1Hot deformation behavior and microstructure evolution of the laser solid formed TC4 titanium alloy显示文摘Hot compressive experiments of the laser solid formed(LSFed)TC4 titanium alloy were conducted at a wide temperature range of 650-950℃and strain rate of 0.01-10 s^(-1).The Arrheniustype constitutive models of the LSFed TC4 alloy were established at the temperature range of 800-950℃and of 650-800℃,respectively.The average relative error between the predicted stresses and experimental values in those two temperature ranges are 10.4%and 8.3%,respectively,indicating that the prediction models constructed in this paper are in a good agreement with experimental data.Processing maps were established by the principle of dynamic materials modeling on the basis of the data achieved from the hot compression experiments.The processing parameters corresponding to the stable and unstable regions of material deformation can be determined from the processing maps.The microstructure evolution of the stable and unstable regions of the samples after tests were observed.Finally,the effect of hot compressive parameters on the microstructure were investigated to research the dynamic recrystallization and the texture of the deformed LSFed TC4 alloy.Xiawei YANG Yanying WANG Xiurong DONG Chong PENG Baijin JI Yaxin XU Wenya LI 2021Chinese Journal of Aeronautics2021,34,5:3
2Interfacial characteristics and mechanical properties of additive manufacturing martensite stainless steel on the Cu-Cr alloy substrate by directed energy deposition显示文摘Copper/steel is a typical bimetal functional material,combining the excellent electrical and thermal conductivity of copper alloy and the high strength and hardness of stainless steel.There has been recent interest in manufacturing copper/steel bimetal by directed energy deposition(DED)due to its layer-bylayer method.However,cracks tend to form on the copper/steel interface because of the great difference in thermal expansion coefficient and crystal structure between copper and steel.In this work,interfacial characteristics and mechanical properties of the copper/steel bimetal were studied from one layer to multilayers.The laser power has a great influence on the Cu element distribution of the molten pool,affecting the crack formation dramatically on the solidification stage.Cracks tend to form along columnar grain boundaries because of the Cu-rich liquid films and spherical particles in the cracks.Crack-free and good metallurgical bonding copper/steel interface is formed at a scanning velocity of 800 mm/min and the laser power of 3000 W.The ultimate tensile strength(UTS)and the break elongation(EL)of the vertically combined crack-free copper/steel bimetal are 238.2±4.4 MPa and 20.6±0.7%,respectively.The fracture occurs on the copper side instead of the copper/steel interface,indicating that the bonding strength is higher than that of the Cu-Cr alloy.The UTS of the horizontally combined crack-free copper/steel bimetal is 746.7±22.6 MPa,which is 200%higher than that of the Cu-Cr alloy substrate.The microhardness is 398.6±5.4 HV at the steel side and is 235.3±64.1 HV at the interface,which is400%higher than that of the Cu-Cr alloy substrate.This paper advances the understanding of the interfacial characteristics of heterogeneous materials and provides guidance and reference for the fabrication of multi-material components by DED.Wenqi Zhang Hailong Liao Zhiheng Hu Shasha Zhang Baijin Chen Huanqing Yang Yun Wang Haihong Zhu 2021Journal of Materials Science & Technology2021,,31:2
3Relationship between the exposure dose of high intensity focused ultrasound and the heated necrosis element显示文摘Heated necrosis element of tissues is the basis for treating tumor by high intensity focused ultrasound (HIFU), so study on the relationship between the HIFU exposure dose and the heated necrosis element of tissues is of great significance for both HIFU clinical applications and basic research. Using Pennes and Rayleigh formulae the relationship between the exposure dose of an annular focused transducer and the heated necrosis element (i.e. biological focused region) in a bovine liver in vitro at a depth of 2 cm is simulated in an exposure dose range with radiation sound intensity of 7000~25400 W/cm 2 and exposure time of 0~20 s. The theoretical results appear to be in quite good agreement with the experimental ones. It is shown that on the basis of existing theories the heated necrosis element produced by HIFU exposure can be predicted. 