| 1 | Live cell imaging and proteomic profiling of endogenous NEAT1 lncRNA by CRISPR/ Cas9-mediated knock-in显示文摘In mammalian cells,long noncoding RNAs(lncRNAs)form complexes with proteins to execute various biological functions such as gene transcription,RNA processing and other signaling activities.However,methods to track endogenous lncRNA dynamics in live cells and screen for lncRNA interacting proteins are limited.Here,we report the development of CERTIS(CRISPR-mediated Endogenous lncRNA Tracking and Immunoprecipitation System)to visualize and isolate endogenous lncRNA,by precisely inserting a 24-repeat MS2 tag into the distal end of lncRNA locus through the CRISPR7Cas9 technology.In this study,we show that CERTIS effectively labeled the paraspeckle lncRNA NEAT1 without disturbing its physiological properties and could monitor the endogenous expression variation of NEAT1.In addition,CERTIS displayed superior performance on both short-and long-term tracking of NEAT1 dynamics in live cells.We found that NEAT1 and paraspeckles were sensitive to topoisomerase I specific inhibitors.Moreover,RNA Immunoprecipitation(RIP)of the MS2-tagged NEAT1 lncRNA successfully revealed several new protein components of paraspeckle.Our results support CERTIS as a tool suitable to track both spatial and temporal lncRNA regulation in live cells as well as study the lncRNA-protein interactomes. | Bohong Chen Shengcheng Deng Tianyu Ge Miaoman Ye Jianping Yu Song Lin Wenbin Ma Zhou Songyang | 2020 | Protein & Cell2020,11,9: | 4 |

| 3 | 3D hierarchical architecture collaborating with 2D/2D interface interaction in Ni Al-LDH/Ti_(3)C_(2) nanocomposite for efficient and selective photoconversion of CO_(2)显示文摘Photocatalytic conversion of CO_(2)into a special chemical fuel with high yield and selectivity is still a major challenge.Herein,a 3 D hierarchical Ni Al-LDH/Ti_(3)C_(2)MXene(LDH/TC)nanocomposite is constructed through in situ loading of Ti_(3)C_(2)nanosheets on the Ni Al-LDH scaffold during the hydrothermal process.The formation of a uniform and well-defined 2 D/2 D heterogeneous interface can be realized by optimizing the ratio of Ti_(3)C_(2)and the precursors for Ni Al-LDH.The 3 D hierarchical scaffold with high specific surface area contributes to the favourable photon adsorption and utilization.The intimate contact between Ti_(3)C_(2)and Ni Al-LDH with numerous interfaces effectively promotes the separation of the photoinduced electron-hole pairs in Ni Al-LDH.Together with the highly exposed oxidation-reduction active sites and the enhanced CO_(2)capture and activation.The maximum photocatalytic CO production rate on Ni AlLDH/Ti_(3)C_(2)reaches 11.82 lmol g^(-1)h^(-1)with 92%selectivity and superior stability.This work provides an effective approach for the development of an ideal photocatalyst by collaborative utilization of materials with different dimensionalities. | Qunrong Shi Xiaoyue Zhang Yong Yang Junjie Huang Xiaolong Fu Tianyu Wang Xiaodong Liu Aiwu Sun Jianhua Ge Jinyou Shen Yong Zhou Zuliang Liu | 2021 | Journal of Energy Chemistry2021,30,8: | 1 |
| 4 | Colloidal quantum-dot light emitting diodes with bias-tunable color显示文摘Although the performance of quantum-dot-based light emitting diodes(QLEDs)has been significantly enhanced over the past years,conventional full-color QLED displays still rely on the side-by-side pattern techniques of red(R)/green(G)/blue(B)quantum dots(QDs).Such lateral integration of multi-color pixels imposes technological difficulty in the development of high-resolution displays due to limited pixel density and fill factors.Herein,we demonstrate the development of full-color QLEDs with bias-tunable emission spectra by engineering mixed R/G/B QDs as light emitting layers.In Commission Internationale de l'Eclairage(CIE)chromaticity coordinates,QLEDs with bias-tunable color exhibit wide color variation ranging from red(0.649,0.330)to green(0.283,0.305)to blue(0.255,0.264)upon increasing voltages and can be tuned to emit white light(0.316,0.325).More importantly,the fabricated multi-color QLEDs show high luminance approaching 103cd m^(-2)and superior external quantum efficiency of 13.3%.Benefitting from the wide spectral tunability and light emitting efficiency,we believe the proposed multi-color QLEDs have great application prospects for both displays and lighting. | GE MU TIANYU RAO MENGLU CHEN YIMEI TAN QUN HAO XIN TANG | 2022 | Photonics Research2022,10,7: | 0 |
