3Aerial observations of sea ice and melt ponds near the North Pole during CHINARE2010显示文摘An aerial photography has been used to provide validation data on sea ice near the North Pole where most polar orbiting satellites cannot cover. This kind of data can also be used as a supplement for missing data and for reducing the uncertainty of data interpolation. The aerial photos are analyzed near the North Pole collected during the Chinese national arctic research expedition in the summer of 2010(CHINARE2010). The result shows that the average fraction of open water increases from the ice camp at approximately 87°N to the North Pole, resulting in the decrease in the sea ice. The average sea ice concentration is only 62.0% for the two flights(16 and 19 August 2010). The average albedo(0.42) estimated from the area ratios among snow-covered ice,melt pond and water is slightly lower than the 0.49 of HOTRAX 2005. The data on 19 August 2010 shows that the albedo decreases from the ice camp at approximately 87°N to the North Pole, primarily due to the decrease in the fraction of snow-covered ice and the increase in fractions of melt-pond and open-water. The ice concentration from the aerial photos and AMSR-E(The Advanced Microwave Scanning Radiometer-Earth Observing System) images at 87.0°–87.5°N exhibits similar spatial patterns, although the AMSR-E concentration is approximately 18.0%(on average) higher than aerial photos. This can be attributed to the 6.25 km resolution of AMSR-E, which cannot separate melt ponds/submerged ice from ice and cannot detect the small leads between floes. 4Characteristics of oral methicillin-resistant Staphylococcus epidermidis isolated from dental plaque显示文摘The oral microbial community is widely regarded as a latent reservoir of antibiotic resistance genes. This study assessed the molecular epidemiology, susceptibility profile, and resistance mechanisms of 35 methicillin-resistant Staphylococcus epidermidis(MRSE) strains isolated from the dental plaque of a healthy human population. Broth microdilution minimum inhibitory concentrations(MICs) revealed that all the isolates were nonsusceptible to oxacillin and penicillin G. Most of them were also resistant to trimethoprim(65.7%) and erythromycin(54.3%). The resistance to multiple antibiotics was found to be largely due to the acquisition of plasmid-borne genes. The mec A and dfr A genes were found in all the isolates, mostly dfr G(80%), aac A-aph D(20%), aad D(28.6%), aph A3(22.9%), msr A(5.7%), and the erm C gene(14.3%). Classical mutational mechanisms found in these isolates were mainly efflux pumps such as qac A(31.4%), qac C(25.7%), tet K(17.1%), and nor A(8.6%). Multilocus sequence type analysis revealed that sequence type 59(ST59) strains comprised 71.43% of the typed isolates, and the e BURST algorithm clustered STs into the clonal complex 2-II(CC2-II). The staphyloccoccal cassette chromosome mec(SCCmec) type results showed that 25(71.43%) were assigned to type IV. Moreover, 88.66% of the isolates were found to harbor six or more biofilm-associated genes. The aap, atl E, embp, sdr F, and IS256 genes were detected in all 35 isolates. 5双膦酸盐对骨转移癌骨痛治疗及血清钙磷影响(英文)显示文摘Objective: Bisphosphonates were widely used with the rate of malignant tumor with bone metastases increasing rapidly. The aim of this study was to evaluate zoledronic acid and pamidronic acid on treating ostealgia of malignant tumor with bone metastases and serum calcium, phosphorus and alkaline phosphorus movement. Methods: One Hundred cancer patients with bone metastases were diagnosed according to pathological and imaging methods. After zoledronic acid or pamidronic acid intravenous infusion one and two months, ostealgia degree, serum calcium, serum phosphorus and alkaline phosphorus were observed before bisphosphonates treatment, one and two months after treatment, respectively. Results: Ostealgia degree did not change significantly after diphosphonates treatment from one to two months (P > 0.05). The linear correlation between serum calcium and serum phosphorus on bone metastases cancer patients before therapy was dismissed after bisphosphonates treatment one month and did not recover in two months. The incidence of hypo-calcium after diphosphonate treatment one month (54%) and two months (56%) were significantly increased than that before treatment (36%) (χ2 = 6.55, P = 0.011; χ2 = 8.05, P = 0.005). Serum calcium and serum phosphorus were both decreased after treatment one month (t = 4.39, P = 0.000; t = 2.50, P = 0.014) and two months (t = 4.32, P = 0.000; t = 2.49, P = 0.010). There had no difference between zoledronic acid and pamidronic acid on treating of ostealgia and serum calcium and phosphorus changing. Conclusion: Zoledronic acid and pamidronic acid can relief ostealgia of cancer patients with bone metastases and induce hypo-calcium, break the linear relationship between calcium and phosphorus. 