein & Cell2018,9,12:2
7植物生长调节剂对百日草生长及开花的影响显示文摘以百日草(Zinnia elegans Jacq.)为试材,采用不同浓度的赤霉素(GA3)、油菜素内酯(BR)和不同稀释倍数的的花粉酵素对百日草进行2次全株喷施处理,探讨对百日草花期及开花质量的影响。结果表明,在一定范围内喷施赤霉素可使百日草开花提前、延长花期,而喷施油菜素内酯、花粉酵素可不同程度地推迟百日草开花;三者均可促进花径、叶面积、株高、单株花朵数的增加,提高百日草的观赏价值。综合考虑,3种植物生长调节剂中以0.02 mg/L的油菜素内酯喷施处理的效果最好;其次是花粉酵素800倍稀释液处理;赤霉素喷施浓度以100 mg/L为宜。孙晨 王克凤 董然 钱英 孙晓丹 李春鹏 2018湖北农业科学2018,57,4:2

10杀线虫活性菌的筛选、分离与初步鉴定显示文摘从河南南阳宝天曼森林土壤样品中分离纯化细菌,以对线虫Caenorhabditis elegans的杀灭作用为筛选模型,从128株细菌中共筛选到3株杀线虫活性较高的细菌,编号分别为B、C、D。当作用于线虫24 h后,它们对线虫的致死率分别为20%、25%、97%。然而发现当B和C共同作用于线虫时,致死率大为提高(约为90%),说明二者在杀线虫方面有协同作用。通过培养观察菌落形态,革兰氏染色;提取各菌种基因组,扩增16S rDNA测序,系统发育分析,确定这三株菌的鉴定结果为:菌株B与Bacillus subtilis strain xfchu2的同源性较高,为96.46%,菌株C与Stenotrophomonas maltophilia strain QT24的同源性较高,为92.75%。菌株D与荧光假单胞菌属的同源性较高。张林 牛秋红 周晓丽 2012南阳理工学院学报2012,4,2:1

For age determination, the diameter growth rate of lichens is usually taken as a constant. Alternatively, the growth curve of lichens is derived by dating objects of known age in some area, and it can then be used for age determination in the same area or areas of similar environment. Unfortunately it is very difficult to obtain such growth curves in many places. The growth curves differ in different environments. 12Expression of hepatitis B virus surface antigens induces defective gonad phenotypes in Caenorhabditis elegans显示文摘AIM To test whether a simple animal, Caenorhabditis elegans(C. elegans), can be used as an alternative model to study the interaction between hepatitis B virus antigens(HBs Ag) and host factors. METHODS Three plasmids that were able to express the large, middle and small forms of HBs Ags(LHBs Ag, MHBs Ag, and SHBs Ag, respectively) driven by a ubiquitous promoter(fib-1) and three that were able to express SHBs Ag driven by different tissue-specific promoters were constructed and microinjected into worms. The brood size, egglaying rate, and gonad development of transgenic worms were analyzed using microscopy. Levels of m RNA related to endoplasmic reticulum stress, enpl-1, hsp-4, pdi-3 and xbp-1, were determined using reverse transcription polymerase reaction(RT-PCRs) in three lines of transgenic worms and dithiothreitol(DTT)-treated wild-type worms. RESULTS Severe defects in egg-laying, decreases in brood size, and gonad retardation were observed in transgenic worms expressing SHBs Ag whereas moderate defects were observed in transgenic worms expressing LHBs Ag and MHBs Ag. RT-PCR analysis revealed that enpl-1, hsp-4 and pdi-3 transcripts were significantly elevated in worms expressing LHBs Ag and MHBs Ag and in wild-type worms pretreated with DTT. By contrast, only pdi-3 was increased in worms expressing SHBs Ag. To further determine which tissue expressing SHBs Ag could induce gonad retardation, we substituted the fib-1 promoter with three tissue-specific promoters(myo-2 for the pharynx, est-1 for the intestines and mec-7 for the neurons) and generated corresponding transgenic animals. Moderate defective phenotypes were observed in worms expressing SHBs Ag in the pharynx and intestines but not in worms expressing SHBs Ag in the neurons, suggesting that the secreted SHBs Ag may trigger a cross-talk signal between the digestive track and the gonad resulting in defective phenotypes. 18Ingestion of Carbon Nanotubes and Mesoporous Silica Nanospheres by Caenorhabditis Elegans显示文摘The toxic effects to microorganism induced by nanomaterials have received considerable attentions in the past decades [1]. Herein, two diverse nanomaterials i.e. multi-walled carbon nanotubes (MWCNTs) and mesoporous silica nanospheres (MSNs) were prepared to investigate their deleterious effects on Caenorhabditis. elegans (C. elegans)[2-3]. As shown in Figure 1A, histidine functionalized MWCNTs (his-MWCNTs) were in length of ~500 nm with outer diameter ~20 nm, while fluorescein isothiocyanate dyed MSNs (FITC-MSNs) were in an average diameter of ~70 nm (Figure 1B). Microscopic images display his-MWCNTs having been ingested into intestine of C. elegans after co-incubation for 2 h, as arrowed in Figure 1C and 1E. In contrast, no MSNs were observed to be ingested after co-incubating in the same liquid medium.
