3Cotton GhBRC1 regulates branching,flowering,and growth by integrating multiple hormone pathways显示文摘Cotton architecture is partly determined by shoot branching and flowering patterns.Gh BRC1 was previously identified by RNA-seq analysis of nulliplex-branching and normal-branching cotton.However,the roles of Gh BRC1 in cotton remain unclear.In the present study,investigations of nuclear localization and transcriptional activity indicated that Gh BRC1 has characteristics typical of transcription factors.Gene expression analysis showed that Gh BRC1 was highly expressed in axillary buds but displayed different expression patterns between the two branching types.Overexpression of Gh BRC1 in Arabidopsis significantly inhibited the number of branches and promoted flowering.In contrast,silencing Gh BRC1 in cotton significantly promoted seedling growth.Gh BRC1 was induced by multiple hormones,including strigolactones,which promoted seedling growth and seed germination of Arabidopsis plants overexpressing Gh BRC1.Consistent with these findings,RNA-seq analysis of virus-induced gene silencing treated cotton revealed that a large number of genes were differentially expressed between Gh BRC1-silenced and control plants,and these genes were significantly enriched in plant hormone signalling pathways.Together,our data indicates that Gh BRC1 regulates plant branching and flowering through multiple regulatory pathways,especially those regulating plant hormones,with functions partly differing from those of Arabidopsis BRC1.These results provide insights into the molecular mechanisms controlling plant architecture,which is important for breeding cotton with ideal plant architecture and high yield.Quan Sun Yuanhui Xie Huimin Li Jinlei Liu Rui Geng Ping Wang Zongyan Chu Ying Chang Guanjun Li Xiao Zhang Youlu Yuan Yingfan Cai 2022The Crop Journal2022,10,1:0
4Chromosome sorting and its applications in common wheat (Triticum aestivum) genome sequencing显示文摘The large genome size (~17000 Mb) and complicated DNA structures of common wheat (Triticum aestivum) hamper its genome sequencing.By means of flow cytometry,systematic investigations on individual chromosome sorting have been carried out to construct chromosome-specific bacterial artificial chromosome (BAC) libraries since the 1980s.Several wheat chromosome-specific BAC libraries,such as chromosome 3B,three D genome chromosomes (1D,4D and 6D),and the short arm of chromosome 1B,have been developed,and the physical map of chromosome 3B was established in 2008.The same chromosome-based strategy is being employed by the International Wheat Genome Sequencing Consortium (IWGSC) to establish the physical maps of the other 20 common wheat chromosomes (cv.Chinese Spring).Several projects on wheat genome sequencing are currently underway.The availability of new sequencing technologies provides new choices for sequencing of gene space of the wheat genome.The applications of flow-sorted chromosomes in wheat genome studies present some examples to analyze the complex genomes of cereals.WU SuoWei XIAO Yang ZHENG Xu CAI YingFan DOLEZEL Jaroslav LIU Bing Hua YANG Li SONG MeiFang ZHOU Peng ZHOU Yang MENG FanHua WANG ShanHong LIU HongWei HAI HuQu YANG JianPing 2010Chinese Science Bulletin2010,55,15:0
