| 1 | Roles of Chemokine Receptor 4 (CXCR4) and Chemokine Ligand 12 (CXCL12) in Metastasis of Hepatocellular Carcinoma Cells显示文摘Chemokines are involved in human hepatocellular carcinoma (HCC) carcinogenesis. However, the exact mechanism of chemokines in HCC carcinogenesis remains unknown. Here we investigated the roles of chemokine receptor 4 (CXCR4) and chemokine ligand 12 (CXCL12) in the metastasis of HCC. We found that the expression levels of CXCR4 mRNA in HCC tissues, MHCC97 cells, and HUVEC cells were 2.52 ± 1.13, 2.34 ± 1.16 and 1.63 ± 1.26, respectively and that the CXCR4 protein levels were 1.38 ± 0.13, 1.96 ± 0.32 and 1.86 ± 0.21, respectively. In contrast, CXCR4 was not detected in normal hepatic tissues. In 78 HCC patients, we also found that the concentration of CXCL12 in cancerous ascitic fluid was 783-8,364 pg/ml and that CXCL12 mRNA level in HCC metastasis portal lymph nodes was 1.21 ± 0.87 but undetectable in normal hepatic tissues. Finally we discovered that recombinant human CXCL12 could induce MHCC97 cells and HUVEC cells to migrate with chemotactic indexes (CI) of 3.9 ± 1.1 and 4.1 ± 1.6, respectively. Cancerous ascitic fluid could also induce the migration of MHCC97 cells with a CI of 1.9 ± 0.8. | 3 | BARRIER LAYER IN THE NORTHEASTERN SOUTH CHINA SEA AND ITS FORMATION MECHANISM显示文摘Robust evidence for the barrier layer (BL) in the northeastern South China Sea (SCS) (16°-25°N, 112°-124°E) is presented. The occurrence rate of the BL peaks in the autumn (45.7%) and then the summer (31.1%) and the spring (23.3%), se-quently. It is estimated that the annual occurrence rate of the BL reaches about 40.0% in the central northeastern SCS (18°-22°N, 112°-120°E) and the Luzon Strait. Stratification-formed (Rain-formed) mechanism is the major factor responsible for the occurrence of the BL in the northeastern SCS in the spring (the summer and autumn), respectively. | 4 | Mesoscale eddy effects on the wintertime vertical mixing in the formation region of the North Pacific Subtropical Mode Water显示文摘Mesoscale eddy effects on the wintertime verti-cal mixing in the formation region of the North Pacific Sub-tropical Mode Water (NPSTMW) are studied using hydro-graphic data from Argo profiling floats deployed in the Ku-roshio recirculation region in February and March of 2001. Anticyclonic (warm) eddy enhances the wintertime vertical mixing and results in the deep mixed layer and the deep thermocline. Consequently, a large volumetric water mass with low potential vorticity corresponding to the prototype of NPSTMW tends to be formed. By contrast, cyclonic (cold) eddy is unfavorable for the vertical mixing process and halts the deepening of the mixed layer and thus the formation of mode water. | 5 | Responses of the Zhe-Min coastal current adjacent to Pingtan Island to the wintertime monsoon relaxation in 2006 and its mechanism显示文摘In conjunction with synchronous remotely sensed winds and sea surface temperature(SST),the spatiotemporal features of the Zhe-Min coastal current(ZMCC),especially responses of the ZMCC adjacent to Pingtan Island(PT) to the wintertime monsoon relaxation in 2006 and corresponding mechanism are investigated based on the field observations.In situ data are acquired from Conductivity-Temperature-Depth(CTD) cruise and Bottom-Mounted Moorings(BMM),which are conducted during a comprehensive survey for the Chinese Offshore Investigation and Assessment Project in winter 2006.It is revealed that the ZMCC is well mixed vertically in winter 2006.The ZMCC(<14°C) recedes during the relaxation of the wintertime monsoon and is accompanied by the enhanced northward shift of the warm,saline Taiwan Strait Mixed Water(TSMW,higher than 14°C and is constituted by the Taiwan Strait Warm Water and the Kuroshio Branch Water).And greatly enhanced southward intrusion of the ZMCC can be detected when the wintertime monsoon restores.Correspondingly,the thermal interface bounded by the ZMCC and the TSMW moves in the northwest/southeast direction,leading to periodic warm/cold reversals of the near-seabed temperature adjacent to the PT.By EOF(Empirical Orthogonal Function) analysis of the large-scale wind fields and wavelet power spectrum analysis of the water level,ocean current and the near-seabed temperature,responses of the ZMCC off the PT to wintertime monsoon relaxation are suggested to be attributed mainly to the southward propagating coastally trapped waves triggered by the impeding atmospheric fronts.As a result,ocean current and near-seabed temperature demonstrate significant quasi-5 d and quasi-10 d subtidal oscillations.By contrast,the onshore/offshore water accumulation resulted from Ekman advection driven by the local winds has minor contributions. | PAN AiJun WAN XiaoFang GUO XiaoGang JING ChunSheng | 2013 | Science China Earth Sciences2013,56,3: | 5 |
