| 1 | AXL is a candidate receptor for SARS-CoV-2 that promotes infection of pulmonary and bronchial epithelial cells显示文摘The current coronavirus disease 2019(COVID-19)pandemic presents a global public health challenge.The viral pathogen responsible,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),binds to the host receptor ACE2 through its spike(S)glycoprotein,which mediates membrane fusion and viral entry.Although the role of ACE2 as a receptor for SARS-CoV-2 is clear,studies have shown that ACE2 expression is extremely low in various human tissues,especially in the respiratory tract.Thus,other host receptors and/or co-receptors that promote the entry of SARS-CoV-2 into cells of the respiratory system may exist.In this study,we found that the tyrosine-protein kinase receptor UFO(AXL)specifically interacts with the N-terminal domain of SARS-CoV-2 S.Using both a SARS-CoV-2 virus pseudotype and authentic SARS-CoV-2,we found that overexpression of AXL in HEK293T cells promotes SARS-CoV-2 entry as efficiently as overexpression of ACE2,while knocking out AXL significantly reduces SARS-CoV-2 infection in HI 299 pulmonary cells and in human primary lung epithelial cells.Soluble human recombinant AXL blocks SARS-CoV-2 infection in cells expressing high levels of AXL.The AXL expression level is well correlated with SARS-CoV-2 S level in bronchoalveolar lavage fluid cells from COVID-19 patients.Taken together,our findings suggest that AXL is a novel candidate receptor for SARS-CoV-2 which may play an important role in promoting viral infection of the human respiratory system and indicate that it is a potential target for future clinical intervention strategies. | Shuai Wang Zongyang Qiu Yingnan Hou Xiya Deng Wei Xu Tingting Zheng Peihan Wu Shaofang Xie Weixiang Bian Chong Zhang Zewei Sun Kunpeng Liu Chao Shan Aifu Lin Shibo Jiang Youhua Xie Qiang Zhou Lu Lu Jing Huang Xu Li | 2021 | Cell Research2021,31,2: | 39 |
| 2 | Correlation of human epidermal growth factor receptor 2 expression with clinicopathological characteristics and prognosis in gastric cancer显示文摘AIM:To investigate human epidermal growth factor receptor 2(HER2) gene amplification and protein expression in Chinese patients with resectable gastric cancer and the association with clinicopathological characteristics and survival.METHODS:One hundred and ninety-seven gastric cancer patients who underwent curative surgery procedures were enrolled into this study.HER2 gene amplification and protein expression were examined using fluorescence in-situ hybridization(FISH) and immunohistochemistry(IHC) analysis on formalin-fixed paraffinembedded gastric cancer samples from all patients.For scoring,Hofmann's HER2 gastric cancer scoring system was adopted.All cases showing IHC3+ or FISH positiv-ity were defined as HER2 positive.Patient clinicopathological data and survival information were collected.Finally,χ 2 statistical analysis was performed to analyze the HER2 positivity rate amongst the subgroups with different clinicopathological characteristics including;gender,age,tumor location,Lauren classification,differentiation,TNM staging,depth of invasion,lymph node metastases and distant metastasis.The probability of survival for different subgroups with different clinicopathological characteristics was calculated using the Kaplan-Meier method and survival curves plotted using log rank inspection.RESULTS:According to Hofmann's HER2 gastric cancer scoring criteria,31 cases(15.74%) were identified as HER2 gene amplified and 19 cases(9.64%) were scored as strongly positive for HER2 membrane staining(3+),25 cases(12.69%) were moderately positive(2+) and 153 cases(77.66%) were HER2 negative(0/1+).The concordance rate between IHC and FISH analyses was 88.83%(175/197).Thirty-six cases were defined as positive for HER2 gene amplification and/or protein expression,with 24 of these cases being eligible for Herceptin treatment according to United States recommendations,and 29 of these cases eligible according to EU recommendations.Highly consistent results were detected between IHC3+,IHC0/1 and FISH(73.68% and 95.42%),but low consistency was observed between IHC2+ and FISH(40.00%).The positivity rates in intestinal type and well-differentiated gastric cancer were higher than those in diffuse/mixed type and poorly-differentiated gastric cancer respectively(28.57% vs 13.43%,P = 0.0103;37.25% vs 11.64%,P < 0.0001),but were not correlated with