1Analysis of DNA methylation in different maize tissues显示文摘DNA methylation plays an important role in gene expression regulation during biological development and tissue differentiation in plants. This study adopted methylation-sensitive Amplified fragment length polymorphism (AFLP) to compare the levels of DNA cyto- sine methylation at CCGG sites in tassel, bracteal leaf, and ear leaf from maize inbred lines, 18 White and 18 Red, respectively, and also examined specific methylation patterns of the three tissues. Significant differences in cytosine methylation level among the three tissues and the same changing tendency in two inbred lines were detected. Both MSAP (methylation sensitive amplification polymorphism) ratio and full methylation level were the highest in bracteal leaf, and the lowest in tassel. Meanwhile, different methylation levels were ob- served in the same tissue from the inbred lines, 18 White and 18 Red. Full methylation of internal cytosine was the dominant type in the maize genome. The differential methylation patterns in the three tissues were observed. In addition, sequencing of nine differentially me- thylated fragments and the subsequent blast search revealed that the cytosine methylated 5′-CCGG-3′sequences were distributed in re- peating sequences, in the coding and noncoding regions. Southern hybridization was used to verify the methylation polymorphism. These results clearly demonstrated the power of the MSAP technique for large-scale DNA methylation detection in the maize genome, and the complexity of DNA methylation change during plant growth and development. 3Spatial Interpolation and Sample Size Optimization for Soil Copper(Cu) Investigation in Cropland Soil at County Scale Using Cokriging显示文摘Knowledge on spatial distribution and sampling size optimization of soil copper(Cu) could lay solid foundations for environmetal quality survey of agricultural soils at county scale.In this investigation,cokriging method was used to conduct the interpolation of Cu concentraiton in cropland soil in Shuangliu County,Sichuan Province,China.Based on the original 623 physicochmically measured soil samples,560,498,and 432 sub-samples were randomly selected as target variable and soil organic matter(SOM) of the whole original samples as auxiliary variable.Interpolation results using Cokriging under different sampling numbers were evaluated for their applicability in estimating the spatial distribution of soil Cu at county sacle.The results showed that the root mean square error(RMSE) produced by Cokriging decreased from 0.9 to 7.77%,correlation coefficient between the predicted values and the measured increased from 1.76 to 9.76% in comparison with the ordinary Kriging under the corresponding sample sizes.The prediction accuracy using Cokriging was still higher than original 623 data using ordinary Kriging even as sample size reduced 10%,and their interpolation maps were highly in agreement.Therefore,Cokriging was proven to be a more accurate and economic method which could provide more information and benefit for the studies on spatial distribution of soil pollutants at county scale.PANG Su,LI Ting-xuan,WANG Yong-dong,YU Hai-ying and LI Xi College of Resource and Environmental Science,Sichuan Agricultural University,Ya’an 625014,P.R.China 2009Agricultural Sciences in China2009,8,11:10
4The Quantitative Analysis of Land Use Structure Characteristics of County in Mountainous Areas in Sichuan Province of China——A Case Study of Rong County显示文摘Based on the data concerning detailed survey on land in the year 2009 and land change in the year 2010 in Rong County,a mountainous region of Sichuan Province,by using quantitative geographical model,we conduct quantitative analysis of the status quo of land use in Rong County in terms of land use diversification,land use centralization,land use combination type,land use geographic significance and comprehensive use degree of land use.The results show some characteristics as follows.Firstly,land use in study area displays prominent characteristic of diversification;there is a high degree of completeness land use type;there is a great similarity among towns.Secondly,there is limited combination number of land use type;mostly the combination number of the towns is 2-3;the holistic function of regional land is fragile;the proportion of farmland areas is big,reaching 40.09%;the land use type of 21 towns is farmland.Thirdly,the towns with prominent characteristic of diversification of land use in Rong County,have low degree of centralization of land use and relatively big combination number of land use type,and vice versa.Fourthly,the type and quantity of agricultural land resources with geographical significance are relatively complete with nothing missing;it abounds in untapped land,but the overall index values of land use are all smaller than 300,with low overall use degree of land.ZHOU Wei1,2,WANG Chen1,2,SONG Xing1,2,DENG Liang-ji1,2 1.College of Resource and Environment,Sichuan Agricultural University,Ya’an 625014,China 2.Key Laboratory of Land Information in Sichuan Province,Ya’an 625014,China 2011Asian Agricultural Research2011,3,5:6
