Reactive capillary hemangiomas: a novel dermatologic toxicity following anti-PD-1 treatment with SHR-1210显示文摘Objective: SHR-1210 is a new and promising anti-PD-1 agent for solid tumors. During the phase I study of SHR-1210, we encountered a novel but prevalent immune-related dermatologic toxicity: reactive capillary hemangiomas(RCHs). Thus we tried to summarize the features of RCHs and estimate their relationship with tumor response. Methods: This prospective observational study systematically enrolled 98 patients with advanced solid tumors from April 27th,2016 to June 8th, 2017 in the context of the phase I clinical study of SHR-1210. This report focused on the skin toxicities. Patients underwent entire skin inspection every two weeks while taking medication. The clinical course of RCHs was recorded and their association with tumor response was estimated. The data cut-off date was November 15th, 2017.Results: After a median follow-up of 242(range, 29–567) days, RCHs were observed in 85.7%(84/98) of patients on cutaneous/mucosal surfaces; 84.5%(71/84) of the RCHs were evaluated as grade 1 adverse events. No grade 3 or 4 RCHs were observed. The time of onset of RCHs was dose dependent and shortest in the 400 mg-dose cohort(P < 0.001). Spontaneous and complete regression of RCHs was observed both during and after treatment. The objective response rate of tumors for patients with RCHs was 28.9%(24/83). However, no responders were observed among the patients without RCHs.Conclusions: RCHs were prevalent but manageable during treatment with SHR-1210. It might add to the expanding literature regarding immune-related dermatologic adverse events. | Xuelian Chen Lanying Ma Xi Wang Hongnan Mo Dawei Wu Bo Lan Dong Qu Hongtu Zhang Jing Huang Binghe Xu | 2019 | Cancer Biology & Medicine2019,16,1: | 53

Peripheral clearance of brain-derived Aβ in Alzheimer's disease: pathophysiology and therapeutic perspectives显示文摘Alzheimer’s disease(AD)is the most common type of dementia, and no disease-modifying treatments are available to halt or slow its progression.Amyloid-beta(Aβ)is suggested to play a pivotal role in the pathogenesis of AD,and clearance of Aβfrom the brain becomes a main therapeutic strategy for AD.Recent studies found that Aβclearance in the periphery contributes substantially to reducing Aβaccumulation in the brain. Therefore, understanding the mechanism of how Aβis cleared in the periphery is important for the development of effective therapies for AD.In this review,we summarized recent findings on the mechanisms of Aβefflux from the brain to the periphery and discuss where and how the brain-derived Aβis cleared in the periphery.Based on these findings,we propose future strategies to enhance peripheral Aβclearance for the prevention and treatment of AD.This review provides a novel perspective to understand the pathogenesis of AD and develop interventions for this disease from a systemic approach. | Yuan Cheng Ding-Yuan Tian Yan-Jiang Wang | 2020 | Translational Neurodegeneration2020,9,2: | 12
Study on the micro-replication of shark skin显示文摘Direct replication of creatural scarfskins to form biomimetic surfaces with relatively vivid morphology is a new attempt of the bio-replicated forming technology at animal body. Taking shark skins as the replication templates, and the micro-embossing and micro-molding as the material forming methods, the micro-replicating technology of the outward morphology on shark skins was demonstrated. The preliminary analysis on replication precision indicates that the bio-replicated forming technology can replicate the outward morphology of the shark scales with good precision, which validates the application of the bio-replicated forming technology in the direct morphology replication of the firm creatural scarfskins. | HAN Xin ZHANG DeYuan | 2008 | Science China(Technological Sciences)2008,51,7: | 12