gender,age,tumor location or TNM stage,depth of invasion,lymph node metastases and distant metastasis.In poorly-differentiated gastric cancer patients,those without lymph node metastasis showed a higher HER2 positivity rate than those with lymph node metastasis(26.47% vs 7.14%,P = 0.0021).This association was not present in thosepatients with well-differentiated gastric cancer(28.57% vs 43.33%,P = 0.2832).Within our patient cohort,26 cases were lost to follow-up.The median survival time for the remaining 171 patients was 18 mo.The median survival times of the HER2 positive and negative groups were 17 and 18.5 mo respectively.Overall survival was not significantly different between HER2-positive and negative groups(χ 2 = 0.9157,P = 0.3386),but in patients presenting well-differentiated tumors,the overall survival of the HER2-positive group was significantly worse than that of the HER2-negative group(P = 0.0123).In contrast,patients with poorly differentiated and diffuse/mixed subtype gastric cancers showed no significant differences in overall survival associated with HER2.Furthermore,the median survival time of the HER2 positive group did not show any statistically significant differences when compared to the subgroups of gender,age,tumor location,TNM classification,lymph node metastases and distant metastasis.CONCLUSION:Patients with intestinal type gastric cancer(GC),well-differentiated GC and poorly-differentiated GC without lymph node metastasis,may all represent suitable candidates for targeted therapy using Herceptin. | Chao He Xue-Yi Bian Xing-Zhi Ni Dan-Ping Shen Yan-Ying Shen Hua Liu Zhi-Yong Shen Qiang Liu | 2013 | World Journal of Gastroenterology2013,19,14: | 28 |
| 3 | The second natural gas hydrate production test in the South China Sea显示文摘Clayey silt reservoirs bearing natural gas hydrates(NGH)are considered to be the hydrate-bearing reservoirs that boast the highest reserves but tend to be the most difficult to exploit.They are proved to be exploitable by the first NGH production test conducted in the South China Sea in 2017.Based on the understanding of the first production test,the China Geological Survey determined the optimal target NGH reservoirs for production test and conducted a detailed assessment,numerical and experimental simulation,and onshore testing of the reservoirs.After that,it conducted the second offshore NGH production test in 1225 m deep Shenhu Area,South China Sea(also referred to as the second production test)from October 2019 to April 2020.During the second production test,a series of technical challenges of drilling horizontal wells in shallow soft strata in deep sea were met,including wellhead stability,directional drilling of a horizontal well,reservoir stimulation and sand control,and accurate depressurization.As a result,30 days of continuous gas production was achieved,with a cumulative gas production of 86.14×104 m3.Thus,the average daily gas production is 2.87×10^4 m^3,which is 5.57 times as much as that obtained in the first production test.Therefore,both the cumulative gas production and the daily gas production were highly improved compared to the first production test.As indicated by the monitoring results of the second production test,there was no anomaly in methane content in the seafloor,seawater,and atmosphere throughout the whole production test.This successful production test further indicates that safe and effective NGH exploitation is feasible in clayey silt NGH reservoirs.The industrialization of hydrates consists of five stages in general,namely theoretical research and simulation experiments,exploratory production test,experimental production test,productive production test,and commercial production.The second production test serves as an important step from the exploratory production test to experimental production test. | Jian-liang Ye Xu-wen Qin Wen-wei Xie Hai-long Lu Bao-jin Ma Hai-jun Qiu Jin-qiang Liang Jing-an Lu Zeng-gui Kuang Cheng Lu Qian-yong Liang Shi-peng Wei Yan-jiang Yu Chunsheng Liu Bin Li Kai-xiang Shen Hao-xian Shi Qiu-ping Lu Jing Li Bei-bei Kou Gang Song Bo Li He-en Zhang Hong-feng Lu Chao Ma Yi-fei Dong Hang Bian | 2020 | China Geology2020,3,2: | 22 |
