| 1 | 中国野生大豆的遗传多样性和生态特异性分析显示文摘野生大豆(Glycine soja)是栽培大豆的祖先,为东亚特有种,大部分分布在中国。我们采用52对SSR引物和10个植物学性状,以遗传丰富度和Simpson多样性指数为指标,对来自中国3个地理生态区域涉及24个省区的196份野生大豆所构成的代表性样本的遗传变异进行了研究,以期从分子水平和表型水平两个层面上揭示中国野生大豆遗传多样性和地理生态特异性。结果表明:中国野生大豆群体SSR位点的等位基因平均丰富度(NA)和平均Simpson多样性指数(H)分别为16.1和0.852,高于栽培大豆(NA=11.4,H=0.773),野生群体的遗传多样性明显高于栽培群体。3个地理生态群体中南方群体多样性最高(NA=12.9,H=0.842),黄淮海群体最低(NA=11.4,H=0.805),东北群体居中(NA=12.5,H=0.834)。群体间存在遗传分化,不同群体具有不同的特异等位基因,位点AW132402(A2连锁群)、Satt522(F)、satt150(M)、Sat_332(D1a)、Satt046(K)、sct_190(K)等的一些等位基因只在特定群体出现,表现出群体分化后的生态特异性。中国野生大豆植物学性状的群体变异丰富,平均Simpson多样性指数为0.710。地理群体间存在分化,最明显的是生育期性状的分化,反映了地理、光照和温度等生态因子的选择作用,其中南方地理群体多样性最高(H=0.671)。SSR分子标记和植物学性状所获结果相对一致,表明中国野生大豆地理群体间性状分化有其遗传分化的基础。 | 丁艳来 赵团结 盖钧镒 | 2008 | 生物多样性2008,16,2: | 49 |

| 6 | Shade stress decreases stem strength of soybean through restraining lignin biosynthesis显示文摘Lodging is the most important constraint for soybean growth at seedling stage in maize-soybean relay strip intercropping system.In the field experiments,three soybean cultivars Nandou 032-4(shade susceptible cultivar; B1),Jiuyuehuang(moderately shade tolerant cultivar; B2),and Nandou 12(shade tolerant cultivar; B3) were used to evaluate the relationship between stem stress and lignin metabolism in the stem of soybean.Results showed that the intercropped soybean was in variable light condition throughout the day time and co-growth stage with maize.The xylem area and cross section ratio played a main role to form the stem stress.The B3 both in intercropping and monocropping expressed a high stem stress with higher xylem area,lignin content,and activity of enzymes(phenylalanine ammonia-lyase(PAL),4-coumarate: CoA ligase(4CL),cinnamyl alcohol dehydrogenase(CAD),and peroxidase(POD)) than those of B1 and B2.Among the soybean cultivars and planting pattern,lignin content was positively correlated with stem stress.However,a negative correlation was found between lignin content and actual rate of lodging.In conclusion,the shade tolerant soybean cultivar had larger xylem area,higher lignin content and activities of CAD,4CL,PAL,and POD than other soybean cultivars in intercropping.The lodging in maize-soybean intercropping can be minimized by planting shade tolerant and lodging resistant cultivar of soybean.The lignin content in stem could be a useful indicator for the evaluation of lodging resistance of soybean in intercropping and activities of enzymes were the key factors that influence the lignin biosynthesis. | LIU Wei-guo Sajad Hussain LIU Ting ZOU Jun-lin REN Meng-lu ZHOU Tao LIU Jiang YANG Feng YANG Wen-yu | 2019 | Journal of Integrative Agriculture2019,18,1: | 8 |
| 7 | Protective effects of glycine pretreatment on brain-death donor liver显示文摘BACKGROUND: Morphological and functional changes commonly occur in livers of brain-death donors. Preven- tion of liver injury from brain-death will benefit the results of transplantation. This study was conducted to evaluate the protection effects of glycine on the liver of brain-death donor. METHODS: Fourty-two male Wistar rats were randomly divided into brain-death donor (BDD) group (B), glycine pretreatment group with BDD (G), and strychnine pre- treatment group with BDD(S). For these groups, brain death model was established in donor rats and liver trans- plantation was performed subsequently utilizing microsur- gical techniques. After the establishment of the model and during cold rinsing of liver donors or liver reperfusion of recipients, glycine was given at a dose of 0. 