1Combination of anti-PD-1 antibody with P-GEMOX as a potentially effective immunochemotherapy for advanced natural killer/T cell lymphoma显示文摘Advanced natural killer/T cell lymphoma(NKTL)has demonstrated poor prognosis with currently available therapies.Here,we report the efficacy of anti-programmed death 1(PD-1)antibody with the P-GEMOX(pegaspargase,gemcitabine,and oxaliplatin)regimen in advanced NKTL.Nine patients underwent six 21-day cycles of anti-PD-1 antibody(day 1),pegaspargase 2000 U/m^(2)(day 1),gemcitabine 1 g/m^(2)(days 1 and 8)and oxaliplatin 130 mg/m^(2)(day 1),followed by anti-PD-1 antibody maintenance every 3 weeks.Programmed death-ligand 1(PD-L1)expression and genetic alterations were determined in paraffin-embedded pretreatment tissue samples using immunohistochemistry and next-generation sequencing(NGS)analysis.Responses were assessed using 18F-fluorodeoxyglucose positron emission tomography(18FDG-PET)and computed tomography or magnetic resonance imaging.Eight patients exhibited significant responses,comprising of seven complete remissions and one partial remission(overall response rate:88.9%).After a median follow-up of 10.6 months,6/9 patients(66.7%)remained in complete remission.The most common grade 3/4 adverse events were anemia(33.3%),neutropenia(33.3%),and thrombocytopenia(33.3%);all of which were manageable and resolved.Immunochemotherapy produced a high response rate in patients with positive PD-L1 expression(5/6,83.3%).NGS analysis suggested that STAT3/JAK3/PD-L1 alterations and ARID1A mutation were associated with immunochemotherapy efficacy.Mutation in DDX3X and alteration in epigenetic modifiers of KMT2D,TET2,and BCORL1 might indicate a poor response to immunochemotherapy.In conclusion,the anti-PD-1 antibody plus P-GEMOX regimen demonstrated promising efficacy in advanced NKTL.PD-L1 expression combined with specific genetic alterations could be used as potential biomarkers to predict therapeutic responses to immunochemotherapy.Jun Cai Panpan Liu Huiqiang Huang Yajun Li Shuyun Ma Hui Zhou Xiaopeng Tian Yuchen Zhang Yan Gao Yi Xia Xuanye Zhang Hang Yang Lirong Li Qingqing Cai 2020Signal Transduction and Targeted Therapy2020,5,1:6
2Insights into Hepatitis B Virus DNA Integration-55 Years after Virus Discovery显示文摘Hepatitis B virus(HBV),which was discovered in 1965,is a threat to global public health.HBV infects human hepatocytes and leads to acute and chronic liver diseases,and there is no cure.In cells infected by HBV,viral DNA can be integrated into the cellular genome.HBV DNA integration is a complicated process during the HBV life cycle.Although HBV integration normally results in replication-incompetent transcripts,it can still act as a template for viral protein expression.Of note,it is a primary driver of hepatocellular carcinoma(HCC).Recently,with the development of detection methods and research models,the molecular biology and the pathogenicity of HBV DNA integration have been better revealed.Here,we review the advances in the research of HBV DNA integration,including molecular mechanisms,detection methods,research models,the effects on host and viral gene expression,the role of HBV integrations in the pathogenesis of HCC,and potential treatment strategies.Finally,we discuss possible future research prospects of HBV DNA integration.Kaitao Zhao Andrew Liu Yuchen Xia 2020The Innovation2020,1,2:3

5Kidney injury molecule-1 is a potential receptor for SARS-CoV-2显示文摘COVID-19 patients present high incidence of kidney abnormalities,which are associated with poor prognosis and mortality.The identification of SARS-CoV-2 in the kidney of COVID-19 patients suggests renal tropism of SARS-CoV-2.However,whether there is a specific target of SARS-CoV-2 in the kidney remains unclear.Herein,by using in silico simulation,coimmunoprecipitation,fluorescence resonance energy transfer,fluorescein isothiocyanate labeling,and rational design of antagonist peptides,we demonstrate that kidney injury molecule-1(KIM1),a molecule dramatically upregulated upon kidney injury,binds with the receptor-binding domain(RBD)of SARS-CoV-2 and facilitates its attachment to cell membrane,with the immunoglobulin variable Ig-like(Ig V)domain of KIM1 playing a key role in this recognition.The interaction between SARS-CoV-2 RBD and KIM1 is potently blockaded by a rationally designed KIM1-derived polypeptide AP2.In addition,our results also suggest interactions between KIM1 Ig V domain and the RBDs of SARS-CoV and MERS-CoV,pathogens of two severe infectious respiratory diseases.Together,these findings suggest KIM1 as a novel receptor for SARS-CoV-2 and other coronaviruses.We propose that KIM1 may thus mediate and exacerbate the renal infection of SARS-CoV-2 in a‘vicious cycle’,and KIM1 could be further explored as a therapeutic target.Chen Yang Yu Zhang Xia Zeng Huijing Chen Dong Yang Yuchen Chen Ziwei Shen Xiaomu Wang Xinran Liu Mingrui Xiong Hong Chen Kun Huang 2021Journal of Molecular Cell Biology2021,13,3:2