| 4 | Pharmacokinetics of radioimmunotherapeutic agent of direct labeling mAb ^(188)Re-HAb18显示文摘AIM: To labed Anti-hepatoma monoclonal antibody (mAb)fragment HAb18 F (ab')2 was labeled with 188 Re for thepharmacokinetic model of 188 Re-HAb18 F (ab')2 and toevaluate its pharmacokinetic parameters in hepatoma-bearing nude mice.METHODS: HAb18 F(ab')2 was directly labeled with 188Reusing 2-mercaptoethanol (2-ME) as reducing agents.Labeling efficiency and immunoreactivity of 188 Re-HAb18 F( ab')2 were evaluated by Whatman 3MM paperchromatography and live cell assay, respectively.Biodiatribution analysis was also conducted in nude micebearing human hepatoma in which animals were sacrificed atdifferent time points(1, 4, 18, 24 and 24h) after 188Re-HAb18F(ab' )2 was injected through tail-vein into hepatoma-bearingnude mice. The blood and radioactivity of organs and masswere measured. The concentrations of 188 Re-HAb18 F(ab')2were evaluated with a pharmacokinetic 3P97 software.RESULTS: The optimum labeling efficiency andimmunoreactive fraction were 91.7% and 0.78%,respectively. The parameters of 188Re-HAb18 F(ab')2 were:T1/2, 2.29h; Vd, 1.49 × 10-9L@ Bq- 1; AUC, 20.49 × 109Bq@ h@L-1 ;CL, 0.45 × 10-3L@ h- 1. 188Re- HAb18 F(ab')2 could locatespecially in hepatoma with high selective reactivity of HAb18F(ab')2. 188 Re-HAb18 F(ab')2 was mainly eliminated bykidney. | 7 | Effects of planting methods on yield and quality of different types of japonica rice in northern Jiangsu plain, China显示文摘Mechanical transplanting with carpet seedlings(MC) and mechanical direct seeding(MD) are newly developed planting methods, which increase in popularity and planted area each year. Knowing the difference for yield and rice quality under different planting methods is of great importance for the development of high quality and yield cultivation techniques under mechanical conditions. Therefore, three kinds of japonica rice including hybrid japonica rice, inbreed japonica rice, and soft rice were adopted as materials. And the differences in the quality of processing, appearance, cooking and eating quality, nutrition, and the rapid viscosity analyzer(RVA) profile were studied to reveal the effects of planting methods on yield and quality of different types of japonica rice. Results showed that the milled rice and head rice rates under MC was significantly higher than those under MD, and the processing quality of inbreed japonica rice was the most stable. Compared with MC, length/width ratio of rice under MD was significantly increased, and chalkiness rate, size, and degree were significantly decreased. The protein content under MD was lower than that under MC. MC showed higher peak viscosity and breakdown value than MD. The taste value was the greatest for soft rice, followed by inbreed japonica rice, and then by japonica hybrid rice, with no significant differences resulting from planting methods. Compared with MC, MD significantly improved the appearance quality, though processing quality and nutritional quality were decreased. And there was no significant difference in cooking and eating quality between MC and MD. Under different planting methods, the appearance quality of inbreed japonica rice changed the most and the processing quality was the most stable. The nutritional, cooking and eating quality of soft rice changed the least. | 8 | Quantification of pancreatic exocrine function of chronic pancreatitis with secretin-enhanced MRCP显示文摘AIM:To obtain reference values for pancreatic flow output rate(PFR)and peak time(PT)in healthy volunteers and chronic pancreatitis(CP);to correlate quantification of secretin enhanced magnetic resonance cholangiopancreatography(SMRCP)of pancreatic fluid output following secretin with fecal elastase-1(FE-1)tests.METHODS:The present study includes 53 subjects comprised of 17 healthy individuals and 36 patients with CP from April 2011 to January 2013.The 36 patients with CP were divided into three groups of mild CP(n=14),moderate CP(n=19)and advanced CP(n=3)by M-ANNHEIM classification for CP..Fifty-three cases underwent FE-1 test and magnetic resonance imaging using 3.0 T-device(Signa EXCITE,GE Healthcare).Coronal T2-weighted single-shot turbo spin-echo,spiratory triggered,covering the papillae,duodenum and small bowel.MRCP was performed with a heavily T2-weighted fat-suppressed long TE HASTE sequence (thick slab 2D MRCP sequence),repeated every 2 min up to 11 min after 0.1 mL/kg secretin injection(Secrelux,Sanochemia,Germany).FE-1 test used sandwich enzyme-linked immunosorbent assay(ELISA)test(ScheBo.Tech,Germany).RESULTS:A good linear correlation showed between the calculated volume and the actual volume by