6 mmol, 25 μmol and 25 μmol in the group G, and a same dose of gly- cine and strychnine (1000 :1) was prescribed for the group S, but nothing for the group B. Before cold rinsing at 2 and 6 hours after portal vein(PV) reperfusion, blood sam- ples were taken from infrahepatic vena cava (IHVC) to de- termine the levels of alanine aminotransferase (ALT), as- partate aminotransferase (AST), tumor necrosis factor al- pha (TNF-α) and hyaluronic acid (HA). At 6 hours after PV reperfusion, graft samples were fixed for morphological observation and apoptosis of hepatocytes was detected using the TUNEL method. RESULTS: Before liver cold rinsing and at 2 and 6 hours af- ter PV reperfusion, the serum levels of ALT, AST, TNF-α, HA and apoptosis index (AI) in the groups B and S were significantly higher than those in the group G (P <0.05). There was no significant difference between the groups B and S (P>0.05). Electron microscopy showed that Kupffer cells were activated and hepatic cells injured more obvious- ly in the groups B and S than in the group G. | 8 | Effects of Lipoxygenase Null Genes of Soybean in Controlling Beany-flavor of Soymilk and Soyflour显示文摘The flavor of the soymilk and soyflour obtained from the lipoxygenase mutant isolines was concentrated by simultaneous distillation and extraction (SDE), and its constituents were identified by gas chromatography (GC) and gas chromatography-mass spectrometry. Results showed that the same 24 flavor constituents were isolated in both soymilk and soyflour, and most of them were aldehydes and alcohols. Lox2 was most responsible for the production of the volatile and beany-flavor components, and Lox1 less responsible.Lox3 was least responsible and can reduce the yield of hexanal. Either Lx1 or Lx2 could significantly reduce the volatile and beany-flavor, and Lx3 could significantly increase the yield of hexanal. Primary and secondary interactions existed among the null mutant genes, and the major effects and interactions could be affected by processing conditions. The isoline with triple lipoxygenase null genes yielded the least volatile and beany-flavor components, and the isoline without the lipoxygenase gene Lx3 produced the greatest amount of the volatile and beany-flavor components. The amounts of volatile and beany-flavor components produced by the other isolines were between that of the isoline with triple lipoxygenase null genes and the isoline without lipoxygenase gene Lx3. According to the correlation analysis, the hexanal amount could be used as an index in evaluating the importance of lipoxygenase isozymes in the yield of beany-flavor compounds, and the effects of the different types of lipoxygenase null mutants in controlling beany-flavor compounds. | 9 | Mapping and validation of a dominant salt tolerance gene in the cultivated soybean(Glycine max) variety Tiefeng 8显示文摘Salt is an abiotic stress factor that strongly affects soybean growth and production. A single dominant gene has been shown to confer salt tolerance in the soybean cultivar Tiefeng 8.The objective of the present study was to genetically map the salt-tolerance gene in an F2:3population and a recombinant inbred line(RIL) population derived from a cross between two cultivated soybeans, Tiefeng 8(tolerant) and 85-140(sensitive). The F2:3families and RILs were treated with 200 mmol L-1Na Cl to evaluate salt tolerance. The F2:3population showed 1(42 tolerant): 2(132 segregating): 1(65 sensitive) segregation, indicating a single dominant gene for salt tolerance in Tiefeng 8. A sequence-characterized amplified region(SCAR) marker from a previously identified random amplified polymorphic DNA(RAPD)marker and four insertion/deletion polymorphism(In Del) markers were developed within the mapping region. Using these markers along with SSR markers, the salt-tolerance gene was mapped within 209 kb flanked by SCAR marker QS08064 and SSR marker Barcsoyssr_3_1301 on chromosome 3. Three markers that cosegregated with the salt tolerance gene and SCAR marker QS08064 were used to genotype 35 tolerant and 23 sensitive soybean accessions. These markers showed selection