| 6 | The spatio-temporal variation of wintertime subtidal currents in the western Taiwan Strait显示文摘A new data set of observations by six cruises of ship-mounted acoustic doppler current profiler(SADCP) and three 40 d long bottom-mounted ADCPs(BADCPs) is employed to reveal the spatiotemporal variability of tidal and subtidal currents in the western Taiwan Strait(TWS) during winter season. The results confirm the existence of intense cotidal lines for M_2 tidal current, which is located north of 25°N. In this case, no existence of an amphidromic point can be identified. It is also revealed that the counter-wind current(CWC) can extend through the whole western TWS and even occupy the entire water column during winter monsoon relaxation. However,this CWC is observed to be thoroughly overwhelmed by the downwind China coastal current(CCC) during the two big monsoon bloom events in the winter of 2007, and the CCC consequently extends southward throughout the western TWS instead. | 7 | Responses of Guangdong coastal upwelling to the summertime typhoons of 2006显示文摘The effects of typhoon intrusion on the Guangdong coastal upwelling system were investigated on the basis of in situ CTD (conductivity-temperature-depth) cruise observations and especially upward-looking ADCP (Acoustic Doppler Current Profil- ers) measurements obtained from a comprehensive survey of the Guangdong coastal region carried out by the Chinese Off-shore Investigation and Assessment Project in the summer of 2006. It was found that northeastward geostrophic advection driven by the summer monsoon has a significant near-seabed onshore component adjacent to Shantou, which in conjunction with upper-level offshore Ekman flow, constitutes the canonical Guangdong coastal upwelling system. Further analyses suggested that the Guangdong coastal upwelling system is sensitive to subtle changes in the typhoon intensity and migration pathway. On one hand, as a typhoon approaches from north of the upwelling system (e.g. Typhoon 0604 (Bilis) and Typhoon 0605 (Kaemi)) in the early phase of intrusion, the enhanced southwesterly leads to exceptional enhancement of the onshore flow; i.e., enhanced upwelling. Afterward, irrespective of the forced ocean responses resulting from the stronger local winds (Typhoon 0604) or the moderate typhoon-induced inertial oscillations (Typhoon 0605), the situation is not conducive to sustaining a stable, persistent upwelling system. On the other hand, when there is typhoon intrusion south of the upwelling system (e.g. Typhoon 0606 (Prapiroon)), the favorable southwesterly tends to be substituted by an anomalous northeasterly, which destroys the traditional coastal upwelling pattern. | 8 | Cyclin-dependent Kinase 18 Promotes Oligodendrocyte Precursor Cell Differentiation through Activating the Extracellular Signal-Regulated Kinase Signaling Pathway显示文摘The correct differentiation of oligodendrocyte precursor cells(OPCs) is essential for the myelination and remyelination processes in the central nervous system.Determining the regulatory mechanism is fundamental to the treatment of demyelinating diseases. By analyzing the RNA sequencing data of different neural cells, we found that cyclin-dependent kinase 18(CDK18) is exclusively expressed in oligodendrocytes. In vivo studies showed that the expression level of CDK18 gradually increased along with myelin formation during development and in the remyelination phase in a lysophosphatidylcholine-induced demyelination model, and was distinctively highly expressed in oligodendrocytes. In vitro overexpression and interference experiments revealed that CDK18 directly promotes the differentiation of OPCs, without affecting their proliferation or apoptosis. Mechanistically, CDK18 activated the RAS/mitogen-activated protein kinase kinase1/extracellular signal-regulated kinase pathway, thus promoting OPC differentiation. | 9 | Subtropical Mode Water in the Northwestern Pacific显示文摘Based on the in situ XBT and other data sets, by analyzing the seasonal cycle of the mixed layer depth (MLD)and using the conservative potential vorticity (PV) as a tool, a clear description of