High-frequency ultrasound features of basal cell carcinoma and its association with histological recurrence risk显示文摘Background: Due to advances in high-frequency ultrasound technology, it is easier to detect fine structures of skin lesions. The aim of this study was to examine the ultrasonographic features and use recurrence risk stratification to assess the diagnostic performance of pre-operative ultrasound examination of basal cell carcinoma (BCC). Methods: This was a retrospective study. Forty-six BCC lesions underwent pre-operative ultrasound examination using 50- and 20- MHz probes. Ultrasonographic shape, margin, internal echoes, hyper-echoic spots, posterior echoes, and depth of the lesion were evaluated and correlated with the risk of recurrence based on histological features. Results: Forty-two patients had 46 skin lesions in total. The high-risk (n = 6) and low-risk (n = 40) groups exhibited considerable overlap in the ultrasonographic manifestations and no significant difference in margin (χ^2 = 3.231, P = 0.072), internal echo (χ^2 = 1.592, P = 0.207), or posterior echo (P = 0.169). However, high-risk BCCs tended to be irregular in shape than low-risk lesions (χ^2 = 4.313, P = 0.038). Both types presented hyper-echoic spots (χ^2 = 1.850, P = 0.174). Additionally, 78% of low-risk lesions were confined to the dermis (31/40), and 100% of high-risk lesions infiltrated into the sub-cutaneous tissue, resulting in a significant difference between the two groups (χ^2 = 10.951, P = 0.001). Ultrasound detected sub-clinical lesions in five patients. Conclusions: High-frequency ultrasound can provide important information for pre-operative evaluation of risk in BCC foci and reveal hidden lesions. The technique may play a crucial role in guiding therapeutic options for BCC. | Shi-Qi Wang Jie Liu Qing-Li Zhu Chen-Yang Zhao Tao Qu Feng Li Ximena Wortsman Hong-Zhong Jin | 2019 | Chinese Medical Journal2019,,17: | 12
Electronic Skin from High-Throughput Fabrication of Intrinsically Stretchable Lead Zirconate Titanate Elastomer显示文摘Electronic skin made of thin, soft, stretchable devices that can mimic the human skin and reconstruct the tactile sensation and perception offers great opportunities for prosthesis sensing, robotics controlling, and human-machine interfaces.Advanced materials and mechanics engineering of thin film devices has proven to be an efficient route to enable and enhance flexibility and stretchability of various electronic skins;however, the density of devices is still low owing to the limitation in existing fabrication techniques. Here,we report a high-throughput one-step process to fabricate large tactile sensing arrays with a sensor density of 25 sensors/cm^(2) for electronic skin, where the sensors are based on intrinsically stretchable piezoelectric lead zirconate titanate(PZT)elastomer. The PZT elastomer sensor arrays with great uniformity and passive-driven manner enable highresolution tactile sensing, simplify the data acquisition process, and lower the manufacturing cost. The high-throughput fabrication process provides a general platform for integrating intrinsically stretchable materials into large area, high device density soft electronics for the next-generation electronic skin. | Yiming Liu Huanxi Zheng Ling Zhao Shiyuan Liu Kuanming Yao Dengfeng Li Chunki Yiu Shenghan Gao Raudel Avila Pakpong Chirarattananon Lingqian Chang Zuankai Wang Xian Huang Zhaoqian Xie Zhengbao Yang Xinge Yu | 2020 | Research2020,,1: | 11

Helicobacter pylori and skin autoimmune diseases显示文摘Autoimmune skin diseases are characterized by dysregulation of the immune system resulting in a loss of tolerance to skin self-antigen(s). The prolonged interaction between the bacterium and host immune mechanisms makes Helicobacter pylori(H. pylori) a plausible infectious agent for triggering autoimmunity. Epidemiological and experimental data now point to a strong relation of H. pylori infection on the development of many extragastric diseases, including several allergic and autoimmune diseases. H. pylori antigens activate cross-reactive T cells and induce autoantibodies production. Microbial heat shock proteins(HSP) play an important role of in the pathogenesis of autoimmune diseases because of the high level of sequence homology with human HSP. Eradication of H. pylori infection has been shown to be effective in some patients with chronic autoimmune urticaria, psoriasis, alopecia areata and Schoenlein-Henoch purpura. There is conflicting and controversial data regarding the association of H. pylori infection with Beh et's disease, scleroderma and autoimmune bullous diseases. No data are available evaluating the association of H. pylori infection with other skin autoimmune diseases, such as vitiligo, cutaneous lupus erythematosus and dermatomyositis. The epidemiological and experimental evidence for a possible role of H. pylori infection in skin autoimmune diseases are the subject of this review. | Eli Magen Jorge-Shmuel Delgado | 2014 | World Journal of Gastroenterology2014,20,6: | 10