5Pathogenic and pathological characteristic of new type gosling viral enteritis first observed in China显示文摘AIM To study the purifying method and characteristics of neW gosling viral enteritis virus (NGVEV), the etiological agent of new gosling viral enteritis (NGVE) which was first recognized in China, as well as the pathomorphological davelapment in goslings infected artificially with NGVEV.``of treatment with chloroform and ammonium sulfate precipitation, dialysis to remove the sulfate radical and ammonium ion and separation by gel filtration Sichuan goslings were orally administered with NGVEV and 24hr later 2 birds were randomly selected and killed at 24hr intervals Until death occurred. Specimens(duodenum, ileum,liver, heart, kidney, spleen, lung, proventriculus, pancreas,esophagus, and the intestinal embolus) were taken until all birds in this group died and were sectioned and stained with hemotoxylin and ensin and studied by light microscope.``RESULTS NGVEV shared the typical characteristics of Adenovirus and which structural proteins consisted of 15polypsptides. Necrosis and sloughing of the epithelial cells covering the villus tips of the duodanum were first observed in goslings 2 days postinfectien artificially with NGVEV. With the progress of infection, this lesion rapidly occurred in the epithelium at the base of the villus and with infiltration of the inflammatory cells, the jejunum tended to be involved. With the intensification of rnucnsa necrosis and inflammatory exudation of the small intestine, fibrinenecrotic enteritis was further developed and embolus composed of either intestinal contents wrapped by pseudomembrane or of the mixture of fibrous exudate and necrotic intestinal rnucoes were observed in the middle-lower part of the small intestine. This structure occluded the intestinal tract and made the intestine dilated in appearance. The intestinal glandular cells underwent degeneration, necrosis and might be found sloughed into the lumen. Hemorrhage and hyperemia could be observed on the lung and kidney . Epithelial cells of the renal tubular underwent degeneration. In some cases, granular degeneration and fatty degeneration could be found in the liver and in some cases at a later stage of this disease the epithelial ceils of trachea and proventriculus might be found sioughed. In some cases at an early stage of this disease,cardiac hyperemia and hemorrhage could be observed.Esophaagus, pancreas and brain were found normal. 6Operation Risk of Rural Cooperative Economic Organization Based on Principal Component Analysis——A Case of Sichuan Province,China显示文摘Based on the previous research,a total of 10 risk indices are selected according to the specific situation in Sichuan Province,such as natural risk,resource constraint risk,internal management risk,capital risk,technical input and output risk,market risk,contract credit risk,property risk and policy support risk.According to the questionnaire survey by cooperative economic organization of Sichuan Province,principal component analysis is used to analyze the importance and influence degree of the operation risk of rural cooperative economic organization.Result shows that in the operation of cooperative economic organization,management risk has greater influence degree and importance degree,which should be paid special attention to.Capital,property,price fluctuation,and quality of members have great degree of risk and are paid little attention to.The technical input and output risk and the contract credit risk have relatively low impact on the operation risk of cooperative economic organization and their importance is relatively low.Therefore,we should continue to maintain its advantages.Nature,resource constraint and policy support have relatively high risk awareness and low influence degree,belonging to the factors with priority attention which are controlled to some extent.Finally,suggestions are put forward,such as enhancing the management level of cooperative economic organizations,strengthening the farmer's risk awareness,and improving the risk aversion ability of cooperative economic organization.JIA Xian-wei,FENG Ling School of Economics and Management,Sichuan Agricultural University,Yaan 625014,China 2010Asian Agricultural Research2010,2,3:3