the formation process of the North PacifNPSTMW reflects well the ventilation process of the isotherms of the permanent thermocline. The formation process can be the sea surface from October, the mixed layer deepens and correspondingly, the water mass with low PV emerges and sinks.After continual cooling from October to March, the mixed layer reaches its maximum value (>300 m) in March. Then, in the second phase (April-June), the mixed layer shoals rapidly from April, a large part of the low PV water mass is sheltered from further air-sea interaction by the emerging seasonal thermocline, and thus forms new NPSTMW. Further analysis indicatesthat the formation region of warm NPSTMW (17-18℃) is limited between 140°-150°E, while the relatively cold NPSTMW (16-17℃) originates in a wider longitude range (140°-170°E).Climate features of NPSTMW are presented with the use of climatological Levitus (1994a, b) dataset. It is shown that NPSTMW lies in the region of(130°-170°E, 22°-34°N) with core temperature ranging from about 16-19℃ and potential density around 25-25.8σ0. | 10 | Conceptual model about the interaction between El Nio/ Southern Oscillation and Quasi-Biennial Oscillation in far west equatorial Pacific显示文摘Interaction between the Quasi-Biennial Oscillation in far west equatorial Pacific (QBOWP) and the El Ni?o/Southern Oscillation (ENSO) is studied using a new conceptual model. In this con- ceptual model, the QBOWP effects on ENSO are achieved through two ways: (1) the oceanic Kelvin wave along equatorial Pacific, and (2) the Atmospheric Walker Circulation anomaly, while ENSO ef- fects on QBOWP can be accomplished by the atmospheric Walker Circulation anomaly. Diagnosis analysis of the model results shows that the Atmospheric bridge (Walker circulation) plays a more important role in interaction between the ENSO and QBOWP than the oceanic bridge (oceanic Kelvin wave along equatorial Pacific); It is found that by the interaction of the ENSO and QBOWP, a free ENSO oscillation with 3-5 years period could be substituted by a oscillation with the quasi-biennial period, and the dominant period of SST anomaly and wind anomaly in the far west equatorial Pacific tends to be prolonged with enhanced ENSO forcing. | 12 | Tocilizumab in patients with moderate or severe COVID-19: a randomized, controlled, open-label, multicenter trial显示文摘Tocilizumab has been reported to attenuate the“cytokine storm”in COVID-19 patients.We attempted to verify the effectiveness and safety of tocilizumab therapy in COVID-19 and identify patients most likely to benefit from this treatment.We conducted a randomized,controlled,open-label multicenter trial among COVID-19 patients.The patients were randomly assigned in a 1:1 ratio to receive either tocilizumab in addition to standard care or standard care alone.The cure rate,changes of oxygen saturation and interference,and inflammation biomarkers were observed.Thirty-three patients were randomized to the tocilizumab group,and 32 patients to the control group.The cure rate in the tocilizumab group was higher than that in the control group,but the difference was not statistically significant(94.12%vs.87.10%,rate difference 95%CI−7.19%–21.23%,P=0.4133).The improvement in hypoxia for the tocilizumab group was higher from day 4 onward and statistically significant from day 12(P=0.0359).In moderate disease patients with bilateral pulmonary lesions,the hypoxia ameliorated earlier after tocilizumab treatment,and less patients(1/12,8.33%)needed an increase of inhaled oxygen concentration compared with the controls(4/6,66.67%;rate difference 95%CI−99.17%to−17.50%,P=0.0217).No severe adverse events occurred.More mild temporary adverse events were recorded in tocilizumab recipients(20/34,58.82%)than the controls(4/31,12.90%).Tocilizumab can improve hypoxia without unacceptable side effect profile and significant influences on the time virus load becomes negative.For patients with bilateral pulmonary lesions and elevated IL-6 levels,tocilizumab could be recommended to improve outcome. | Dongsheng Wang Binqing Fu Zhen Peng Dongliang Yang Mingfeng Han Min Li Yun Yang Tianjun Yang Liangye Sun Wei Li Wei Shi Xin Yao Yan Ma Fei Xu Xiaojing Wang Jun Chen Daqing Xia Yubei Sun Lin Dong Jumei Wang Xiaoyu Zhu Min Zhang Yonggang Zhou Aijun Pan Xiaowen Hu Xiaodong Mei Haiming Wei Xiaoling Xu | 2021 | Frontiers of Medicine2021,15,3: | 2 |
