| 2 | Exploring gut microbes in human health and disease:Pushing the envelope显示文摘Humans have coevolved with their microbes over thousands of years,but this relationship,is now being dramatically affected by shifts in the collective human microbiome resulting from changes in the environment and societal norms.Resulting perturbations of intestinal host-microbe interactions can lead to miscues and altered host responses that increase the risk of pathogenic processes and promote“western”disorders such as inflammatory bowel diseases,cancers,obesity,diabetes,autism,and asthma.Given the current challenges and limitations in gene therapy,approaches that can reshape the gut microbiome represent a reasonable strategy for restoring the balance between host and microbes.In this review and commentary,we highlight recent progress in our understanding of the intestinal microbiome in the context of health and diseases,focusing on mechanistic concepts that underlie the complex relationships between host and microbes.Despite these gains,many challenges lie ahead that make it difficult to close the gap between the basic sciences and clinical application.We will discuss the potential therapeutic strategies that can be used to manipulate the gut microbiota,recognizing that the promise of pharmabiotics(“bugs to drugs”)is unlikely to be completely fulfilled without a greater understanding of enteric microbiota and its impact on mammalian physiology.By leveraging the knowledge gained through these studies,we will be prepared to enter the era of personalized medicine where clinical inventions can be custom-tailored to individual patients to achieve better outcomes. | Jun Sun Eugene B.Chang | 2014 | Genes & Diseases2014,1,2: | 17 |

| 4 | Alfalfa-containing diets alter luminal microbiota structure and short chain fatty acid sensing in the caecal mucosa of pigs显示文摘Background: Pork produced by outdoor-reared pigs raised mostly on alfalfa pastures attracts increasing population of consumer from most of the world. In China, pigs were raised with alfalfa-containing diets to seek for good quality pork.However, the influence of dietary alfalfa involving high level of insoluble dietary fiber(IDF) on pig intestinal luminal microbiota composition remains unclear. The objective of this study was to investigate the effects of alfalfa on luminal microbiota and short chain fatty acids(SCFA) production, and gene expressions involved in SCFA sensing, transporting and absorbing in pig caecal mucosa.Results: Twenty-four growing pigs were randomly allotted to four diets containing 0%, 5%, 10% and 15% alfalfa meal for a 28-d experiment. Ingestion of alfalfa meal-contained diets significantly increased the ratio of body weight gain to feed consumption. Illumina MiS eq sequencing of the V3 region of the 16 S r RNA genes showed that alfalfa-containing diet significantly decreased the relative abundance of genera Turicibacter, Acidiphilium, Paracoccus, Propionibacterium,Corynebacterium, Pseudomonas, Acinetobacter, and Staphylococcus, and increased the relative abundance of genera Lachnospira, Marvinbryantia, and Desulfovibrio in the caecal digesta. Butyrate concentration was significantly increased in the hindgut by the supplementation of alfalfa meal in diets. | 6 | Supplementation of milk formula with galacto-oligosaccharides improves intestinal micro-flora and fermentation in term infants显示文摘Background Oligosaccharides in human milk may protect infants by improving the intestinal micro-flora and fermentation. This study was to investigate effects of infant formula milk consisting of galacto-oligosaccharide (GOS) on intestinal microbial populations and the fermentation characteristics in term infants in comparison with that of human milk. Methods The test formula (Frisolac H,Friesland,Netherland) was supplemented with GOS at a concentration of 0.24 g/dl. Human milk and another formula without oligosaccharides (Frisolac H,Friesland,Netherland) were used as positive and negative control respectively. Growth,stool characteristics,and side effects of the recruited infants were recorded after 3 and 6 months’ follow-up,and the fecal species were collected for the analysis of intestinal micro-flora,short chain fatty acid (SCFA) and pH.Results At the end of 3- and 6-month feeding period,intestinal Bifidobacteria and Lactobacilli were significantly increased in infants fed with GOS supplemented formula and human milk when compared with infants fed with negative