7Single nucleotide polymorphisms in chicken lmbr1 gene were associated with chicken growth and carcass traits显示文摘Lmbr1 is the key candidate gene controlling vertebrate limb development, but its effects on animal growth and carcass traits have never been reported. In this experiment, lmbr1 was taken as the candi-date gene affecting chicken growth and carcass traits. T/C and G/A mutations located in exon 16 and one A/C mutation located in intron 5 of chicken lmbr1 were detected from Silky, White Plymouth Rock broilers and their F2 crossing chickens by PCR-SSCP and sequencing methods. The analysis of vari-ance (ANOVA) results suggests that T/C polymorphism of exon 16 had significant association with eviscerated yield rate (EYR), gizzard rate (GR), shank and claw rate (SCR) and shank girth (SG); A/C polymorphism of intron 5 was significantly associated with SCR, liver rate and head-neck weight (HNW), while both sites had no significant association with other growth and carcass traits. 11Amino Acids Analysis in Different Antigens and Relationship with Immunoreactivity in Cysticercus cellulosae显示文摘The six antigens of Cysticercus cellulosae: CFag, CSag, CBWag, CWag, UCWag and TSag were prepared respectively. The amino acids of the antigens were determined quantitative by automatic amino acid analyzer. The relationship of original reaction was confirmed between amino acids and antigens. The results showed that there were 17 amino acids among all of the antigens. There were no significant difference (P >0.05) of Asp, Glu, Lys, His in composition of all antigens by data analysis software SPSS. There are also no significant difference ( P > 0.05) in the composition of all amino acids between CSag and CBWag. The composition of Thr, Ser, Tyr, Ved, Pro in UCWag shows significant difference (P < 0.05) with other antigens.The sensitivity and peculiarity of UCWag are higher than that of the others. 12Relationship of the Interests between the Dairy Farmer and the Company in Ecological Agriculture——A Case of the Dairy Industry in Sichuan Province,China显示文摘Development status of dairy industry in Sichuan Province,China is analyzed,indicating that how to ensure the high-quality and ecological production of peasant households has become an important issue for the safety and healthy of the masses and the development of dairy industry.Game model is established and the solution for the model is obtained.Based on analyzing the relationship of the interests between the dairy farmer and the company in ecological agriculture,two mixed strategies of Nash equilibrium are obtained.One is that when the probability is equal to α*1,there are no differences between purchasing or not purchasing;if the probability of company purchase is equal to β*1,there are no differences between producing high-quality milk and poor-quality milk.The other is that if the probability of dairy farmer under normal transaction is equal to α*2,there are no differences for the company to select high-quality inspection or low-quality inspection;if the probability of high-quality inspection is equal to β*2,passing off inferior milk as high-grade one has no differences with normal transaction.Therefore,moral risk of the dairy farmer should be prevented by probability β*1 and high-quality inspection.Finally,countermeasures are put forward,such as reducing the risk of transaction,ensuring the basic interest of the dairy farmer engaged in ecological production,intensifying the supervision,correctly preventing the moral risk of dairy farmer,improving the laws and regulations,and coordinating interests between dairy farmer and company in industrial chain.CHEN Jia-wei,QI Yan-bin College of Economics and Management,Sichuan Agricultural University,Yaan 625014,China 2010Asian Agricultural Research2010,2,8:1
13Preparation of giant unilamellar CdTe quantum dot vesicles and their metabolic pathway in vivo显示文摘Glutathione (GSH) capped CdTe quantum dots (QDs) with photoluminescence quantum yields of 61% and the maximum emitting at 601.2 nm were prepared in water phase.Giant unilamellar CdTe quantum dot vesicles (GUVs-CdTe),with diameters larger than 1.5 μm,were obtained using lower-pressure evaporation techniques with soybean lecithin.Compared with other QD liposomes,the entrapment efficiency of GUVs-CdTe for QDs has been significantly improved to 86.3%.After GUVs-CdTe were injected into mice through the tail vein,the fluorescence microscopy of tissue sections showed that GUVs-CdTe could not pass through the blood-brain barrier and air-blood barrier,which were removed mostly by the reticuloendothelial system and were widely distributed in the spleen and the liver.This behavior is the same as the character of the metabolic pathway of giant unilamellar vesicles by intravenous injections in mice.WANG XianXiang1,3,YANG ZhongKe2,SAN Zhi1,PEN Xi2 & HUANG QianMing1 1College of Biology and Science,Sichuan Agricultural University,Ya’an 625014,China 2College of Veterinary Medicine,Sichuan Agricultural University,Ya’an 625014,China 3College of Chemistry,Sichuan University,Chengdu 610041,China 2010Science China Chemistry2010,53,8:1
