1Structural effects of three-dimensional angle-interlock woven composite undergoing bending cyclic loading显示文摘This paper reports the structural effects of three-dimensional(3-D)angle-interlock woven composite(3DAWC)undergoing three-point bending cyclic loading from experimental and finite element analysis(FEA)approaches.In experiment,the fatigue tests were conducted to measure the bending deflection and to observe the damage morphologies.By the FEA approach,a micro-structural unit-cell model of the 3DAWC was established at the yarn level to simulate the fatigue damage.The stress degradation at the loading condition of constant deformation amplitude was calculated to show the degradation of mechanical properties.In addition,the stress distribution,fatigue damage evolution and critical damage regions were also obtained to qualitatively reveal the structural effects and damage mechanisms of the 3DAWC subjected to three-point bending cyclic loading.JIN LiMin YAO Yao YU YiMin ROTICH Gideon SUN BaoZhong GU BoHong 2014Science China(Physics,Mechanics & Astronomy)2014,57,3:5
2Live cell imaging and proteomic profiling of endogenous NEAT1 lncRNA by CRISPR/ Cas9-mediated knock-in显示文摘In mammalian cells,long noncoding RNAs(lncRNAs)form complexes with proteins to execute various biological functions such as gene transcription,RNA processing and other signaling activities.However,methods to track endogenous lncRNA dynamics in live cells and screen for lncRNA interacting proteins are limited.Here,we report the development of CERTIS(CRISPR-mediated Endogenous lncRNA Tracking and Immunoprecipitation System)to visualize and isolate endogenous lncRNA,by precisely inserting a 24-repeat MS2 tag into the distal end of lncRNA locus through the CRISPR7Cas9 technology.In this study,we show that CERTIS effectively labeled the paraspeckle lncRNA NEAT1 without disturbing its physiological properties and could monitor the endogenous expression variation of NEAT1.In addition,CERTIS displayed superior performance on both short-and long-term tracking of NEAT1 dynamics in live cells.We found that NEAT1 and paraspeckles were sensitive to topoisomerase I specific inhibitors.Moreover,RNA Immunoprecipitation(RIP)of the MS2-tagged NEAT1 lncRNA successfully revealed several new protein components of paraspeckle.Our results support CERTIS as a tool suitable to track both spatial and temporal lncRNA regulation in live cells as well as study the lncRNA-protein interactomes.Bohong Chen Shengcheng Deng Tianyu Ge Miaoman Ye Jianping Yu Song Lin Wenbin Ma Zhou Songyang 2020Protein & Cell2020,11,9:4
3Hybrid superhydrophilic-superhydrophobic micro/nanostructures fabricated by femtosecond laserinduced forward transfer for sub-femtomolar Raman detection显示文摘Raman spectroscopy plays a crucial role in biochemical analysis.Recently,superhydrophobic surface-enhanced Raman scattering(SERS)substrates have enhanced detection limits by concentrating target molecules into small areas.However,due to the wet transition phenomenon,further reduction of the droplet contact area is prevented,and the detection limit is restricted.This paper proposes a simple method involving femtosecond laser-induced forward transfer for preparing a hybrid superhydrophilic–superhydrophobic SERS(HS-SERS)substrate by introducing a superhydrophilic pattern to promote the target molecules to concentrate on it for ultratrace detection.Furthermore,the HS-SERS substrate is heated to promote a smaller concentrated area.The water vapor film formed by the contact of the solution with the substrate overcomes droplet collapse,and the target molecules are completely concentrated into the superhydrophilic region without loss during evaporation.Finally,the concentrated region is successfully reduced,and the detection limit is enhanced.The HS-SERS substrate achieved a final contact area of 0.013mm2,a 12.1-fold decrease from the unheated case.The reduction of the contact area led to a detection limit concentration as low as 10−16 M for a Rhodamine 6G solution.In addition,the HS-SERS substrate accurately controlled the size of the concentrated areas through the superhydrophilic pattern,which can be attributed to the favorable repeatability of the droplet concentration results.In addition,the preparation method is flexible and has the potential for fluid mixing,fluid transport,and biochemical sensors,etc.Xiaodan Ma Lan Jiang Xiaowei Li Bohong Li Ji Huang Jiaxing Sun Zhi Wang Zhijie Xu Liangti Qu Yongfeng Lu Tianhong Cui 2019Microsystems & Nanoengineering2019,5,1:3
4Comparative study on bearing mechanism and design parameters of confined concrete arch joints in deep soft rock roadway显示文摘Square confined concrete arch is increasingly used in deep soft rock roadway support because of its advantages of high strength and construction convenience.However,the design of confined concrete arch in underground engineering still remains in experience-based method and lacks quantitative analysis.As a connecting component between arch sections,the connection joints have an important influence on the internal force distribution and failure mechanism of support arch.Therefore,a reasonable design of arch joints is the premise of rational support design.Taking Liangjia Coal Mine,a typical deep soft rock mine in China,as research background,this paper fully compared the most widely used joint types of confined concrete arch as analytical objects:flange joints and casing joints.The main failure modes of these two kinds of joints under bending moment are defined.Laboratory and numerical tests are carried out to study the mechanical characteristics of joints.Based on the M-θ curve,the influence law of different design parameters is analyzed,and the design principles of joints are proposed.The research results could provide a theoretical basis for the design and application of confined concrete arch in related projects.Wei Lu Qi Wang Bei Jiang Shuo Xu Bohong Liu Peng Zhang Zhenhua Jiang 2019International Journal of Coal Science & Technology2019,6,4:3