7Investigation on the genetic-inconsistent paternity cases using the MiSeq FGx system显示文摘Mutations might challenge the paternity index calculation in forensic identification.While many studies have focussed on the autosomal short tandem repeats(A-STR),the mutation status of sex chromosomes and single nucleotide polymorphism(SNP)remain blank.Next generation sequencing(NGS),known as high throughput and large sequence polymorphism,is a promising tool for forensic genetics.To describe the mutation landscapes in the paternity cases with genetic inconsistencies,a total of 63 parentage confirmed paternity cases contained at least one mismatched locus have been collected.The mutations were subsequently evaluated using Verogen’s MPSForenSeqTM DNASignature Kit and a microsatellite instability(MSI)detection kit.The result showed 98.41%(62/63)of the cases had no additional autosomal mutations even when the number of A-STRs increased to 27.As for the sex chromosomes,about 11.11%(7/63)of the cases exhibited either X-STR or Y-STR mutations.D2S1338,FGAand Penta Ewere the most frequent altered STRs,which suggested they might be the mutation hotspots.In addition,a male with sex chromosome abnormality was observed accidently,whose genotype might be 47,XXY,rather than MSI.Nearly 56.90%of the STR loci possessed isoalleles,which might result in higher STR polymorphisms.No Mendelian incompatibility was detected among the SNP markers,which indicated that SNP was a more reliable genetic marker in the genetic-inconsistent paternity cases.Anqi Chen Ruiyang Tao Chengtao Li Suhua Zhang 2022Forensic Sciences Research2022,7,4:0

9Multi-stage Coordinated Robust Optimization for Soft Open Point Allocation in Active Distribution Networks with PV显示文摘To optimize the placement of soft open points(SOPs)in active distribution networks(ADNs),many aspects should be considered,including the adjustment of transmission power,integration of distributed generations(DGs),coordination with conventional control methods,and maintenance of economic costs.To address this multi-objective planning problem,this study proposes a multi-stage coordinated robust optimization model for the SOP allocation in ADNs with photovoltaic(PV).First,two robust technical indices based on a robustness index are proposed to evaluate the operation conditions and robust optimality of the solutions.Second,the proposed coordinated allocation model aims to optimize the total cost,robust voltage offset index,robust utilization index,and voltage collapse proximity index.Third,the optimization methods of the multiand single-objective models are coordinated to solve the proposed multi-stage problem.Finally,the proposed model is implemented on an IEEE 33-node distribution system to verify its effectiveness.Numerical results show that the proposed index can better reveal voltage offset conditions as well as the SOP utilization,and the proposed model outperforms conventional ones in terms of robustness of placement plans and total cost.Anqi Tao Niancheng Zhou Yuan Chi Qianggang Wang Guangde Dong 2023Journal of Modern Power Systems and Clean Energy2023,11,5:0
10Mutations in the DNA polymerase binding pathway affect the immune microenvironment of patients with small-cell lung cancer and enhance the efficacy of platinum-based chemotherapy显示文摘Background:Small-cell lung cancer(SCLC)is characterized by its high malignancy and is associated with a poor prognosis.In the early stages of the disease,platinum-based chemotherapy is the recommended first-line treatment and has demonstrated efficacy.However,SCLC is prone to recurrence and is generally resistant to chemotherapy in its later stages.Methods:Here,we collected samples from SCLC patients who received platinum-based chemotherapy,performed genomic and transcriptomic analyses,and validated our results with publicly available data.Results:SCLC patients with DNA polymerase binding pathway mutations had an improved prognosis after platinum chemotherapy compared with patients without such mutations.Patients in the mutant(MT)group had higher infiltration of T cells,B cells,and M1 macrophages compared with patients without DNA polymerase binding pathway mutations.Conclusions:DNA polymerase binding pathway mutations can be used as prognostic markers for platinum-based chemotherapy in SCLC.Anqi Lin Weiming Mou Lingxuan Zhu Tao Yang Chaozheng Zhou Jian Zhang Peng Luo 2023Cancer Innovation2023,2,6:0