6SRO1 regulates heavy metal mercury stress response in Arabidopsis thaliana显示文摘Heavy metals in the environment are harmful limiting factors for the normal growth and development of plants.Here,we isolated and identified an Arabidopsis thaliana T-DNA insertion mutant,named sro1-1,which showed a hyper-sensitive response to HgCl2.The SRO1protein contains a WWE domain that mediates proteinprotein interactions.Under HgCl2treatment,when compared with the wild-type plants,the growth of sro1-1 was repressed dramatically and the number of true leaves was reduced and etiolated.The electrolyte leakage rates showed that cell membrane integrity in sro1-1 was damaged more severely than in the wild type.DAB(3,5-diaminobenzidine)staining and confocal microscopy showed that Hg2?stress induced more hydrogen peroxide accumulation in sro1-1 than in the wild type.The qRT-PCR results indicated that the expression of some abiotic stress-induced genes,such as L-ascorbate peroxidase(APX1),was reduced under oxidative or Hg2?stress.Transgenic plants containing a GFP::SRO1 fusion protein showed that SRO1was localized in the nucleus of the cells.SRO1 was shown to be expressed in various tissues,and was most highly expressed in the vigorous tissues.Our results suggest thatSRO1 may play an important role in the stress response of A.thaliana to heavy metals.Xiaoli Zhang Xiaoliang Zhao Baozhu Li Jinchan Xia Yuchen Miao 2014Chinese Science Bulletin2014,59,25:2

8Three-stage Nonlinear Thermal Model for Microspeakers in Mobile Phones显示文摘Since microspeakers are widely used in mobile phones,high power is usually applied to obtain sufficient output sound pressure.However,the electric power is almost converted into heat,leading to the thermal problem in microspeakers.Compared with large loudspeaker,microspeakers are smaller and the under yoke is relatively closed,causing the heat transfer more complex.In this study,a three-stage nonlinear thermal model was proposed for analyzing the thermal behavior in microspeakers.The inside air is a buffer area between the voice coil and magnet,and modeled as a middle stage of the heat transfer.The forced convection is still significant in microspeakers while the eddy current can be ignored.In order to obtain the thermal parameters of the model,a corresponding parameter identification method was put forward.The basic linear parameters and forced convection parameters were all obtained by measuring and fitting the temperature curves of voice coil at different single tones.A series of experiments were conducted to verify the proposed model and parameter identification method,and the results showed good aggrement between the measured and predicted temperature curves for different input signals.The proposed model was valid and accurate,and may be helpful for the design and application of microspeakers.Zhiliang Zhang Yong Shen Jie Xia Yuchen Shen 2018Sound & Vibration2018,52,5:1

13Antibiotic-induced gut bacteria depletion has no effect on HBV replication in HBV immune tolerance mouse model显示文摘Commensal microbiota is closely related to Hepatitis B virus(HBV)infection.Gut bacteria maturation accelerates HBV immune clearance in hydrodynamic injection(HDI)HBV mouse model.However,the effect of gut bacteria on HBV replication in recombinant adeno-associated virus(AAV)-HBV mouse model with immune tolerance remains obscure.We aim to investigate its role on HBV replication in AAV-HBV mouse model.C57BL/6 mice were administrated with broad-spectrum antibiotic mixtures(ABX)to deplete gut bacteria and intravenously injected with AAV-HBV to establish persistent HBV replication.Gut microbiota community was analyzed by fecal qPCR assay and 16S ribosomal RNA(rRNA)gene sequencing.HBV replication markers in blood and liver were determined by ELISA,qPCR assay and Western blot at indicated time points.Immune response in AAV-HBV mouse model was activated through HDI of HBV plasmid or poly(I:C)and then detected by quantifying the percentage of IFN-γ^(+)/CD8^(+)T cells in the spleen via flow cytometry as well as the splenic IFN-γmRNA level via qPCR assay.We found that antibiotic exposure remarkably decreased gut bacteria abundance and diversity.Antibiotic treatment failed to alter the levels of serological HBV antigens,intrahepatic HBV RNA transcripts and HBc protein in AAV-HBV mouse model,but contributed to HBsAg increase after breaking of immune tolerance.Overall,our data uncovered that antibiotic-induced gut bacteria depletion has no effect on HBV replication in immune tolerant AAV-HBV mouse model,providing new thoughts for elucidating the correlation between gut bacteria dysbiosis by antibiotic abuse and clinical chronic HBV infection.Yanan Bu Kaitao Zhao Zaichao Xu Yingcheng Zheng Rong Hua Chuanjian Wu Chengliang Zhu Yuchen Xia Xiaoming Cheng 2023Virologica Sinica2023,38,3:0