14Analysis on the Offside and Vacancy in the Administration of the Village Party Branch and the Villagers′ Committee in Sichuan Province——Based on Villagers′ Autonomy显示文摘In the paper, the researches on the administration scope, the leadership style of the village party branch, the core status of village party branch and the autonomy of villagers′ committee are expounded. On the basis of the empirical investigation of countryside in Sichuan Province, the offside and vacancy of the administration of the 'two committees in village' in the villagers′ autonomy are studied tentatively. The results show that the offside and vacancy of the 'two committees in village' can be divided into three types, covering the village party branch-dominated, the villagers′ committee-dominated type and the antagonism of the two village committees. The reasons for the offside and vacancy of administration of the 'two committees in village' are analyzed, including the fragility of the two committees of the village; the imbalanced power sources of the two committees of the village; the imbalance of the competition for interests of the two committees and the negative effects of traditional politics and culture. 15Suitability Evaluation on the Information Service System of 'Agricultural Science and Technology 110' in Sichuan Province, China显示文摘Method of fuzzy comprehensive evaluation is used to carry out suitability evaluation on the information service system of 'Agricultural Science and Technology 110' in Sichuan Province, China. Analytic hierarchy process (AHP) and expert consultation method are used to determine the evaluation index system and index weight. Suitability effect of the information service system of 'Agricultural Science and Technology 110' is taken as the target layer. The five indices at criterion layer are the organizational mode of agricultural information service, the support system of agricultural information resources, the agricultural information transfer system, the capital source of agricultural information service, and the support system of agricultural information service. And the index layer includes the talent team of agent service, the interaction between the subject and object, the accuracy of agricultural information, the convenience of agricultural information transfer, the adequacy of fund, the efficiency in the use of fund, the status of information infrastructure and so on. Evaluation result shows that the information service system of 'Agricultural Science and Technology 110' in Sichuan Province is suitable for the rural economic development at present. The major factors restricting the information service system are the lack of continuity, the weak infrastructure of agricultural information infrastructure, and the relatively low education level of agricultural producers. 16Cytoplasmic Male Sterility in Maize显示文摘14 isoplasmic and allonuclear cytoplasmic male sterile lines were used as female parents, 8 tester lines as male parents, 101 F1 progenies were obtained. Fertility restoration response of 101 F1 progenies were investigated through field observation and pollen stainability examination under microscope. 14 isoplasmic and allonuclear cytoplasmic male sterile lines were developed by repeated backcross with recurrent male parent lines for more than 8 generations. The result shows: tester line Zifeng1 not only restored the isoplasmic and allonuclear sterile lines of group C backcrossed with Mo17, Yu30 and Heer, but also completely restored the isoplasmic and allonuclear cytoplasm male sterile lines of group T backcrossed with Mo17, HZS , 1792 ,292 and Yu30. Therefore, nuclear background limits the use of Zifeng1 as a tester for identification of cytoplasmic male sterility. Furthermore RFLPs of mitochondrial DNA of 6 isonuclear and alloplasmic cytoplasmic male sterile lines were analyzed with Bam H Ⅰ and Hind Ⅲ restriction endonuclease and mitochondrial DNA probes pBcmH3 and Cox Ⅱ. The same RFLPs were found within sterile cytoplasm of group C, including C,Chuan G, Lei 2 and Lei 3, but a different RFLP pattern was observed among sterile cytoplasm of group S, C,T and the normal cytoplasm. 20Study on Haploid Inducing and Its Meiotic Abnormality in Maize显示文摘The haploid-inducing line Stock 6 was used to produce haploid maize and expected to obtain maize haploid plants successfully. The detailed meiotic studies on selected haploid maize (n = x = 10) were conducted. Cytogenetic analysis revealed a high frequency of meiotic abnormality occurred in both meiosis Ⅰ and meiosis Ⅱ. During the prophase Ⅰ, univalents were common configurations, and there were bivalents or trivalents in some pollen mother cells, however, a few cells containing five bivalents were also observed. After prophase Ⅰ, chromosomes did not congregate in a single metaphase plate but they were scattered in the cytoplasm. At anaphase Ⅰ, the chromosome distribution was highly irregular with almost all possible combinations. In some cells, chromosomes were grouped into the three or four masses and several spindles appeared. At the tetrad stage of meiosis Ⅱ, cytokinesis splitting abnormality occurred, and a variety of diad, triad, tetrad, pentad, hexad, as well as decury microspores were easily observed.