| 5 | Large scale air pollution prediction with deep convolutional networks显示文摘Although considerable success has been achieved in urban air quality prediction(AQP) with machine learning techniques, accurate and long-term prediction is still challenging. One of the key issues for existing AQP approaches is that air quality monitoring stations are sparsely distributed, typically with around ten monitoring stations per city. As air quality may change abruptly in a local area, it is difficult to perform AQP accurately in areas that are far away from observation points. In addition, due to the large distance between every two monitoring stations, we cannot effectively leverage spatial relations among them to improve the AQP accuracy. In this paper, thanks to the development of low-cost air quality sensors, we are now able to collect a large-scale air quality dataset with 393 deployed air quality monitoring stations in a 120 km × 70 km region, which is more than ten times denser than existing AQP datasets. Further,we present a novel method to handle the data effectively. Specifically, we first convert the observed data from irregularly distributed monitoring stations into a regular image-like pollution map, which can then be processed with advanced deep convolutional networks. | 6 | EHMT2 promotes tumorigenesis in GNAQ/11-mutant uveal melanoma via ARHGAP29-mediated RhoA pathway显示文摘Constitutive activation of GNAQ/11 is the initiative oncogenic event in uveal melanoma(UM).Direct targeting GNAQ/11 has yet to be proven feasible as they are vital for a plethora of cellular functions.In search of genetic vulnerability for UM,we found that inhibition of euchromatic histone lysine methyltransferase 2(EHMT2)expression or activity significantly reduced the proliferation and migration capacity of cancer cells.Notably,elevated expression of EHMT2 had been validated in UM samples.Furthermore,Kaplan-Meier survival analysis indicated high EHMT2 protein level was related to poor recurrence-free survival and a more advanced T stage.Chromatin immunoprecipitation sequencing analysis and the following mechanistic investigation showed that ARHGAP29 was a downstream target of EHMT2.Its transcription was suppressed by EHMT2 in a methyltransferasedependent pattern in GNAQ/11-mutant UM cells,leading to elevated RhoA activity.Rescuing constitutively active RhoA in UM cells lacking EHMT2 restored oncogenic phenotypes.Simultaneously blocking EHMT2 and GNAQ/11 signaling in vitro and in vivo showed a synergistic effect on UM growth,suggesting the driver role of these two key molecules.In summary,our study shows evidence for an epigenetic program of EHMT2 regulation that influences UM progression and indicates inhibiting EHMT2 and MEK/ERK simultaneously as a therapeutic strategy in GNAQ/11-mutant UM. | Yongyun Li Tianyu Zhu Jie Yang Qianqian Zhang Shiqiong Xu Shengfang Ge Renbing Jia Jianming Zhang Xianqun Fan | 2024 | Acta Pharmaceutica Sinica B2024,14,3: | 0 |

| 8 | Comparison of protective effects of electroacupuncture and moxibustion at Zusanli(ST 36)on perinatal nicotine exposure-induced lung phenotype in rat offspring显示文摘Objective:To compare the effects of electroacupuncture(EA)and moxibustion at Zusanli(ST 36)on the lung phenotype of rat offspring exposed to nicotine during the perinatal period.Methods:Pregnant Sprague-Dawley rats were randomly divided into 4 groups:the control group(saline only),the model group(nicotine only),the EA group(nicotine+EA at ST 36 acupoints bilaterally),and the moxibustion group(nicotine+moxibustion at ST 36 acupoints bilaterally).n=6 rats per group.On