| 2 | Bioreductive prodrugs as cancer therapeutics:targeting tumor hypoxia显示文摘Hypoxia, a state of low oxygen, is a common feature of solid tumors and is associated with disease progression as well as resistance to radiotherapy and certain chemotherapeutic drugs. Hypoxic regions in tumors, therefore, represent attractive targets for cancer therapy. To date, five distinct classes of bioreactive prodrugs have been developed to target hypoxic cells in solid tumors. These hypoxia-activated prodrugs, including nitro compounds, N-oxides, quinones, and metal complexes, generally share a common mechanism of activation whereby they are reduced by intracellular oxidoreductases in an oxygensensitive manner to form cytotoxins. Several examples including PR-104, TH-302, and EO9 are currently undergoing phase II and phase III clinical evaluation. In this review, we discuss the nature of tumor hypoxia as a therapeutic target, focusing on the development of bioreductive prodrugs. | 3 | Three-dimensional DEM-CFD analysis of air-flow-induced detachment of API particles from carrier particles in dry powder inhalers显示文摘Air flow and particle-particle/wall impacts are considered as two primary dispersion mechanisms for dry powder inhalers(DPIs).Hence,an understanding of these mechanisms is critical for the development of DPIs.In this study,a coupled DEM-CFD(discrete element method-computational fluid dynamics)is employed to investigate the influence of air flow on the dispersion performance of the carrier-based DPI formulations.A carrier-based agglomerate is initially formed and then dispersed in a uniformed air flow.It is found that air flow can drag API particles away from the carrier and those in the downstream air flow regions are prone to be dispersed.Furthermore,the influence of the air velocity and work of adhesion are also examined.It is shown that the dispersion number(i.e.,the number of API particles detached from the carrier)increases with increasing air velocity,and decreases with increasing the work of adhesion,indicating that the DPI performance is controlled by the balance of the removal and adhesive forces.It is also shown that the cumulative Weibull distribution function can be used to describe the DPI performance,which is governed by the ratio of the fluid drag force to the pull-off force. | Jiecheng Yang Chuan-Yu Wu Michael Adams | 2014 | Acta Pharmaceutica Sinica B2014，4，1： | 4 |
| 4 | Dynamics of algae growth and nutrients in experimental enclosures culturing bighead carp and common carp: Phosphorus dynamics显示文摘This is the third paper of the series about 'Dynamics of algae growth and nutrients in experimental enclosures culturing bighead carp and common carp'.In this paper,phosphorus dynamics were investigated under the condition of culturing bighead carp and common carp with added fish food(nitrogen dynamics is discussed in the second paper because their behaviors are so different from each other).Nearly fifty days' observation results indicated that the reservoir water was typical of 'phosphorus limited' water,and soluble reactive phosphorus(SRP) was the main constituent of measured total phosphorus(TP).The presence of fish food resulted in significantly higher SRP,dissolved total phosphorus(DTP) and TP concentrations in contrast with the reservoir water.Moreover,continuous supply of fish food led to the decline of total nitrogen to total phosphorus(TN:TP) from more than 100 to less than5.Variations in the ratio of TN to TP favored the growth of blue-green algae.Fish species affected phosphorus concentrations,and culturing bait-eating common carp contributed more to reducing the SRP,DTP and TP concentrations than culturing planktivorous bighead carp.0.5%,4.1%and 3.1%TP can be removed in enclosures with culturing bighead carp,common carp and mixed bighead carp and common carp,respectively.Abundant phosphorus in the fish culturing activities may be present as the uneaten food,algae cells,and within the water column and sediment,which should be taken into serious consideration for the target of future water eutrophication prevention and safety of the drinking water supply. | Suiliang Huang Min Wu Changjuan Zang Shenglan Du Joseph Domagalski Magdalena Gajewska Feng Gao Chao Lin Yong Guo Baoyan Liu Shaoming Wang Yang Luo Adam Szymkiewicz Romuald Szymkiewicz | 2016 | International Journal of Sediment Research2016，31，2： | 3 |
