| 3 | SPECT/CT同机融合显像和CT在估测口腔恶性肿瘤侵犯下颌骨范围的应用价值(英文)显示文摘Objective: The aim of our study was to compare the value of computed tomography (CT) and 99mTc-methylene-diphosphonate (MDP) SPECT (single photon emission computed tomography)/CT fusion imaging in determining the extent of mandibular invasion by malignant tumor of the oral cavity. Methods: This study had local ethical committee approval, and all patients gave written informed consent. Fifty-three patients were revealed mandibular invasion by malignant tumor of the oral cavity underwent CT and SPECT/CT. The patients were divided into two groups: group A (invasion-periphery-type) and group B (invasion-center-type). Two radiologists assessed the CT images and two nuclear medicine physicians separately assessed the SPECT/CT images in consensus and without knowledge of the results of other imaging tests. The extent of bone involvement suggested with an imaging modality was compared with pathological findings in the surgical specimen. Results: With pathological findings as the standard of reference, Group A: The extent of mandibular invasion by malignant tumor under-went SPECT/CT was 1.02 ± 0.20 cm larger than that underwent pathological findings. And the extent of mandibular invasion underwent CT was 1.42 ± 0.35 cm smaller than that underwent pathological examination. There were significant difference among the three methods (P < 0.01). Group B: The extent of mandibular invasion by malignant tumor underwent SPECT/CT was 1.3 ± 0.39 cm larger than that underwent pathological examination. The extent of mandibular invasion underwent CT was 2.55 ± 1.44 cm smaller than that underwent pathological findings. There were significant difference among the three methods (P < 0.01). The extent of mandibular invasion underwent SPECT/CT was the extent which surgeon must excise to get clear margins. | 5 | Antibiotic-induced gut bacteria depletion has no effect on HBV replication in HBV immune tolerance mouse model显示文摘Commensal microbiota is closely related to Hepatitis B virus(HBV)infection.Gut bacteria maturation accelerates HBV immune clearance in hydrodynamic injection(HDI)HBV mouse model.However,the effect of gut bacteria on HBV replication in recombinant adeno-associated virus(AAV)-HBV mouse model with immune tolerance remains obscure.We aim to investigate its role on HBV replication in AAV-HBV mouse model.C57BL/6 mice were administrated with broad-spectrum antibiotic mixtures(ABX)to deplete gut bacteria and intravenously injected with AAV-HBV to establish persistent HBV replication.Gut microbiota community was analyzed by fecal qPCR assay and 16S ribosomal RNA(rRNA)gene sequencing.HBV replication markers in blood and liver were determined by ELISA,qPCR assay and Western blot at indicated time points.Immune response in AAV-HBV mouse model was activated through HDI of HBV plasmid or poly(I:C)and then detected by quantifying the percentage of IFN-γ^(+)/CD8^(+)T cells in the spleen via flow cytometry as well as the splenic IFN-γmRNA level via qPCR assay.We found that antibiotic exposure remarkably decreased gut bacteria abundance and diversity.Antibiotic treatment failed to alter the levels of serological HBV antigens,intrahepatic HBV RNA transcripts and HBc protein in AAV-HBV mouse model,but contributed to HBsAg increase after breaking of immune tolerance.Overall,our data uncovered that antibiotic-induced gut bacteria depletion has no effect on HBV replication in immune tolerant AAV-HBV mouse model,providing new thoughts for elucidating the correlation between gut bacteria dysbiosis by antibiotic abuse and clinical chronic HBV infection. | Yanan Bu Kaitao Zhao Zaichao Xu Yingcheng Zheng Rong Hua Chuanjian Wu Chengliang Zhu Yuchen Xia Xiaoming Cheng | 2023 | Virologica Sinica2023,38,3: | 0 |
| 6 | A recent retrotransposon insertion of J caused E6 locus facilitating soybean adaptation into low latitude显示文摘Soybean(Glycine max) is an important legume crop that was domesticated in temperate regions.Soybean varieties from these regions generally mature early and exhibit extremely low yield when grown under inductive short-day(SD) conditions at low latitudes. The long-juvenile(LJ) trait, which is characterized by delayed flowering and maturity,and improved yield under SD conditions, allowed the cultivation of soybean to expand to lower latitudes. Two major loci control the LJ trait: J and E6. In the current study, positional cloning, sequence analysis, and transgenic complementation confirmed that E6 is a novel allele of J, the ortholog of Arabidopsis thaliana EARLY FLOWERING 3(ELF3). The mutant allele e6^(PG), which carries a Ty1/Copia-like retrotransposon insertion, does not suppress the legume-specific flowering repressor E1, allowing E1 to inhibit Flowering Locus T(FT) expression and thus delaying flowering and increasing yields under SD conditions. The e6^(PG)allele is a rare allele that has not been incorporated into modern breeding programs.The dysfunction of J might have greatly facilitated the adaptation of soybean to low latitudes.