14Clinical outcomes of sildenafil application in patients of poor endometrial development显示文摘Objective:To determine whether sildenafil has an efect on pregnancy outcomes in patients with poor endometrial development.Mathods:This study included 472 infertility patients who underwent in viro fer tilization/intracytoplasmadc spemm injection and frozen-thawed embryo tr ansfer(IVF/ICSI-FET)and subsequently sufered from poor endo-metral dewelopment during homnone replacement cycle(HRC)from April 2017 to July 2019.The patients were divided into two groups:the sildenafl group(n=88)and the control group(n=384).We analyzed endometrial thicknuess and types on endometrial transformation day,as well as pregnancy outcomes after FET(biochemical pregnancy,clinical pregnancy,early abortion,late abortion,and live birth rates)between the two groups.Results:After adjusting for confounding factors,we found no significant differences in endometrial thicknesses and types on endometrial transfomation day between the sildenafil group and the control group(0.79±0.08 vs 0.81±0.09,P=0.144;79.76%vs83.87%,P=0.402).There were also no statistcally significant differences in biochemical pregnancy rate(75.0%vs76.8%,P=0.892),clinical pregnancy rate(59.09%vs 69.53%,P=0.087),early abortion rate(17.31%vs 14.61%,P=0.557),late abortion rate(3.85%vs 4.49%,P=0.859),or live birth rate(45.45%vs 55.47%,P=0.101)between the two groups.In subgroup analysis,the application of sildenafil was unable to improve endometrial thickness(group one:0.80±0.08 cm vs 0.82±0.08 cm;group two:0.78±0.08 cm vs 0.80±0.10 cm;group three:0.75±0.11 cm vs 0.77±0.08 cm,p>0.05)and type endometrium on transfomation day(group one:78.57%vs 86.78%;group two:80.65%vs 77.78%;group three:81.82%ws.83.78%,p>0.05).Moreover,sildenafil use was not closely associated with clinical pregnancy outcomes,clinical pregnancy rate,early abortion rate,late abortion rate,and live birth rate(p>0.05).Conclusions:Sildenafl did not benefit endometrial development and pregnancy outcomes in patients with poor endometr:ial development during the hormone replacement cycle.Meiling Guo Yuchen Yan Jianan Lv Hua Xin Wei Zhou Na Yu Mingdi Xia Jing Li Qian Zhang Junhao Yan 2022Gynecology and Obstetrics Clinical Medicine2022,2,1:0

16Electronic and Optical Properties of B- and/or In-Doped GaAs Calculated Using Many-body Green's Function Theory显示文摘Using ab initio many-body Green's Function theory, the electronic and optical properties of pure BAs,InAs as well as B- and/or In-doped GaAs supercells were studied. The results show that the calculated quasiparticleband gaps of BAs and InAs are in agreement with the experiments. The electronic and optical absorption properties of B-and/or In-doped GaAs are very sensitive to the lattice constant models used in the calculations, which can explainthe controversies in previous theoretical and experimental works. Under the lattice constant condition based on theVegard's Law, the reduced band gap and the redshift of the first excited state E0 can be observed for InyGa1-y As withIn concentrations(atomic fractions) ranging from 0 to 15.625%, while the band gap increase and the blueshift of stateE0 can be realized for BxGal-xAs. However, an opposite trend can be found when the lattice constant is fixed to theexperimental value, i.e., In doping leads to a band gap increase and a blueshift of the E0 state, while B doping resultsin a band gap decrease and a redshift of the E0 state. For BxGal-x-y InyAs, the values of the band gap variation and theshift of state E0 are well located between the B and In mono-doping cases. 18Wideband polarization-independent anomalous reflection metasurface with multiple resonance modes显示文摘An ultra-thin metasurface is proposed to realize wideband polarization-independent anomalous reflection. 19Recent advances in bacterial therapeutics based on sense and response显示文摘Intelligent drug delivery is a promising strategy for cancer therapies.In recent years,with the rapid development of synthetic biology,some properties of bacteria,such as gene operability,excellent tumor colonization ability,and host-independent structure,make them ideal intelligent drug carriers and have attracted extensive attention.By implanting condition-responsive elements or gene circuits into bacteria,they can synthesize or release drugs by sensing stimuli.Therefore,compared with traditional drug delivery,the usage of bacteria for drug loading has better targeting ability and controllability,and can cope with the complex delivery environment of the body to achieve the intelligent delivery of drugs.This review mainly introduces the development of bacterial-based drug delivery carriers,including mechanisms of bacterial targeting to tumor colonization,gene deletions or mutations,environment-responsive elements,and gene circuits.Meanwhile,we summarize the challenges and prospects faced by bacteria in clinical research,and hope to provide ideas for clinical translation.Zhuo Feng Yuchen Wang Haiheng Xu Yunfei Guo Wen Xia Chenxuan Zhao Xiaozhi Zhao Jinhui Wu 2023Acta Pharmaceutica Sinica B2023,13,3:0