5Water molecules on the liquid superlubricity interfaces achieved by phosphoric acid solution显示文摘The superlubricity mechanism of phosph olipid molecules on biological surfaces and interfaces has been receiving much attention.Since the superlubricity can be achieved with a phosphoric acid aqueous solution,the interaction of the phosphate group with the water molecule is considered to be the key to bio-lubrication.However,no direct evidence has been offered to distinctly interpret the underlying mech anism of superlubricity,due to the limitations in the method for the detection of interfacial molecules,especially water molecules on the interface.By using sum-freq uency g en eration(SFG)vibrational spectroscopy,which is extremely sensitive to interfacial molecules,the authors were able to explore the underlying microscopic mechanism for superlubricity in a phosphoric acid system,which might be related to interfacia I water molecules.By trapping the phosphoric acid aqueous solution between silicon nitride and silica surfaces,they discovered that phosphoric acid could exercise a great influence on the ordered structure of interfacial water molecules,contributing to SFG spectral changes in the broad O-H bond stretching region.This work directly revealed the interfacial interaction between water and phosphoric acid molecules in liquid superlubricity interfaces,which could shed new light on the microscopic mechanism of liquid superlubricity.Yuan Gao Liran Ma Yong Liang Bohong Li Jianbin Luo 2018Biosurface and Biotribology2018,4,3:3
6Salivary microbiota may predict the presence of esophageal squamous cell carcinoma显示文摘The aim is to explore the predictive value of salivary bacteria for the presence of esophageal squamous cell carcinoma (ESCC). Saliva samples were obtained from 178 patients with ESCC and 101 healthy controls, and allocated to screening and verification cohorts, respectively. In the screening phase, after saliva DNA was extracted, 16S rRNA V4 regions of salivary bacteria were amplified by polymerase chain reaction (PCR) with high-throughput sequencing. Highly expressed target bacteria were screened by Operational Taxonomic Units clustering, species annotation and microbial diversity assessment. In the verification phase, the expression levels of target bacteria identified in the screening phase were verified by absolute quantitative PCR (Q-PCR). Receiver operating characteristic (ROC) curves were plotted to investigate the predictive value of target salivary bacteria. LEfSe analysis revealed higher proportions of Fusobacterium, Streptococcus and Porphyromonas, and Q-PCR assay showed significantly higher numbers of Streptococcus salivarius, Fusobacterium nucleatum and Porphyromonas gingivalis in patients with ESCC, when compared with healthy controls (all P < 0.05). The areas under the ROC curves for Streptococcus salivarius, Fusobacterium nucleatum, Porphyromonas gingivalis and the combination of the three bacteria for predicting patients with ESCC were 69%, 56.5%, 61.8% and 76.4%, respectively. The sensitivities corresponding to cutoff value were 69.3%, 22.7%, 35.2% and 86.4%, respectively, and the matched specificity were 78.4%, 96.1%, 90.2% and 58.8%, respectively. 7Map-based cloning of a novel QTL qBN-1 influencing branch number in soybean[Glycine max(L.)Merr.]显示文摘Branch number(BN)is an important agronomic attribute related to the plant architecture,adaptability,and yield of soybean.To date,few studies ofBNhave been conducted to elucidate its genetic background.We aimed to localize genetic factors affecting BN using segregating populations derived fromthe high-branching cultivar‘Kennong24’(KN24)and the low-branching cultivar‘Kenfeng19’(KF19).Composite interval mapping analysis detected a QTL(qBN-1)on chromosome 6 between the SSR markers BARCSOYSSR_06_0993 and BARCSOYSSR_06_1070 using an F2 population.To fine-map qBN-1,a RIL population was developed and genotyped with 14 SSRmarkers located in the QTL region.qBN-1 was localized to a 115.67-kb interval flanked by markers BARCSOYSSR_06_1048 and BARCSOYSSR_06_1053.The QTL was further confirmed using backcross populations of size 1305(BC2F2 with KN24 as a recurrent parent)and 1712(BC3F2 with KF19 as a recurrent parent).The fine-mapping region of qBN-1 contained only two candidate genes,Glyma.06G208800 and Glyma.06G208900,whose expression patterns were investigated by qRT-PCR.Compared to Glyma.06G208800 gene expression,Glyma.06G208900 showed the highest expression of the two genes and showed a significant difference in expression between high-and low-branching genotypes in either axillary meristem or shoot apical meristem,and showed opposite expression patterns in the two tissues at V4 and R1 stages.These results identify Glyma.06G208900 as a novel candidate gene controlling BN.Taken together,the results of this study provide a foundation for cloning and functional analysis of the qBN-1 gene and for the improvement of BN bymarker-assisted selection in soybean breeding.Sobhi F.Lamlom Yong Zhang Bohong Su Haitao Wu Xia Zhang Jindong Fu Bo Zhang Li-Juan Qiu 2020The Crop Journal2020,8,5:2

17Analyzing Student Behavior in Online Programming Courses显示文摘Rather than maintaining the classic teaching approach, a growing number of schools use the blended learning system in higher education. The traditional method of teaching focuses on the result of students' progress. However, many student activities are recorded by an online programming learning platform at present. In this paper, we focus on student behavior when completing an online open-ended programming task. First, we conduct statistical analysis to examine student behavior on the basis of test times and completed time. By combining these two factors, we then classify student behavior into four types by using k-means algorithm. The results are useful for teachers to enhance their understanding of student learning and for students to know their learning style in depth.