| 13 | North-south difference of water mass properties across the Lembeh Strait, North Sulawesi, Indonesia显示文摘Two field observations were conducted around the Lembeh Strait in September 2015 and 2016, respectively.Evidences indicate that seawater around the Lembeh Strait is consisted of North Pacific Tropical Water(NPTW),North Pacific Intermediate Water(NPIW), North Pacific Tropical Intermediate Water(NPTIW) and Antarctic Intermediate Water(AAIW). Around the Lembeh Strait, there exist some north-south differences in terms of water mass properties. NPTIW is only found in the southern Lembeh Strait. Water mass with the salinity of 34.6 is only detected at 200–240 m between NPTW and NPTIW in the southern Lembeh Strait, and results from the process of mixing between the saltier water transported from the South Pacific Ocean and the lighter water from the North Pacific Ocean and Sulawesi Sea. According to the analysis on mixing layer depth, it is indicated that there exists an onshore surface current in the northern Lembeh Strait and the surface current in the Lembeh Strait is southward.These dramatic differences of water masses demonstrate that the less water exchange has been occurred between the north and south of Lembeh Strait. | 20 | Prevention of urinary tract infection using a silver alloy hydrogel-coated catheter in critically ill patients: A single-center prospective randomized controlled study显示文摘Background:A new type of silver alloy hydrogel-coated(SAH)catheter has been shown to prevent bacterial adhesion and colonization by generating a microcurrent,and to block the retrograde infection pathway.However,these have only been confirmed in ordinary patients.This study aims to evaluate the effectiveness of a SAH catheter for preventing urinary tract infections in critically ill patients.Methods:This was a prospective single-center,single-blind,randomized,controlled study.A total of 132 patients requiring indwelling catheterization in the intensive care unit(ICU)of the First Affiliated Hospital of the University of Science and Technology of China between October 2022 and February 2023 and who met the study inclusion/exclusion criteria were randomly divided into two groups.Patients in the SAH catheter group received a SAH catheter,while patients in the conventional catheter group received a conventional siliconized latex Foley catheter.The main outcome measure was the incidence of catheter-associated urinary tract infections(CAUTIs).Secondary outcome indicators included urine positivity for white blood cells and positive urine cultures on 3 days,7 days,10 days,and 14 days after catheterization,number of viable bacteria in the catheter biofilm on day 14,pathogenic characteristics of positive urine cultures,length of ICU stay,overall hospital stay,ICU mortality,and 28-day mortality.All the data were compared between the two groups.Results:A total of 68 patients in the conventional catheter group and 64 patients in the SAH catheter group were included in the study.On day 7 after catheter placement,the positivity rate for urinary white blood cells was significantly higher in the conventional catheter group than in the SAH catheter group(33.8%vs.15.6%,P=0.016).On day 10,the rates of positive urine cultures(27.9%vs.10.9%,P=0.014)and CAUTIs(22.1%vs.7.8%,P=0.023)were significantly higher in the conventional catheter group than in the SAH catheter group.On day 14,the numbers of viable bacteria isolated from the catheter tip([3.21±1.91]×10^(6) colony-forming units[cfu]/mL vs.[7.44±2.22]×10^(4) cfu/mL,P<0.001),balloon segment([7.30±1.99]×10^(7) cfu/mL vs.[3.48±2.38]×10^(5) cfu/mL,P<0.001),and tail section([6.41±2.07]×10^(5) cfu/mL vs.[8.50±1.46]×10^(3) cfu/mL,P<0.001)were significantly higher in the conventional catheter group than in the SAH catheter group.The most common bacteria in the urine of patients in both groups were Escherichia coli(n=13)and Pseudomonas aeruginosa(n=6),with only one case of Candida in each group.There were no significant differences between the two groups in terms of ICU hospitalization time,total hospitalization time,ICU mortality,and 28-day mortality.Conclusion:SAH catheters can effectively inhibit the formation of catheter-related bacterial biofilms in critically ill patients and reduce the incidence of CAUTIs,compared with conventional siliconized latex Foley catheters;however,regular replacement of the catheter is still necessary. | Menglong Zhao Shike Geng Lei Zhang Xiaoqin Fan Fei Tong Xianlin Meng Tianfeng Wang Xiaowei Fang Qing Mei Aijun Pan | 2024 | Journal of Intensive Medicine2024,4,1: | 0 |