Stiffness requirement of flexible skin for variable trailing-edge camber wing显示文摘The method for analyzing the deformation of flexible skin under the air loads was developed based on the panel method and finite element method. The deformation of flexible skin under air pressures and effects of the local deformation on the aerodynamic characteristics were discussed.Numerical results show that the flexible skin on the upper surface of trailing-edge will bubble under the air loads and the bubble has a powerful effect on the aerodynamic pressure near the surface of local deforma-tion.Then the stiffness requirements for flexible skin of variable trailing-edge were given by using the Jacobs rule,i.e.,the maximum displacement of skin is not greater than 0.1% of wing chord. Results show that the in-plane stiffness can be reduced by increasing the ratio of bending stiffness to in-plane stiffness.Although the deformation of flexible skin increases with the in-plane stiffness decreasing,it depends on the bending stiffness. When the bending stiffness exceeds critical value, the deformation of flexible skin only depends on the bending stiffness and has nothing to do with the in-plane stiffness. The conclusions can be used for the structural design of flexible skin. | YIN WeiLong Center for Composite Materials and Structures and Postdoctoral Station of Mechanics,Harbin Institute of Technology,Harbin 150080,China | 2010 | Science China(Technological Sciences)2010,53,4: | 9
Lotus seed skin proanthocyanidin extract exhibits potent antioxidant property via activation of the Nrf2-ARE pathway显示文摘Lotus seed is well known as traditional food and medicine, but its skin is usually discarded.Recent studies have shown that lotus seed skin contains a high concentration of proanthocyanidins that have multi-functions,such as antioxidation,anti-inflammation,and anti-cancer effects. In the present study,we aimed to isolate and purify the proanthocyanidins from lotus seed skin by acetone extraction and rotary evaporation, identify their chemical structures by HPLC-MS-MS and NMR,and further investigate the antioxidant properties of the extract purified by macroporous resin (PMR) from lotus seed skin both in vitro and in vivo. The results showed that PMR mainly contained oligomeric proanthocyanidins, especially dimeric procyanidin B1 (PB1),procyanidin B2 and procyanidin B4.Although it had limited ability to directly scavenge radicals in vitro,PMR could significantly enhance the expressions of antioxidant proteins via activation of nuclear factory-related factor 2 (Nrf2)-antioxidant response element (ARE) pathway in HepG2 cells.Molecular data revealed that PB1,a major component in PMR,stabilized Nrf2 by inhibiting the ubiquitination of Nrf2,which led to subsequent activation of the Nrf2-ARE pathway, including the enhancements of Nrf2 nuclear translocation,Nrf2-ARE binding and ARE transcriptional activity. Moreover, the in vivo results in high fat diet-induced mice further verified the powerful antioxidant property of PMR.These results revealed that lotus seed skin is a promising resource for functional food development. | Tao Li Qili Li Weiguo Wu Yong Li De-xing Hou Hua Xu Baodong Zheng Shaoxiao Zeng Yang Shan Xiangyang Lu Fangming Deng Si Qin | 2019 | Acta Biochimica et Biophysica Sinica2019,51,1: | 8
Skin bioprinting:the future of burn wound reconstruction?显示文摘Burns are a significant cause of trauma, and over the years, the focus of patient care has shifted from just survival to facilitation of improved functional outcomes.Typically, burn treatment, especially in the case of extensive burn injuries, involves surgical excision of injured skin and reconstruction of the burn injury with the aid of skin substitutes.Conventional skin substitutes do not contain all skin cell types and do not facilitate recapitulation of native skin physiology.Three-dimensional(3D)bioprinting for reconstruction of burn injuries involves layer-by-layer deposition of cells along with scaffolding materials over the injured areas.Skin bioprinting can be done either in situ or in vitro. Both these approaches are similar except for the site of printing and tissue maturation. There are technological