control formula; however,there was no statistically significant difference between GOS supplemented formula and human milk groups. Stool characteristics were influenced by the supplement and main fecal SCFA (acetic),and stool frequency were significantly increased in infants fed with GOS supplemented formula and human milk,while the fecal pH was significantly decreased as compared with that of negative control ( P <0.05). Supplementation had no influence on incidence of side effects (including crying,regurgitation and vomiting). | 13 | Eggshell and environmental bacteria contribute to the intestinal microbiota of growing chickens显示文摘Background: The initial intestinal microbiota acquired from different sources has profound impacts on animal health and productivity. In modern poultry production practices, the source(s) of the establishing microbes and their overall contribution during development of gastrointestinal tract communities are still unclear. Using fertilized eggs from two independent sources, we assessed the impact of eggshell-and environmental-associated microbial communities on the successional processes and bacterial community structure throughout the intestinal tract of chickens for up to 6 weeks post-hatch.Results: Culturing and sequencing techniques identified a viable, highly diverse population of anaerobic bacteria on the eggshell. The jejunal, ileal, and cecal microbial communities for the egg-only, environment-only, and conventionally raised birds generally displayed similar successional patterns characterized by increasing community richness and evenness over time, with strains of Enterococcus, Romboutsia, and unclassified Lachnospiraceae abundant for all three input groups in both trials. Bacterial community structures differed significantly based on trial and microbiota input with the exception of the egg-exposed and conventional birds in the jejunum at week 1 and the ileum at week 6. Cecal community structures were different based on trial and microbiota input source, and cecal short-chain fatty acid profiles at week 6 highlighted functional differences as well.Conclusion: We identified distinct intestinal microbial communities and differing cecal short-chain fatty acid profiles between birds exposed to the microbiota associated with either the eggshell or environment, and those of conventionally hatched birds. Our data suggest the eggshell plays an appreciable role in the development of the chicken intestinal microbiota, especially in the jejunum and ileum where the community structure of the eggshellonly birds was similar to the structure of conventionally hatched birds. | 19 | The association between the metabolites of intestinal flora and diabetes mellitus显示文摘Objective To contrast and analyze the difference of metabolites of intestinal flora between patients with type 2diabetes and normal blood glucose human in Xinjiang Uygur and Kazak.Methods Uygur fecal samples and Kazak fecal samples were collected,and short-chain fatty acids(SCFA)were extracted by water from feces,then the content of SCFA in fecal samples was analyzed by gas-chromatography mass-spectrometry(GC-MS).Results Both Uygurs and Kazaks,in their feces,the content of SCFA in the feces showed significantly decreased in patients with type 2diabetes,and a significant negative linear correlation was shown between the fasting plasma glucose and the concentration of SCFA in the fecal samples.But,there was no statistical differences between uygurs and kazaks,in not only normal blood glucose group,but also type 2 diabetes group.Conclusion The feces of patients with type 2diabetes has an decreased concentration of SCFA.Between Uygurs and Kazaks,the metabolic changes of intestinal flora was similar in patients with type 2diabetes. | WANG Ye WANG Chang-hui YU Xin-lan LI Ge LIU Jing-bao GAO Bing MAO Xin-min LI Lin-lin | 2010 | 新疆医科大学学报2010,33,9: | 0 |
| 20 | 梯度乙醇沉淀法提取杏鲍菇多糖及其抗氧化和免疫刺激活性研究显示文摘食用菌多糖具有多种生理活性,这些活性与其组成及结构等有直接的关系。杏鲍菇在我国栽培面积大,对其深度开发具有重要的经济价值。本文的目的是提取杏鲍菇多糖,并通过不同浓度的乙醇沉淀获得各级醇沉多糖,并评价其体内免疫调节活性。在温和条件下,从杏鲍菇子实体中提取多糖,并通过20%、.40%、60%80%梯度乙醇沉淀法或传统的80%乙醇沉淀获得不同的多糖组分(分别命名为PP20、PP40、PP60、PP80和TPP),通过高效液相色谱(HPLC)进行定性和定量分析;然后将多糖给小鼠灌胃。评估了杏鲍菇多糖的抗氧化能力及其对小鼠免疫活性和肠道短链脂肪酸(SCFA)的调节作用。结果表明,PP60和TPP中含有岩藻糖成分,而在PP80和TPP中含有核糖。PP60和PP80处理小鼠体内中超氧阴离子的清除率显著高于PP40和正常对照组(p<0.01)。PP80组的血清IL-6浓度明显低于正常对照组(p<0.01)。PP60组的血清TNF-α浓度降至82.13 pg·mL^-1,明显低于PP40组和空白对照组(p<0.01)。所有经过多糖处理的组在粪便中均显示出较高的短链脂肪酸(SCFA)含量。 | 孙庆申 赵越龙 王鑫 刘欣洋 韩晓云 李梦洋 | 2020 | 黑龙江大学自然科学学报2020,37,4: | 0 |
