1Tissue-specific transcription reprogramming promotes liver metastasis of colorectal cancer显示文摘Metastasis,the development of secondary malignant growths at a distance from a primary tumor,is the cause of death for 90%of cancer patients,but little is known about how metastatic cancer cells adapt to and colonize new tissue environments.Here,using clinical samples,patient-derived xenograft(PDX)samples,PDX cells,and primary/metastatic cell lines,we discovered that liver metastatic colorectal cancer(CRC)cells lose their colon-specific gene transcription program yet gain a liver-specific gene transcription program.We showed that this transcription reprogramming is driven by a reshaped epigenetic landscape of both typical enhancers and super-enhancers.Further,we identified that the liver-specific transcription factors FOXA2 and HNF1A can bind to the gained enhancers and activate the liver-specific gene transcription,thereby driving CRC liver metastasis.Importantly,similar transcription reprogramming can be observed in multiple cancer types.Our data suggest that reprogrammed tissue-specific transcription promotes metastasis and should be targeted therapeutically.Shuaishuai Teng Yang Eric Li Ming Yang Rui Qi Yiming Huang Qianyu Wang Yanmei Zhang Shanwen Chen Shasha Li Kequan Lin Yang Cao Qunsheng Ji Qingyang Gu Yujing Cheng Zai Chang Wei Guo Pengyuan Wang Ivan Garcia-Bassets Zhi John Lu Dong Wang 2020Cell Research2020,30,1:11
2Multicenter phaseⅡstudy of apatinib single or combination therapy in HER2-negative breast cancer involving chest wall metastasis显示文摘Objective:Breast cancer(BC)with chest wall metastasis(CWM)usually shows rich neovascularization.This trial explored the clinical effect of apatinib on human epidermal growth factor receptor 2(HER2)-negative advanced BC involving CWM.Methods:This trial involved four centers in China and was conducted from September 2016 to March 2020.Patients received apatinib 500 mg/d[either alone or with endocrine therapy if hormone receptor-positive(HR+)]until disease progression or unacceptable toxicity.Progression-free survival(PFS)was the primary endpoint.Results:We evaluated 26 patients for efficacy.The median PFS(mPFS)and median overall survival(mOS)were4.9[range:2.0-28.5;95%confidence interval(95%CI):2.1-8.3]months and 18(range:3-55;95%CI:12.9-23.1)months,respectively.The objective response rate(ORR)was 42.3%(11/26),and the disease-control rate was76.9%(20/26).In the subgroup analysis,HR+patients compared with HR-negative patients had significantly improved mPFS of 7.0(95%CI:2.2-11.8)months vs.2.3(95%CI:1.2-3.4)months,respectively(P=0.001);and mPFS in patients without or with chest wall radiotherapy was 6.4(95%CI:1.6-19.5)months vs.3.0(95%CI:1.3-4.6)months,respectively(P=0.041).In the multivariate analysis,HR+status was the only independent predictive factor for favorable PFS(P=0.014).Conclusions:Apatinib was highly effective for BC patients with CWM,especially when combined with endocrine therapy.PFS improved significantly in patients with HR+status who did not receive chest wall radiotherapy.However,adverse events were serious and should be carefully monitored from the beginning of apatinib treatment.Huiping Li Cuizhi Geng Hongmei Zhao Hanfang Jiang Guohong Song Jiayang Zhang Yaxin Liu Xinyu Gui Jing Wang Kun Li Zhongsheng Tong Fangyuan Zhao Junlan Yang Guoliang Chen Qianyu Liu Xu Liang 2021Chinese Journal of Cancer Research2021,33,2:4
3Highly aligned lithiophilic electrospun nanofiber membrane for the multiscale suppression of Li dendrite growth显示文摘Using inorganic fibrous membranes as protective layers has yielded success in suppressing dendrite growth.However,conventional fibrous membranes usually have large voids and low affinity for Li,promoting inhomogeneous charge distribution and allowing some dendrites to grow.Herein,we introduce a highly aligned TiO_(2)/SiO_(2)(A-TS)electrospun nanofiber membrane as a protective layer for the Li metal anode.The A-TS membrane is fabricated by a custom-made electrospinning system with an automatic fiber alignment collector that allows control of the fibers’orientation.At the scale of the individual fibers,their high binding energies with Li can attract more“dead”Li by reacting with the SiO_(2) component of the composite,avoiding uncontrollable deposition on the metal anode.At the membrane scale,these highly ordered structures achieve