and regulatory challenges that need to be overcome for clinical translation of bioprinted skin for burn reconstruction. However, the use of bioprinting for skin reconstruction following burns is promising;bioprinting will enable accurate placement of cell types and precise and reproducible fabrication of constructs to replace the injured or damaged sites.Overall,3D bioprinting is a very transformative technology, and its use for wound reconstruction will lead to a paradigm shift in patient outcomes. In this review,we aim to introduce bioprinting, the different stages involved,in vitro and in vivo skin bioprinting, and the various clinical and regulatory challenges in adoption of this technology. | Mathew Varkey Dafydd O.Visscher Paul P.Mvan Zuijlen Anthony Atala James J.Yoo | 2019 | Burns & Trauma2019,7,1: | 8
Epidermal stem cells and skin tissue engineering in hair follicle regeneration显示文摘The reconstitution of a fully organized and functional hair follicle from dissociated cells propagated under defined tissue culture conditions is a challenge stillpending in tissue engineering. The loss of hair follicles caused by injuries or pathologies such as alopecia not only affects the patients' psychological well-being, but also endangers certain inherent functions of the skin. It is then of great interest to find different strategies aiming to regenerate or neogenerate the hair follicle under conditions proper of an adult individual. Based upon current knowledge on the epithelial and dermal cells and their interactions during the embryonic hair generation and adult hair cycling, many researchers have tried to obtain mature hair follicles using different strategies and approaches depending on the causes of hair loss. This review summarizes current advances in the different experimental strategies to regenerate or neogenerate hair follicles, with emphasis on those involving neogenesis of hair follicles in adult individuals using isolated cells and tissue engineering. Most of these experiments were performed using rodent cells, particularly from embryonic or newborn origin. However, no successful strategy to generate human hair follicles from adult cells has yet been reported. This review identifies several issues that should be considered to achieve this objective. Perhaps the most important challenge is to provide threedimensional culture conditions mimicking the structure of living tissue. Improving culture conditions that allow the expansion of specific cells while protecting their inductive properties, as well as methods for selecting populations of epithelial stem cells, should give us the necessary tools to overcome the difficulties that constrain human hair follicle neogenesis. An analysis of patent trends shows that the number of patent applications aimed at hair follicle regeneration and neogenesis has been increasing during the last decade. This field is attractive not only to academic researchers but also to the companies that own almost half of the patents in this field. | María Eugenia Balaná Hernán Eduardo Charreau Gustavo Jose Leirós | 2015 | World Journal of Stem Cells2015,7,4: | 7
A role for the Tgf-β/Bmp co-receptor Endoglin in the molecular oscillator that regulates the hair follicle cycle显示文摘The hair follicle is a biological oscillator that alternates growth, regression, and rest phases driven by the sequential activation of the proliferation differentiation programs of resident stem cell populations. The activation of hair follicle stem cell niches and subsequent entry into the growing phase is mainly regulated by Wnt/β-catenin signalling, while regression and resting phases are mainly regulated by Tgf-β/Bmp/Smad activity.A major question still unresolved is the nature of the molecular switch that dictates the coordinated transition between both signalling pathways. Here we have focused on the role of Endoglin(Eng),a key co-receptor for members of the Tgf-β/Bmp family of growth factors.Using an Eng haploinsufficient mouse model,we report that Eng is required to maintain a correct follicle cycling pattern and for an adequate stimulation of hair follicle stem cell niches. We further report thatβ-catenin binds to the Eng promoter depending on Bmp signalling. Moreover,we show thatβ-catenin interacts with Smad4 in a Bmp/Eng-dependent context and both proteins act synergistically to activate Eng promoter transcription. These observations point to the existence of a growth/rest switching mechanism in the hair follicle that is based on an Eng-dependent feedback cross-talk between Wnt/β-catenin and Bmp/Smad signals. | Maria I.Calvo-Sanchez Sandra Fernandez-Martos Elisa Carrasco Gema Moreno-Bueno Carmelo Bernabeu Miguel Quintanilla Jesus Espada | 2019 | Journal of Molecular Cell Biology2019,11,1: | 7