Phantom experiments.Fifty-three paired Quantification of secretin enhanced magnetic resonance cholangiopancreatography(MRCPQ)and FE-1 data sets were analyzed.The mean FE-1 of 53 cases was 525.41±94.44μg/g for 17 healthy volunteers,464.95±136.13μg/g for mild CP,301.55±181.55μg/g for moderate CP,229.30±146.60μg/g for advanced CP.Also,there was statistically significant difference in FE-1(P=0.0001)between health and CP.The mean values of PFR and PT were 8.18±1.11 mL/min,5.76±1.71 min for normal;7.27±2.04 mL/min,7.71±2.55 min for mild CP;4.98±2.57 mL/min,9.10±3.00 min for moderate CP;4.13±1.83 mL/min,12.33±1.55 min for advanced CP.Further,statistically significant difference in PFR(P=0.0001)and PT(P=0.0001)was observed between health and CP.Besides,there was correlation(r=0.79)and consistency(K=0.6)between MRCPQ and ELISA Test.It was related between M-ANNHEIM classification and PFR(r=0.55),FE-1(r=0.57).CONCLUSION:SMRCP can provide a safe,non-invasive and efficient method to evaluate the exocrine function of the pancreas. | Yun Bian Li Wang Chao Chen Jian-Ping Lu Jia-Bao Fan Shi-Yue Chen Bing-Hui Zhao | 2013 | World Journal of Gastroenterology2013,19,41: | 11 |

| 10 | A 2.5D finite element approach for predicting ground vibrations generated by vertical track irregularities显示文摘Dynamic responses of track structure and wave propagation in nearby ground vibration become significant when train operates on high speeds.A train-track-ground dynamic interaction analysis model based on the 2.5D finite element method is developed for the prediction of ground vibrations due to vertical track irregularities.The one-quarter car model is used to represent the train as lumped masses connected by springs.The embankment and the underlying ground are modeled by the 2.5D finite element approach to improve the computation efficiency.The Fourier transform is applied in the direction of train's movement to express the wave motion with a wave-number.The one-quarter car model is coupled into the global stiffness matrix describing the track-ground dynamic system with the displacement compatibility condition at the wheel-rail interface,including the irregularities on the track surface.Dynamic responses of the track and ground due to train's moving loads are obtained in the wave-number domain by solving the governing equation,using a conventional finite element procedure.The amplitude and wavelength are identified as two major parameters describing track irregularities.The irregularity amplitude has a direct impact on the vertical response for low-speed trains,both for short wavelength and long wavelength irregularities.Track irregularity with shorter wavelength can generate stronger track vibration both for low-speed and high-speed cases.For low-speed case,vibrations induced by track irregularities dominate far field responses.For high-speed case,the wavelength of track irregularities has very little effect on ground vibration at distances far from track center,and train's wheel axle weights becomes dominant. | Xue-cheng BIAN Chang CHAO Wan-feng JIN Yun-min CHEN | 2011 | Journal of Zhejiang University-Science A(Applied Physics & Engineering)2011,12,12: | 10 |
| 11 | Economic optimization for configuration and sizing of micro integrated energy systems显示文摘Based on analysis of construction and operation of micro integrated energy systems(MIES), this paper presents economic optimization for their configuration and sizing. After presenting typical models for MIES, a residential community MIES is developed by analyzing residential direct energy consumption within a general design procedure. Integrating with available current technologies and local resources, the systematic design considers a prime mover, fed by natural gas, with wind power, photovoltaic generation, and two storage devices serving thermal energy and power to satisfy cooling, heating and electricity demands. Control strategies for MIES also arepresented in this study. Multi-objective formulas are obtained by analyzing annual cost and dumped renewable energy to achieve optimal coordination of energy supply and demand. According to historical load data and the probability distribution of distributed generation output,clustering methods based on K-means and discretization methods are employed to obtain typical scenarios representative of uncertainties. The modified non-dominated sorting genetic algorithm is applied to find the Pareto frontier of the