homogeneous contact and charge distribution on the Li metal surface,leaving no vulnerable areas to nucleate dendrite formation.Additionally,the excellent mechanical and thermal stability properties of the A-TS membrane prevent any potential puncturing by dendrites or thermal runaway in a battery.Hence,an A-TS@Li anode exhibits stable cycling performance when used in both Li-S and Li-NCM811 batteries,highlighting significant reference values for the future design and development of high-energy-density metal-based battery systems.Jianan Wang Qianyue Ma Shiyi Sun Kai Yang Qiong Cai Emilia Olsson Xin Chen Ze Wang Amr MAbdelkader Yinshi Li Wei Yan Shujiang Ding Kai Xi 2022eScience2022,2,6:2
4The interferon-stimulated exosomal hACE2 potently inhibits SARS-CoV-2 replication through competitively blocking the virus entry显示文摘Since the outbreak of coronavirus disease 2019(COVID-19),it has become a global pandemic.The spike(S)protein of etiologic severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)specifically recognizes human angiotensin-converting enzyme 2(hACE2)as its receptor,which is recently identified as an interferon(IFN)-stimulated gene.Here,we find that hACE2 exists on the surface of exosomes released by different cell types,and the expression of exosomal hACE2 is increased by IFNα/β treatment.In particular,exosomal hACE2 can specifically block the cell entry of SARS-CoV-2,subsequently inhibit the replication of SARS-CoV-2 in vitro and ex vivo.Our findings have indicated that IFN is able to upregulate a viral receptor on the exosomes which competitively block the virus entry,exhibiting a potential antiviral strategy.Junsong Zhang Feng Huang Baijin Xia Yaochang Yuan Fei Yu Guanwen Wang Qianyu Chen Qian Wang Yuzhuang Li Rong Li Zheng Song Ting Pan Jingliang Chen Gen Lu Hui Zhang 2021Signal Transduction and Targeted Therapy2021,6,6:2
5A rice XANTHINE DEHYDROGENASE gene regulates leaf senescence and response to abiotic stresses显示文摘Xanthine dehydrogenase, a member of the molybdenum enzyme family, participates in purine metabolism and catalyzes the generation of ureides from xanthine and hypoxanthine. However, the mechanisms by which xanthine dehydrogenase affects rice growth and development are poorly understood. In the present study, we identified a mutant with early leaf senescence and reduced tillering that we named early senescence and less-tillering 1(esl1). Map-based cloning revealed that ESL1 encodes a xanthine dehydrogenase, and it was expressed in all tissues. Chlorophyll content was reduced and chloroplast maldevelopment was severe in the esl1 mutant. Mutation of ESL1 led to decreases in allantoin, allantoate, and ABA contents. Further analysis revealed that the accumulation of reactive oxygen species in esl1 resulted in decreased photosynthesis and impaired chloroplast development, along with increased sensitivity to abscisic acid and abiotic stresses. 14Nanoparticle accumulation in liver may induce resistance to immune checkpoint blockade therapy显示文摘Despite immune checkpoint blockade(ICB)therapy has transformed cancer treatment,only 20.2%of these patients achieved a response.Understanding resistance mechanisms to ICB is important for the treatment of a wider population.In this work,we occasionally found that the silica nanoparticles(SiO_(2)NPs)accumulated in the liver can induce resistance to following ICB therapy to a subcutaneous tumor in mice.By analysis of T cells frequency,we uncovered that SiO_(2)NPs in the liver resulted in a siphoning of T cells from circulation to the liver by produced chemokines.In addition,liver immunosuppressive cells further inhibit the function and induce apoptosis of recruited T cells,leading to a systemic loss and reduced tumor infiltration of T cells,which contributes to poor responses to ICB therapy.However,such effect is not observed in poly(lactic-co-glycolic acid)(PLGA)NPs treated mice under the same conditions,likely due to their much lower immunogenicity in perturbing the liver immune microenvironment,indicating that cancer is not a local disease but an ecosystem that is linked to the distal environment.We further provide a new mechanism insight into ICB resistance induced by liver accumulation of nanoparticles.Huaxing Dai Qianyu Yang Rong Sun Yue Zhang Qingle Ma Yifan Shen Beilei Wang Yitong Chen Jialu Xu Bo Tian Fang Xu Chao Wang 2023Nano Research2023,16,4:1