Research on proximity effect of electromagnetic railgun显示文摘The rails of electromagnetic railgun can be ablated by the temperature rise due to current concentration. The current distributions on the rails and armature are not only affected by the skin effect, but also influenced by the proximity effect which is rarely mentioned. This paper illustrated the difference between skin effect and proximity effect, and the influencing factors of proximity effect were investigated. Results show that the current is concentrated on the surface around rails due to the skin effect, and the proximity effect exacerbates the current density on the inner surfaces of rails.Decrease in distance from rails enhances the proximity effect, but has nothing to do with the skin effect, which also augments the rail resistance, resulting in temperature rise.It can explain the reason why the ablation is often detected in the small caliber railgun.Research results in this paper can provide support for design and optimization of electromagnetic railgun. | Yu-tao LOU Hai-yuan LI Bao-ming LI | 2016 | Defence Technology(防务技术)2016,12,3: | 7

Stem cell therapy for chronic skin wounds in the era of personalized medicine:From bench to bedside显示文摘With the significant financial burden of chronic cutaneous wounds on the healthcare system, not to the personal burden mention on those individuals afflicted,it has become increasingly essential to improve our clinical treatments. This requires the translation of the most recent benchtop approaches to clinical wound repair as our current treatment modalities have proven insufficient. The most promising potential treatment options rely on stem cellbased therapies.Stem cell proliferation and signaling play crucial roles in every phase of the wound healing process and chronic wounds are often associated with impaired stem cell function.Clinical approaches involving stem cells could thus be utilized in some cases to improve a body’s inhibited healing capacity. We aim to present the laboratory research behind the mechanisms and effects of this technology as well as current clinical trials which showcase their therapeutic potential.Given the current problems and complications presented by chronic wounds,we hope to show that developing the clinical applications of stem cell therapies is the rational next step in improving wound care. | Elam Coalson Elliot Bishop Wei Liu Yixiao Feng Mia Spezia Bo Liu Yi Shen Di Wu Scott Du Alexander J.Li Zhenyu Ye Ling Zhao Daigui Cao Alissa Li Ofir Hagag Alison Deng Winny Liu Mingyang Li Rex C.Haydon Lewis Shi Aravind Athiviraham Michael J.Lee Jennifer Moriatis Wolf Guillermo A.Ameer Tong-Chuan He Russell R.Reid | 2019 | Genes & Diseases2019,6,4: | 6
Choroidal and cutaneous metastasis from gastric adenocarcinoma显示文摘Choroidal or cutaneous metastasis of gastric cancer is rare. Gastrointestinal cancer was found in only 4% in patients with uveal metastasis. Choroidal metastasis from gastric cancer was reported in two cases in earlier literature. The frequency of gastric cancer as a primary lesion was 6% in cutaneous metastasis of men, and cutaneous metastasis occurs in 0.8% of all gastric cancers. We report a patient with gastric adenocarcinoma who presented with visual disorder in his left eye and skin pain on his head as his initial symptoms. These symptoms were diagnosed to be caused by choroidal and cutaneous metastasis of gastric adenocarcinoma. Two cycles of chemotherapy consisted of oral S-1 and intravenous cisplatin (SPIRITS regimen); this was markedly effective to reduce the primary gastric lesion and almost all the metastatic lesions. | Shoichiro Kawai Tsutomu Nishida Yoshito Hayashi Hisao Ezaki Takuya Yamada Shinichiro Shinzaki Masanori Miyazaki Kei Nakai Takayuki Yakushijin Kenji Watabe Hideki Iijima Masahiko Tsujii Kohji Nishida Tetsuo Takehara | 2013 | World Journal of Gastroenterology2013,19,9: | 6