constructed multi-objective formulas. In addition, aiming to explore the Pareto frontier, the dumped energy cost ratio is defined to check the energy balance in different MIES designs and provide decision support for the investors. | 13 | Comparative analysis on grain quality and yield of different panicle weight indica-japonica hybrid rice(Oryza sativa L.) cultivars显示文摘Indica-japonica hybrid rice(Oryza sativa L.) cultivars showed high yield potential and poor tasting quality when compared with common japonica rice cultivars. Large panicle is a prominent factor of high yield for indica-japonica hybrid rice cultivars, and the panicle weight varies greatly among different indica-japonica hybrid rice cultivars. It is important to research on yield and grain quality of different panicle weight indica-japonica hybrid rice cultivars. In this study, two different panicle types indica-japonica hybrid cultivars were used to research on the relation of yield and grain quality. The yields of two heavy panicle weights indica-japonica hybrid cultivars were significantly higher than that of two medium panicle weight rice cultivars. The cooking and eating quality and starch properties of different panicle type cultivars were evaluated. Yongyou 6715(medium panicle) and Yongyou 1852(heavy panicle) got the relatively higher cooking and eating quality. Rice cultivars with medium panicle weight had more large starch granules and higher relative crystallinity than cultivars with heavy panicle weight. Transition temperature and retrogradation enthalpy(ΔHret) of medium panicle type cultivars were significantly higher than that of heavy panicle type cultivars. There was no significant difference in amylose content among different panicle type cultivars. Protein content of heavy panicle type cultivar was higher than that of medium panicle type cultivar, and protein content is the main factor affect cooking and eating quality in this study. The cultivar Yongyou 6715 got the highest taste value with the lowest protein content. Thus, it is suggested that the emphasis on improving rice cooking and eating quality of indica-japonica hybrid rice cultivars is how to reduce the protein content in rice grain. | 14 | Three-dimensional dynamic contrast-enhanced MR angiography for evaluating recipient vessels in orthotopic liver transplantation显示文摘BACKGROUND:With the improvement of MR technology, three-dimensional dynamic contrast-enhanced MR angiography(3D-DCE-MRA)may be the optimal vascular imaging method for preoperative evaluation of liver transplantation candidates.This study was undertaken to determine the value of 3D-DCE-MRA in the assessment of recipient vessels in orthotopic liver transplantation(OLT). METHODS:The surgical and pathological records were taken as the'gold standards'.Eighteen cases of OLT were retrospectively analyzed to assess the image quality of MRA,including the signal-to-noise ratio(SNR)in arteries and veins,depiction of vascular variation and vessel disease, and the accuracy of vascular diameter measurement. RESULTS:3D-DCE-MRA of 34 cases was carried out before OLT.The rates(excellent and good)showing hepatic arteries and portal vein for 3D-DCE-MRA were 94.1%(32/34)and 88.2%(30/34),respectively.The SNRs of the celiac axis and portal vein measurements from 3D-DCE-MRA were 20.58±3.74 and 13.43±4.12, and the mean diameters were 3.4±0.3 mm and 13.1±3.2 mm,respectively.There were 5 cases of vessel variation according to the Michel's classification.Of the 34 patients, 18 were compared radiologically and pathologically.The accuracy of depiction of the hepatic artery and portal vein with 3D-DCE-MRA was 100%for both;3D-DCE-MRA precisely assessed 4 cases of more than moderate stenosis in hepatic arteries,2 cases of small-caliber hepatic artery 3 cases of venous stenosis at the second porta hepatis, 6 cases of collateral vasculature,1 case of portal vein thrombosis and 1 case of portal vein aneurysm;all were confirmed pathologically.CONCLUSION:3D-DCE-MRA may be the first choice for recipient vascular assessment before OLT. | Bian, Jie Sha, Lin Yang, Chao Sun, Chuan-Shu | 2008 | Hepatobiliary & Pancreatic Diseases International2008,7,5: | 8 |