4The interferon-stimulated exosomal hACE2 potently inhibits SARS-CoV-2 replication through competitively blocking the virus entry显示文摘Since the outbreak of coronavirus disease 2019(COVID-19),it has become a global pandemic.The spike(S)protein of etiologic severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)specifically recognizes human angiotensin-converting enzyme 2(hACE2)as its receptor,which is recently identified as an interferon(IFN)-stimulated gene.Here,we find that hACE2 exists on the surface of exosomes released by different cell types,and the expression of exosomal hACE2 is increased by IFNα/β treatment.In particular,exosomal hACE2 can specifically block the cell entry of SARS-CoV-2,subsequently inhibit the replication of SARS-CoV-2 in vitro and ex vivo.Our findings have indicated that IFN is able to upregulate a viral receptor on the exosomes which competitively block the virus entry,exhibiting a potential antiviral strategy.Junsong Zhang Feng Huang Baijin Xia Yaochang Yuan Fei Yu Guanwen Wang Qianyu Chen Qian Wang Yuzhuang Li Rong Li Zheng Song Ting Pan Jingliang Chen Gen Lu Hui Zhang 2021Signal Transduction and Targeted Therapy2021,6,6:2

8Enhancement of CAR-T cell activity against cholangiocarcinoma by simultaneous knockdown of six inhibitory membrane proteins显示文摘Background:Existing treatments for cholangiocarcinoma have poor efficacy.However,chimeric antigen receptor-T(CAR-T)cells are emerging as a potential therapeutic strategy.Solid tumors possess multiple adverse factors in an immunosuppressive microenvironment that impair CAR-T cell infiltration and function.This study aimed to improve the function of CAR-T cells through knock down immune checkpoints and immunosuppressive molecular receptors.Methods:We evaluated the expression of epidermal growth factor receptor(EGFR)and B7 homolog 3 protein(B7H3)antigens in cholangiocarcinoma tissues using immunohistochemistry and screened specific immune checkpoints in the cholangiocarcinoma microenvironment via flow cytometry.Subsequently,we engineered CAR-T cells targeting EGFR and B7H3 antigens.We simultaneously knocked down immune checkpoints and immunosuppressive molecular receptors in CAR-T cells by constructing two clusters of small hairpin RNAs and evaluated the engineered CAR-T cells for antitumor activity both in vitro,using tumor cell lines and cholangiocarcinoma organoid models,and in vivo,using humanized mouse models.Results:We observed high expression of EGFR and B7H3 antigens in cholangiocarcinoma tissues.EGFR-CAR-T and B7H3-CAR-T cells demonstrated specific anti-tumor activity.We found an abundance of programmed cell death protein 1(PD-1),T cell immunoglobulin and mucin domain-containing protein 3(Tim-3),and T cell immunoglobulin and ITIM domain(Tigit)on infiltrated CD8^(+)T cells in the cholangiocarcinoma microenvironment.We then decreased the expression of these 3 proteins on the surface of CAR-T cells,named PTG-scFV-CAR-T cells.Furthermore,we knocked-down the expression of transforming growth factor beta receptor(TGFβR),interleukin-10 receptor(IL-10R),and interleukin-6 receptor(IL-6R)of PTG-scFV-CAR-T cells.Those cells,named PTG-T16R-scFVCAR-T cells,potently killed tumor cells in vitro and promoted apoptosis of tumor cells in a cholangiocarcinoma organoidmodel.Finally,the PTG-T16R-scFv-CART cells showed greater inhibitory effect on tumor growth in vivo,and were superior in prolonging the survival of mice.Conclusions:Our results revealed that PTG-T16R-scFV-CAR-T cells with knockdown of sextuplet inhibitory molecules exhibited strong immunity against cholangiocarcinoma and long-term efficacy both in vitro and in vivo.This strategy provides an effective and personalized immune cell therapy against cholangiocarcinoma.Yidan Qiao Jie Chen Xuemei Wang Shumei Yan Jizhou Tan Baijin Xia Yongjian Chen Keming Lin Fan Zou Bingfeng Liu Xin He Yiwen Zhang Xu Zhang Hui Zhang Xiangyuan Wu Lijuan Lu 2023Cancer Communications2023,43,7:0
9Chiral structures of 6,12-dibromochrysene on Au(111)and Cu(111)surfaces显示文摘Nanoscale low-dimensional chiral architectures are increasingly receiving scientific interest,because of their potential applications in many fields such as chiral recognition,separation and transformation.Using 6,12-dibromochrysene(DBCh),we successfully constructed and characterized the large-area two-dimensional chiral networks on Au(111)and one-dimensional metal-liganded chiral chains on Cu(111)respectively.The reasons and processes of chiral transformation of chiral networks on Au(111)were analyzed.We used scanning tunneling spectroscopy(STS)to analyze the electronic state information of this chiral structure.This work combines scanning tunneling microscopy(STM)with non-contact atomic force microscopy(nc-AFM)techniques to achieve ultra-high-resolution characterization of chiral structures on low-dimensional surfaces,which may be applied to the bond analysis of functional nanofilms.Density functional theory(DFT)was used to simulate the adsorption behavior of the molecular and energy analysis in order to verify the experimental results.Shijie Sun Baijin Li Boyu Fu Zilin Ruan Hui Zhang Wei Xiong Yong Zhang Gefei Niu Jianchen Lu Xiaoqing Zuo Lei Gao Jinming Cai 2022Chinese Chemical Letters2022,33,12:0