postnatal day 21,the body weight,lung weight,and pulmonary function were determined and lung morphometry was performed.Peroxisome proliferator-activated receptor gamma andβ-catenin levels in the lung tissue of offspring were also determined.Results:Perinatal nicotine exposure(PNE)results in decreased body and lung weights of offspring rats,abnormal lung tissue morphology,and significantly altered pulmonary function,showing an increase in total airway resistance and a decrease in tidal volume,minute ventilation,total airway compliance,and peak expiratory flow.Bilateral EA at ST 36 acupoints could block all of these perinatal nicotine-induced effects.Although moxibustion also had protective effects in nicotine-induced offspring lungs,some of these effects did not reach statistical significance,e.g.,protection against the upregulation ofβ-catenin,the downregulation of PPARγsignaling,and the increase in peak expiratory flow.Conclusion:Maternal EA at ST 36 blocked the PNE-induced changes in key developmental signaling pathways,prevented the PNE-induced changes in lung morphology,and protected pulmonary function.Moxibustion at ST 36 showed similar but weaker protective effects against the PNE-induced changes in the exposed offspring.It is important to note that the mechanism underlying the protective effects of moxibustion at ST 36 may be different from those of EA at ST 36,and further research is needed to understand these differences. | Yunpeng Ge Yitian Liu Guozhen Zhao Reiko Sakurai Yana Xie Tianyu Shi Yang Fang Jiajia Wang Virender K.Rehan Bo Ji | 2023 | Journal of Traditional Chinese Medical Sciences2023,10,3: | 0 |
| 9 | Allotetraploidization event of Coptis chinensis shared by all Ranunculales显示文摘Coptis chinensis Franch.,also named Chinese goldthread is a member of Ranunculaceae in the order Ranunculales and represents an important lineage of early eudicots with traditional medicinal value.In our study,by using syntenic analysis combined with phylogenomic analysis of C.chinensis and four other representative genomes from basal and core eudicots,we confirmed that the WGD event in C.chinensis was shared by Aquilegia coerulea and Papaver somniferum L.and quickly occurred after Ranunculales diverged from other eudicots,likely a Ranunculales common tetraploidization(RCT).The synonymous nucleotide substitutions at synonymous sites distribution of syntenic blocks across these genomes showed that the evolutionary rate of the P.somniferum genome is faster than that of the C.chinensis genome by approximately 13.7%,possibly due to Papaveraceaes having an additional special tetraploidization event(PST).After Ks correction,the RCT dated to 115—130 million years ago(MYA),which was close to the divergence of Ranunculaceaes and Papaveraceaes approximately115.45—130.51 MYA.Moreover,we identified homologous genes related to polyploidization and speciation and constructed multiple sequence alignments with different reference genomes.Notably,the event-related subgenomes in the basal genomes all showed genomic fractionation bias,suggesting a likely allopolyploid nature of the RCT,PST and T-Alpha and T-Beta events in Tetracentron sinense.In addition,we detected that the sixteen P450 subfamilies were markedly expanded in the genomes of Ranunculales,and most of them were related to the RCT and PST events.We constructed a new platform for Early Eudicot Comparative Genomic Research(http://www.cgrpoee.top/index.html)to store more information.In summary,our findings support the WGD of C.chinensis shared by Ranunculales,which is likely an allotetraploidization event.This present effort offered new insights into the evolution of key polyploidization events and the genes related to secondary metabolites during the diversification of early eudicots. | Yan Zhang Weina Ge Jia Teng Yanmei Yang Jianyu Wang Zijian Yu Jiaqi Wang Qimeng Xiao Junxin Zhao Shaoqi Shen Yishan Feng Shoutong Bao Yu Li Yuxian Li Tianyu Lei Yuxin Pan Lan Zhang Jinpeng Wang | 2024 | Horticultural Plant Journal2024,10,1: | 0 |