Necroptosis was found in a rat ischemia/reperfusion injury flap model显示文摘Background:Necroptosis is a new form of cell death that has been identified as a third pathway causing cell death. In this study,necrostatin-1 (Nec-1) was used to determine whether necroptosis exists in a rat ischaemia/reperfusion injury flap model. Methods:In this study, twenty male Sprague-Dawley rats were divided randomly into two groups:a control group (CTL group) and a Nec-1 group. Each abdominal skin flap underwent 3 h of ischaemia and then reperfusion.Fifteen minutes before and after reperfusion, phosphate buffer saline (PBS) was administered intraperitoneally to the CTL group, while Nec-1 was administered intraperitoneally to the Nec-1 group.Twenty-four hours after reperfusion, the whole flap was divided equally into 54 sections.Flap blood perfusion was measured. One sample was taken randomly from each row.Morphological changes,apoptosis,receptor-interacting protein-1 (RIP-1) expression and caspase-3 activity were observed and detected. The measurements between the two groups were compared with the independentt-test,and aPvalue of <0.05 was considered statistically significant. Results:Compared to flaps in the CTL group, flaps in the Nec-1 group showed longer survival rates, better blood perfusion and less inflammatory infiltration. The total flap area considered to have survived was 70.88 ± 10.28% in the CTL group, whereas 80.56 ± 5.40% of the area was found to be living in the Nec-1 group (Nec-1 vs.CTL,t= –2.624,P<0.05).For some rows, there were significant differences in cell apoptosis between the two groups, the apoptosis index (AI) in rows '9 cm','7 cm','6 cm' and '5 cm' was significantly lower in the Nec-1 group than that in the CTL group (Nec-1 vs.CTL,P<0.05).RIP-1 expression was much lower in the Nec-1 group than that in the CTL group in rows '5 cm' to '9 cm' (Nec-1 vs.CTL,P<0.05).No significant differences in caspase-3 activity were found. Conclusion:According to the results, necroptosis was present in a rat abdominal ischaemia/reperfusion injury flap model. | Hao Uu Ming-Zi Zhang Yi-Fang Liu Xin-Hang Dong Yan Hao You-Bin Wang | 2019 | Chinese Medical Journal2019,,1: | 6
Successful rescue of acute liver failure and hemophagocytic lymphohistiocytosis following varicella infection: A case report and review of literature显示文摘Herein we report a case of acute liver failure(ALF) and hemophagocytic lymphohistiocytosis(HLH) induced by varicella infection, successfully rescued by a combination therapy of acyclovir, supportive care, and immunosuppression with dexamethasone and etoposide. A previously healthy 16-year-old boy presented with generalized rash, fever, severe abdominal pain, and abnormal liver function within 4 d. Chickenpox was suspected, and acyclovir and intravenous immunoglobulin were started on admission. However, the patient's condition deteriorated overnight with soaring transaminases, severe coagulopathy and encephalopathy. On the fourth day of admission, pancytopenia emerged, accompanied by hypofibrinogenemia and hyperferritinemia. The patient was diagnosed with ALF. He also met the diagnostic criteria of HLH according to the HLH-2004 guideline. Polymerase chain reaction(PCR) amplifications of varicella-zoster virus(VZV) were positive, confirming that VZV was a causative trigger for ALF and HLH. In view of the devastating immune activation in HLH, immunosuppression therapy with dexamethasone and etoposide was administered, in addition to high dose acyclovir. The patient's symptoms improved dramatically and he finally made a full recovery. To our knowledge, this is only the second report of a successful rescue of ALF associated with HLH, without resorting to liver transplantation. The first case was reported in a neonate infected by herpes simplex virus-1. However, survival data in older children and adults are lacking, most of whom died or underwent liver transplantation. Our report emphasizes the clinical vigilance for the possible presence of HLH, and the necessity of extensive investigation for underlying etiologies in patients presenting with indeterminate ALF. Early initiation of specific therapy targeting the underlying etiology, and watchful immunosuppression such as dexamethasone and etoposide, together with supportive therapy, are of crucial importance in this life-threatening disorder. | Li-Na Zhang Wei Guo Ji-Hong Zhu Yang Guo | 2018 | World Journal of Clinical Cases2018,6,13: | 6