efficiencies of 76.2% to94.2%. | 12 | Density functional theory study of 1:1 glycine–water complexes in the gas phase and in solution显示文摘We present a systematic study of 1:1 glycine-water complexes involving all possible glycine conformers. The complex geometries are fully optimized for the first time both in the gas phase and in solution using three DFT methods (B3LYP, PBE1PBE, X3LYP) and the MP2 method. We calculate the G3 energies and use them as the reference data to gauge hydrogen bond strength in the gas phase. The solvent effects are treated via the integral equation formalism-polarizable continuum model (IEF-PCM). Altogether, we locate fifty-two unique nonionized (N) structures and six zwitterionic (Z) structures in the gas phase, and fifty-five N structures and thirteen Z structures in solution. Both correlation and solvation are shown to be important in geometry determination. We found that in the gas phase, a water molecule binds more strongly to the carboxylic acid group of glycine than to its amine group, whereas in solution phase the reverse is true. | 15 | Expression analysis of gdcsP promoter from C_3-C_4 intermediate plant Flaveria anomala in transgenic rice显示文摘The gdcsP promoter isolated from C3-C4 intermediate plant Flaveria anomala was fused to the β-glu-curonidase (GUS) gene. The chimeric gene was inserted into the binary vector pBin19 and introduced into the rice (Oryza saliva L.) cv. 8706 by Agrobacterium-mediated gene transfer. GUS activity can be detected in leaf, leaf sheath, stem and root tissues via fluorometric GUS assay. However, no GUS activity was found in mature endosperm. Histochemical localization revealed that GUS expression was exclusively restricted to vascular tissues in transgenic plants. This promoter also showed spatial-temporal expression patterns that GUS expression declined significantly with the maturity of plants. | 20 | Map-based cloning of a novel QTL qBN-1 influencing branch number in soybean[Glycine max(L.)Merr.]显示文摘Branch number(BN)is an important agronomic attribute related to the plant architecture,adaptability,and yield of soybean.To date,few studies ofBNhave been conducted to elucidate its genetic background.We aimed to localize genetic factors affecting BN using segregating populations derived fromthe high-branching cultivar‘Kennong24’(KN24)and the low-branching cultivar‘Kenfeng19’(KF19).Composite interval mapping analysis detected a QTL(qBN-1)on chromosome 6 between the SSR markers BARCSOYSSR_06_0993 and BARCSOYSSR_06_1070 using an F2 population.To fine-map qBN-1,a RIL population was developed and genotyped with 14 SSRmarkers located in the QTL region.qBN-1 was localized to a 115.67-kb interval flanked by markers BARCSOYSSR_06_1048 and BARCSOYSSR_06_1053.The QTL was further confirmed using backcross populations of size 1305(BC2F2 with KN24 as a recurrent parent)and 1712(BC3F2 with KF19 as a recurrent parent).The fine-mapping region of qBN-1 contained only two candidate genes,Glyma.06G208800 and Glyma.06G208900,whose expression patterns were investigated by qRT-PCR.Compared to Glyma.06G208800 gene expression,Glyma.06G208900 showed the highest expression of the two genes and showed a significant difference in expression between high-and low-branching genotypes in either axillary meristem or shoot apical meristem,and showed opposite expression patterns in the two tissues at V4 and R1 stages.These results identify Glyma.06G208900 as a novel candidate gene controlling BN.Taken together,the results of this study provide a foundation for cloning and functional analysis of the qBN-1 gene and for the improvement of BN bymarker-assisted selection in soybean breeding. | Sobhi F.Lamlom Yong Zhang Bohong Su Haitao Wu Xia Zhang Jindong Fu Bo Zhang Li-Juan Qiu | 2020 | The Crop Journal2020,8,5: | 2 |