| 15 | Urinary metabolomics analysis identifies key biomarkers of different stages of nonalcoholic fatty liver disease显示文摘AIM To identify a panel of biomarkers that can distinguish between non-alcoholic fatty liver disease(NAFLD) and non-alcoholic steatohepatitis(NASH), and explore molecular mechanism involved in the process of developing NASH from NAFLD.METHODS Biomarkers may differ during stages of NAFLD. Urine and blood were obtained from non-diabetic subjects with NAFLD and steatosis, with normal liver function(n = 33), from patients with NASH, with abnormal liver function(n = 45), and from healthy age and sex-matched controls(n = 30). Samples were subjected to metabolomic analysis to identify potential non-invasive biomarkers. Differences in urinary metabolic profiles were analyzed using liquid chromatography tandem mass spectrometry with principal component analysis and partial least squares-discriminate analysis.RESULTS Compared with NAFLD patients, patients with NASH had abnormal liver function and high serum lipid concentrations. Urinary metabonomics found differences in 31 metabolites between these two groups, including differences in nucleic acids and amino acids. Pathway analysis based on overlapping metabolites showed that pathways of energy and amino acid metabolism, as well as the pentose phosphate pathway, were closely associated with pathological processes in NAFLD and NASH.CONCLUSION These findings suggested that a panel of biomarkers could distinguish between NAFLD and NASH, and could help to determine the molecular mechanism involved in the process of developing NASH from NAFLD. Firstly,the dynamic equations of a flexible manipulator with a flexible joint and a flexible link are derived. Secondly,the relationship and property between the flexible joint and the flexible link are analyzed. It shows that the flexible joint's amplitude will increase abruptly,thereby the dynamic singularity occurs if the frequency of a flexible joint is near or equal to some natural frequency of a flexible link. Finally,some numerical simulations which will verify the correctness of the theoretical analysis,are carried out. In modern clinics, YCHD is mostly used for hepatic diseases. This review summarizes the biological activities of YCHD and its medical applications. The main active compounds of YCHD are chlorogenic acid, rhein, geniposide, emodin, and scoparone. The pharmacological actions of YCHD include inhibition of hepatic steatosis, apoptosis, necrosis, anti-inflammation, and immune regulation. | 19 | Involvement of NF-κB and the CX3CR1 Signaling Network in Mechanical Allodynia Induced by Tetanic Sciatic Stimulation显示文摘Tetanic stimulation of the sciatic nerve(TSS)triggers long-term potentiation in the dorsal horn of the spinal cord and long-lasting pain hypersensitivity. CX3CL1-CX3CR1 signaling is an important pathway in neuronalmicroglial activation. Nuclear factor κB(NF-κB) is a key signal transduction molecule that regulates neuroinflammation and neuropathic pain. Here, we set out to determine whether and how NF-κB and CX3CR1 are involved in the mechanism underlying the pathological changes induced by TSS. After unilateral TSS, significant bilateral mechanical allodynia was induced, as assessed by the von Frey test. The expression of phosphorylated NF-κB(pNF-κB) and CX3CR1 was significantly up-regulated in the bilateral dorsal horn. Immunofluorescence staining demonstrated that pNF-KB and NeuN co-existed, implying that the NF-κB pathway is predominantly activated in neurons following TSS. Administration of either the NF-κB inhibitor ammonium pyrrolidine dithiocarbamate or a CX3CR1-neutralizing antibody blocked the development and maintenance of neuropathic pain. In addition, blockade of NF-κB downregulated the expression of CX3 CL1-CX3CR1 signaling,and conversely the CX3CR1-neutralizing antibody also down-regulated pNF-κB. These findings suggest an involvement of NF-κB and the CX3CR1 signaling network in the development and maintenance of TSS-induced mechanical allodynia. The first step of analyzing user behaviors is to extract information of user actions from HTTP traffic data by multi-pattern URL matching. However, the efficiency is a huge problem when performing this work on massive network traffic data. To solve this problem, we propose a novel and accurate algorithm named Multi-Pattern Parallel Matching(MPPM) that takes advantage of HashMap in data searching for extracting user behaviors from big network data more effectively. Extensive experiments based on real-world traffic data prove the ability of MPPM algorithm to deal with massive HTTP traffic with better performance on accuracy, concurrency and efficiency.