11Comprehensive bioinformatics analysis and experimental validation:An anoikis-related gene prognostic model for targeted drug development in head and neck squamous cell carcinoma显示文摘We analyzed RNA-sequencing(RNA-seq)and clinical data from head and neck squamous cell carcinoma(HNSCC)patients in The Cancer Genome Atlas(TCGA)Genomic Data Commons(GDC)portal to investigate the prognostic value of anoikis-related genes(ARGs)in HNSCC and develop new targeted drugs.Differentially expressed ARGs were screened using bioinformatics methods;subsequently,a prognostic model including three ARGs(CDKN2A,BIRC5,and PLAU)was constructed.Our results showed that the model-based risk score was a good prognostic indicator,and the potential of the three ARGs in HNSCC prognosis was validated by the TISCH database,the model’s accuracy was validated in two independent cohorts of the Gene Expression Omnibus database.Immune correlation analysis and half-maximal inhibitory concentration were also performed to reveal the different landscapes of TIME between risk groups and to predict immuno-and chemo-therapeutic responses.Potential small-molecule drugs for HNSCC were subsequently predicted using the L1000FWD database.Finally,in vitro experiments were used to verify the database findings.The relative ARG mRNA expression levels in HNSCC and surrounding normal tissues remained consistent with the model results.BIRC5 knockdown inhibited anoikis resistance in WSU-HN6 and CAL-27 cells.Molecular docking,real-time PCR,cell counting kit-8(CCK-8),plate clone,and flow cytometry analyses showed that small-molecule drugs predicted by the database may target the ARGs in the prognostic model,inhibit HNSCC cells survival rate,and promote anoikis in vitro.Therefore,we constructed a new ARG model for HNSCC patients that can predict prognosis and immune activity and identify a potential small-molecule drug for HNSCC,paving the way for clinically targeting anoikis in HNSCC.LIN QIU ANQI TAO XIAOQIAN SUN FEI LIU XIANPENG GE CUIYING LI 2023Oncology Research2023,31,5:0

13A telomere-to-telomere genome assembly of Zhonghuang 13,a widely-grown soybean variety from the original center of Glycine max显示文摘Soybean(Glycine max)stands as a globally significant agricultural crop,and the comprehensive assembly of its genome is of paramount importance for unraveling its biological characteristics and evolutionary history.Nevertheless,previous soybean genome assemblies have harbored gaps and incompleteness,which have constrained in-depth investigations into soybean.Here,we present Telomere-to-Telomere(T2T)assembly of the Chinese soybean cultivar Zhonghuang 13(ZH13)genome,termed ZH13-T2T,utilizing PacBio Hifi and ONT ultralong reads.We employed a multi-assembler approach,integrating Hifiasm,NextDenovo,and Canu,to minimize biases and enhance assembly accuracy.The assembly spans 1,015,024,879 bp,effectively resolving all 393 gaps that previously plagued the reference genome.Our annotation efforts identified 50,564 high-confidence protein-coding genes,707 of which are novel.ZH13-T2T revealed longer chromosomes,421 not-aligned regions(NARs),112 structure variations(SVs),and a substantial expansion of repetitive element compared to earlier assemblies.Specifically,we identified 25.67 Mb of tandem repeats,an enrichment of 5S and 48S rDNAs,and characterized their genotypic diversity.In summary,we deliver the first complete Chinese soybean cultivar T2T genome.The comprehensive annotation,along with precise centromere and telomere characterization,as well as insights into structural variations,further enhance our understanding of soybean genetics and evolution.Anqi Zhang Tangchao Kong Baiquan Sun Shizheng Qiu Jiahe Guo Shuyong Ruan Yu Guo Jirui Guo Zhishuai Zhang Yue Liu Zheng Hu Tao Jiang Yadong Liu Shuqi Cao Shi Sun Tingting Wu Huilong Hong Bingjun Jiang Maoxiang Yang Xiangyu Yao Yang Hu Bo Liu Tianfu Han Yadong Wang 2024The Crop Journal2024,12,1:0
